1.The study on the influence of blood specimen collection to the hospital inspection results

Objective To investigate the blood specimen collection site,inspection time and hemolysis influ-ence on biochemical test results.Methods The specimens of 2586 cases outpatients and hospitalized patients in our hospital who accepted blood routine and blood coagulation four check were analyzed.The hemolysis and qualified specimens of biochemical test results were compared.Results 2 586 specimens found unqualified blood samples in 18 cases,and the incidence rate was 0.70% (18 /2 586).Samples of different section and source were compared and found there were no significant difference (F =0.36,P >0.05).In 18 cases of unqualified blood samples due to technical problems and unqualified sample accounted for 88.89% (16 /18).The technical factors that lead to unqualified samples was significantly higher than that of the instrument factor or fixed factors,and the difference was statistically significant(χ2 =152.63,98.52,all P <0.05).In addition,hemolysis caused the most of the unqualified samples for the technical factors.To choose potassium infusion on the side ipsilateral to the site or from the opposite side of the site collected blood samples.Sodium,chlorine,creatinine,uric acid,glucose detection found in effect of blood on the same side of the infusion of the biochemical test results in varying degrees.Potassium,chlorine,sodium, creatinine decreased significantly,and uric acid,glucose and other indicators were significantly increased (t =4.51, 4.98,5.50,3.25,10.18,15.25,all P <0.05).Gamma transpeptidase,creatine kinase,sodium and uric acid and other index of hemolytic samples were significantly lower than those of normal samples,and alanine aminotransferase (ALT),direct bilirubin,potassium,phosphorus,serum creatinine and creatine kinase were significantly higher than those of the normal samples (t =14.85,21.21,7.15,4.86,10.33,4.02,3.11,8.20,7.54,5.11,all P <0.05).This may cause the depth in erythrocytes and some related substances.The time of collecting blood to sent to the room had obvious influence on the biochemical test results,alanine aminotransferase,aspartate amino transferase,glucose, creatine kinase and hydroxybutyrate and project them 1 h after submission were significantly lower than those of the standard inspection,and 1 hour after the submission of lactate dehydrogenase and creatine kinase isoenzyme index were significantly higher than those of the standardize inspection means (t =2.95,4.82,2.88,5.44,4.05,3.98, 4.89,all P <0.05).Conclusion Sampling sites,hemolysis and inspection time had effect on blood specimen,so for the collection of blood samples must be strictly in accordance with the requirements of the operation.
2.Reversed island flap pedicled with nutrient vessels of sural nerve for repairing soft tissue defect of lower leg,ankle joint and foot dorsum in 12 cases

From March 2002 to February 2006, 12 patients with soft tissue defects of lower third of leg, ankle and dorsum of foot, who were repaired with reversed island flap pedicled with nutrient vessels of sural nerve, were enrolled at Department of Plastic and Burn Surgery, Affiliated Hospital of Luzhou Medical College. Of the 12 patients, 6 cases were achilles tendon exposure caused by soft tissue necrosis after trauma, 6 cases chronic ulcer in leg ,ankle and dorsum of foot, respectively. All patients knew and agreed to participate in the experiment. The areas of soft tissue defect from 6 cm?7.5 cm-10 cm?12 cm. The maximal area of flap was 12 cm?14 cm. After debridement, the flap which designed based on the size and shape of defect, the line from fibular head to the middle point of lateral malleolus and achilles as axes was transfered to repair the defect, taking 6 cm on lateral malleolus as swivel. Eleven flaps survived postoperatively, and raw surface was primary healing. Margin necrosis was occurred in 1 patient, and healed after changing drugs. Follow-up was performed from 6-18 months in all cases. The texture and color of skin flap is similar to that normal skin, and the function and shape of recipient and donor site were satisfied. No secondary ulcer or functional disturbance, wear-resisting were found. The patients could walk and load normally. The reversed island flap pedicled with nutrient vessels of sural nerve is ideal donor site for soft tissue defect repairing of lower third of leg, heel, dorsum of foot and ankle.

Objective To construct an effective expression system for secretory canstatin, and to study its biological effects. Methods Canstatin cDNA was ligated with the cDNA of CEA signal peptide by SOE PCR, and the new fused fragment was cloned into eukaryotic expression vector pCMV-Script. The recombination vector pCMV-CEAS-Cans was transferred into human umbilical vein endothelial cells (HUV-EC) and human lung adeno-carcinoma cell line A549 by cationic liposome. The expression of canstatin mRNA together with CEAS mRNA in the transformed cells were detected by real time PCR method. Results Canstatin mRNA was detected in both transformed cells. Objective To study the expression of a new drug resistance-related gene cDNA fragment from human lung adenocarcinoma cell line in several types of lung cancer and their adjacent normal tissues. Abstract Objective:To study GST-? expression in breast cancer and axillary lymphnodes.Methods: The expression of GST-Tr proteinand mRNA was detected by means of immunohistochemistry and RT- PCR. Results: In normal breast tissue, cancer tissue , HE positive lymph-nodes and HE negative lymphnodes , GST-? protein expression rate is 0,60% ,77% ,43%; GST-? mRNA expression rate is 0,88% ,71%and 39% , respectively. Conclusion; GST-?gene whose expression is upregulated as tumor occurs and develops can be used as a tumor biomar-ker; GST-? expression in HE negative lymphnodes suggests the poor prognosis; drug-resistance response is more sensitive in lymphnode than inprimary cancer. The expression of GST-? gene may be regulated by RNA or before RNA transcription.
7.Canstatin high level expression system and its biology effects on lung cancer

