1.Establishment of animal model Of acute suprachoroidal haemorrhage and its pathology

Background Expulsive choroidal haemorrhage is a severe complication during the intraocular operation.To create a feasible animal model of expulsive choroidal haemorrhage is very important for the study of its nature disease course and therefore prevention and treatment.Objecfive Present study was to modify the method of establishing acute choroidal haemorrhage animal model and explore the risk factor and pathogenesis mechanism.Methods The suprachoroidal explusive haemorrhage animal models were created by modifying the Zauberman method to ligate 2-4 vortex veins and rapidly intravenous injection of 250 ml normal solution to elevate the intraocular pressure(IOP),and then low the IOP by opening the anterior chamber via limbal incision.The bleeding of choroid and detachment of retina were examined by B-type ultrasound immediately,1,2 and 4 weeks after operation to assess the disease alteration with lapse of time.The samples of eyeballs were prepared in mentioned above time points for the histopathologieal evaluation of retina tissue.Results The animal models of suprachoroidal explusive haemorrhage were established bv this modified method with the successful rate 100%,and the manifestation resembled a clinical disease course.The shape,range and location of choroidal bleeding and retinal detachment were determined,and the diagnosis and change of suprachoroidal explusive haemorrhage with time were clarified using B-uhrasonography.The histopathological change revealed the breaking of Brueh's membrane and retinal pigmentary layer.It was verified that the dominant pathological change of retina was retinal destroy in 1 week after operation,and in 4 weeks after operation,the main pathological mechanism was proliferation of fibroblasts and collagen fiber. Conclusion The consequence procedure of modified Zauberman method,quickly intravenous injection of normal solution and puncture of the anterior chamber can create an ideal animal model of suprachoroidal explusive haemorrhage.The rapid IOP descent is a main risk factor of supraehoroidal explusive haemorrhage.The results of supraehoroidal explusive haemorrhage is severe destroy of retina and ehoroid.This study implies that supraehoroidal explusive haemorrhage is likely to be prevented and treated in the early stage.
2.Serum excitatory amino acids concentrations in patients with amnestic mild cognitive impairment or Alzheimer's disease

Objective To figure out changes of serum excitatory amino acids (EAAs) levels in patients with amnestic mild cognitive impairment (aMCI) and Alzheimer's disease (AD).Methods The levels of serum EAAs was assessed in 34 cognitively normal control subjects,30 patients with aMCI,and 32 patients with AD using high performance liquid chromatography (HPLC).Results ①Higher serum concentrations of glutamate((39.6?22.1) ?mol/L),alanine((282.5?71.3) ?mol/L) were found in the aMCI patients (P=0.044,P=0.007),and higher serum concentrations of glutamate ((42.2?21.8) ?mol/L),glycine ((464.2?142.6) ?moL/L) were found in the AD patients than in the control subjects (P=0.010,P=0.010).②No statistically significant difference of EAAs level between the aMCI and AD groups was found.③A close and positive correlation between the serum concentrations of glutamate, aspartate and the mini-mental status examination scores were found in AD patients:the 2 amino acid levels were higher in patients with mild dementia((42.1?21.3),(55.0?29.0) ?mol/L) than those with moderate or severe dementia ((25.4?9.2) ?mol/L,P=0.023;(34.6?11.1) ?mol/L,P=0.036). Conclusion EAAs,correlating with the severity of the condition,play a significant role in AD,while aMCI patients also have disturbance of metabolism of EAAs,indicating that it has similar pathogenesis to AD.
3.Chemical constituents from polarity part in roots of Angelica dahurica var. formosana cv. Chuanbaizhi.

The chemical constituents from polarity part in the roots of Angelica dahurica var. formosana cv. Chuanbaizhi were studied in this paper. The compounds were separated and purified by repeated column chromatographic methods on silica gel and HPLC, and the chemical structures of compounds were determined by spectral data analyses. Fourteen compounds were obtained and identified as tert-O-β-D-glucopyranosyl-(R)-byakangelicin (1), (2"S) -3"-O-β-D-glucopyranosyl-oxypeucedanin hydrate (2), marmesinin (3), sec-O-β-D-glucopyranosyl-byakangelicin (4), isofraxidin-7-O-β-D-glucopyranoside (5), benzyl-O-β-D-glucopyranoside (6), 8-O-β-D-glycopyranosylxanthotoxol (7), prenyl-O-β-D-glucopyranoside (8), scopolin (9), (2' R) -5'-hydroxymarmesin-5'-O-β-D-glucopyranoside (10), (2'S,3'R) -3'-hydroxymarmesinin (11), skimmin (12), benzyl-O-β-D-apiofuranosyl-(1"--> 6')-β-D-glucopyranoside (13), and decuroside IV (14). Among them, compounds 2, 5, 6, 8, and 10-13 were obtained from the roots of title plant for the first time.

5.Study on relationship between expression of PKC? in glomeruli and development of nephropathy in diabetic rats

AIM: To observe the dynamic changes of expression of PKC? , TGF-? 1 and ?-SMA in glomeruli of diabetic rats induced by the alloxon and to invesitigate their roles in the diabetic nephropathy(DN). METHODS: Rats were randomly divided into four groups: normal control group (group A), diabetic group of one week (group B), diabetic group of one month (group C), diabetic group of two months (group D). Immunohistochemistry and Western blotting were used to detect the expression of PKC?, TGF-? 1 and ?-SMA in renal tissue of all groups. Blood glucose, triglycerides, cholesterol, creatinine and urine protein were analysed by chemical methods. The morphological changes of renal tissue were checked through microscopy. 7.Clinical significance of T cell subgroups and NK cells' detection in peripheral blood of diffuse large B-cell lymphoma patients

