1.Expression and significance of N-myc downstream regulated gene 1 in renal cell carcinoma

Objective To study the expression of N-myc downstream regulated gene 1 (NDRG1) in renal cell carcinoma and its relationship with microvessel density (MVD) and clinicopathologic parameters.Methods Immunohistochemical study for NDRG1 and CD34 was performed on paraffin sections of cases of renal cell carcinoma and adjacent non-neoplastic renal parenchymal tissue. MVD was analyzed by CD34immunostaining.Results Immunohistochemical study showed that non-neoplastic proximal convoluted tubule, distal convoluted tubule and collecting ducts was positive for NDRG1 (membranous and cytoplasmic). The expression rate of NDRG1 in renal cell cancer was 51%, which was significantly lower than that in normal renal tissues of 100 % (P <0.05).Clinicpathologically, the result also showed a close relation between lower NDRG1 expression and higher pathologic grade (χ2 =9.968, P =0.007), later clinic stage (χ2= 6.437,P =0.011), lymph node metastases (χ2=5.800, P =0.016) and higher MVD of cancer (t =2.235, P =0.030)whereas no relation with other factors. Conclusion NDRG1 might play as a cancer suppressor gene in renal cell carcinoma in that it could be correlated with tumor invasion and metastasis.It might also suppress tumor growth and metastasis by regulation of tumor angiogenesis.
2.Construction of a human Flt3-ligand gene containing eukaryotic expressing vector and its expression in COS-7 cells

Objective:To construct an eukaryotic expressing vector pIRES1neo/hFL and express hFL in COS 7 cell.Methods:The cDNA encoding soluble human Flt3 ligand(FL) was obtained by RT PCR from the TF 1 cell lines and was inserted into eukaryotic expressing vector pIRES1neo between EcoR I and BamH I sites after sequencing,and then COS 7 cells were transfected with the recombinant by liposome.The transcription and expression of hFL in the transfected COS 7 cells were assayed by RT PCR,ELISA and the experiment of the human umbilical blood CD34 + cell multiplication,respectively,at 72 hours after transfection.Results:It showed that hFL cDNA cloned in our laboratory was 546 bp in length encoding soluble human FL which was in accordance with the report previously.FL gene was transcripted in transfectants,and FL protein with obvious biological activity was highly expressed with 251 ng/(10 6 cell?d) in the supernatant of the transfectants.Conclusion:Human FL in the recombinant vector is proved to be expressed in COS 7 cells and obvious biological activity of hFL in the supernatant of the transfectants was detected.
3.Treatment of hematomas in operation area after anterior approach surgery for cervical spondylosis

Objective To investigate the treatment and prevention strategies of hematomas in operation area after anterior approach surgery for cervical spondylosis.Methods A retrospective review was conducted on 12 with hematoma compression in operation area out of 785 patients managed by anterior cervical surgery from January 2007 to July 2013,including 10 males and 2 females at age ranging from 40-71 years (mean 56.8 years).Surgery method was anterior cervical corpectomy and interbody fusion using titanium mesh cage plus plate and intraoperative blood loss was 300-1 200 ml.Primary clinical manifestations were neurological dysfunction in 5 patients,dyspnea in 6,and both neurological dysfunction and dyspnea in 1.There were 10 patients with the presence of symptoms at postoperative 0.5-22 hours,1 at postoperative 73 hours,and 1 at postoperative 74 hours.All the 12 patients underwent a second anterior cervical exploration.Results There were 5 patients with epidural hematoma,6 with subcutaneous hematoma,and 1 with both hematomas.After surgical interventions,the patients presented improvement in respiratory and neurological function,with inapparent respiratory abnormality and improved neurological function at discharge.One patient was died of cardiovascular-associated disease after being discharged from hospital.The left 11 patients were followed up for mean 19.8 months (range,6-43 months),with improved Japanese Orthopedic Association (JOA) score at final follow-up.Conclusions Hematoma took place frequently in the early period,especially within 24 hours in operation area after anterior approach to cervical disorders and close attention should be paid to respiratory and limb sensation and motion functions.Early detection and early surgical interventions are the key countermeasures to avoiding the severe results.
4.Clinical analysis of cytogenetic features in acute myeloid leukemia and its relationship with early responses after induction therapy

