1.Cloning and analysis of psaB cDNA of Dunaliella salina.

One pair of degenerate primer was designed according to conserved motifs of the psaB (A2 subunit of photosystem I) of Chlamydomonas reinhardtii, Chlamydomonas moewusii, Chlorella vulgaris and Mesostigma viride, and a total RNA of Dunaliella salina (D. salina) was extracted with TRIzol reagent. A cDNA fragment, about 1.8kb in length, from green algal D. salina was obtained through RT-PCR method. The resulting PCR product was cloned into T-vector and screened to determine its sequence. Homologous analysis of the deduced amino acid sequence was performed by BLAST and subsequeqtly compared with GenBank data. The obtained cDNA sequence was 1815 bp long, which encodes 605 amino acids (GenBank accession number: AY820754). The sequence shared high homologue with the following psaB: Chlamydomonas reinhardtii 92%, Chlamydomonas moewusii 91%, Chlorella vulgaris 86%, Mesostigma viride 85%, Physcomitrella patens subsp. Patens 85% and Nephroselmis olivacea 84%. It can be concluded that the cloned sequence is psaB cDNA fragment from D. salina.

2.Effect of psychological intervention to anxiety-depression and T lymph cells in breast cancer patients during the peri-operation

Objective To study the affect of psychological intervention to anxiety-depression emotion and T Lvmlcih cells in breast cancer patients during the peri-operation. Methods We divided 60 cases into two groups at random. the experiment group(30) and the control group(30).The psychological situation Was evaluated according to the scores of Self-Rating Anxiety Status Inventory(SAS)and Depression Status Inventory (SDS).2 ml of venous blood Was taken to measure the level of T lymph cells. Then the experiment group Was given the specific and generalized psychological interventions. while the control group was given only the common nursing care. Results The scores of anxiety(8.65±3.69)and depression(31.70±5.79)after the intervention in the experiment group were lower than those before the intervention。and had significant difference (P<0.05).After the operation,the average value of CD3,CD4,CD4/CD8 in the experiment group was higber than that of the control group,and the values of CD3 and CD4 had statistical meaning(P<0.05).After the intervention,the vatues of CD3,CD4,CD4/CD8 of the experiment group were higher than those before the intervention. and the values of CD3 and CD4 had obvious difference(P<0.05).Conclusions In this study, a suitable and effective psychological intervention can improve the status of anxiety and depression of patients with breast cancer during the peri-operative period, and at the same time can modulate the immune status.
3.Hypertension situations and strategy in middle school teachers in Beijing area

Objective To investigate the hypertension situations and nursing strategy of middle school teachers in Beijing area.Methods We have done a healthy check-up to 2 114 middle school teachers in Beijing,and then we made an investigation on the hypertension situation of these teachers,and summarized and analyzed the results of the physical examination.Results In the 2 114 middle school teachers,644 cases have hypertension problems.The hypertension sicken rate is 30.5%,and the prevalent rate was different in male and female (34.1% vs 24.9% ; x2 =10.052,P <0.01),and in people with different age and BMI (P <0.01).Along with the growth of the age,hypertension rates show an obvious ascendant trend,with the increase of body mass index (BMI),hypertension rates is also increasing.Conclusions The situation about hypertension in middle school teachers is not very optimistic.Middle school teachers often have more mental works to do,their work heavy,mental strain,psychological pressure big,physical exercises less,diet style is not reasonable.All of these are leading to high blood pressure.We should pay more attention to these,and adopt active actions as early as possible to change the unhealthy lifestyles and reduce hypertension.
4.Molecular scanning of candidate mtDNA gene fragment in diabetic pedigrees.

OBJECTIVETo explore novel pathogenic mutation in the mitochondrial DNA gene in diabetic pedigree.
METHODSTwenty-eight suspected mitochondrial DNA diabetic families were recruited. The gene fragment was produced by PCR, and mutation was detected by direct sequencing.
RESULTSIn one pedigree, the proband and her mother were found carrying the most common nt3243 A --> G mutation and another 16S rRNA 3205C --> T mutation. But only 3205C --> T was found in her affected brother. All the two patients were deaf and developed diabetes in early age, characterized by impaired beta cell function and low body mass index (BMI). The proband had relatively higher lactic acid concentration than normal individuals. A novel ND1 gene 3434 A --> G(TAT --> TGT) mutation was explored in another proband with deafness and her affected family members.
CONCLUSION16SrRNA 3205C --> T mutation was found in a mitochondrial diabetes mellitus pedigree, implying its potential pathogenic role in diabetes. Another novel ND1 3434 A --> G mutation was found in another diabetic pedigree. Because this mutation causes amino acid change (Tyr --> Cys) and is co-segregated with diabetes, it may be diabetogenic.
