Objective To investigate the relationship between oxidative stress and cell apoptosis in the kidney of rabbit induced by high energy shock wave(HESW).Methods Forty healthy male rabbits were surgically managed to the mono-nephron models and randomized into 4 groups consisting of ten each: control group,1 000 HESW group,2 000 HESW group and allopurinol pretreated plus 2 000 HESW group.The rabbit kidney tissues were obtained.The activity of superoxide dismutase(SOD) and malondialdehyde(MDA) levels in the renal tissue were measured 24 hours after HESW application.The cell apoptosis was detected by TdT-mediated dUTP nick end labelling(TUNEL) and flow cytometry.Results Compared with the control group,the MDA level and the rate of apoptosis in the HESW groups increased,and the activity of SOD reduced significantly(P
2.Effects of intrathecally administered morphine and ketamine on nitric oxide synthase activity and nitric oxide production in the spinal cord

Objective To study the effects of intrathecally administered morphine and ketamine on nitric oxide synthase (NOS) activity and nitric oxide content in the spinal cord.Methods Thirty-two male Wistar rats weighing 220-260 g were anesthetized with intraperitoneal chloral hydrate 300 mg?kg-1 . A catheter was implanted in subarachnoid space at lumbal region. Sciatic constriction injury (SCI) was produced by loose ligation of right sciatic nerve trunk with 4-0 cutgut. On the 4th postoperative day the animals were randomly divided into four groups of 8 animals :(1) control group (C); (2) morphine group (M);(3) ketamine group (K) and morphine-ketamine group (KM) . Morphine 20 ?g / ketamine 20 ?g / morphine 10 ?g + ketamine 10 ?g were injected intrathecally every day for 7 consecutive days in group M, K and KM. In control group normal saline was injected intrathecally instead of morphine and / or ketamine. The withdrawal latenvies to radiant heat focused on plantar surface were measured as radiant heat threshold before intrathecal administration of the analgesic (baseline) and 30 min after intrathecal administration of ketamine and / or morphine every day for 7consecutive days. The percentage of maximal possible effect ( MPE % ) was calculated : MPE % = (latency after intrathecal administration-baseline latency) / (radiant heat cut-off time-baseline latency) X 100% . After 7 days of intrathecal administration the animals were decapitated and the spinal cord was immediately removed and the lumbal spinal cord was dissected on ice. 6.Comparison of single-versus double-injection techniques forsciatic nerve block

Objective To compare the single-and double-injection techniques for sciatic nerve block.Methods Sixty ASA physical status Ⅰ or Ⅱ patients of both sexes,aged 18-64 yr,weighing 48-72 kg,undergoing elective unilateral foot and ankle surgery,were randomly divided into 2 groups (n =30 each) using a random number table:single-injection group (group S) and double-injection group (group D).Labat-winnie approach to sciatic nerve block was performed under the guidance of a nerve stimulator.When ankle dorsal or plantar flexion developed,group S received a single injection of 0.5% ropivacaine 30 ml.When ankle dorsal and plantar flexion developed,0.5% ropivacaine 15 ml was injected each time in group D.The failure of location was recorded.The time spent performing the procedure,onset time and duration of sensory and motor blockade,and the total time for the block were recorded.The effectiveness of block (success,failure) was assessed.Results The failure rate of location was 10% in group D.The success rate of block was 93% in group S,and was 96% in group D,and there was no significant difference in the success rate of block between the two groups.Compared with group S,the onset time of sensory and motor blockade was significantly shortened,the time spent performing the procedure was prolonged,and no significant change was found in the total time for the block,duration of sensory and motor blockade in group D.Conclusion Single-injection technique is recommended for the sciatic nerve block in the patients undergoing lower extremity surgery.

