Objective To explore the protective mechanism of fructose-1, 6-disphophate on the hypoxic-ischemic brain damage of neonatal rats.Methods Seven-day-old Wistar rats were randomly divided into HIBD control group,saline-treated group, FDP-treated group. FDP-treated group was redivided into groups of high, middle and low dose.The expression of caspase-3mRNA was detected at 24h after HIBD with RT-PCR. Results There were no sifnificant differences of the caspase-3 mRNA expression among the saline-treated group, HIBD group and low dose FDP-treated group(P >0.05).The expressions of caspase-3 mRNA in the middle and high dose FDP-treated groups decreased significantly as campared to the above three groups (P < 0.05), but there was no significant difference between the two groups (P > 0.05). Conclusion The neuronal protective mechanism of FDP might be related to dewnregulate the gene expression of caspase-3 in brain tissue,and decrease the apoptosis of neuron after HIBD.

Methods The cellular distribution of IGF-Ⅱin normal and PIH placenta was determined by immunohistochemistry. Trophoblasts were digested,separated,and purified from placenta. IGF-ⅡmRNA expression in trophoblasts was detected by RT-PCR. The mean density was determined by image analysis technique. Results (1) IGF-Ⅱwas mostly found in syncytiotrophoblast,cytotrophoblast and amnio-chorion. The strongest immunoreactivity was observed in the syncytiotrophoblast and cytotrophoblast. Objective To investigate the expression of matrix metalloproteinases(MMP) related to oncogene C-myc and tumor differentiation of cutaneous squamous cell carcinoma (CSCC). Methods Expression of MMP2,MMP9 and C-myc was detected by immunohistochemistry in 68 paraffin embedded specimens of CSCC. Results Distribution of MMP2 and MMP9 was demonstrated within cytoplasm of tumor and stromal cells, especially for those cells at the interface of tumor and interstitial tissues. No expression of MMP2 and MMP9 was observed in the horny centers of tumor cell nests. Expression of MMP2 and MMP9 increased along with the decreasing of the differentiation of tumor tissue and the increasing of C-myc expression. Conclusions Expression of MMP2 and MMP9 might be a common event for both tumor and stromal cells but parenchymal tumor cells might predominate over the activation of MMP2 and MMP9. Oncogene C-myc protein might promote the secretion of MMP2 and MMP9 by tumor cells.

Objective To study the expression of vascular endothelial growth factor(VEGF) mRNA in neonatal rats with HIBD and to explore the function of VEGF in HIBD. Methods Fourty-eight Wistar rats of 7-day-old were assigned randomly to the control or HIBD groups. The left common carotid artery of rats in HIBD group were isolated and ligated, then exposed to hypoxic environment (8%O 2) for two hours. In control group the rat left common carotid artery were isolated, but not ligated and not exposed to hypoxic environment. Animals were killed in 3 h, 12 h, 24 h and 3 d after HIBD. The expression of VEGF mRNA in hippocampus were examined by RT-PCR.Results VEGF164 was the most prominent transcript of VEGF mRNA splice variants. Objective To detect the serum levels of carbohydrate antigen 199(CA199),carbohydrate antigen 125 (CA125)and carcino-embryonic antigen(CEA)in different population,and to explore their significances in diagnosis of pancreatic cancer.Methods The serum levels of CA199,CA125 and CEA in 56 patients with pancreatic cancer (pancreatic cancer group ), 5 6 patients with benign pancreatic disease (benign pancreatic disease group ) and 56 healthy volunteers(normal control group)were detected,then the positive rates of CA199,CA125 and CEA and the sensitivities and specificities of CA199,CEA and CA125 for the diagnosis of pancreatic cancer were compared among three groups.Results The levels of serum CA199,CA125 and CEA of the patients in pancreatic cancer group were significantly higher than those in benign pancreatic disease group and normal control group (P<0.05). The positive rates of CA199,CEA and CA125 in pancreatic cancer group were 87.50 %,71.42%,and 60.71%, respectively. The positive rates of the three tumor markers of the patients in pancreatic cancer group were obviously higher than those in other two groups (P<0.05).The sensitivity of CA199(87.50%)was the highest,while the specificity of CEA (9 1.9 6%)was the highest. The method of parallel combined testing increased the diagnostic sensitivity to 95.12% and negative predictive value to 90.47%,but its specificity and the positive predictive value were decreased. Serial combined examination increased the diagnostic specificity to 92.86% and the positive predictive value to 89.19%,but its sensitivity and negative predictive value were decreased.Conclusion Combined detection of serum levels of CA199,CA125 and CEA has higher sensitivity and specificity than single-marker detection in the diagnosis of pancreatic cancer.
