2.Sesquiterpenoids from Solanum lyratum.

Ten compounds were isolated and purified by column chromatography over silica gel, preparative TLC, and Sephadex LH-20 from the whole plant of Solanum lyratum. The structures were elucidated on the basis of physico-chemical properties and spectral data as 1beta-hydroxy-1 ,2-dihydro-alpha-santonin (1) , boscialin (2) , blumenol C (3), 3beta-hydroxy-5alpha, 6alpha-epoxy-7-megastigmen-9-one(4), dehydrovomifoliol(5) , blumenol A(6), (1'S,2R,5S, 10R) -2-(1', 2'-dihydroxy-l1'-methylethyl) -6,10-dimethylspiro[4,5] dec-6-en-8-one(7) , (1'R,2R,5S,10R)-2-( 1',2'-dihydroxy-l '-methylethyl) -6,1 l0-dimethylspiro[4,5]dec-6-en-8-one( 8) , 2-(1',2'-dihydroxy-1 '-methylethyl) -6,1 0-dimethyl-9-hydroxyspiro [4,5] dec-6-en-8-one (9) , and grasshopper ketone (10). Compounds 1-10 were isolated from this plant for the first time.

3.Optimization of Fermentation Medium for S-adenosylmethionine Production by Candida sp.YQ5

The optimized cultural medium of fermentation for Candida sp.mutant strain YQ5 to produce S-adenosylmethionine was studied.The results of single factor experiment showed that the most favorable pH value,carbon source,nitrogen source organic nutrient is 6.0,8% sucrose,1.5% tryptone and 2% yeast extract,respectively.As to inorganic salts,MgSO4?7H2O,CaCl2,FeSO4?7H2O,CoCl2,CuSO4?5H2O,H3BO3 could improve accumulation of the intercellular SAM.The ingredients of the culture medium are also opti-mized by the orthogonal experiment.Fermentation for 48 h under the optimal condition resulted in AdoMet production at 1740.0 mg/L.

Objective To explore the association of Gly71Arg mutation in gene of bilirubin uridine 5'-diphosphate-glucuronosyltransferase(UGT1A1)and neonatal jaundice in Beijing city Han population.Methods The genotypes and alleles of the Gly71 Arg polymorphism for UGT1A1 gene were identified by polymerase chain reaction-restricted fragment length polymorphism assay in infants of Beijing city Han population of China,including 96 infants with neonatal jaundice[serum bilirubin(307.06?38.5)?mol/L,indirect bilirubin(292.9?35.9)?mol/L] and 101 healthy control infants [serum bilirubin(131.2?42.1)?mol/L,indirect bilirubin(126.3?39.7)?mol/L].The genotypes and allele frequencies of the polymorphism were compared between infants with neonatal jaundice group and healthy infant group(control group).The effect of polymorphism in infants with neonatal jaundice group on serum bilirubin level were analyzed.Results There were significant differences in genotypes distribution in Gly71Arg polymorphism for UGT1A1 gene between the 2 groups(?2=9.47 P=0.002).Compared with control group,neonatal jaundice group had significantly higher Arg allele frequency in the polymorphism for UGT1A1 gene(?2=10.34 P=0.001).There were independent effects of Gly71Arg mutation in the gene on serum bilirubin level in neonatal jaundice group,at the carriers of homozygote of the Arg allele of Gly71Arg polymorphism had higher serum bilirubin levels compared to carriers of heterozygote of the Arg allele of the polymorphism and non-carriers of the Arg allele of the polymorphism(Pa
5.Genetic polymorphisms of 9 X-chromosome short tandem repeat loci in a Inner Mongolia Ewenki population and their forensic evaluation.

