1.Outcome of Neonatal Respiratory Distress Sydrome Treated with High Freguency Ventilation

Objective To study the outcome of NRDS treated with HFV. Methods The study was a prospective controlled clinical trial. Twenty-five newborns with NRDS were treated with HFV and another 25 treated with normal frequency ventilation were served as control. Results Twenty newborn infants survived in HFV group, and complicated with neonatal pneumonia in 12, intraventricular hemorrhage in 3, neonatal pulmonary hemorrhage in 4, patent ductus arteriosus in 4, broncho-pulmonary dysplasia in 2, and air Leak in 3. Fourteen newborn infants were survived in control group, and complicated with neonatal pneumonia in 12, patent ductus arteriosus in 8, intraventricular hemorrhage in 4, neonatal pulmorary hemorrhage in 11, broncho-pulmonany in 6, and air Leak in 3. The mean time of mechanical ventalation was 88.6 hrs in survivors of HFV group vs. 154.7 hrs in that of control group. Conclusions The HFV can reduce the mean time used for mechanical ventilation, and it is as safe as normal-frequency ventilation.
2.HPLC Determination of Aloin and Aloeemodin in Barbodos Aloe

Objective To isolate and determine Aloin and Aloeemodin in Barbodos Aloe by ionic liquid-based ultrasonic-assisted extraction coupled with high performance liquid chromatography, with 1-butyl-3-methylimidazolium chloride ([BMIM]Br) solution as the extraction solvent. Methods The separation was performed on Phenomenex C18 column (250 mm×4.6 mm, 5 μm) with detection wavelength of 360 nm. The mobile phase was consisted of methanol-0.3% acetic acid solution (65∶35) with the flow rate of 0.80 mL/min, and the column temperature at 35 ℃. Results The calibration curves for Aloin and Aloeemodin were liner within 0.000 336-1.68 μg (r=0.999 96) and 0.000 608-3.04 μg (r=0.999 76), respectively. The limit of detection (LOD) was 0.05 06 ng/mL and 0.262 ng/mL, respectively. The average recovery was 95.99% and 95.80%, respectively. Conclusion The method is simple, rapid, accurate, sensitive, low cost and environment-friendly, thus it provides an effective means for assaying anthraquinones in Barbodos Aloe.
3.Determination of Puerarin, Daidzin and Daidzein in Puerariae Lobatae Radix by ILDLPME Coupled with HPLC

Objective To develop a sensitive and specific HPLC method to simultaneously determine the contents of puerarin, daidzin and daidzein in Puerariae Lobatae Radix. Methods The three compounds were obtained by ionic liquid based on dispersive liquid phase microextraction. The determination was carried out on a Phenomenex C18 column (250 mm×4.6 mm, 5μm) with a mixture of methanol-0.2%acetic acid (volume ration 45∶55) as mobile phase at a flow rate of 0.8 mL/min. The UV detective wavelength was 250 nm, and the column temperature was set at 35 ℃. Results The linear response ranged from 6.24×10-6-37.44 μg for puerarin (r=0.999 71), 5.44×10-6-27.20μg for daidzin (r=0.999 85), and 5.60×10-6-28.00μg for daidzein (r=0.999 94), respectively. Conclusion The method is quick, simple and repeatable for simultaneous determination of the contents of puerarin, daidzin and daidzein in Puerariae Lobatae Radix.
4.Content Determination of Chlorogenic Acid, Quercetin and Kaempferol inPyrrosia Lingua (Thumb.) Farwell by HPLC

Objective To establish an HPLC method for simultaneous separation and determination of chlorogenic acid, quercetin and kaempferol inPyrrosia lingua (Thumb.) Farwell. Methods The separation was performed on Phenomenex C18 column (250 mm×4.6 mm, 5μm) with the mobile phase of methanol-acetic acid (pH=3.0) solution and gradient elution. Flow rate was 1.0 mL/min;the UV detection wavelength was 254 nm;column temperature was 35℃.Results The calibration curves for chlorogenic acid, quercetin, and kaempferol were in good linearity in the range of 0.000 24-3.00μg (r=0.999 9), 0.000 16-2.00μg (r=0.999 9), and 0.000 16-2.00μg (r=0.999 9), respectively. The limits of detection (S/N=3) were 3.29, 0.43 and 0.33 ng/mL, respectively. The average recovery rates were 97.73%, 98.07% and 96.92%, respectively. ConclusionThe method is simple, precise and sensitive. It provides scientific proof for separation and determination of chlorogenic acid, quercetin and kaempferol inPyrrosia lingua (Thumb.) Farwell.
5.The change of α-melanocyte-stimulating hormone(α-MSH)in the serum of patients multipleinjuries

