1.Impulsion therapy of children's nephrotic syndrome

Objctive To search for the therapeutic method of children's nephrotic syndrome.Methods Sixty-six cases oe children's nephrotic syndrome were randomly divided into 2 groups,impulsion group (34 cases) and control group (32 cases). Dexamethasone (1.5~3 )/mg (kg?d) added into (100~150)ml 10%GS solution, intravenous drip in impulsion group, one time a day, totat 3 days, the fourth day stoped. The fifth day started again and used one time evcry two days, total 6 times. Prednisone(1.5~3)mg/(kg?d) were taken next day and total 4 weeks, then grandually decreased the dose. Only prodnisone was used in control group, the method and dose were the same as impulsion group.Results Complete remission. partial remission inefficacy ere 23, 7 and 4 cases respectiye1y in impulsion group and 22, 5 and 5 cases respectively in control group, the effective rates of the 2 group are 88.23%and 84.38% (P>0.05). The times of state of illness stabilization are respectively 11.3?7.2 and 10.48?6.34 months in the 2 groups. The side effect of impulsion group is bigger than that of control group.Conclusion Children's primary nephrotic syndrome should be treated for 8 weeks by routine hormone induction therapy, if no remission, impulsion therapy could be used.
2.Endoscopic treatment of benign ureterointestinal anastomotic strictures in patients with urinary diversion

Objective To evaluate the clinical efficacy of endourologic treatment of benign uret-erointestinal anastomotic strictures in patients with urinary diversion. Methods Nine cases of benign ureterointestinal anastomotic strictures with a length of 1-3 cm following radical cystectomy and uri-nary diversion accepted endourologic treatment. 8 cases were treated by antegrade percutaneous ap-proach, 1 case by retrograde ureteroscopic approach. The strictures received balloon dilation, and ure-teral stents indewelled. Results In a follow up of 0.5-5.0 years, 1 case received percutaneous ne-phrostomy for complete ureterointestinal anastomotic atresia and refused to open operation reconstruc-tion. 5 cases had no recurrence after 2-3 endoscopic sessions. 3 cases needed long time ureteral stents indwelled. Conclusion Endourological technique for ureterointestinal strictures following urinary di-version avoided the disadvantages of open operation.
3.Research progress on musk secretion mechanism of forest musk deer.

Forest musk deer (Moschus berezovskii), a rare wild medicinal animal, is listed under the category of the state key protected wildlife list of China. Musk, secreted by the musk glands, is with high economic and medicinal value and used as precious traditional medicine in China. In order to meet the needs of musk in Chinese traditional medicine, forest musk deer farming was conducted in 1950s, but the research progress on musk secretion mechanism was slow. Therefore, by reviewing the histological and anatomical structure of forest musk deer musk gland, the relationship between sex hormones and the musk secretion process, and the molecular mechanism of the musk secretion, the existing problems in investigating the musk secretion mechanism were analyzed and the development trends in this field were also discussed, in order to provide a reference for further studies on the musk secretion mechanism and improve musk production of forest musk deer.

7.Effect of adult-onset hypothyroidism on neurogranin expression in hippocampus of adult male mice

Adult male Kunming mice were divided into normal control group,propylthiouracil(PTU) treated group and PTU+T_4 treated group.Neurogranin(Ng)protein expression in the hippocampus after 8 weeks was assayed by Western blotting and immunohistochemical method.Results showed that Ng protein was decreased in the hippocampus of male mice with adult-onset hypothyroidism,suggesting that decreased Ng,which is correctable by T_4,is involved in cognitive dysfunction in adult-onset hypothyroid male mice.
8.Expression of transforming growth factor β1/Smad4 signal pathway in rats with nonalcoholic fatty liver disease

