1.Effects of atorvastatin in different dosage on plasma level of fibrinogen in patients with acute cerebral infarction

Objective To investigate whether intervention with two different dosage of atorvastatin may benefit to plasma level of fibrinogen (Fg) in patients with acute cerebral infarction. Methods One hundred and two patients with acute cerebral infarction were randomly assigned into three groups:the control group (33 cases) treated without lipid-lowering drugs, 10 rag (30 cases) and 20 mg (39 cases) atorvastatin group with 3 days of treatment. Before and after the treatment, the plasma levels of Fg and lipids were detected.Results The treatment of 10 mg atorvastatin did not significantly decrease the level of Fg. Compared to the control group, the level of Fg was markedly reduced by 20 mg atorvastatin (21.6% vs 3.2%,P< 0.05). No changes of lipid levels were observed in any group before and after the treatment, whilst there was no relation between the decreasing percentage of Fg and that of TC(r = 0.125 ,P= 0.618), TG(r = 0.147,P = 0.573) or LDL- C (r = -0.279, P = 0.235 ), HDL-C (r = -0.021, P = 0.157). Conclusion Treatment with high dose atorvastatin (20 mg) for 3 days could reduce the level of Fg, and improve the function of vascular endothehum,which may promote the stabilization of atherosclerotic plaque.
2.Therapeutic effect of heat-sensitive moxibustion plus medications for senile osteoporosis and its effect on serum BMP-2 and OPG levels

Objective: To observe the clinical efficacy of heat-sensitive moxibustion plus medications on senile osteoporosis (SOP), and to explore the related mechanisms. Methods: A total of 70 elderly participants with osteoporosis were randomly divided into an observation group and control group, with 35 cases in each group. The control group was treated with conventional drugs, and the observation group was treated with heat-sensitive moxibustion on the basis of the conventional drugs. Both groups were treated for 3 months. Before and after treatment, assessed the visual analog scale (VAS) and Oswestry disability index (ODI) scores, determined the bone mineral density of the participants' lumbar spine (L2-L4) and left femoral neck, and detected the participants' serum bone morphogenetic protein-2 (BMP-2) and osteoprotegerin (OPG) levels. Results: After treatment, the VAS scores of both groups were lower than before treatment (both P<0.05), and the VAS score of the observation group was significantly lower than that of the control group (P<0.05). After treatment, the bone mineral density values of the lumbar spine and left femoral neck in both groups were significantly higher than before treatment (both P<0.05), and the bone mineral density values of the observation group were higher than those of the control group (P<0.05). After treatment, the ODI scores of the two groups were lower than those before treatment (both P<0.05), and the ODI score of the observation group was lower than that of the control group (P<0.05). After treatment, the serum BMP-2 and OPG levels in the observation group were significantly higher than those in the control group (both P<0.05). Conclusion: Heat-sensitive moxibustion plus medications for SOP can significantly relieve patients' pain, improve dysfunction, and increase bone density, which may be related to the improvement of the serum BMP-2 and OPG levels.
3.Influence on cultured human keratocytes by liposome

AIM:To observe the effects on human keratocytes by cationic liposome LipofectamineTM 2000(LF2000).to investigate the efficiency and safe range applied in human keratocytes,and establish basis for gene therapy of human keratocytes.METHODS: Human keratocytes cultured in vivo within 3 to 5 passages were used in experiment after being identified.The effects on proliferation of cultured human keratocytes by LF2000 with different concentrations and time were evaluated By MTT:the effects of LF2000 on the survival rate and its relation with 5,10,20,40.80mg/L concentration and time were detected by trypan blue staining.related with concentration and time.The cellular proliferation and survival rate declined when concentration of LF2000 was above certain level,and this effect increased as time became longer.LF2000 had no effect with concentration under 40mg/L for 24 hours. CONCLUSION:LF2000 did ont cause cytotoxicity during a concentration range"tested",and it is hoped to play an important role in gene therapy of human keratocytes.