OBJECTIVETo investigate the alleles and genotypes frequency of 9 short tandem repeat (STR) loci on the X chromosome (DXS6789, DXS101, DXS8378, DXS7132, DXS7133, DXS7423, DXS6804, DXS6799, HPRTB) of Ewenki individuals living in Inner Mongolia Autonomous Region of China.
METHODSThe 9 X-chromosomal STR loci were analyzed with polymerase chain reation (PCR), followed by polyacylamide gel electrophoresis and silver staining. Software SPSS13.0, Genepop, Fstat and Powerstats were used to evaluate their polymorphism diversity and potential forensic application.
RESULTSAllele frequencies and genotype frequencies of 99 unrelated Ewenki individuals were obtained. Among the 9 loci, DXS6789, HPRTB showed less polymorphism and diversity in the population. The diversity of DXS7132 has no statistical difference between Ewenki population and other 4 Asian populations.
CONCLUSIONExcept DXS6789, HPRTB, the other 7 X-chromosomal STR loci are appropriate for individual identification, paternity test involving a female child, and studies on related disease. DXS7132 should be excluded when being used to distinguish diversity difference among populations.

6.Expression of NFkappaB p65 and its target genes in gastric cancer and precancerous lesions.

OBJECTIVETo study the expression of NFkappaB p65 and its target genes in intestinal metaplasia (IM), dysplasia (Dys), gastric cancer (GC) infected with Helicobacter pylori (Hp) and explore the mechanism of infection by cytotoxin-associated antigen A expressing Hp (CagA(+)Hp) in the development of gastric cancer.
METHODSCagA antibody in blood sample of 289 patients was determined by ELISA. Hp was detected by rapid urease test and Warthin starry staining. Expression of NFkappaB p65 and its target genes in IM, Dys and GC was examined by immunohistochemistry.
RESULTSIn IMI approximately II, IMIII, DysI, DysII approximately III and GC, the expression of NFkappaB p65 was significantly higher in patients with CagA(+)Hp infection than those without CagA Hp infection. In IMIII and DysII approximately III, the expression of NFkappaB p65, c-myc, CyclinD(1) and bcl-xl was significantly higher in patients with CagA Hp infection than those without CagA Hp infection. In gastric cancer infected with CagA(+)Hp, the expression of NFkappaB p65, c-myc, CyclinD(1) and bcl-xl was significantly higher in intestinal type than in diffuse type.
CONCLUSIONThere are different mechanisms in intestinal type and diffuse type in the development of gastric cancer. The occurrence of intestinal type gastric cancer is associated with CagA(+)Hp infection which by NFkappaB p65 upregulating the expression of c-myc, CyclinD(1),bcl-xl in patients with IMIII, DysII approximately III. It may be an effective method to prevent gastric cancer by inhibiting NFkappaB p65.

7.Growth status of children under 7 years in Wuzhong City, Ningxia Hui Autonomous Region, China and its influential factors.

OBJECTIVETo investigate the growth status of children under 7 years in Wuzhong City, Ningxia Hui Autonomous Region, China and its influential factors, and to provide a basis for related intervention measures.
METHODSChildren under 7 years were selected from two county-level districts in Wuzhong by stratified cluster sampling, and their growth status were evaluated by the Z score method.
RESULTSThe prevalence rates of growth retardation, underweight, and wasting were 12.58%, 5.71%, and 5.55% respectively. The height-for-age Z score, weight-for-age Z score, and weight-for-height Z scores were -0.26±2.50, 0.29±4.54. and 0.65±3.02 respectively. There were significant differences in the prevalence rate of wasting among children of different ethnic groups (P<0.05); also, there were significant differences in the prevalence rates of growth retardation and underweight among children from different regions and with different age (P<0.05). The main influential factors for growth retardation were region (OR=0.369, P<0.001), ethnic groups (OR=1.694, P=0.027), and age (OR=1.143, P=0.002). The main influential factors for underweight were region (OR=0.453, P=0.001) and age (OR=1.204,P=0.002). The main influential factor for wasting was nation (OR=1.735, P=0.024).
CONCLUSIONSIn Wuzhong, children under 7 years have poor growth status, which are related to ethnic groups, region, and age.

