In order to develop characteristic folk medicine resources in Jiangxi, a pharmacognostical study was systematically performed for four different origin plants of Sikuaiwa, the result of study provides the microscopic features of powder and tissue of the crude drug. The research provided reference for the identification of Sikuaiwa, as well as a theoretical basis for the further development and the formulation of quality standards.

3.Molecular identification of Tibetan medicine Qianghuoyu by CO I.

The CO I gene sequences of Qianghuoyu, Pachytriton labiatus and Gehyra mutilata were achieved by PCR amplification and bi-directional sequencing. Furthermore, a pair of specific primers SJYW1 and SJYW2 in the non-conservative district were designed through sequence alignment. The PCR reaction condition was established by changing the annealing temperature and cycle numbers. The results showed that 350 bp DNA fragment was amplified from Qianghuoyu in PCR with annealed temperature at 54 °C and the cycle number was 25 cycles, whereas not any DNA fragment was amplified from P. labiatus and G. mutilata under the same reaction condition. This method is well-performed in the identification of Qianghuoyu for its excellent specificity and repeatability.

4.Analysis of the reconstruction of binocular visual function in 76 cases of pediatric ocular trauma

AIM: To evaluate the effect of binocular visual function training in pediatric ocular trauma. METHODS: There were 76 patients (76 eyes) that were hospitalized with a primary diagnosis of ocular injury at the Affiliated Hospital of Medical College, Qingdao University between January 2006 and December 2009. Binocular visual function training was given after primary wound repair. Far stereopsis function were checked using AIT-1000 synoptophore fusion, and near stereopsis function was checked using Titmus stereogram.Binocular visual function was compared before and after training. RESULTS: Before binocular visual function training,26 eyes(34%)had no binocular vision, after training there were only 16 eyes(21%) without binocular vision. Before undertaking binocular visual function training with fusion, only 27 eyes (36%) had binocular vision, after the training there were 48 eyes (63%) with binocular vision. Before undertaking binocular visual function training with far stereopsis ,there were 23 eyes (30%) with binocular vision, after the training there were 29 eyes (38%). Before binocular visual function training with near stereopsis, there were 14 eyes (18%) with binocular vision, after the training there were 33 eyes (43%) with binocular vision. There was a significant difference in the number of patients with binocular vision before and after binocular visual function training. CONCLUSION:The training is useful for the reconstruction of binocular visual function in pediatric ocular trauma.
5.Discussion about the Acupuncture and Needling Sensations

This article aimed to discuss the definition of the ancient characterZhen (needle) from three aspects: the origin of the character, the meaning of hexagramXian, and application of interaction in acupuncture. The writers hold thatZhen means feeling, and the definition ofZhen reveals that the key to achieving acupuncture efficacy is needling sensation. In clinic, when practicing acupuncture, the writers should especially pay attention to the use of interactions between the left and the right, the upper and the lower, the anterior and the posterior, and intersections among meridians, which will benefit the enhancement the clinical efficacy of acupuncture-moxibustion therapy.
6.Preliminary Exploration of Sawada's Thought of Acupuncture Diagnosis and Treatment from "The Essence of Acupuncture"

By studying doctor Sawada's theoretical thought related to clinical experience in "The Essence of Acupuncture", it is found that the data recorded by Daita Bunji according to what he saw and heard provide many returning-to-nature treasures for mining. That plays an important guiding role in expanding the idea of acupuncture treatment and improving clinical therapeutic effects. This article outlines doctor Sawada's thought of acupuncture treatment from the theory of "one original, two opposite, three large, four spiritual and five distributed", the theory of abdominal"卍"symbol, and the development and application of tri-jiao theory. It is intended to make that well known. Posterity is lucky if it can be carried forward.
7.Chromosomal localization of primary gout susceptibility gene in a Chinese pedigree

Objective To determine chromosomal localization of the primary gout susceptibility gene in a pedigree.Methods The clinical data and the peripheral blood samples were collected in the pedigree members and the genomic DNA was extracted from peripheral blood.A genome-wide screening was performed using 400 micro-satellite DNA markers in this family,and linkage analysis was used to determine the chromosomal location of the primary gout susceptibility gene.Results Linkage analysis showed that the maximum LOD score reached 1.50 at marker D4S1572 (at recombination fraction?=0.00).Conclusion Since D4S1572 is localized at 4q25,the primary gout susceptibility gene of this pedigree is localized at 4q25.
8.PFGE Genotyping of MRSA Outbreak in a Burn Unit