Objective To explore the inhibitory effect of rosiglitazone of peroxisome proliferator-activated receptor-?(PPAR?) agonist on aldosterone-induced mesangial cell(MC) proliferation.Methods Mouse primary MC were cultured and treated with aldosterone(100 nmol/L) in the presence or absence of rosiglitazone(1.0,2.5,5.0,10.0 ?mol/L).The incorporation of 3H-thymidine(3H-TdR) and cell count were used as the measure of MC proliferation.Cyclin D1 and cyclin A expression,PI3K and Akt phosphorylation were determined by Western blot analysis.Results 1.Aldosterone induced MC proliferation,as assessed by 3H-TdR incorporation and cell number,which were increased by 2.46-and 2.14-fold,respectively,in aldosterone-treated cells.Aldosterone-induced MC proliferation was inhibited by PPAR? agonist rosiglitazone in dose-dependent manner in mouse MC.2.Aldosterone induced cyclin D1 and cyclin A expression.Rosiglitazone reduced aldosterone-induced cyclin D1 and cyclin A expression in dose-dependent manner.3.Aldosterone induced PI3K/Akt activation in dose-dependent manner,incubation with 100 nmol/L aldosterone for 60 min,phosphorylation PI3K and Akt expression increased by above 3.0-fold.4.PI3K inhibitor LY294002 and Akt inhibitor significantly inhibited aldosterone-induced cyclin D1 and cyclin A expression.5.Rosiglitazone significantly inhibited aldosterone-induced PI3K/Akt activation,10 ?mol/L rosiglitazone almost completely blocked aldosterone-induced PI3K/Akt activation.Conclusion Rosiglitazone can block aldosterone-induced MC proliferation via inhibition of PI3K/Akt activation.
9.Effect of SP600125 on AngⅡ-Induced Transforming Growth Factor-1 and Fibronectin Expression in Human Mesangial Cells

Objective To investigate the effect of c-Jun N-terminal kinase(JNK) specific inhibitor SP600125 on Angiotensin Ⅱ(AngⅡ)-induced transforming growth factor-?1(TGF-?1) and fibronectin (FN) expression in human mesangial cells (MC).Methods Human MC were isolated and cultured in vitro and were treated with AngⅡ in the presence or absence of JNK specific inhibitor SP600125.The protein was isolated or the supernate of medium was collected at the end of experiment.JNK,extracellular signal-regulated kinase(ERK1/2),and p38 mitogen-activated protein kinase(MAPK) activity were determined by Western blot method.TGF-?1 and FN were determined by enzyme linked immunosorbent assay(ELISA).Results SP600125 inhibited AngⅡ-induced Ser63 phosphorylation of c-Jun in a concentration-dependent manner,and JNK activity was reduced by 75% at 10 ?mol/L and by 90% at 20 ?mol/L.SP600125 had no effect on AngⅡ-induced ERK1/2 and p38 activity.TGF-?1 and FN protein were constitutively produced in MC,and production was significantly stimulated for 8 to 48 h after addition of AngⅡ.Preincubation of cells with SP600125(20 ?mol/L) significantly inhibited AngⅡ-induced TGF-?1 and FN production during this time period.SP600125 inhibited AngⅡ-induced production of TGF-?1 and FN in a concentration-dependent manner.Conclusion SP600125 inhibited AngⅡ-induced JNK activation and TGF-?1 and FN expression in human MC and may serve as the novel approach for the treatment of patients with chronic kidney disease.
10.Middle cerebral artery blood flow changes at different tilting angles in patients with cervical and thoracic spinal cord injury

ObjectiveTo study the speciality of hemodynamics in middle cerebral artery(MCA) by means of transcranial color Doppler ultrasound(TCCD) at different tilting angles in patients with cervical and thoracic spinal cord injury.Methods48 patients with spinal cord injury were involved. The velocity of peak systolic(PSV), end diastolic(EDV) and mean velocity(MV) of MCA on the tiltable at 0°,30°,60°,80° were measured.ResultsThere was no significant difference in PSV between 0°and 30°(P>0.05), while EDV and MV showed very significant (P<0.01)from 0°to 30°,60°and 80° position. From 0°to 60°,80°,there were differences not only at EDV and MV, but also PSV(P<0.01).ConclusionThe velocities of MCA were changed at different tilting angles in the patients with spinal cord injury.