Objective To investigate the expression of transforming growth factor β1,transforming growth factor beta receptor(TBR)Ⅰ,TβR Ⅱ,Smad4 and C-Jun in rats with nonalcoholic fatty liver disease(NAFLD)and to find out the mechanisms of liver fibrosis in patients with NAFLD.Methods A total of 18 male SD rats were randomly divided into normal control group(n=9)and model group(n=9).The rats in control group were fed with normal diet,and those in model group were fed with fat-rich diet(consisted of 10%lard oil+2%cholesterol).An rats were sacrificed at the 20th week.The levels of TGFβ1,TβR Ⅰ and TβR Ⅱ mRNA were examined by RT-PCR.The expressions of TGFβ1 and Smad4 in liver tissue were detected by immunohistochemistry.The expression of C-Jun protein was detected by Western blotting.Results The NAFLD model was successfully established.The immunohistochemistry examination revealed that TGFβ1 and Smad4 were expressed weekly in control group,but strongly expressed in model group.RT-PCR showed that A values of TGFβ1,TβR Ⅰ and TβR Ⅱ mRNA were 0.46±0.12,5.z4±2.70 and 3.35±1.95,respectively,in model group,which were higher than those in control group(0.21±0.09,1.36±0.77 and 0.52±0.19,all P values<0.01).The Western blotting results demonstrated that the expression of C-Jun protein in model group(0.93±0.41)was higher than that in control group (0.32±0.25,P=0.001).Conclusion TGFβ1/Smad4 signal pathway might be involved in the development of hepatic fibrosis in NAFLD.Blocking TGFβ1/Smad4 signal pathway will be helpful in treatment of NAFLD.
9.Prokaryotic Expression and Acetylation Assays of Histone Acetyltransferase PCAF

P300/CBP-associated factor(PCAF),an important member of histone acetyltransferase family(HATs) within eukaryotic cells,is capable of inducing the acetylation of histone,promoting the transcription of specific genes and involving in many biological effects.In the present study,full-length cDNA of PCAF was inserted into plasmid pGEX-5x-1,then the soluble protein GST-PCAF was expressed in E.coli BL21(DE3) after the optimization of inducing conditions.The recombinant protein was further purified with affinity chromatography and tested the activity by in vitro acetylation assays.High efficient PCAF protein produced by this method could serve for the study on the role of PCAF in gene regulation and the interaction between PCAF and other proteins.
10.Correlations of blood lead levels in infant, in maternal blood and in breast milk.

OBJECTIVETo understand the correlation of blood lead levels in infant, in maternal blood and in breast milk as to providing evidence for prevention of potential infant hazard from lead.
METHODSLead levels were measured by using graphite stove atom absorption spectrographic methods in maternal breast milk, maternal blood and infant blood in infants aged 0 to 11 months and their mothers between November and December 2002 in Xiamen City. Blood samples were collected from both mother and infant's fingertips. Questionnaires were also used to collect information about childbirth, mothers, families and other related environmental factors.
RESULTSAll 177 infants and their mothers were enrolled in the study. Infant blood lead levels reached a range from 0.12 micromol/L to 1.36 micromol/L, with a geometric mean (GM) of 0.37 micromol/L. There were 46 infants (21.64%) having blood lead levels over 0.48 micromol/L. And the maternal blood lead levels ranged from 0.21 micromol/L to 2.38 micromol/L (GM = 0.50 micromol/L). Among the 177 infants, 160 (93.8%) were breastfed; breast milk was collected from 105 (63.3%) of these mothers. Infant blood lead level was significantly correlated with the levels of maternal blood lead and breast milk lead, which indicated that maternal blood lead level might influence the infant blood lead levels through the breast milk. Blood lead levels in infants living in old business district and the breast milk lead levels of their mothers were higher than those in any other areas (P < 0.01); partial correlation analysis showed that infant blood lead levels were positively associated with the maternal blood lead level, infant's age and mother's work, and negatively associated with mother's height.
CONCLUSIONThe infant blood lead levels should not only relate to the maternal blood lead and the breast milk lead levels, so regards should be had to the other environmental factors, when selecting the feeding pattern and family rearing behaviors.
