1.Detection of growth hormone(GH) in urine with immuno-PCR

Objective:To establish a highly sensitive method by immuno PCR for measurement of GH in Urine.Methods:Sandwich ELISA was operated in PCR tubes which treated with glutarakdehyde,and PCR was tested in the same PCR tube.The PCR production were electrophoresis in Agarose gel and Scan for gel.Results:The results show that ELISA could be correctly reacted in PCR tubes which treated with glutarakdehyde,more than 0.1 pg/ml GH could be detected.Results of Scan showed that the GH values in urine overnight have significantly deference between 30 normal children and 10 GHD patients which were diagnosed by GH provocative test.Conclusion:The sensitivity of Immuno PCR is 10 3~10 4 times higher than that of ELISA,Immuno PCR could be used in detection of GH in urine of children.
2.Occupational exposure to HIV and causes of HIV infection among nursing staff

Objective To analyze the infection among nursing staff after occupational exposure to human immunodeficiency virus(HIV)and status of HIV-infected nursing staff,provide basis for prevention of HIV infection among nursing staff.Methods Nurses who sustained HIV occupational exposure in a hospital between January 2004 and June 2015,nurses with positive anti-HIV and admitted to this hospital,consulted in outpatient department,as well as consulted outside this hospital between January 2001 and December 2015 were and analyzed,HIV infection status among nurse's mate was also analyzed.Results 76 nurses who sustained HIV occupational exposure didn't infect with HIV after received standard treatment and follow-up.Among 13 nurses infected with HIV,3 might be with occupational exposure infection previously,5 were with blood transfusion-related infection in the 1990s,4 were with sexually transmitted infection(including 2 male nurses who had sex with man,MSM),1 case with infection of unknown transmission route;secondary transmission occurred in 2 cases;9 nurses are still alive.The infected nurses have received effective antiretroviral treatment and then work at the logistic or administrative department.Conclusion Nurses often suffer from occupational exposure to HIV,occupational exposure infection may occur in the absence of the implementation of standardized management,implementation of standardized management is essential to avoid the occupational exposure of HIV.Sexual transmission(including MSM)has become an important transmission route for HIV infection among nurses.
4.Comparative study of EEG and CT in early youth with cerebral cysticercosis

Objective To investigate the diagnostic value of EEG and CT in early youth with cerebral cys- ticercosis.Methods The EEG and CT manifestations were studied in 240 early youth with cerebral cysticercosis. Results The abnormal rate of EEG was 86.7 % in children with cysticercosis,which mainly showed the diffuse or focal irregularity complex slow waves in frontal lobe,central lobe and anterior lobe.The abnormal rate of CT was 70.8 % in all patients,and flaky and circular focus were chief manifestations.There was a significant difference be- tween the rates of EEG and CT(P
5.Effect of Ginkgo biloba Tablet on the Expression of Scavenger Receptor A of the Aortic Wall in Atherosclerotic Rats.

OBJECTIVETo observe the expression of Ginkgo biloba Tablet (GbT) on scavenger receptor A (SRA) of the aortic wall and changes of serum inflammatory factors in atherosclerotic rats, and to explore its new mechanism for fighting against atherosclerosis (AS).
METHODSTotally 45 male Wistar rats were randomly divided into the control group, the model group, the GbT group, 15 rats in each group. Levels of blood glucose, blood lipids, blood calcium, serum C-reactive protein (CRP), soluble intercellular adhesion molecule-1 (slCAM-1), and soluble vascular cell adhesion molecule-1 (sVCAM-1) were measured in all rats. The expression of SRA in the aortic wall of atherosclerotic rats was observed by immunohistochemical assay. The correlation between the expression of SRA and levels of in-flammatory factors was also observed.
RESULTSCompared with the control group, blood glucose and blood calcium obviously increased (P < 0.05); levels of TG, TC, and LDL-C were significantly elevated (P < 0.01); neointimal areas were significantly thickened, increased intima percentage was significantly enlarged, narrowed lumen index was significantly reduced; levels of CRP, sICAM-1, and sVCAM-1 were significantly elevated in the model group (all P < 0.01). Compared with the model group, blood glucose and blood calcium obviously decreased (P < 0.05); levels of TG, TC, and LDL-C significantly decreased (P < 0.01) in the GbT group. Aortic lumens were obviously narrower in the model group than in the GbT group (P < 0.05). SRA expressed at the aortic wall. The aforesaid 3 indices were significantly improved in the GbT group than in the model group (P < 0.01). Serum levels of CRP, sICAM-1, and sVCAM-1 were significantly decreased in the GbT group than in the model group (P < 0.01). Serum levels of CRP, sICAM-1, and sVCAM-1 were positively correlated with the percentage of SRA positive expression area (r = 0.701, 0.604, 0.581, all P < 0.01).
CONCLUSIONSSerum levels of inflammatory factors in atherosclerotic rats were elevated, and the expression of SRA in the aortic wall was enhanced. The expression of SRA was closely correlated with serum levels of inflammatory factors. GbT could decrease serum levels of inflammatory factors and inhibit the expression of SRA.

