1.Study on anoxia-reoxygenation injury and effects of copper and zinc in heart cell cultures

The effect of various concentrations of copper and zinc on anoxia-reoxygenation in the neonatal rat heart cell cultures was studied by determining the activity of superoxide dismutasc (SOD), glutathione peroxidase (GSH-px), the content of malondialodchyde (MDA) and the release of lactate dehydrogenasc (LDH) The results showed that teh activity of SOD decreased and the activity of GSH-px, the content of MDA and the release of LDH increased following anoxia and reoxygenation. The actiwty or SOD appeared to be increased by either copper or zinc to varying degrees, but the effect of copper showed to be stronger than that of zinc. The release of LDH was reduced at the presence of lower amounts of copper or zinc, but the effect of zinc was better than that of copper. The increased release of LDH occured when higher amounts of copper or zinc were added to the medium. In addition, copper increased the activity of GSH-px while reducing the content of MDA significantly.
2.Expression of toll-like receptor 4 in hepatic ischemia-reperfusion injury

Objective To analyze the expression and the role of Toll-like receptor 4 (TLR4) and the effect of N-acetylcysteine (NAC) on hepatic ischemia- reperfusion (I/R) injury.Methods The rat model of 45 min hepatic ischemia followed by 24 h reperfusion was used. Animals were divided into 3 groups: the pseudoperated control group (P), I/R group and treated group with NAC at a dose of 300 mg/kg at 5 min before reperfusion (I/R-NAC). Plasma concentrations of ALT, AST and lipopolysaccharide (LPS) at various time points were measured. The expression of TLR4 protein and mRNA was detected in liver tissue by using immunohistochemistry and RT-PCR.Results The TLR4 expression, plasma ALT, AST and LPS levels were markedly increased during 45 min hepatic ischemia and 24 h reperfusion, but they were lower in I/R-NAC group and no alteration almost in group P. The plasma concentrations showed a significant correlation between ALT, AST and LPS at different time points in I/R group and I/R-NAC group.Conclusion LPS and TLR4 play a prominent role in the pathophysiological processes of I/R and NAC can be of therapeutic potential for reducing the expression of TLR4 mRNA and alleviating endotoxemia derived from gut.
3.The effects of autologous blood transfusion and acute normovolemic hemodilution inflammatory response in patients with spinal surgery

Objective To investigate the effects of autologous blood transfusion and acute normovolemic hemodilution inflamma‐tory response in patients with spinal surgery .Methods 60 patients with spinal surgery ,were randomly divided into the control group ,autologous blood transfusion group ,acute normovolemic hemodilution group ,20 cases in each group .The control group in in‐traoperative bleeding time and blood input variants of inventory .Autologous blood transfusion group was used in combination with intraoperative autologous blood recovery unit ,lose banked blood hemoglobin was low .Acute normovolemic hemodilution group via peripheral vein input must first crystal liquid or gel liquid ,then through internal jugular vein slowly pulled out the body′s blood into the special bags containing anticoagulant ,swing machine through blood kept shaking ,input when appropriate .Preoperative(T1) , After surgery 2 h(T2) ,6 h(T3) ,12 h(T4) and 24 h(T5) blood at each time point 5 mL ,detect the WBC ,IL‐6 ,TNF‐α.Records with and without postoperative complications .Results WBC in autologous blood transfusion group and acute normovolemic he‐modilution group were higher than that of preoperative ,difference was statistically significant(P<0 .05);In T2 -T5 ,serum IL‐6 , TNF‐a and the WBC concentration compared with the basis of their respective value(T1) increased significantly ,but were signifi‐cantly lower than control group ,the difference was statistically significant(P< 0 .05) .Postoperative follow‐up of the autologous blood transfusion group and acute normovolemic hemodilution group and no complications .Conclusion The autologous blood trans‐fusion group and acute normovolemic hemodilution group could effectively reduce the intraoperative and postoperative systemic in‐flammatory response ,which obviously save blood resources .
4.Effect of Total Flavonoids of Hedysarum Polybotry on Inducing Differentiation of Leukemic Cells

Objective:To investigate the effect and mechanism of total flavonoids of Hedysarum polybotry on induction of differentiation in human leukemia HL-60 cells. Method After the treatment of HL-60 cells with total flavonoids of Hedysarum polybotry, the cell differentiation was detected with NBT reduction method. Cell cycle, CD11b and C-fos were analysed by the flow cytometry. Result The positive rate of NBT reduction and the expression of CD11b were significantly increased. Similar, the expression of C-fos gene was upregulated. The growth of HL-60 cells was arrested at G0/G1 and G2/M phase. Conclusion Total flavonoids of Hedysarum polybotry could induce differentiation of HL-60 cells. Its molecular mechanism might be related to the modulation of gene expressions associated with the proliferation and differentiation, which leads to the inhibition of DNA synthesis.
5.Lateral Lung Volume Reduction with Small Incision and Ligation for COPD:Report of 36 Cases

