1.The killing efficiency of arilin and contact lens solutions on Acanthamoeba cultured in vitro

AIM:To analyze the killing efficiency of six kinds of contact lens solutions and solutions with arilin on free living Acanthamoeba culturedin vitroMETHODS:Six kinds of contact lens solutions were added into 96-well microtiter plates,respectively,with each care solutions used 48 holes of them.Suspension of Acanthamoeba were added into 24 of these holes.and arilin gutta and suspension of Acanthamoeba were added into the other 24 holes.After standing in room ternperature for 8 hours,the morphologic change and quantity of the remnant Acanthamoeba were observed under the jnverted microscope.The remnant Acanthamoeba were cultivanted in peptone-yeast extract-glucose (PYG)-culture medium for 5 days.Their variation of appearance,activity and reproductive activity were observed.RESULTS:In the six experimental groups using contact lens solutions only.the detection rate of Acanthamoeba of were 0%,80.3%,29.1%,41.7%,62.5% and 79.2%,respectively.After arilin was added,the detection rate of Acanthamoeba of the six groups were 0%,0%,4.2%,8.3%,16.7% and 16.7%,respectively.From group 3 to group 6,after arilin was added,the differences of the killing efficiency of contact lens solutions have statistical significance(X2=3.75,7.11,10.54 and 18.78;P<0.05).Cultivation of the remnant Acanthamoeba showed a reduction in the activity and proliferative ability.CONCLUSION:The killing efficiency of some contact lens solutions on free-living Acanthamoeba were not satisfying.Arilin can improve the killing efficiency of contact lens solutions.
2.Comparison of the adherent ability of Acanthamoeba to three kinds of contact lens and the study of their easy cleansing methods

AIM: To compare the adherent ability of Acanthamoeba to three kinds of contact lens (CL) and survey the effect to remove the Acanthamoeba from the surface of contact lens after blowing with stroke-physiological saline solution (SPSS) repeatedly.METHODS: Three kinds of contact lens were chosen in this experiment, including: rigid gas permeable (RGP) contact lens, soft contact lens (SCL) and colored contact lens. They were co-cultivated with Acanthamoeba suspension for 16 hours. Then the number of Acanthamoeba adhered on CL was compared. Comparison was also made between different zones of colored CL. We also compared the number of Acanthamoeba adhered on CL in three groups after blowing with SPSS.RESULTS: The number of Acanthamoeba adhered to colored CL was more than RGP group and SCL group (P<0.05). The differences between RGP group and SCL group had no statistical significance (P>0.05). In colored CL group, the number of Acanthamoeba adhered to the colored zone was more than the Uncolored zone (P<0.01). In all the three groups, after blowing with SPSS, there was statistically significant decrease of the number of Acanthamoeba adhered to CL (P<0.01).CONCLUSION: Compared with RGP group and SCL group, the colored contact lens was more vulnerable to be adhered by Acanthamoeba. After being blown by SPSS repeatedly, the effect to eliminate Acanthamoeba has been improved greatly.
3.Progress in Elimination of Elemental Sulfur by the Acidophilic Sulfur-oxidizing Bacteria

Metal sulfides are chemically attacked by Fe~ 3+ and H~+, resulting in the formation of elemental sulfur via polysulfides or thiosulfate pathway. Elemental sulfur may aggregate and even form a layer on the metal sulfide surface, which will inhibit leaching metals from the sulfides minerals. Elimination of inert elemental sulfur in a typical acidic environment can exclusively be by way of oxidation of acidophilic sulfur-oxidizing bacteria, such a way includes the attachment, transport and oxidation process of elemental sulfur by acidophilic sulfur-oxidizing bacteria. On the basis of analysis on the pertinent researches, the molecular mechanism of sulfur elimination by the acidophilic bacteria is far away from elucidated, and to attain that target, there are still much work to be done for elucidating the molecular mechanism on the attachment, transport and oxidation process of elemental sulfur by the acidophilic sulfur-oxidizing bacteria.

