1.Effects of sodium arsenite on melanin metabolism of A375 and G361 cell lines

Objective To study the impact of sodium arsenite(NaAsO2) exposure on melanoma cells A375 (hereinafter referred to as the A375) and G361 (hereinafter referred to as the G361) pigment production and tyrosinase (TYR) activity and the differences of pigment metabolism capacity between the cell lines.Methods A375 and G361 cells were exposed to sodium arsenite at concentrations of 0.0(control),0.1 and 1.0 μmol/L for 72hours.Cell viability was measured by Alamar Blue assay.Melanin levels and TYR activity were measured at the same time.Results After exposure for 72 hours,the cells of 0.1 μmol/L dose groups of both of the two cell lines [A375:(103.32 + 1.26)%; G361:(104.10 + 1.76)%] showed a slightincrease of proliferation without significant differences compared with those of the control[A375:(100.00 ± 1.08)%; G361:(100.00 + 1.79)%,all P < 0.05] ;while cell viability of the 1.0 μmol/L dose group of both of the two cell lines[A375:(75.32 ± 1.59)%; G361:(78.26 ± 2.10)%] were significantly lower than those of the control (all P < 0.05).Melanin levels of G361 cell line [(7.19 ± 0.35),(7.34 ± 0.83),(8.19 ± 0.86)pg/cell] were significantly higher than that of A375[(4.35 ± 0.72),(4.54 ± 0.01),(4.60 + 0.59)pg/cell,all P < 0.05] in all the three groups.TYR activity of G361 cell line [(54.13 ± 1.21),(54.56 ± 0.21),(56.25 ± 0.85)Bq] were also markedly higher than that of A375 cell[(42.00 ±0.21),(42.90 ± 0.54),(42.91 ± 0.01)Bq,all P < 0.05] in all the three groups.The melanin levels and TYR activities of both of the two cells lines showed an increase tendency along with increased doses of arsenic exposure,but without significant differences when compared with those of the three groups (all P > 0.05).Conclusions Arsenic related pigment disorder may be associated with increased melanin levels and TYR activities induced by arsenic exposure; individual difference of pigment metabolism may be associated with different basal melanin levels and TYR activity between different individuals.
2.A comparative study of dispatcher-assisted telephone-cardiopulmonary resuscitation and smart phone APPs for conducting cardiopulmonary resuscitation

Objective The purpose of our study was to assess the onset effect of bystander CPR assisted by a smart phone APP or by a dispatcher.Methods 74 volunteers,without any experience in basic life support measures,was randomly divided into three groups:no-assisted group (n=13),dispatcher-assisted group (n=34) and smart phone APP-assisted group (n=27).Under the condition of a simulative SCA,a QCPR mannequin was used to record the hands-off time,hand placement,rate,depth and complete chest recoil after each compression of the three different groups to make analysis.The no-assisted group didn't receive any CPR instruction;the dispatcher-assisted group was instructed to do CPR through the voice from the telephone;the smartphone APP-assisted group was instructed by a preset Android smart phone CPR APP which guided the volunteers to do CPR step by step by words,voice and animations.Rank sum test analysis of each group using SPSS19.0 software.Results In aspects of hands-off time (1.0 second vs.1.5 seconds vs.5.0 seconds),mean rate of compression (121.0 per minute vs.127.5 per minute vs.58.0 per minute),percentage of correct compression rate (99.0% vs.98.0% vs.0.0%) and mean depth of compression (33.0 cm vs.26.5 cm vs.16.0 cm),smart phone APP-assisted group performed as well as dispatcher-assisted group (P>0.05) and was superior to no-assisted group (P<0.05).Conclusion In terms of increasing the quality of CPR,the smart phone APP can play the same role as dispatcher's telephone guidance,so as to enhance the effect of cardiopulmonary resuscitation.

4.Effect of testosterone on mitotic orientation in rat prostate epithelial cells

Objective To investigate the effect of testosterone on mitotic orientation in rat prostate epithelial cells and the relative differential gene expression.Methods Twenty SPF male SD rats were divided into 2 groups at random and then subjected to castration.One group of rats was administrated with testosterone 3.7 mg daily for 30 days and the control group was only injected with olive oil.Microscopic analysis was performed using immunohistochemistry.Differential gene expression analysis was conducted by gene microarray and RT-PCR techniques.Results In the testosterone-adminis-trated group, there was a significant mitosis orientation parallel to the basement membrane.But in the control group, mito-sis orientation was oriented perpendicular to the basement membrane.Using the gene microarray and RT-PCR techniques, the cell proliferation genes such as Ran, Tgm4 and Wnt2 in Wnt signal pathway were up-regulated in the testosterone group.Conversely, suppressor cell proliferation genes such as Dkk3 and Fas were down-regulated.Conclusions Mitotic orientation of prostate epithelia cells is changed after testosterone administration.Wnt signal pathway and AR singling path-way also have an influence on the mitosis orientation and cell proliferation.
5.Effects and mechanisms of Shaoqiduogan on immunological hepatic fibrosis