Objective To study the cytogenetic features of acute myeloid leukemia (AML) and analyze the association with cytogenetic features and early responses after induction therapy.Methods The karyotypes of 395 patients who had been newly diagnosed with AML were analyzed.These patients were divided into three groups (low-risk,intermediate-risk and high-risk),according to the AML NCCN guidelines.The incidence of different karyotypes in these three groups and the complete remission (CR) rate after the first cycle of induction therapy were analyzed.Results The incidence rates of karyotypes in high-risk,intermediate-risk and low-risk groups were 50.56 % (180/356),39.89 % (142/356),9.55 % (34/356),respectively.All patients with t(15;17) who completed induction therapy reached CR.There was significant difference in the CR rates of t(8;21) groups with or without additional karyotypes [92.00 %(23/25) vs 50.00 %(11/22)] (x2 =10.317,P =0.001).There was no significant difference in the CR rates between normal and-Y karyotype group [61.90 % (39/63) vs 58.82 % (10/17)] (x2 =0.054,P =0.817).Complex cytogenetics ascribed to the low-risk group,of which monosomal karyotype was common,nine of ten patients with monosomal karyotype were associated with an inferior CR rate.Conclusion The cytogenetic features of AML are different from previous reports by other centers.The cytogenetic features of AML patients not only influence the long-term survival,but also the CR rates of induction therapy.
5.Magnetic Beads-Based Biological Mass Spectrometry Technology for Evaluating Therapeutic Efficacy of Acute Leukemia

A serum method for evaluating therapeutic efficacy of acute leukemia has been built by magnetic beads-based biological mass spectrometric technology. After analysis of quality control sample, the reproduci-)bility) of within-run assay and between-run assay was obtained and consistent with the previous reports. This result) manifested that the method used for serum peptides analysis was stable and reliable. Serum samples from 30 patients with primary acute leukemia and those with complete remission after treatment were extracted by metal-chelated magnetic beads and detected by MALDI-TOF-MS. Peaks at the m/z range 1000-10000 were analyzed by FlexAnalysis 2.4 software and Biosun_MS software. Approximately 27 differential peaks that were of statistical significance(p<0.05) were obtained. Ten peaks were up-regulated) and 17 peaks were down-regulated) after complete remission, respectively. The diagnostic model based on these differential peaks achieved) 100% sensitivity and 90% specificity. The results indicated that the model based on magnetic beads extraction and MALDI-TOF-MS detection acquires high sensitivity and specificity. Further sequence identification of differential) peaks is useful for evaluation of therapeutic efficacy and mechanism research on acute leukemia.)

7.Effects of rich environmental rehabilitation training on the physical and neurobehavioral development of neonates with hypoxic-ischemic encephalopathy

Objective:To observe the effect of rich environmental rehabilitation training on the physical system and ner-vous system development of newboms with hypoxic-ischemic encephalopathy(HIE). Method:Sixty-one HIE newboms were divided into the observation group(n=31)and the control group(n=30).The control group was given the routine clinical treatment and care,and the observation group was supplemented with enriched environmental rehabilitation training on the basis of the control group for 3 weeks.The body length and weight,and the head circumference of the two groups were measured to assess the physical development at admis-sion and 3 weeks after the intervention,and the neonatal behavioral neurological assessment(NBNA)and test of in-fant motor performance(TIMP)were used to evaluate the neurological development and motor function. Result:After the intervention,the changes of body length and weight changes in the observed group were bet-ter than the control group(P<0.05),no significant difference in head circumference(P>0.05),and the NB-NA score and TIMP score were higher than those of the control group(P<0.05). Conclusion:The application of rich environmental rehabilitation training can promote the rapid development of physique and nervous system of the newboms with HIE and improve the motor function.
8.Repetitive transcranial magnetic stimulation can improve the effectiveness of constraint-induced movement therapy for hemiplegic children with cerebral palsy

