2.Inhibitory effect of RNAi on the expression of AQP1 and the migration ability of Hep-2

3.Treatment of 30 Cases of Prolapse of Lumbar Intervertebral Disc by Electroacupuncture plus Tuina

Purpose: To observe the clinical therapeutic effect of electro acupuncture plus Tuina on treating prolapse of lumbar intervertebral disc. Methods: Electro -acupuncture at Jiaji(Ex-B 2) acupoints plus reduction manipulations were used to treat 30 cases of prolapse of lumbar intervertebral disc. Results: After 2 courses of treatment, among the 30 cases, 8 cases were cured and 22cases got improvements, and the effective rate was 100%.Conclusion: Electroacupuncture plus reduction manipula -tions had a satisfactory effect on treating this disease;and it can significantly improve the clinical symptoms of the patients.
4.STUDIES ON THE THERAPEUTIC EFFECT OF TRADITIONAL CHINESE MEDICINE“RE DU QING”ON SILICOSIS:Cytochemical and Scanning Electron Microscope Studies of the Protective Effect on the Macrophages Phagocytosed Silica Dusts

In order to investigate the therapeutic effect of traditional chinese medicine "Re Du Qing" on silicosis, the cultured and purifiel peritoneal macrophages in five groups obtained from mice were observed dynamically with cytochemical me- thods and scanning electron microscope. The results showed that the survival times of cell cultures were 2-3 weeks, 24-48, 72, 48-72 and 48-72 hr in normal, silica, "Re Du Qing", P204 and joint action group respectively in vitro. The characteristics of cell morphology with a series of cell surface ultrastructural changes in different times of culturd and the stages of various function were different in five groups. The cell surface ultrastructural changes of "Re Du Qing" group were similar to the normal group. The macrophages in the silica group phagocytosed silica dusts rapidly died and broken down much earlier than the cells in "Re Du Qing" group. The cell surface ultrastructural changes in P204 group were less than that of the cells in silica group, whereas the eell surface ultrastructural changes in joint action group were between the cells in "Re Du Qing" and P204 groups. The activities of intracellular acid phosphatase (AcP) and succinic dehydrogenase (SDH) in silica group were also much lower than that of "Re Du Qing" one. This study suggested that traditional chinese medicine "Re Du Qing" is evidently more effective on therapy of experimental silicosis.
5.Influence of mild hyperuricemia on the function of glomerular endothelial cells and vascular smooth muscle cells in rats

Objective To discuss whether mild hyperuricemia can lead to kidney damage and the protection of decreased uric acid,through observing that hyperuricemia did damage to glomerulus endothelial function and cell proliferation of vascular smooth muscle in rats. Methods Fifty-four male SD rats were divided into four groups,the control group,model group (Oxonate),allopurinol group and Oxonate+allopurinol group.Rats were administered on a low sodium diet and their systolic blood pressure (SBP) were measured each 10 days.ELISA was used to detect rat plasma markers of endothelial function damage [nitric oxide (NO),type-1 plasminogen activator inhibitor (PAI-1),endothelin 1 (ET-1)] and cell proliferation of vascular smooth muscle[plateletderived growth factor (PDGF),cycloxygenase 2 (COX2),monocyte chemotactic protein-1 (MCP-1)],and the markers of inflammatory reaction[interleukin-18 (IL-18),tumor necrosis factor α(TNF-α)].PDGF and nitric oxide synthase (NOS) levels of rats were detected by immunohistochemical method.Renal tissue pathology of rats was observed. Results Compared to the control group,the plasmic concentration of COX2,ET-1,IL-18,PAI-1,PDGF,TNF-o,MCP-1 increased,and NO decreased significantly in rats of model group (all P<0.05),expression of NOS significantly reduced and PDGF increased (all P<0.05).Under light microscope,vascular wall thickening,intimal proliferation and lumen slight stricture without uric acid crystals in renal tissue were found in model group,which were obviously improved by using allopurinol. Conclusion Mild hyperuricemia can do damage to endothelial function of glomerulus and lead to vascular cell proliferation,which can be improved through decreasing uric acid.
6.The exPression of forkhead box 2 in invasive ductal carcinoma of breast and the relationshiP with delta-like Ligand 4

