1. Multiple autoantibody detection in esophageal cancerous or precancerous serum using tumor-associated antigens microarray

Objective: To identify serum autoantibody biomarkers by an tumor-associated antigens (TAAs)-based microarray in the patients with esophageal precancerous lesions (EPLs) and patients with esophageal cancer (EC). Methods: The antigens microarray with 55 TAAs was used to detect the serum autoantibodies of the residency-, age-, and sex-matched 26 pairs, which included 26 cases with EC, 26 cases with EPLs and 26 normal controls. The cut-off value was defined as mean ± 2 standard deviation of the expression levels of the antibodies in the normal control group. The value corresponding to the expression level of the antibody beyond the range of cut-off value was considered seropositive. Results: Five autoantibodies (CYFRA21-1, CA72-4, NY-ESO-1, GAGE-7 and SCCA) were significantly up-regulated, and others were down-regulated. Twenty-five autoantibodies had significant difference among EC, EPLs and normal controls which were found by multi-factor analysis of variance. Serum antibodies could be detected by 9 antigens, including CA72-4, CCNB1, CDKN2A, NY-ESO-1, CYFRA21-1, E2F1, ERBB2, GAGE-7 and SCCA. The positive detection rates of CYFRA21-1 were the highest in EC and EPLs, which were 61.54% and 60.00%, respectively. Furthermore, other antigens with higher seropositive rates were NY-ESO-1 (50.00% for EC, 52.00% for EPLs), SCCA (46.15% for EC, 48.00% for EPLs), GAGE-7 (46.15% for EC, 24.00% for EPLs) and CA72-4 (34.62% for EC, 16.00% for EPLs) in turn. The seropositive rates of these 9 antigens in combined detection panel were 88.46% and 84.00% for EC and EPLs, respectively. Conclusion: CYFRA21-1 is possibly used as an autoantibody indicator for early screening of EC and EPLs. The combined detection of the 9 antigens is likely to improve the sensitivity of autoantibody detection in EC and EPLs serum samples. Copyright © 2013 by TUMOR.

Quality of ulcer healing (QUH) was applied to explore the mechanism of the recurrence of peptic ulcer. The relationship of QUH and epidermal growth factor, prostaglandins, heat shock proteins, immune function and Helico-bacter pylori was assayed. Mechanism of Chinese herbal medicine for the recurrence of peptic ulcer was presented as follows: improving blood circulation and barrier function of gastric mucosa, eliminating Helicobacter pylori and increas ing QUH.
3.STUDY ON THE ANTIFUNGAL ACTIVITIES OF ENDOPHYTIC FUNGI ISOLATED FROM SEVERAL PHARMACEUTICAL PLANTS

One hundred and seventy-two strains of endophytic fungi were isolated from Taxus mairei,Cephalotaxus fortunei and Torreya grandis cv.merrillia.The result of the antifungal assay shows that ninety strains of the fungi have antagonism against one or more botanical pathogenic fungi,such as Neurospora sp.,Trichoderma sp.,Fusarium sp.etc.The percentage of antifungal strains to tested strains are as follows:40% Cephalotaxus fortunei,54.2% Taxus mairei,57.1% Torreya grandis cv.merrillia.Thirty-five strains have high antifungal activities,and their inhibition zone diameter is at least 15mm.The active endophytic fungi were identified as 18 genera,most of which belong to Paecilomyces and Fusarium etc.

5.Effects of Vitamin B_6 Injection on Small Intestinal Peristalsis in Mice and Its Mechanisms

Objective To study the effects of vitamin B_6(VitB_6) injection on small intestinal peristalsis in mice and its mechanisms.Methods The mice were divided into 12 groups:calcium chloride injection group(1 mg/10 g),neostigmine methylsulfate injection group(0.001 5 mg/10 g),atropine sulfate injection group(0.005 mg/10 g),their combination with VitB_6 injection and high/low dose treated groups,high dose VitB_6 injection group(5 mg/10 g),low dose VitB_6 injection group(0.5 mg/10 g) and physiologic saline group(0.1 mL/ 10 g ).After administration 30 minutes,mice were intragastric administration Indian ink(0.1 mL/g),and they were luxated and put to death 20 minutes later.The mice belly were cut open,the length of intestine and distance of Indian ink that had moved were measured,and then the ink progradation rate were calculated.Results Compared with control group,the high dose VitB_6 injection could inhibit normal intestinal peristalsis of mice markedly(P0.05).Conclusions VitB_6 injection can inhibit hyperanakinesia of small intestine in mice,especially high dose.And this will be provided as theory foundation on enterospasm treatment.

Omics docking study for polygenic inheritance tumors has become an important strategy in oncology research. This review focuses on the conceptions and technologies of omics, and puts forward the central contents and omics docking for polygenic inheritance tumor to reveal the role of molecular changes at different stages of polygenic inheritance tumor at multidisciplinary and multilayer level. It is a new strategy to explore the mechanism of tumor carcinogenesis, and to regulate the network, key molecules, and drug target by combined biology effects.

