Glycyrrhiza uralensis Fish Polysacchoride(GPS)was observed to have mitogenic activity on murine spleen cell proliferation in vitro,The results showed that this mitogenic activity of GPS may be directed to B Iymphocytes,i.e.GPS is a new mitogen of murine B lymphocytes.Furthermore,it was found that B lymphocytes stimulated by GPS was M? depedent and T cell-indepedent.On the contrary,antibodies and IL-2 production was inhibited by GPS in vivo.
2.The effect of amygdaloid nuci ei on immunogical function induced by stress

Rat with amputation operation stress possessed a decline of NK activity and splenocyte pro-liferation to mitogen ConA.Amygdaloid nuclei lesion could antagonize the decrease of those ofcell-mediated immunity.Further results showed that serum corticosterone and methionine-enkephahn was high in stress' rats.Amugdaloid Nuclei lesion could antagonize the increase ofserum corticosterone level,but seem no dffeet on serum methionine-enkephalin level,thereforesuggesting that it is related to ratio of corticosterone to enkephalin changed by decrease of corti-costerone that amygdaloid neuclei lesion antagonized the decline of immunogical function inducedby stress.

The levels of c-fos mRNA in U937 cell line were determined with in situ hybridization.Theresults showed that TPA induced the expression of c-fos gene with a relationship of dosage-ef-fect.It was also proved that GM-CSF,TNF,IL-9,IL-3 and IL-6 increased the expression of c-fos gene.

The systemic anaphylaxis murine model was induced by the injection of oval albumin (OVA)in aluminum hydroxide gel adjuvant. The death rate varied with the mouse species: 40-90% in Swiss and 69% in average,no death in C57BL/6 and 615 but some symptoms. developed. Swiss mice was adopted in this experiment.90% of mice pretreated with Lx remained living after allergen challenge,but only 20% in normal control. For elucidation the possible mechanism of Lx action,the study was carried out on detection of anti-OVA IgE specific antibody and histamin concentration in lung tissues. Passive cutaneous anaphylaxis(PCA)titers for anti-OVA IgE antibody in serum ranged from undilution to 160 in control group and not detectable in group pretreated with Lx. In addition, after absorption of PCA positive sera with anti-IgG antibody.the PCA activity was not affected. The initial study of the tumour DNA fingerprints using MYO minisatellite DNA probe wascarried out,and then,by means of DNA recombinant techniques,the fragment of MYO min-isatellite DNA probe obtained from plasmid pUC19-MYO was inserted into plasmid pGEM-4Z containing RNA polymerase promotor,thus a sub-clone refferred as pGEM-4Z-MYOwas constucted.That made an offer of the conditions of preparing RNA probe in order to in-cerase the sensitivities of DNA fingerprinting and laid a foundation for raised the efficiency of de-tecting the polymorphism of the minisatellite DNA.
7.Clinical effects of steel plate fixation for the failure of arthrodesis of ankle by screw fixation.

OBJECTIVETo observe the therapeutic effects of steel plate fixation after the failure of arthrodesis of ankle by screw fixation.
METHODSFrom August 2001 to October 2011, 15 patients were with steel plate fixation after failure of arthrodesis of ankle by screw fixation. Among patients, 9 cases were males and 6 cases were females,ranging age from 40 to 65 years old with the average of 56 years old. Ten cases were in left and 5 cases were in right. Screws were removed and steel plate was fixed intraoperatively, and plaster external fixation for postoperation. Clincal effect were evaluated according to AOFAS scoring system from pain, waliking ability and aligment before and after operation, and X-ray was used to evaluate joint fusion after operation.
RESULTSAll patients were followed up, and the duration ranged from 4 months to 4 years with an average of 2 years. The incison were healed in stage I. No ankle pain,injury of blood vessel and nerve,infection and farilure of internal fixation occuerred. The AOFAS score increased from 36.86 +/- 8.32 preoperatively to 85.09 +/- 4.65 (t = -26.366, P = 0.000).
CONCLUSIONSteel plate fixation after the failure of ankle arthrodesis of screw fixation has the advantages of rigid stability, simple manipulation and high success rate, less pain, perfect recovery.

8.Studies on anti-proliferation and inducing apoptosis effects of paeonol on human esophageal cancer cell line Eca-109 in vitro and in vivo

Aim To investigate the inhibitory effect of paeonol (Pae) on the human esophageal cancer cell line Eca-109 in vitro and in vivo and its effect on apoptosis.Methods Cytotoxic effect of Pae on Eca-109 cells cultured in vitro was measured by MTT assay.Anti-tumor activity was performed on BALB/c nude mice xenografts model.The morphologic changes of tumor tissue were observed under light microscope and transmission electron microscope.The apoptosis index was assessed by TUNEL.Results Pae had significant in- hibitory effect on the proliferation of Eca-109 cells,and the IC50 value was 0.342 mmol?L-1.In the model of human esophageal cancer xenografts in BALB/c nude mice,the inhibitory rates of Pae group (25、50、100、200 mg?kg-1) were 10.67%、23.54%、27.91% and 34.46% respectively.In vivo administration of Pae 100 mg?kg-1 combined with cisplatin 5 mg?kg-1 resulted in a significant inhibition of Eca-109 tumor growth with the inhibitory rate of 77.91%,compared with cisplatin used alone (58.71%).The more apoptotic tumor cells could be seen under light microscope in every theraperutic groups than those in control.Changes of ultrastructure of tumor cells including concentration and side accumulation of the nuclear chromatin,and the fragmentation of the nuclear was observed under transmission electronic microscope.Apoptosis body was also found.The apoptosis indexes of every theraperutic groups were significantly different from the control.Conclusion Pae can inhibit the cell growth and induce apoptosis in human esophageal cancer cell line Eca-109 in vitro and in vivo.
9.Subdiaphragmatic vagotomy reduces the responses of fever and discharge of neurons in PVN to LPS.

AIMTo study the possibility that responses of fever and discharge of neurons in PVN to intraperitoneal administration of LPS are mediated by vagal afferents.
METHODSRectal temperature of rat was detected by digital temperature detecting instrument. Glass micropipette placed in PVN was used to record unit discharges of neurons in it, before and after LPS was injected into PVN in normal rats and vagotomy rats.
RESULTSThe rectal temperature change value in vagotomy LPS group was significantly decreased compared with that in sham LPS group, and there was striking difference between them (P < 0.05). The discharges of neurons in PVN was increased in the normal rat in response to LPS. The discharges of neurons in PVN had no significant change in the vagotomy rats in response to LPS.
CONCLUSIONThe results indicate that vagus nerve may be one of the pathways of peripheral LPS signal communicating to CNS.
