1.Development and application of new-type animal dental stent

Objective To develop a new-type animal dental stent for the experiment of canine pulp regeneration.Methods The stent was composed of an upper support frame,an oral limit bracket and a lower support frame,which were aligned from top to bottom.A sleeve joint was made for the connection structure of the bracket,and the height and the width both could be adjusted to adapt to different animals' mouths.The rubber dam could be spread into tension and concave form by rotating the upper and lower support frames' ends.Results By resizing the animal dental stent based on the structure of animal facies maxillaris,medical staffs could proceed the examination and treatment to animals effectively,including the throat examination,oral examination,oral care,dental treatment (such as root canal therapy and dental filling) and other medical behaviors.Conclusion The experimental result shows the feasibility of utilizing the stent in the process of animal experiment or diagnosis and treatment.Moreover,the stent proves the capacity in protecting the soft tissue,widening the vision of operation field,getting rid of the saliva contamination,which is worthy popularizing in animal experiments and animal medical care.
2.Single nucleotide polymorphisms research of osteopontin gene in Zhuang populations in Guangxi

Objective To investigate the genotype and allele frequencies of osteopontin gene single nucleotide polymorphisms (SNP) rs11728697and rs9138 in Zhuang populations in Guangxi ,and to compare the distribution of osteopontin polymorphisms a‐mong different races .Methods The osteopontin gene rs11728697 and rs9138 polymorphisms were detected by SNaPshot SNP gen‐otyping technique in 150 Zhuang populations in Guangxi ,the genotype and allele frequencies of osteopontin gene rs 11728697 and rs9138 polymorphisms were analyzed in Zhuang populations compared with the other four populations (HapMap‐CEU ,HapMap‐YRI ,HapMap‐JPT ,HapMap‐HCB) from HapMap database .Results The most common genotype and allele of osteopontin gene rs11728697 polymorphism in Zhuang populations in Guangxi were CC(42 .7% ) and C(62 .7% ) ,and the most common genotype and allele of osteopontin gene rs9138 polymorphism were CA (51 .3% ) and C(63 .0% ) .There were no significant differences in the gen‐otype and allele frequencies of osteopontin gene rs11728697 and rs9138 polymorphisms between male and female groups ( P >0 .05) .The genotype and allele frequencies of osteopontin gene rs11728697 polymorphism were significantly differenct compared with HapMap‐CEU ,HapMap‐JPT and HapMap‐YRI(P< 0 .05) ,but were not significantly different compared with HapMap‐HCB (P> 0 .05) .The genotype and allele frequencies of osteopontin gene rs9138 polymorphism were significantly differenct compared with HapMap‐CEU and HapMap‐YRI(P < 0 .05) ,but had no significantly different compared with HapMap‐JPT and HapMap‐HCB(P> 0 .05) .Conclusion There are significant differences in the genotype and allele frequencies of osteopontin gene rs 11728697 and rs9138 polymorphisms between Zhuang populations and other ethnic populations ,and this variation might contribute for a varie‐ty of clinical manifestation and morbidity of some osteopontin related diseases .
3.Median effective dose of 0.5% ropivacaine for ultrasound-guided femoral nerve block: dose per unit femoral nerve cross-sectional area

Objective:To determine the median effective dose (ED 50) of 0.5% ropivacaine based on femoral nerve cross-sectional area for ultrasound-guided femoral nerve block. Methods:Patients of both sexes, aged 18-64 yr, of American Society of Anesthesiologists physical status I or Ⅱ, with body mass index of 20-30 kg/m 2, scheduled for elective open reduction and internal fixation for patella fracture or removal of patella fracture by internal fixation, were enrolled in this study.Ultrasonic localization of femoral nerve was performed for measurement of the femoral nerve cross-sectional area, and 0.5% ropivacaine was injected based on the area.ED 50 was determined by Dixon′s up-and-down sequential method.The initial dose was 0.22 ml/mm 2, and the difference between the two successive doses was 0.02 ml/mm 2.The effective block was defined as complete loss of pain sensation in the areas of anterior skin of knee joint, skin on the inner side of the calf and dorsal medial skin of the foot and the degree of motor block was in stages 1-3 assessed using Brunnstrom motor function within 30 min after nerve block.Nerve block was considered ineffective if pain occurred in any nerve distribution area mentioned above.The study was terminated if 7 effective and ineffective alternating waves occurred.ED 50 and 95% confidence interval (CI) were calculated using Probit analysis. Results:Twenty-seven patients were enrolled in the study with the femoral nerve cross-sectional area (75±5) mm 2.ED 50 (95%CI) of 0.5% ropivacaine for ultrasound-guided femoral nerve block was 0.106 (0.069-0.125) ml/mm 2. Conclusion:ED 50 of 0.5% ropivacaine based on femoral nerve cross-sectional area for ultrasound-guided femoral nerve block is 0.106 ml/mm 2.
