1.Chemical constituents in root and rhizome of Polygonum cuspidatum

Objective To study the chemical constitients of Polygonum cuspidatum.Methods The compounds were isolated through column chromatography and their structures were elucidated through physicochemical and spectral analyses.Results Seven compounds were obtained from the diethyl ether fraction of ethanol extract and identified as physcion(Ⅰ),emodin(Ⅱ),ambrettolide(Ⅲ),?-sitosterol(Ⅳ),oleanolic acid(Ⅴ),coumarin(Ⅵ),and 2-ethoxy-8-acetyl-1,4-naphthoquinone(Ⅶ).Conclusion Compound Ⅶ is a new compound named as cuspidatumin A,and compound Ⅲ is obtained from P.cuspidatum for the first time.
2.Experimental study of cleft palate repair by rhBMP-2/Co/PLGA composite membrane

Objective:To study whether the guided bone regeneration (GBR) technique can be used to achieve good bone formation in the defects of palate. Methods:Bone defect in the shape of triangle with the height of 4 cm and base line of 1.5 cm was created in the middle of palate in each of 16 adult dogs. The dogs were equally divided into experimental group(A) and control group(B). rhBMP-2/Co/ PLGA membrane was implanted in the defects in group A and the Co/ PLGA membrane in group B. The maxillary bone were observed by CT scan at 4, 8, 12 and 24 weeks after operation. The defect repair was observed by histological examination. Results:CT and histological examinations showed that new bone formation in group A was better than that in group B. 24 weeks after operation the defects in group A were completely repaired by new bone, those in group B partly repaired. Conclusion:The rhBMP-2/Co/PLGA composite membrane can be used to repair the defects in palate.
3.Chemical constituents in Dioscorea septemloba

Objective To study the chemical constituents of Dioscorea septemloba.Methods The compounds were isolated by GC-MS and chromatography on silica gel column and identified on the basis of physico-chemical constants and spectral analysis.Results Thirteen compounds were isolated and their structures were identified as 4,8-dimethyl-1,7-nonadiene(Ⅰ),5-isopropyl-2,8-dimethylcyclodeca(Ⅱ),3,7-dimethyl-6-octen-1-ol-formate(Ⅲ),dodecanoic acid methyl easter(Ⅳ),N,N'-dinitro-1,2 cyclohexanddiamine(Ⅴ),1-octyn-4-ol(Ⅵ),?-sitosterol(Ⅶ),palmitic acid(Ⅷ),stigmasterol(Ⅸ),6,7-dihydroxy-2-methoxy-1,4-phenanthrenedione(Ⅹ),diosgenin(Ⅺ),ruscogenin(ⅩⅡ),and stigmasterol-3-O-?-D-glucoside(ⅩⅢ).Conclusion Compounds Ⅰ—Ⅵ,Ⅸ,Ⅹ,ⅩⅡ,and ⅩⅢ are obtained from this plant for the first time.
4.Study on exons 3 and 4 of alpha-synuclein gene in Chinese familial Parkinson disease patients.

OBJECTIVETo study the relationship of Chinese familial Parkinson disease with alpha-synuclein gene.
METHODSPolymerase chain reaction-single strand conformational polymorphism (PCR-SSCP) and polymerase chain reaction-heteroduplex analysis(PCR-HA) were employed to detect the abnormal mobilization in the familial Parkinson disease and sporadic Parkinson disease patients, then it was verified by gene sequencing.
RESULTSNo mutation was found in alpha-synuclein gene exons 3 and 4 by PCR-SSCP together with PCR-HA. An inserted c and an inserted t were found in intron 4, position 23 and position 67 respectively.
CONCLUSION(1) Exons 3 and 4 of alpha-synuclein gene are not the mutational hot spots of Chinese familial Parkinson disease. (2) Two polymorphisms were found in intron 4 of alpha-synuclein gene. They are 23 ins c and 67 ins t.