Objective To study the expression of forkhead box 2(FOXC2)and delta-like liGand 4 (DLL4)in invasive ductal carcinoma(IDC)of breast and the clinical siGnificance. Methods The expression of FOXC2 and DLL4 in l22 cases of IDC(Grade Ⅰ33,Grade Ⅱ40,GradeⅢ 49)were observed by tissue chip and immunohistochemistry. The relationship of the expression with clinicopatholoGical characteristics and between FOXC2 and DLL4 were statistically analyzed. PCR experiment was performed in normal breast tissue,ductal carcinoma in situ(DCIS)IDC Grade Ⅰ,Grade Ⅱ and Grade Ⅲ(l0 cases respectively). Results The positive rate of FOXC2 and DLL4 in IDC was 77. 87% and 74. 59% respectively. A hiGher expression was observed in GradeⅢ than in GradeⅠand Ⅱ( P<0. 05 ). The expression of FOXC2 was related to the neGative expression of ER. The expression of DLL4 was related to the tumour size,clinical staGe and lymph node metastasis( P<0. 05). The RCR of FOXC2 and DLL4 were Gradually increased in normal breast tissue,DCIS,IDC GradeⅠ, Grade Ⅱ and Grade Ⅲ(P<0. 05). Moreover the expression of FOXC2 was related to the expression of DLL4(r=0. 233,P=0. 0l0). Conclusion FOXC2 and DLL4 miGht toGether have influence on the proGression and outcome of breast carcinoma,and could be important markers of proGnosis. DLL4 miGht be reGulated by variety of factors includinG FOXC2 at the same time.
7.Priliminary study of the effect of tumor-associated fibroblasts on the infiltration and metastasis of papillary thyroid carcinoma

Objective:To detect the expression of fibroblast-specific protein 1 (FSP1/S100A4) , ɑ-smooth-muscle actin (ɑ-SMA) and fibroblast-activated protein (FAP-ɑ) in tumor-associated fibroblasts (TAFs) in papillary thyroid carcinoma (PTC) , and to investigate its relationship with the origination and development of PTC.Methods:The expression of FSP1/S100A4, ɑ-SMA and FAP-ɑ in normal thyroid and PTC was determined by SP method of immunohistochemistry, and the relationship between these indicators and important clinicopathological parameters were analyzed.Results:The positive expression of FSP1/S100A4, ɑ-SMA and FAP-ɑ was observed in PTC, but not detected in the follicular epithelium or stromal cells of normal thyroid. In addition, the expression of FAP-ɑ was significantly related to tumor size, lymph node metastasis and TNM classification ( χ2=6.833, P<0.05; χ2=10.296, P<0.05; χ2=4.910, P<0.05) . The expression of ɑ-SMA was positively related to the invasion of capsule and lymph node metastasis ( χ2=6.008, P<0.05; χ2=11.766, P<0.05) . The expression of FSP1/S100A4 was negatively related to the clinicopathological parameters above ( P>0.05) in PTC. Conclusion:TAFs in PTC may indicate the infiltration and metastasis, which provideds new thinking for the treatment strategies of papillary thyroid carcinoma.
8.Application of self-management-oriented network platform in depression rehabilitation