Objective To detect the variations of the serum α-MSH and TNF-α in multiple-trauma patients and discuss their role in severity of casualties.Methods Fifty casualties were divided into two groups for study.There were 30 casualties with moderate severe trauma(ISS 16 ~ 25 point)and 20 patients with extreme severe trauma(ISS > 25 point),and another 15 healthy subjects were enrolled as controls.The blood samples were obtained within 24 hours,and 3 days,5 days,7 days after admission.The serum levels of α-MSH and TNF-α in casualties with multiple injuries were determined by using enzyme-linked immunosorbent double antibody sandwich method(ELISA).The data were expressed in((x)± s),and analyzed with chi-square test and repetitive measures of ANOVA by using SPSS 13.0 package.P value less than 0.05 indicated statistical significance Results The serum α-MSH levels of casualties within 24 hours,and 3 days,5 days,7 days after injury in the two groups were much lower than those in the control group (P < 0.01),while the serum TNF-α levels of casualties were much higher than those in the control group (P <0.01).The serum α-MSH levels of casualties with extreme severe traumawere lower,and the TNF-αlevels of casualties with extreme severe trauma were higher than those in patients with moderate severe trauma(P <0.01,respectively).There were negative correlations between two biomarkers 24 hours,5d and 7d after injury.Conclusions In casualties,the serum levels of α-MSH decreased and the serum levelsof TNF-α increased,and the degrees of changes were closely depended on the severity of trauma,the more severe the more significant changes.There was a negative correlation between two biomarkers.
6.HPLC method for determination of uric acid in plasma of hyperuricemia model mice

Objective To establish an efficient HPLC method for the determination of uric acid in plasma of hyperuricemia model mice,and the evaluation of uric acid lowering effect of Lesinurad.Methods The Laballiance Series Ⅲ HPLC system was adopted with Kromasil C18 column (100 mm × 4.6 mm,5 μm).The mobile phase consisted of methanol-0.5% acetic acid (10:90) for isocratic elution with a flow rate of 0.4 mL/min.The detection wavelength was set at 283 nm.The established HPLC method was used to detect the plasma uric acid level of mice at 0.5,1.0,and 2.0 h time points after which being ip injected with 250 and 500 mg/kg uric acid.Lesinurad of 250 and 500 mg/kg was ig given to mice,0.5 h later,mice were ip injected with 500 mg/kg uric acid to establish hyperuricemia model,and 1 h later,the established HPLC method was used to detect the plasma uric acid level of mice.Results There was a good linear relationship between peak area and the concentration of plasma uric acid in the range of 7.5-150 μg/mL (r =0.997).The specificity,repeatability,precision,stability,and recovery of the established HPLC method was in accordance with the guiding rules of biological sample determination.Compared with the endogenous serum uric acid concentration of control group mice,serum uric acid concentration of 250 mg/kg dose group was significantly increased 0.5 h after ip administration with uric acid (P < 0.01),and serum uric acid concentration of 500 mg/kg dose group was significantly increased 0.5,1.0,and 2.0 h after ip administration with uric acid.Compared with model group,the concentration of uric acid in plasma decreased significantly in low dosage group administered with Lesinurad (P < 0.05),while decreased more significantly in high dosage group (P < 0.01).Conclusion This convenient,rapid,and accurate method can be applied to the determination of uric acid in mouse plasma and the evaluation of relative drugs,which provide an efficient analysis way for establishing hyperuricemia model and screening relative drugs.
7.Correlation Between Plasma Homocysteine Concentration and Transient Ischemic Attack

