1.Screening and cloning of the target genes immuno-regulated by glycyrrhizin using suppression subtractive hybridization technique

Objective To construct a subtractive cDNA library of genes differentially expressed in human lymphoma cell line Jurkat cells treated with glycyrrhizin (GL), and to clone genes associated with its immunological regulation, and to further elucidate the molecular immune mechanism of GL. Methods The mRNA was isolated from Jurkat cells treated with either GL or 0.9 percent sodium chloride as a control, then cDNA was synthesized. Suppression subtractive hybridization (SSH) method was employed to analyze the differentially expressed DNA sequence between the two groups. The cDNA was sequenced and analyzed in GenBank with Blast search after the amplification of the subtractive library by PCR. Results The amplified library contained 28 positive clones. Colony PCR analysis showed that there were 22 clones containing 200-1 000 bp inserts. Sequence analysis was performed, and the full length sequences were obtained with bioinformatics method. Altogether 11 kinds of encoding sequences were achieved, including interleukin-12, interleukin-18, and thymosin ?1, etc. Conclusions A subtractive cDNA library of genes differentially expressed in Jurkat cells treated with GL using SSH technique was constructed successfully, and it might give some new clues for the study of the immune regulation mechanism of GL.

3.Iridoid glycosides from buds of Jasminum officinale L. var. grandiflorum.

The study on the buds of Jasminum officinale L. var. grandiflorum was carried out to look for anti-HBV constituents. The isolation and purification were performed by HPLC and chromatography on silica gel, polyamide and Sephadex LH-20 column. The structures were elucidated on the basis of physicochemical properties and spectral analysis. Six iridoid glycosides were identified as jasgranoside B (1), 6-O-methy-catalpol (2), deacetyl asperulosidic acid (3), aucubin (4), 8-dehydroxy shanzhiside (5), and loganin (6). Jasgranoside B (1) is a new compound. Compounds 2-6 were isolated from Jasminum officinale L. var. grandiflorum for the first time.
4.Effect of systemic chemotherapy on conditions of tumor infiltrating, metastasis and disease-specific survival for advanced retinoblastoma

Objective To observe the effect of systemic chemotherapy on conditions of tumor infiltrating,metastasis and disease-specific survival (DSS) for advanced retinoblastoma (RB).Methods Forty-one patients with advanced RB who received enucleation were enrolled in this study.There were 26 males and 15 females,age at diagnosis was ranged from 2 to 72 months,with a mean of 23.08 months.There were 16 bilateral patients and 25 unilateral patients;13 group D eyes and 28 group E eyes.16 patients received enucleation as the primary treatment (operation group),25 eyes received chemotherapy before enucleation (chemotherapy group).There was no significant statistical difference between two groups for the gender,unilateral and bilateral,international staging or diagnostic age (P>0.05).The histopathology report was performed to assess the risk of postoperative tumor-node-metastasis staging (pTNM) in each patient,and the extent of tumor invasion in the optic nerve,choroid and anterior chamber was divided into 3 levels of low risk,medium risk and high risk.Five deaths were all in the group E with chemotherapy before enucleation.Using R software survival analysis software package survfit function,the application of Kaplan-Meier estimation method,DSS of RB children was calculated from the time of diagnosis,up to the date of the death of patient.DSS differences between chemotherapy,operation group and eye removal time (more than 3 months,less than 3 months) in group E RB children were analyzed.Results The proportion of high risk pTNM stage in chemotherapy group was significantly lower than the operation group.But there was no significant difference between the two groups in the overall risk classification (x2 =3.130,P=0.077).For group D eyes,the overall risk classification in chemotherapy group was significantly lower than the operation group (x2 =5.870,P=0.015).There was no significant difference between the two groups in the overall risk of group E eyes (x2 =0.020,P=0.889).The DSS in chemotherapy group and operation group were 0.71 and 1.00,respectively;the difference was significant (x2 =3.700,P=0.05).The DSS in children whose enucleation delayed for more than 3 months and children whose enucleation performed within 3 months were 0.64 and 1.00,respectively;the difference was significant (x2 =4.800,P=0.028).Conclusion Systemic chemotherapy did not reduce the risk of tumor invasion and metastasis in patients with advanced RB.Instead,it will reduce the DSS in group E eyes of RB.