Objective To investigate the efficacy of lateral lung volume reduction with small incision and ligation for the treatment of serious chronic obstructive pulmonary disease(COPD).Methods From April 1994 to May 2006,36 patients suffering from serious COPD complicated with severe emphysema underwent lateral lung volume reduction in our hospital.Small incision and ligation were used during the operation.Results After the surgery,6 patients developed pulmonary air-leak,and were cured after 180-minute to 5-day treatments(mean,1.6 days).No perioperative death in this series.One year after the operation,the patient had significantly higher FEV1 and PaO2,and lower TLC and RV than those determined preoperation [(839.9?76.0)ml vs(589.0 ?80.5)ml,t=18.451;(72?10)mm Hg vs(62?8)mm Hg,t=4.686;(5167.6?639.9)ml vs(6725.8?976.6)ml,t=8.116;(3062.4?722.0)ml vs(4329.6?669.9)ml,t=7.720;all P=0.000].In addition,they had significantly longer 6MWT than that before the operation [(306.0?24.8)m vs(223.8?36.7)m,t=11.135,P=0.000].Conclusion For the patients with COPD who had indications for lateral lung volume deduction,resection of emphysemic tissues by the procedure using small incision and ligation can improve their symptoms and physiological conditions.
8.Social support status and influencing factors among schizophrenics in remission in Northeast Sichuan

ObjectiveTo investigate the social support status and influencing factors of schizophrenics in remission in Northeast Sichuan, and to provide ideas for improving their social support. MethodsFrom May to September 2020, a total of 533 patients who met the diagnosis criteria of the International Classification of Diseases, tenth edition (ICD-10) for schizophrenics in remission at the mental health institutions in Guangyuan, Bazhong and Dazhou cities were selected for the survey, and patients were assessed by self-made demographic and clinical data inventory and Social Support Rating Scale (SSRS). Then the social support status of schizophrenic in remission and influencing factors were analyzed, meantime, the impact of the second round reimbursement policy of medical insurance benefits on their social support was addressed particularly. Results①The SSRS total score, objective support, subjective support, and utilization of support scores of schizophrenics in remission were lower than those of the national norm (t=5.065~30.382, P<0.01). ②Univariate analysis showed that SSRS score was relatively high among patients with female gender (t=-3.632), retired status (F=5.951), married status (F=5.951), spouse as primary caregiver (F=23.841), annual household income >5 000 yuan (F=15.892), patient's economic income (t=4.083), and outpatient or online follow-up (F=3.954), with statistically significant differences (P<0.05 or 0.01). ③The total and dimensional scores of SSRS in patients with access to the second round medical insurance reimbursement were significantly higher than those without (t=10.195~25.103, P<0.01). ④Multiple linear regression analysis denoted that gender, work status, marital status, primary caregivers, annual family income, economic income, follow-up visits and the second round medical insurance reimbursement were the factors influencing social support status of schizophrenics in remission (β=0.201~2.115, P<0.05 or 0.01). ConclusionThe social support of schizophrenics in remission in Northeast Sichuan is below the national average, furthermore, their social support levels are affected by the gender, work status, marital status, primary caregivers, annual family income, economic income, follow-up visits and the second round medical insurance reimbursement, and the second round medical insurance reimbursement may ameliorate the social support status of patients.
9.Epidemiology and pathogen composition of enterovirus in children with hand, foot and mouth disease in Guangyuan, Sichuan

Objective To investigate the epidemiology and pathogenic characteristics of enterovirus in children with hand, foot and mouth disease (HFMD) in Guangyuan area, and to provide a basis for the development of HFMD prevention and control strategies. Methods A total of 759 children with HFMD clinically diagnosed in Guangyuan area from January 2019 to January 2021 were randomly selected. The seasonal, age, gender, regional distribution and pathogen distribution characteristics of HFMD were analyzed by descriptive epidemiological methods. Results The incidence of HFMD was seasonal and presented a typical bimodal distribution, with the summer peak occurring from April to July (χ2=8.714, P<0.05) and the winter peak from October to December (χ2=7.542, P<0.05). There was a significant difference in the incidence rate between regions, and the incidence rate in urban areas was much higher than that in rural areas (χ2=5.915, P<0.05). P < 0.05). The male-female incidence ratio was 1.63:1. The high incidence group of HFMD was children younger than 5 years old, and the number of reported cases was 730. The high incidence age group was 1-3 years old (χ2=8.455, P<0.05). The incidence rate of HFMD in all age groups ≤5 years old was higher in male than female, and the difference was statistically significant (χ2=5.174, P<0.05). A total of 388 HFMD cases were tested positive for viral nucleic acid, with a positive rate of 51.12%, In mild cases, Cox A16, EV71, and other enteroviruses accounted for 27.64%, 29.54% and 29.54%, respectively. The proportion of EV71 in severe cases was 89.47%. Conclusion HFMD is widely distributed in Guangyuan area, with obvious seasonal and population differences. The prevention and control of HFMD should be strengthened in key areas, high incidence seasons and key populations, and the vaccination of children ≤3 years old should be further promoted to prevent the outbreak of HFMD.
10.DETERMINATION OF SERUM FREE AMINO ACIDS IN NORMAL ADULTS——THE COMPARISON OF THE METHODS FOR PHYSIOLOGICAL FLUID AND PROTEIN HYDROLYZATE ANALYSIS

The free amino acid concentrations in serum were determined in 65 normal adults by use of the physiological fluid analysis in Hitachi 835-50 Model Amino Acid Autoanalyer. 29 specimens were determined by physiological fluid analysis and protein hydrolyzate analysis method simultaneous-ly.The results of Serine, Glycine, Valine, Isoleucine, Leucine, Tyrosine, Phenylalanine, Lysine, Tryptophan, Proline, and the molar ratio of the bran-ched-chain amino acids/aromatic amino acids were very similar. By hy-drolyzate method Aspartic acid was not detected and Threonine, Glutmic acid, Cystine, Methionine and Arginine were higher than physiological fluid analysis.The reasons for such higher values were discussed.