Objective:To investigate the correlation between plasma homocysteine (Hcy) concentration and transient ischemic attack (TIA) and traditional vascular risk factors.Methods:The plasma Hcy concentrations of 112 patients with TIA and 62 controls were measured by fluorescenee polarization immunoassay.Hcy concentrations and related risk factors were analyzed.Results:The risk of TIA was increased significantly in plasma Hcy concentration 10.0 to 14.9 μmol/L group(OR=2.450,95% CI 1.091 to 5.502) and≥15.0 μmol/L group(OR=5.169,95% CI 2.096 to 12.746) compared with plasma Hcy concentration<10.0 μmol/L group.Using TIA as the dependent wariable,various vascular risk factors (including plasma Hcy concentration) as the independent variable,logistic regression was analyzed.The result showed that the risk of TIA was increased significantly in plasma Hcy concentration>10.0 μmol/L group compared with plasma Hcy concentration<10.0 μmol/L group(OR=3.150,95% CI 1.380 to 7.192).Conclusions:Plasma Hcy concentration is an independent risk factor for TIA.
8.The role of raffinose-low potassium dextran lung preservation solution with blood in pulmonary graft preservation

Objective To investigate the potential of a novel preservation solution,raffinose-low potassium dextran lung preservation solution with blood (BR-LPDS) in the preservation of the isolated lungs in rats,when compared to low-potassium dextran solution (LPDS).Methods Fifty donor lungs of Sprague-Dawley rats were randomly flushed with either R-LPDS (R-LPDS group,n =25) or RLPDS modified by the addition of blood (BR-LPDS group,n =25).After reperfusion,the double lungs were kept half inflate and then the pulmonary artery and bronchus were clamped.Then the grafts were flushed with and submerged in cold preservation solution at 4 C for 24 h.And they were excised for measurement of wet-to-dry ratio,as well as histologic examination to evaluate pulmonary edema,inflammation and cleaved caspase-3 expression at 0,2,4,5,6,8,10,12,14,16,24 h.Results As compared with the R-LPDS group,the mean wet-to-dry ratio of lungs in the BR-LPDS group was significantly reduced from 4 to 10 h after reperfusion.Upon histologic examination,less inflammatory cell aggregates and lower caspase-3 activity were seen in the lungs of the BR-LPDS group from 10 to 12 h and 10 to 14 h after reperfusion,respectively.Conclusion The addition of blood to the R-LPDS solution was shown to be more effective in reducing swelling,inflammation and caspase-3 expression of donor lungs than the R-LPDS solution.Further investigation is needed to evaluate lung preservation in BR-LPD solution as a viable option for transplant.
9.Congenital tongue hypoplasia(non-syndromic):A case report and literature review

Congenital tongue dysplasia is rare in clinical practice.This paper reports a case of congenital tongue dysplasia,and analyzes its possible mechanism in detail through literature review,so as to provide a basis for the prevention of this disease in the first trimester.Accord-ing to the current development conditions of the discipline,the author tries to put forward palliative care program suitable for this disease,es-pecially the serial treatment and psychological management of aglossia deformity,in order to improve the life quality of the children with this disease and provide a certain reference for clinical workers.
10.Safety and nosocomial infection control in lung transplantation for a case of COVID-19 with end-stage ARDS

Objective:To explore the safety and nosocomial infection control measures of COVID-19 patients of end-stage respiratory failure after lung transplantation.Methods:Lung transplantation was performed for a COVID-19 patient with end-stage respiratory failure after a negative conversion of 2019-nCoV nucleic acid. Before operation, all medical staff received simulated training on nosocomial infection. The procedures were performed in an operation room with a negative pressure environment. The three-grade preventive strategy was implemented and wearing positive pressure protective mask of electric air supply required. During operation, the patient was managed according to the in-hospital protection process. The environment and medical instruments were disinfected after operation.Results:The operation was completed successfully. The 2019-nCoV nucleic acid test was negative for many times after operation. Participants were placed in medical isolation for 14 days after surgery. During the period, nucleic acid test was negative twice.Conclusions:As an exploratory treatment, lung transplantation is a safe option for end-stage respiratory failure in COVID-19 in operation room with a negative pressure environment and implementations of three-grade preventive strategy.