Objective To investigate peripheral blood T lymphocyte subsets and NK cells changes in the diffuse large B cell lymphoma (DLBCL) patients before and after chemotherapy,and analyse the relationship between the results and treatment.Methods Collect the 47 patients venous blood of the effective treatment DLBCL by pathology.T lymphocyte subsets and NK cells were determined by flow cytometric.Analyse the results statistically significant difference before treatment,the second chemotherapy cycle and the fourth chenmotherapy cycle compared with 50 healthy control persons.Results The levels of CD3+,CD4+,CD4+/CD8+ NK cells in DLBCL patients before chemotherapy [(70.04±8.87)%,(42.79±6.06)%,(1.68±0.59)%,(14.40±6.02)%]were lower than healthy controls [(63.89±6.67)%,(32.72±5.77)%,(0.85±0.25)%,(9.95±5.24)%](P < 0.05),and the level of CD8+ cells is higher than the healthy controls [(27.21 ±6.54)% vs.(39.92±7.11)%](P < 0.05).The levels of CD3+,CD4+,CD4+/CD8+ cells had significant difference between the second and the fourth chemotherapy cycle in DLBCL patients (P < 0.05).The levels of CD3+,CD4+,CD8+,CD4+/CD8+,NK cells had significant difference between the fourth chemotherapy cycle DLBCL patients and the DLBCL patients before chemotherapy (P < 0.05).Conclusion The DLBCL patients exist immunosuppression before chemotherapy.Peripheral T lymphocyte subsets and NK cells can be used as good reflect of the cellular immune function in DLBCL patients.Clinical parameters can be used for the immune function monitoring and providing guidance for the treatment options.
8.Nursing of patients with high grade intraepithelial neoplasia treated by endoscopic submucosal dissection

Objective To explore the experience in nursing the patients with esophageal high-grade intraepithelial neoplasia lesions treated with endoscopic submucosal dissection. Method A retrospective analysis was carried out among the patients with esophageal high-grade intraepithelial neoplasia lesions to look into the nursing care to the patients. Results Thirty patients were successfully stripped, and no surgical complication occurred. After intensive care, they were cured and discharged. Conclusion Professional and meticulous care can effectively improve the success rate of endoscopic submucosal dissection and reduce the incidence of complications, and promote the rehabilitation of patients.
9.Comparison of inhaled nitric oxide and Iloprost in infants with moderate or severe pulmonary hypertension af-ter cardiac surgery:a prospective randomized trial

Objective To compare the efficacy of inhaled Iloprost and nitric oxide( NO)in infants with moderate or severe pulmonary hypertension(PH)after congenital heart disease surgery. Methods This was a prospec-tive randomized study in Children's Hospital of Fudan University from January 2011 to January 2014,including 40 in-fants who suffered from moderate to severe PH after heart surgery. Their ages ranged from 1 to 24 months. Their weight ranged from 3. 2 to 11. 0 kg. They were randomly allocated to inhale NO( NO group,n = 20)or Iloprost( Iloprost group,n = 20). Iloprost group was given Iloprost with 50 ng/(kg·min)for 10 min and then combined with NO 20 × 10 - 6 for 10 min;NO group was first given 20 × 10 - 6 of NO for 10 min,then combined with Iloprost 50 ng/(kg·min) for 10 min. Heart rate,systolic blood pressure,pulmonary artery pressure(PAP)and central venous pressure were recor-ded continuously. At the same time,the concentration of nitrogen dioxide and methemoglobin after inhaling NO was de-tected. Results Inhaled Iloprost and NO caused significant reduction in PAP(t = 4. 670,P = 0. 009;t = 3. 762,P =0. 004)and pulmonary - to - systemic pressure ratio(Pp/ Ps)(t = 16. 974,P = 0. 000;t = 9. 682,P = 0. 000)but signifi-cant increase in oxygen index separately. The combination had no additional effect compared with single application. In term of the reduction of PAP,there was no significant difference between inhaled Iloprost and NO(F = 2. 742,P =0. 129). The levels of nitrogen dioxide and methemoglobin were not above the normal limits. Conclusions Moderate to severe PH after cardiac surgery was significantly reduced by inhaled NO and Iloprost. They had similar effects. However, the combination of both vasodilators failed to prove more potent than either substance alone. Compared with the potential toxicity of NO,the individual differences and the complex transmission,Iloprost may be more reasonable and feasible for the postoperative treatment of PH.
10.The pathological TDP-43 protein expression in the central nervous system of motor neuron disease

Objective To understand pathological TDP-43 features in the central nervous systems of patients with clinically and autopsy confirmed motor neuron disease (MND).Methods The clinical and histopathological features of 4 cases with MND confirmed by autopsy were summarized; anti-ubiquitin (Ub) and anti-TDP-43 immunohistochemical staining were carried out on tissue of brains and spinal cords from 4 cases with MND and 3 control cases without history of neurological disorders.Results These 4 cases presented with typical clinical and histologic features of MND.Ub-positive inclusions were observed in brain and spinal cord from 3 cases with the Ub-positive inclusions of skein-round-and lewy body-like structures.Strong TDP-43 pathological staining in brain and spinal cord was identified in 2 cases with MND presented as neuronal and glial cytoplasmic inclusions with various shapes.The TDP-43 positive inclusions were widely distributed in the motor cortex of brain and the anterior horn of spinal cord.TDP-43 weak staining in the spinal cord tissue was observed in 1 case with MND.No Ub-and TDP-43 positive inclusions were found in 3 control cases.Conclusion There is widespread pathological TDP-43 expression in the central nervous system of MND.TDP-43 positive inclusions in MND have relatively high specificity.It is worth further study on their formation mechanism.