8.Effect of menbranous milkvetch root parenteral solution on insulin resistance of gerontism cerebral infarction in stage of recovery

ObjectiveTo study the effect of menbranous milkvetch root parenteral solution on insulin resistance of gerontism cerebral infarction in recovery stage.Methods66 patients with gerontism cerebral infarction were randomly divided into therapy group(33 cases) which received membranous milkvetch root parenteral solution and control group(33 cases).Both groups adopted routine treatment at the same time, the period was 20 days. Insulin(Ins), free blood sugar(FBS), total cholesterol(CH), triglyceride(TG), hemorheology and insulin resistance(indicating by index of insulin sensitive) of blood on empty stomach were evalutated before and after treatment. ResultsAfter treatment there was decrease in CH, FBS, FINS in therapy group than in control group(P<0.01 or P<0.05); the clinical effect in therapy group was better than in control group(P<0.01).ConclusionMembranous milkvetch root parenteral solution can significantly decrease insulin resistance, blood lipin, blood viscosity in recovery stage of gerontism cerebral infarction, and improve clinical efficiency.
9.Effect analysis of combined immunological blockade mother-to-child transmission of hepatitis B virus in Hainan Province from 2015 to 2020

Abstract: Objective To analyze the influencing factors of mother-to-child transmission of hepatitis B virus after combined immunological blockade, and to evaluate the effect of mother-to-child blockade, and to provide a basis for health policies and health interventions for preventing mother-to-child blockade of hepatitis B virus. Methods A total of 11 363 pairs of HBsAg positive pregnant women and their infants aged 7-12 months in Hainan Province from 2015 to 2020 were included in the study. The general situation, the situation of health care and delivery in this pregnancy and perinatal period, the detection of hepatitis B markers, the situation of antiviral therapy, the general situation of mother and infant during delivery and the implementation of blockade measures for mother-to-child transmission of hepatitis B were collected and analyzed. Results Among the 11 363 pairs of HBsAg positive pregnant women and their infants delivered in hospitals in Hainan province from 2015 to 2020, the positive rate of HBsAg in children at 7-12 months after birth was 1.47 %, and the difference in HBsAg positive rate of infants born in different years was not statistically significant (P>0.05). There were no significant differences in the positive rate of HBsAg among children born to pregnant women with different nationalities, educational levels, occupations, delivery modes, delivery places, obstetric operations and perineal laceration, abnormal perinatal period, children with different genders and premature delivery and perinatal (all P<0.05). There was significant difference in HBsAg positive rate among infants born to pregnant women of different ages, the positive rate of HBsAg of infants born to young pregnant women was higher than that of older pregnant women (P<0.05). The rate of antiviral therapy was low in HBeAg positive pregnant women, and the positive rate of HBsAg in their infants was 2.54%, which was higher than 0.83% in HBeAg negative pregnant women (P<0.05). Conclusions Combined immunological blockade with hepatitis B vaccine and hepatitis B immunoglobulin can effectively prevent the mother-to-child transmission of HBV. HBsAg-positive women can give birth at the right age, and HBeAg-positive pregnant women can be treated with antiviral therapy to block mother-to-child transmission, providing the important basis for the formulation of hepatitis B prevention and control strategies and measures.
10.Expression of Wnt5a gene in breast cancer cell line MCF-7 and its relationship with E-cadherin or vimentin

Objective To investigate the relationship between Wnt5a gene and E-cadherin or vimentin gene in breast cancer cell line MCF-7. Methods RT-PCR was used to detect the mRNA expression of Wnt5a, E-cadherin and vimentin in breast cancer MCF-7 cells and the normal human mammary epithelial cell line MCF-10A, respectively, and their correlation was analyzed. Results The mRNA expression levels of Wnt5a and E-cadherin in cell line MCF-7 were significantly lower than those in cell line MCF-10A [(16.93± 2.97)%vs. (27.47±2.76) %, (12.97±1.35) % vs. (20.43±2.60) %, both P<0.05]. The mRNA expression level of vimentin in cell line MCF-7 was significantly higher than that in cell line MCF-10A [(16.53±0.85)%(6.33± 2.08) %, P<0.05 ]. In cell line MCF-7, the expression of Wnt5a was positively related to E-cadherin (г=0.997, P<0.05), but it was negatively related to vimentin (г=-0.998, P<0.05). Conclusions The expression of Wnt5a in human breast cancer cell line MCF-7 is significantly lower than that in cell line MCF-10A, which indicates that Wnt5a is a cancer suppressor gene in breast cancer. The expression of Wnt5a in cell line MCF-7 is positively related with E-cadherin, and it is negatively related with vimentin. Wnt5a may cause invasion and metastasis of breast cancer cell through the breast epithelial mesenchymal transitions.