Purpose:To study the biological effects of high secreted canstatin on human umbilical vein endothelial cells (HUVEC) and tumor model. Methods:Hypoxic responsive elements (HREs) were inserted in the upper stream of canstatin cDNA of a recombinant vector we constructed in a former research. The new recombinant vector named pCMV-3HRE-CEAS-Cans was transferred into A549 cells by cationic liposome; canstatin mRNA expression in the transformed cells under oxic and anoxic condition was detected by Taqman PCR. Then we designed a co-cultured assay: HUVECs were co-cultured with recombinant vector transformed A549 cells with Transwell plates, and the proliferation and apoptosis of co-cultured HUVECs were evaluated with 3H-thymidine incorporation method, TUNEL method respectively. Finally the vector was introduced into tumor tissues of lung cancer-bearing nude mice, and the biological activity in the tumor tissues was tested by micro-vessel count (MVC). A vector of canstatin we constructed before was used as controls in above experiments.Results:The pCMV-3HRE-CEAS-Cans vector was successfully constructed and transferred into A549 cells. canstatin mRNA was detected both in the recombinant vector transformed A549 cells and the pCMV-CEAS-Cans transformed A549 cells, moreover the expression of canstatin in the new vector transformed A549 was significantly higher than that of the controls under hypoxic condition. ~(3)H-TdR intake rate of HUVECs was decreased markedly, and a large amount of them underwent apoptosis when they were co-cultured with recombinant vector transformed A549 cells. 8.Study on Release of Cardiac Troponin Ⅰ Following Coronary Artery bypass Grafting

Objective To investigate the change of serum cardiac troponin I(cTnI)following coronary artery bypass grafting.Methods 55 OPCABs and 55 cCABGs were performed.Serum cTnI were measured before operation(T1)and immediately after operation(T2)and 1d(T3),2d(T4),3d(T5),4d(T6),5d(T7),6d(T8),7d(T9),10d(T10),14d(T11).Results The serum cTnI level increased differently after OPCAB and cCABG and then returned to normal level gradually.The peak value of serum cTnI level in OPCAB was lower than that in cCABG.There were 9 cases on myocardial infarction,of which OPCAB 4 cases,cCABG 5 case,with serum cTnI level increasing;and there weren't death.Conclusion The serum cTnI in OPCAB and cCABG both increased and then decreased to normal level gradually.Measuring serum cTnI could diagnose myocardial injury and indicate prognosis.
9.The length measurement of adult women abdominal aortic bifurcation to the femoral artery puncture points on interventional methods

Objective To measure the length between the bifurcation of abdominal aorta to the femoral artery puncture point in adult women.Methods 127 patients were enrolled in the study.Femoral artery puncture using of the roberts uterine catheter (RUC) for the diagnosis and treatment of obstetrics and gynecologic disease was conducted.When the RUC bending vertex code located at the bifurcation above, fingers fixed femoral artery puncture point at the mouth of the catheter's sheath and evacuate the catheter and measure the code to the fixed position.Results In this group, the length of the bifurcation of abdominal aorta to the femoral artery puncture point was 24.38-25.33 cm.There was no significant difference in the length with patient age (P>0.05), but there were statistical significances with the patients height and weight(P<0.05).Conclusion The length of adult women abdominal aortic bifurcation to the femoral artery puncture point has no significant difference with patient age.The individual height and weight are very much significant.The length estimation can be done according to the patient's height and weight for low abdominal aortic catheter,on clinic for avording X-ray irradiation or no X-ray equipment condition.
10.A radiological study of dysphagia and rehabilitative outcome in post-stroke patients

Objective To explore the effects of videofluoroscopic swallowing study (VFSS) in the assessment of dysphagia in post-stroke patients. To study the representation and rehabilitative outcome of dysphagia following stroke. Methods Seventy post-stroke patients and 80 normal adults were assigned to study group and control group and underwent VFSS. Abnormal signs on VFSS were compared between 2 groups. Aspiration patients received swallowing therapy for 4 weeks followed by VFSS again. Abnormal signs on VFSS were compared with those of pre-treatment. Results Penetration/aspiration, oral retention and pharyngeal retention occurred respectively at a frequency of 5.0%, 13.4% and 25.3% in control group in mild status, 45.0%,46.5%, 48.9% in study group in moderate to severer status. 10 times of silent aspiration were seen, accounting for 24.4% of all aspiration.