6.Apoptotic Effects and Its Mechanisms on Leukemic K562 Cells Caused by Interferon-alpha Combined with Cytarabine

Objective: To investigate the apoptotic effects and its mechanisms on K562 cells caused by Interferon-alpha (INF-?) and Cytarabine (Ara-C). Methods: The variation of both morphology and inhibitory rate of K562 cells was observed in culture medium with IFN-? and various concentrations of Ara-C at different time in vitro. The variation of telomerase activity and P53 protein expression were detected before and after apoptosis occurred. Results: INF-? and Ara-C used concurrently could cause apoptosis and inhibit the growth of K562 cells as well as decrease the telomerase activity and increase P53 protein expression significantly. All these processes showed both in time- and dose-dependent manner. Conclusions: INF-? and Ara-C used concurrently can inhibit the growth of K562 cells and induce apoptosis, inhibiting the telomerase activity and increasing the expression of P53 protein may be one of the most important mechanisms.
7.Correlation between IL-6,IL-10,TNF-α and disease severity of children with mycoplasma pneumoniae

Objective To study the relationship between the severity of Mycoplasma pneumoniae pneumonia(MPP) and interleukin(IL)-6,IL-10,tumor necrosis factor(TNF)-α level.Methods From Jan.to Jul.2012,according to clinical pulmonary infection score(CPIS),30 children with severe MPP(severe MPP group) and 32 children with mild MPP(mild MPP group) were enrolled,and 30 healthy children were enrolled as control group).Subjects of all groups were detected for the levels of IL-6,IL-10,TNF-α,IgA,IgM and IgG.Besides,children with MPP were also detected for the levels of IL-6,IL-10 and TNF-α after treatment.Results The levels of IL-6,IL-10 and TNF-α in bronchoalveolar lavage fluid(BALF) and serum in severe MPP group were higher than those in mild MPP group and control group(P<0.05).The levels of IL-6,IL-10 and TNF-α in BALF and serum were significantly higher in mild MPP group than those in control group(P<0.05).In all children,the levels of IL-6,IL-10 and TNF-α in BALF were higher than those in serum.After treatment,the levels of IL-6 and TNF-α in BALF in children with severe MPP were significantly decreased,while that of IL-10 were increased(P<0.05).Compared with control group,the IgM level in children with MPP was increased significantly,and the IgG level was decreased notablely(P<0.05).Conclusion There might be significant correlation between levels of cytokines IL-6,IL-10,TNF-α and the disease severity of MPP in children patients,which could be used as the basis for the clinical diagnosis and treatment.
8.Changes of angiogenesis in rectal cancer patients after preoperative radiotherapy

Objective To explore the effect of preoperative radiotherapy on angiogenesis of rectal cancer. Methods Twenty patients with advanced lower rectal carcinoma received preoperative radiation with a dosage of 30~40 Gy each time for a total of 15~20 sessions during a period of 3~4 weeks. The definitive surgery was performed 7~10 days after radiotherapy. Another 20 patients undergoing tumor resection without preoperative radiotherapy served as control. Tumor sample was sent for pathology. The expression of vascular endothelial growth factor (VEGF) and CD34 in the rectal cancer were detected immunohistochemically. The intratumoral microvessel density (MVD) was measured. Results Fifteen patients were found with grade Ⅱ and five patients with grade Ⅲ tissue response in radiotherapy group. Objectives To assess if visual analogue scale(VAS) or Likert scale was relatively more suitable when measuring self-reported clinical outcomes for apoplexy spastic-paralysis patients.Methods Nineteen questions were made from the serf-evaluating instrument based on patient reported outcomes for apoplexy spasticparalysis patients developed previously in cross-sectional study.104 patients answered the questions by both continuous and discrete visual analogue scale(VAS),while other 110 patients by both five-point Likert scales and Likert-typo scales.Cronbach'sαfor internal consistency reliability,spearman for correlation,and exploratory factor analysis for construct validity were applied.Results Except for emotional domain,the α coefficients of physical domain,social domain and overall score with continuous and discrete VAS are higher than that with five-point Likert scales and Likert-type scales.Internal consistency reliability was stronger for VAS.lntraclass correlation coefficients were above 0.4 mostly for each item with its domain,and VAS may produce the higher correlation.For VAS,there were three factors in the exploratory factor analysis,which confirmed the domain hypotheses of physical,mental and social functions,but for the Likert scales,the result was undesiable.Conclusion The more accurate for the response options,the better for the performance.When the patient can complete the measurement without the help of others,VAS seems better suited.