Objective:To explore the effect of combining repetitive transcranial magnetic stimulation (rTMS) with constraint-induced movement therapy (CIMT) in rehabilitating the upper limb functioning of children with hemiplegic cerebral palsy.Methods:Sixty children with hemiplegic cerebral palsy were recruited and randomly divided into a control group ( n=30) and an experimental group ( n=30). Both groups were given conventional rehabilitation therapy and intensive one-to-one CIMT of the affected hand for 30 minutes 5 times a week for 4 successive weeks. The healthy hand was immobilized in a hand cushion for 3 hours. The experimental group was additionally provided with 20 minutes of rTMS at 1.0Hz, 5 times a week for the same 4 weeks. Before and after the treatment, both groups were evaluated using the Peabody Developmental Motor Scales (PDMS-2 FM) and an upper extremities function test (UEFT). Results:After 4 weeks of treatment there was significant improvement in both groups with regard to the average PDMS-2 FM and UEFT scores. The improvement in the experimental group was significantly better.Conclusions:rTMS combined with CIMT can effectively improve upper limb motor function and promote the fine motor development of children with hemiplegic cerebral palsy.
9.A Modified NHL-BFM-95 Regimen Produces Better Outcome ThanHyperCVAD in Adult Patients with T-Lymphoblastic Lymphoma,a Two-Institution Experience

Purpose:
Lymphoblastic lymphoma (LBL) is an invasive neoplasm of precursor T-cell or B-cell lineage.A broadly accepted standard treatment for adult LBL has not yet been defined.
Materials and Methods:
To address this issue, we compared two chemotherapy regimens: a modified non-Hodgkinlymphoma Berlin–Frankfurt–Mu!nster-95 (NHL-BFM-95) regimen and HyperCVAD/MA. Thisretrospective study consecutively enrolled 207 adult LBL patients at two hospitals from2000 to 2018. Univariate and multivariate analysis were used to assess prognostic factors.
Results:
In the present study, most clinical characteristics were similar between the two treatmentgroups except for age and lactate dehydrogenase (LDH) level. Patients treated with modifiedNHL-BFM-95 regimen tended to be younger and with elevated LDH level. The modified NHLBFM-95 regimen produced better treatment outcomes than those with HyperCVAD/MA inpatients with T-LBL or patients < 40 years. Treatment with HyperCVAD/MA, high EasternCooperative Oncology Group scores, and bone marrow involvement were independent riskfactors in T-LBL. No patients interrupted treatment for severe adverse events.
Conclusion
The results suggested that the modified regimen is well-tolerated and can produce the promisingoutcomes in patients with T-LBL or patients < 40 years.
10.Effect of aerobic exercise on core symptoms and executive function in children with attention deficit hyperactivity disorder

ObjectiveTo observe the effect of aerobic exercise on core symptoms and executive function in children with attention deficit hyperactivity disorder (ADHD). MethodsFrom June, 2020 to December, 2021, 64 children with ADHD at outpatient in Dushu Lake Hospital were randomly divided into control group (n = 32) and observation group (n = 32). The control group sat down and watched the cartoon for 20 minutes, and the observation group performed cycling exercise while watching the cartoon, for twelve weeks. The core symptoms were assessed with Conners Parent Symptom Questionnaire (PSQ), while the inhibition, conversion and refresh functions were assessed with the psychological test software E-Prime 2.0 system. ResultsAfter twelve weeks of treatment, the PSQ factor scores decreased in the observation group (t > 4.775, P < 0.001), and were lower than that of the the control group (t > 3.184, P < 0.001). The response time and accuracy of inhibition, conversion, and refresh functions decreased in the observation group (t > 2.259, P < 0.05), and were lower than that of the control group (t > 2.007, P < 0.05). ConclusionAerobic exercise could reduce the core symptoms of poor attention and hyperactivity impulsivity, and improve the executive functioning in children with ADHD.