6.Significance of Multidrug Resistance-Associated Protein Gene and P-gp Expression in Malignant Lymphomas

Objective: This study was designed to investigate the relationship between expression level of multidrug resistance-associated protein (MRP) or P-glycoprotein (P-gp) genes and chemotherapy efficacy or clinical drug resistance in the patients with malignant lymphomas. Methods: Using the methods of semi quantitative reverse transcription polymerase chain reaction (RT-PCR) and flow cytometry (FCM), The authors examined the expression level of MRP and P-gp of 46 lymphoma patients. Results: The expression level and positive rate of P-gp in the recurrent patients was higher than that in untreated patients (P<0.001). There was no difference in MRP gene expression level and positive rate between recurrent and untreated patients (P>0.05). Chemotherapeutic effective rate in P-gp positive patients (26.67% ) was lower than that in P-gp negative patients (83.87% )(P<0.001). While there was no difference between MRP gene positive and negative patients, their chemotherapy effective rate had no difference. Relevant analysis showed that there was no correlation between MRP and P-gp (r-0.0808, P>0.05). Conclusion: P-gp expression correlates to multidrug resistance, and is the major mechanism of clinical drug resistance of lymphomas, whereas, MRP gene does not appears to play any role in that course.
7.The solubilization effect of 2-hydroxypropyl-beta-cyclodextrin on paeonolum.

OBJECTIVETo study the solubilization effect of 2-hydroxypropyl-beta-cyclodextrin(HPCD) on paeonolum at various pH value.
METHODPhase-solubility method was adopted to study the solubilization effect at 25 degrees C and UV spectrohotometer was used to determine paeonolum content.
RESULTSThe apparent solubility of paeonolum was significantly enhanced by increased HPCD concentration. The apparent stability constant of paeonolum compounds was calculated up to 1 425 in which pH was 3 and HPCD concentration was 133.33 mmol x L(-1). The solubility of paeonolum came up to 10 mg x mL(-1).
CONCLUSIONHPCD is an ideal solubilizer for paeonolum.