8.Analysis on the incidence and risk factors of campus violence among college students in Guangzhou.

OBJECTIVETo explore the incidence and risk factors of campus violence in Guangzhou.
METHODS2200 college students in three universities in Guangzhou were selected by cluster sampling method and were interviewed with self-designed questionnaire about the incidence and risk factors of campus violence in 2010. The final analysis sample was 2103. Chi-square test was used to analyze the gender, grade and major distribution of campus violence. Logistic regression method was used to analyze the influencing factors of campus violence in bully and victim.
RESULTSThe incidence of campus violence in Guangzhou was 69.9% (1471/2103). In boys and girls the incidence of campus violence was 75.6% (830/1098) and 63.8% (641/1005) (χ(2) = 34.82, P < 0.05). The incidence of bully and victim of campus violence was 63.6%(1338/2103) and 55.3% (1163/2103). The incidence of bully and victim in boys was 70.9%(778/1098) and 60.0%(659/1098), and in girls was 55.7% (560/1005) and 50.1% (504/1005) (χ(2)(bully) = 51.93, χ(2)(victim) = 20.68, P < 0.01). The incidence of psychological violence was the highest (68.0%, 1430/2103), followed by sexual violence (34.2%, 719/2103), the incidence of physical violence was the lowest (16.5%, 348/2103). Results of logistic regression showed that boys (OR = 1.60), arts (OR = 1.82), with siblings (OR = 1.31), the living expenses was not enough (basic enough OR = 1.35, not enough OR = 1.54), playing the computer games (OR = 1.70), playing tricks such as sliding plate (OR = 2.03), loving violence program (general OR = 1.30, very like OR = 1.44), mother with gamble behavior (OR = 4.29), father's indulgent education style (OR = 1.60), been bullied by others before high school (OR = 1.61), dissatisfaction to the environment and rules of campus (nothing special feeling OR = 1.67, some dissatisfaction OR = 1.89), been treated badly by primary school teacher (one kind OR = 1.35, two kinds and above OR = 1.90)were the risk factors of bully. Boys (OR = 1.23), minority (OR = 1.71), with siblings (OR = 1.39), bad behavior and habit (OR = 1.32), the bad family economic conditions (general OR = 1.51, difficult OR = 1.88), mother with gamble behavior (OR = 2.33), father's indulgent education style (OR = 1.37), occasional physical punishment by mother (OR = 1.35), been bullied by others before high school (sometimes OR = 1.61, often OR = 1.85), high pressure during the study (a little high OR = 1.37, very high OR = 1.40), dissatisfaction to the environment and rules of campus (some dissatisfaction OR = 1.56, completely dissatisfaction OR = 2.04), been treated badly by primary school teacher (one kind OR = 1.70, two kinds and above OR = 2.04)were the risk factors of being victim.
CONCLUSIONThe campus violence in Guangzhou is serious, especially the psychological violence and sexual violence. And the risk factors of campus violence in bully and victim are multifold.

10.Development of MTS/pms colorimetric assay in the proliferation of leukemic cells.

In order to establish a new more rapid, safe and sensitive colorimetric assay for the proliferation of leukemic cells, MTS/pms has been developed. This automated colorimetric assay is based on the characteristic of viable and metabolically active leukemic cells to cleave MTS/pms into a water-soluble product whose optical density is determined at 492 nm by an automated microtiter-plate reader photometer. The results indicated that only active leukemic cells cleaved MTS/pms into product measured, and dead cells did not reduce MTS/pms. A linear relations hip were found between the viable cell number and optical density of MTS/pms cleaved by HL-60 and K562 cell (r = 0.963). Compared with MTT and INT assays, the reduced product of MTS/pms is water-soluble. It is concluded that MTS/pms colorimetric assay is more rapid, accurate and sensitive for the bioassay of proliferation of leukemic cells.