8.Over-expression of SK2 channel in PVN reduces renal sympathetic nerve activity in chronic heart failure rats

[ABSTRACT]AIM:Toinvestigatetheeffectofover-expressionofsmall-conductancecalcium-activatedpotassium channel subtype 2 (SK2) in the paraventricular nucleus (PVN) of hypothalamus on heart rate(HR), mean arterial pres-sure (MAP) and renal sympathetic nerve activity (RSNA) of the rats with chronic heart failure (CHF).METHODS:Adenovirus vector encoding SK2 (pDC316-mCMV-EGFP-rKCNN2) was constructed.The CHF model of the male Sprague Dawley ( SD) rats was established by the ligation of anterior descending coronary artery .Echocardiogram was used at the 6th week after the operation to identify the CHF model .Adenovirus vector encoding SK 2 ( pDC316-mCMV-EGFP-rKCNN2) or control adenovirus vector ( pDC316-mCMV-EGFP) were transfected into the PVN in vivo.The cardiac func-tion was monitored by echocardiogram .The expression of SK2 at mRNA and protein levels was determined by RT-PCR, Western blotting and immunofluorescence .The HR, MAP and RSNA were measured by PowerLab 8/30 in anesthetized rats with bilateral PVN microinjection of SK channel blocker apamin .RESULTS:Treatment with pDC316-mCMV-EGFP-rKC-NN2 significantly decreased the renal sympathetic nerve activity in the rat with CHF .Injection of pDC316-mCMV-EGFP did not change the renal sympathetic nerve activity in the rats in sham group .CONCLUSION:The expression and func-tion of SK channels in PVN in the heart failure rats were decreased , suggesting a reduced negative regulation of sympathetic activity in central neural system .Increased expression of SK 2 in the PVN leads to a reduction in sympathetic outflow under the condition of CHF , which may provide a new target for the treatment of heart failure .
9.Changes of collagen fiber during reattaching of the musseter muscle following different curved osteotomies of prominent mandibular angle: a study in goats

Objective To investigate the changes of the constitution and its ratio of collagen fiber in the process of masseter reattachment following different osteotomies of the prominent mandibular angle so as to offer guidance for the resection of mandibular angle. Methods Sixteen adult goats were randomized into four groups. In group A we performed unilateral curved osteotomy of the mandibular angle. In group B unilateral curved ostectomy was performed with partial masseter resection. In group C unilateral angle splitting ostectomy, while in group D unilateral dissection of the masseter muscle was conducted. The constitution and its ratio of collagen fiber in the interface were observed at 1-month, 2-month, 3-month, and 6-month after operation. Results On the changes of collagen fiber in the process of muscular reattachment, at 1-month post-operation, the constitution of collagen fiber (types Ⅰ and Ⅲ) in groups A and B were significantly different from that of control group (P<0.05). However, both groups C and D had no statistic difference from control group (P>0.05). At 2-month, 3-month and 6-month post-operation, those of all experimental groups had no statistic difference from control group. And with time, the percentage of collagen fiber type Ⅰ increased and type Ⅲ decreased gradually. Conclusion The recovery sequences of masseter muscle reattachment in this study are firstly group C, secondly group A and finally group B. It suggests that the recoveries of mastication and other oral activities are different. Group B turns out to be with a slow muscle reattachment. Thus, we recommend treating different kinds of mandibular hypertrophy with different ostectomies.
10.Correlation of blood glucose, blood pressure and body weight with pancreatic cancer

Objective To investigate the correlation of blood glucose,blood pressure and body weight with pancreatic cancer.Methods From January 2011 to December 2013,110 patients diagnosed with pancreatic cancer in our hospital were selected as the observation group and 110 agematched cases without cancer during the same period were selected as the control group.The percentages of patients with diabetes,hypertension and elevated body mass index (BMI) were analyzed in both groups.Results The number of patients with diabetes was higher in the observation group than in the control group (32 cases or 29.1% vs.16 cases or 14.6%,P<0.05).The proportions of pancreatic cancer patients with diabetes duration ≤ 2 years,2~5 years and 5~10 years were higher in the observation group than in the control group (P<0.05).The proportion of subjects with increased BMI was higher in the observation group than in the control group (24.6% or 27cases vs.10.9% or 12 cases,P<0.05).The proportions of patients with diabetes combined with increased BMI and of patients with hypertension and increased BMI were higher in the observation group than in the control group (17.3% or 19 cases vs.2.7% or 3 cases,10.9% or 12 cases vs.2.7% or 3 cases,respectively,P< 0.05 for both).Conclusions Diabetes,hypertension,and elevated BMI can be considered as risk factors for pancreatic cancer,and it is possible that these factors are involved in the development of pancreatic cancer.