8.Effects of fluorine and aluminum on index of hematologic tests of rats

Objective To study the effects of fluorine and aluminum on index of hematologic tests of rats. Methods According to body mass,56 Wistar rats of 130-200 g were randomly divided into control,low-fluorine (F),middle-F,high-F,low-F + aluminum(Al),middle-F + Al,high-F + Al group,8 rats in each group were given a series of doses of fluoride and aluminum,which were (0 + 0),(100 + 0),(200 + 0),(300 + 0),(100 + 10),(200 + 10),(300 + 10)mg/L After 90-day intragastrie administration,blood samples were collected on eyes of rats to undergo blood routine test,including red blood cell (RBC),lymphocyte (LYM),platelet (PLT),hemoglobin (HGB),white blood cell (WBC),hematocrit (HCT),mean corpuscular hemoglobin (MCH),mean corpuscular-hemoglobin concentration(MCHC),mean corpuscular volume(MCV),and at the same time some blood biochemistry indicators related to functio ns of liver and kidney were determined such as aspartic acid aminotransferase(AST),alanine aminotransferase(ALT),alkaline phosphatase(ALP),Crea(Cr) and Urea. Organ coefficient of liver and kidney were calculated. Results The difference of RBC,HCT,MCV among all groups of rats was statistically significant(F = 3.202,3.316,2.915,P < 0.05). The RBC,HCT of the low-F group[(7.59± 2.40)×10~(12)/L,0.51±0.11],the middle-F group[(8.60±1.16)×10~(12)/L,0.55±0.05],the high-F group[(9.23± 0.60)×10~(12)/L,0.54±0.03],the low-F + Al group[(9.25±0.79)×10~(12)/L,0.53±0.04],the middle-F + Al group[(7.98±2.14)×10~(12)/L,0.49±0.08]and the high-F + Al group[(7.61±3.17)×10~(12)/L,0.49±0.16]were significantly higher than that in the control group[(4.46±3.10)×10~(12)/L,0.31±0.16,P< 0.05 or < 0.01)]. The MCV of the middle-F group[(64.06±6.51)fl],high-F group[(58.67±1.13)fl],low-F + Al group[(57.78± 1.57)fl]and the middle-F + Al group[(63.04±10.64)fl]were significantly higher than the control group[(78.54± 15.57)fl,P < 0.05 or < 0.01]. The difference of AST and Urea among all the groups of mrs serum was statistically significant(F= 2.847,5.549,P < 0.05 or < 0.01). The serum AST of low-F group[(399.00±54.99)U/L],the middle-Fgroup[(465.60±76.99)U/L],the high-F group[(465.80±75.41)U/L],the low-F + Al group[(346.00±69.26) U/L],the middle-F + Al group[(437.40±68.31)U/L]and the high-F + Al group[(403.00±30.61)U/L]were all significantly higher than that in the control group[(336.67±94.34)U/L,P < 0.05],and the high-F group significantly higher than the high-F + Al group(P < 0.05). The serum Urea of the middle-F group[(7.70±0.52)mmol/L],the high-F group[(8.44±1.30)mmol/L],the low-F + Al group[(7.83±0.62)mmol/L],the middle-F + Al group [(7.73±0.47)mmol/L],and the high fluoride + aluminum group[(7.70±0.21)mmol/L]were all significantly higher than that in the control group[(6.55±0.50)mmol/L,P< 0.05 or < 0.01],and the low-F group was significantly lower than the low-F + Al group(P < 0.01),however the high-F group was significantly higher than that in the high-F + Al group(P< 0.05). The liver organ coefficient of the low-F group(2.94±0.36) was higher than the low-F + Al group (2.60±0.15,P < 0.05). Conclusions Fluorine and combination of aluminum and fluorine have toxicity on rats to a certain extent,including the proliferation of crythrocytes of rat,while the cell size gets smaller and the cell quality is deteriorated,meanwhile functions of liver and kidney are impaired. Aluminum shows different joint action in different concentrations of fluorine.
10.Investigation on the negative emotion and coping mode among patients with chronic heart failure and relative nursing intervention

Objective To discuss the coping modes for patients with chronic heart failure (CHF)dealing with their anxiety and depression; and to explore the possible way to guide effective nursing intervention aimed to reduce the patients' hospitalization rate and the length of hospitalization and to improve the quality of life for patients. Methods 120 patients with CHF were investigated by self-rating anxiety scale (SAS), selfrating depression scale and medical coping modes questionnaire (MCMQ). Results The score of patients with CHF in SAS was higher than norm model [(19.48 ± 3.81) vs (29.78 ± 10.07), t = 11. 127, P < 0. 01] and the SDS was also higher than normal model (P < 0. 01). The "facing" score of patients with CHF in MCMQ was statistically lower than that of normal model in China[(16. 08 ± 3.88) vs (19.48 ± 3.81), t = 2. 046, P <0. 05]; Both of the scores on "avoidance" and "surrender" were statistically higher than those of normal model (P < 0. 05 or P < 0. 01). Conclusions Most patients with CHF had the problems involving anxiety, depression and ineffective coping mode; patients' quality of life could be improved when their negative emotion released and their coping mode improved.