Up to now, there is no clear and effective measure to protect the cisplatin-induced ototoxicity in the clinical use of cisplatin studies indicated that autophagy may be involved in the whole process of cisplatin-induced hearing loss. In this review, the relationship between cisplatin ototoxicity and autophagy was reviewed. It is hoped that this study can provide reference for further study of cisplatin ototoxicity and intervention of autophagy with autophagy activator or inhibitor.

7.Model study on CO2 removal by photobioreactor.

The key point of study on CO2 removal by microalgae cultured in a photobioreactor is to improve CO2 removal capability. In this paper, a model of air-lift photobioreactor was developed by combination of conditions including the velocity of flow, the degree of mixing, the gas-liquid mass transfer and the rate of photosynthesis, and two corresponding simplified methods, such as time discretization and lumped parameters were put forward. Using a method of lumped parameters, the model for simulation of time course of DO, pH in the column air-lift photobioreactor and prediction of CO2, O2 concentrations in the outlet gas under different CO2 concentration in the aeration gas was thoroughly discussed. Experimental data were also used to verify the model which could potentially be applied to rational design of the photobioreactor, high-density culture of microalgae and efficient removal of CO2.

8.Characterization of the doxDA Operons of Acidithiobacillus ferrooxidans

Reverse transcriptase-PCR experiments suggest that the two clusters of genes potentially involved in the oxidation of reduced sulfur compounds are organized as operons in strain of the acidophilic, chemolithoautotrophic bacterium Acidithiobacillus ferrooxidans ATCC 23270, the two clusters of genes including such the ORF of putative sulfate-thiosulfate-molybdate binding proteins, the ORF of putative thiosulfate: quinone oxidoreductase and the ORF of the rhodanese-like protein (P21). Bioinformatic analyses have predicted the possible promoters sequences and the possible +1 start site of transcription for the doxDA operons.
9.Immunophenotype of Children with Acute Myeloid Leukemia and Its Clinical Significance

Objective To explore the immunophenotype of children with acute myeloid leukemia(AML) and its clinical significance.Methods Statistics was used to analyze the relationship between the immunophenotype of AML and their French-American-Britain(FAB) classification,complete remission (CR) in one month and 3-years event-free survival(EFS).Results CR rate was 71.6% and 3-years EFS rate was 50.8%. HLA-DR and CD34 absent mainly in M3, associated with higher CR and EFS rate. So did CD33 negative cases, especially in M2. CD13 positive was significantly predictive factor for achieving CR.Co-expression of lymphoid antigens and NK cell antigens(CD56) with M2 which correlated with lower CR and EFS rate.Conclusions The negative of HLA-DR, CD34, CD33,as well as CD13 positive, have relationship with good prognosis. Lymphoid antigens and CD56 are poor prognostic factors.
10.Investigation of Leukemia-Associated Immunophenotyping at Relapse and Treatment Failure in Children with Acute Lymphoblastic Leukemia

Objective To investigate the stability of immunophenotyping in the course of relapse or at treatment failure of patients with acute lymphoblastic leukemia(ALL) and that of immunophenotyping of positive minimal residual disease(MRD).Methods From Aug.2000 to Dec.2007,33 children with ALL who relapsed or treated failure were enrolled. These children were detected MRD by flow cytometry. The immunophenotyping of children who relapsed or treated failure were compared with that of initial therapy;the immunophenotyping of MRD relapsed was compared with that of initial therapy.Results 1.In 23 out of 27 cases (85.18%) with B-ALL,changed at least 1 antigen between diagnosis and relapse.Six children with CD45 down-modulation and 2 children with CD45 up-modulation.Two children with CD19 down-modulation and 1 child with CD19 up-modulation.Six children with CD34 down-modulation and 4 children with CD34 up-modulation. Five children with CD10 down-modulation and 7 children with CD10 up-modulation.2.Six children with T-ALL had the same expression in CD45 between relapse and treatment failure. 3.These were 15 children had the least 1 case MRD,25 cases MRD were detected,these was 1 case up-modulation in CD45,1 case down-modulation in CD19,2 cases up-modulation and 8 cases down-modulation in CD34,3 cases up-modulation and 6 cases down-modulation in CD10.Conclusions Immunophenotyping of children with ALL may change at relapse and treatment failure. The frequency of change in B-ALL is higher than that of in T-ALL,but the change can not impact the detection of MRD.