Aim To investigate the effects of Shaoqiduogan(SQDG)on immunological hepatic fibrosis induced by human albumin in rats as well as its possible mechanisms.Methods The model of immunological hepatic fibrosis induced by human albumin was prepared.The rats were randomly divided into 6 groups,namely normal control group,liver fibrosis model group,SQDG(42.5,85,170 mg·kg~(-1))treated groups and colchicine(0.1 mg·kg~(-1)) treated group.HE staining was used to examine the histopathological change.The activities of transaminase in serum,malondiadehyde(MDA)content,superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)activities,hydroxyproline(Hyp)content in liver homogenate were assayed byspectrophotometry.The levels of hyaluronic acid(HA)and procollagen Ⅲ (PCⅢ)in serum were determined by radioimmunoassay.In vitro,the collagen production of hepatic stellate cell(HSC)-T6 stimulated with transforming growth factor beta1(TGF-β1)was measured with 3H-Proline uptake.Results SQDG had obvious protective effects on human albumin induced hepatic fibrosis in rats.The results showed that the serum ALT and AST decreased by SQDG treatment,but had no significant difference compared with model group.Pathological examination showed that SQDG could remarkably alleviate the hepatic fibrosis.SQDG not only decreased the Hyp content in liver homogenates,but also the elevated level of HA,PCⅢ in serum.SQDG also ameliorated the oxidative stress state of hepatic fibrosis rats,decreased the production of MDA and enhanced the activities of antioxidative enzyme including SOD and GSH-Px.Furthermore,SQDG(20~160 mg·L~(-1))inhibited the collagen production of HSC stimulated with TGF-β1 in vitro.Conclusion sSQDG has protective effect on liver fibrosis rats induced by human albumin.The mechanisms of its anti-fibrotic effects may be associated with its action of ameliorating the oxidative stress in liver,and inhibiting the production of collagen in HSC.

7.Correlation of blood glucose, blood pressure and body weight with pancreatic cancer

Objective To investigate the correlation of blood glucose,blood pressure and body weight with pancreatic cancer.Methods From January 2011 to December 2013,110 patients diagnosed with pancreatic cancer in our hospital were selected as the observation group and 110 agematched cases without cancer during the same period were selected as the control group.The percentages of patients with diabetes,hypertension and elevated body mass index (BMI) were analyzed in both groups.Results The number of patients with diabetes was higher in the observation group than in the control group (32 cases or 29.1% vs.16 cases or 14.6%,P<0.05).The proportions of pancreatic cancer patients with diabetes duration ≤ 2 years,2~5 years and 5~10 years were higher in the observation group than in the control group (P<0.05).The proportion of subjects with increased BMI was higher in the observation group than in the control group (24.6% or 27cases vs.10.9% or 12 cases,P<0.05).The proportions of patients with diabetes combined with increased BMI and of patients with hypertension and increased BMI were higher in the observation group than in the control group (17.3% or 19 cases vs.2.7% or 3 cases,10.9% or 12 cases vs.2.7% or 3 cases,respectively,P< 0.05 for both).Conclusions Diabetes,hypertension,and elevated BMI can be considered as risk factors for pancreatic cancer,and it is possible that these factors are involved in the development of pancreatic cancer.

Objective To study the clonal heterogeneity in differentiation potential of immortalized mesenchymal stem cells in vitro and in rive.Methods The monoclonal cell lines were performed with limiting dilution cloning,and were induced to adipocytic,asteogenic and neuronal differentiation in vitro.After transplanted the monoclonal cell lines into SCID mice,the xenotransplants were removed and evaluated by immunohistochemistry.Results From the parental HMSC-TERT,32 single-cell derived clones were established,of which the differentiation properties varied considerably in vitro.The cells grow in different plating densities during expansion in culture:HMSC-TERT-2 expressed more strongly in LCA,GFAP and vimemin;HMSC-TERT-C19 expressed more strongly in keratinose than HMSC-TERT-2,HMSC-IERT-20,and MSC-H;HMSC-TERT-C2 expressed more strongly in actin than HMSC-TERT-2.Conclusions The HMSC-TERT monoclone cells are heterogeneity in differentiation petential in vitro and in vivo,suggesting that standard in vitro culture and in vivo inoculate procedure phy an important role in the clinical application of stem cells.