To explore the epidemiological character of Methicillin-resistant Stapkylococcus aureus (MRSA) by the phenotyping and genotyping motheds and to investgate the source, transmission, and the spread of nosocomial MRSA infection, consequently, reducing the nosocomial infection of MRSA. In this study, 19 MRSA strains were isolated from patients and environment in a hospital in two months. Patterns of resistantce against 16 antimicrobial agents and pulsed-field gel electrophoresis ( PFGE) of these strains were analyzed to find the relationship among those isolates Clustering analysis was made from the patterns. Some isolates with high homology was found in 19 MRSA, 11 of them belong to type A, and 8 of them belong to the same subtype A1. They were endemic in burn ward, oncological ward and ICU. In addition, 4 isolates were clustered into group B, all found in the same ward of burn unit Thus, our results indicated a outbreak of MRSA ( A type) in this hospital and the potential prevalence of MRSA (B type) , which might be mediated by health care stuff. It is essential to enhance the infection control implementation and to utilize the PFGE genotyping system for the real-time surveillance of MRSA.
9.Application of a self-made steel wire guide in the treatment of avulsion fractures of tibial posterior cruciate ligament.

OBJECTIVETo explore the effect of a self-made guiding needle of steel wire in guiding the wire through the tibial tunnel for the treatment of avulsion fractures of tibial posterior cruciate ligament with open reduction and wire fixation.
METHODSFrom February 2011 to June 2014, a total of 22 patients with avulsion fractures of tibial posterior cruciate ligament underwent surgical treatments were analyzed, including 14 males and 8 females with an average age of 35.6 years old (ranged, 17 to 63 years old). According to Meyers classification, 9 patients were classified as type II, 13 patients were classified as type III. All the patients underwent open reduction and wire fixation with medial knee "L" shape approach. A wire guiding needle was used to guide the wire through the tibial tunnel during operation.
RESULTSWith the assistance of wire guidance needles, wires passed through the tibial tunnel rapidly during the operation in all the 22 patients. All the patients were followed up, X-ray imagings 6 months after operation showed the fractures healed well. The average follow-up time in all patients was 6 months (ranged, 6 to 12 months). The averaged Lysholm knee score in 22 knee was 92.7 +/- 3.4. All patients' posterior drawer test were negative.
CONCLUSIONSelf-made wire guiding needle can simplify the operation procedures in which the wires pass through the tibial tunnel, shorten the operation time, reduce the surgical trauma and complications, and be worthy of clinical application.

10.Isolation of gastric cancer stem cells based on colony morphology and detection of their sensitivity to 5-fluorouracil.

OBJECTIVETo investigate the sensitivity of gastric cancer stem cells (GCSCs) to 5-fluorouracil (5-FU), and to explore the cytological mechanism of drug-resistance of gastric cancer.
METHODSImmunohistochemical staining was employed to examine expression of stem cell marker CD44 and drug-resistance protein thymidylate synthase (TS) in 71 cases with gastric cancer. Based on morphology of cell colonies derived from AGS gastric cell line, colonies consisting of GCSCs were isolated, and expression of CD44 and TS, as well as self-renewal capacity of GCSCs were detected. Sensitivity of GCSCs to 5-FU was examined through CCK-8 assay.
RESULTSThe positive rates of CD44 and TS were 59.2% (42/71) and 56.3% (40/71) in gastric cancer, and expression of CD44 was associated with that of TS (χ(2)=12.76, P<0.01; Kappa=0.41). Serial sections indicated that CD44+ cancer cells simultaneously expressed TS. AGS developed morphologically diverse colonies, and the GCSCs colonies exhibited a tight and regular shape, which were called holoclone. Holoclones expressed CD44 and TS strongly, possessing capacity of robust self-renewal and forming a lot of second passage clones after incubation. Subclones expressed CD44 and TS weakly, forming less second passage clones. Paraclones almost did not express CD44 and TS, forming no second passage clone after incubation. Affected by 5-Fu, three holoclones showed less growth inhibition compared with another colony type and wild-type AGS cells. Furthermore, IC50 of 5-FU for three holoclones was (113.43±5.81), (272.68±25.75) and (118.14±17.75) μmol/L respectively, which were significantly higher than that of one subcolony type [(16.97±1.01) μmol/L] and AGS cells [(27.52±0.59) μmol/L] (all P<0.05).
CONCLUSIONSGCSCs possess lower sensitivity to 5-FU, and may play a critical role in drug-resistance of gastric cancer.