Objective:To explore the application and effects of the self-management-oriented network platform in depression rehabilitation.Methods:Totally 118 patients with depression discharged from a municipal mental health center and the communities that it governs between January and December 2017 were selected by convenient sampling and divided into the study group and the control group by Excel sheets, with 59 cases in each group, of which 2 cases dropped out from the study group. Patients in the control group received routine disease management, while patients in the study group received self-management and education via the depression network management platform after a personalized management plan was formulated upon discharge on this basis. After 6 months of intervention, the improvements in satisfaction, self-efficacy and suicidal risks were compared between the two groups.Results:After intervention, the satisfaction scores of visit time, visit attitude, visit content, and humanistic care of the study group were higher than those of the control group, and the differences were statistically significant ( P<0.05) ; the General Self-Efficacy Scale score of the study group was higher than that of the control group, and the difference was statistically significant ( P<0.05) ; the scores of despair, optimism, and sleep factors in suicidal ideation in the study group were lower than those of the control group, and the differences were statistically significant ( P<0.05) . Conclusions:The self-management-oriented network platform management model for discharged patients with depression can improve patient satisfaction, enhance their sense of self-efficacy, reduce the risk of suicide, and strengthen their self-management capabilities.
9.A facile and universal method to achieve liposomal remote loading of non-ionizable drugs with outstanding safety profiles and therapeutic effect.

Liposomes have made remarkable achievements as drug delivery vehicles in the clinic. Liposomal products mostly benefited from remote drug loading techniques that succeeded in amphipathic and/or ionizable drugs, but seemed impracticable for nonionizable and poorly water-soluble therapeutic agents, thereby impeding extensive promising drugs to hitchhike liposomal vehicles for disease therapy. In this study, a series of weak acid drug derivatives were designed by a simplistic one step synthesis, which could be remotely loaded into liposomes by pH gradient method. Cabazitaxel (CTX) weak acid derivatives were selected to evaluate regarding its safety profiles, pharmacodynamics, and pharmacokinetics. CTX weak acid derivative liposomes were superior to Jevtana® in terms of safety profiles, including systemic toxicity, hematological toxicity, and potential central nerve toxicity. Specifically, it was demonstrated that liposomes had capacity to weaken potential toxicity of CTX on cortex and hippocampus neurons. Significant advantages of CTX weak acid derivative-loaded liposomes were achieved in prostate cancer and metastatic cancer therapy resulting from higher safety and elevated tolerated doses.
10.Comparison on Skin Permeability of Corydalis Rhizoma Total Alkaloid Patches at Shenque Acupoint and Non-acupoint Administration

ObjectiveTo compare the differences in resistance and structure of skin between acupoints and non-acupoints, and to study the difference in skin permeability characteristics of Corydalis Rhizoma total alkaloid patches (CTTP) after administration at Shenque acupoint and non-acupoint, so as to provide experimental support for its clinical acupoint application to prevent and treat chronic pain. MethodTaking corydaline (CD), tetrahydropalmatine (THP) and corydalis L (CDL) as evaluation indexes, and the quantitative analysis was carried out by high performance liquid chromatography (HPLC). The mobile phase was methanol-0.04 mol·L-1 phosphoric acid aqueous solution (70∶30, pH 6.0 adjusted with triethylamine), the detection wavelength was 281 nm. In vitro transdermal test in Franz diffusion cell and in vivo transdermal test were used to study the skin permeability characteristics of CTTP through Shenque acupoint and non-acupoint administration. At the same time, the skin resistance between Shenque acupoint and non-acupoint was measured before and after the administration, and the distribution of the drug in each layer of the skin was compared by freezing sectioning, and visual verification was performed with fluorescence inverted microscope. ResultAfter 24 h of administration, the results of in vivo and in vitro experiments showed that the cumulative permeation and retention of CD, THP and CDL at Shenque acupoint skin were higher than those at non-acupoint skin (P<0.05, P<0.01), the skin resistance of Shenque acupoint was lower than that of non-acupoint at all time points. The fluorescence microscopic observation results showed that the drug content of each layer of the skin was all Shenque acupoint>non-acupoint, indicating that the skin of Shenque acupoint had better effect on drug penetration and storage than non-acupoint. ConclusionThe 24 h cumulative permeation and retention of CTTP in Shenque acupoint skin are higher than those in non-acupoint skin, and the mechanism may be related to the thin skin, low electrical resistance and large number of hair follicle bodies at Shenque acupoint.
