1. Mechanism of PI3K/AKT signaling pathway regulating osteonecrosis of the femoral head

BACKGROUND: In recent years, with the deepening of medical research, it has been found that PI3K/AKT signaling pathway has a regulatory effect on vascular repair regeneration, osteoblast differentiation and proliferation, and osteoclast bone differentiation. This is very important for the treatment of femoral head necrosis. OBJECTIVE: To provide a brief overview of the main research progress in mechanisms of PI3K/AKT signaling pathway regulating femoral head necrosis in recent years, aiming to provide new ideas for the treatment of osteonecrosis of the femoral head in the future. METHODS: PubMed and MEDLINE database, Wanfang, CNKI, WIPO and CBM database were searched from 2012 to 2019, for relevant domestic and foreign literatures, including: (1) epidemiological study of osteonecrosis of the femoral head and related pathogenesis; (2) PI3K/AKT pathway related mechanism; (3) research literature on the effects of PI3K/AKT on factors related to vascular repair and regeneration; (4) research literature on the regulation of PI3K/AKT on osteoblast differentiation and proliferation-related factors; (5) literature on the regulation of PI3K/AKT on functional factors related to osteoclast differentiation. A total of 62 literatures were included for analysis and summary. RESULTS AND CONCLUSION: (1) The PI3K/AKT signaling pathway has been proven to be effective in the regulation of vascular repair regeneration, osteoblast differentiation, proliferation, apoptosis and osteoclast differentiation. After understanding these pathways, research and development of related drugs to improve the success rate of early conservative treatment of osteonecrosis of the femoral head have great development prospects and potential, opening up a new path for future orthopedic surgeons to treat osteonecrosis of the femoral head, and bringing new hope to patients and their families. (2) According to the different osteonecrosis of the femoral head conditions of patients, how to use PI3K/AKT signaling pathway to guide treatment becomes a breakthrough point and challenge of this technology, and more research is needed later.
2.Studies on the quality standard of xingnao tinctures.

OBJECTIVETo establish the method for quality control of Xingnao tinctures.
METHODThe Mentholum, Flos Caryophylli and Borneoum Syntheticum in this medicine were identified by TLC method. The content of Mentholum was determined by GC. Conditions of GC were: With Naphthalene as an internal standard, packed glass column, Shimadzu 7G 2.1 mm x 3.2 m, carrier gas N2, flow rate 50 mL.min-1, column temperature. 145 degrees C, ingector and detector temperature. 180 degrees C, injection way of splitless and injection volume 3 microL.
RESULTThe spots on TLC plates were clear, with no interference in the blank reference. The precision and repeatability of GC were are good respectively. The average recovery was 98.4%, RSD = 0.94%.
CONCLUSIONThis method was fast, simple, accurate and suitable for quality control of Xingnao tinctures.

3. Association of workplace loneliness, job burnout and conscientiousness in teachers of special education in Guangxi

OBJECTIVE: To explore the association of workplace loneliness, job burnout and conscientiousness in teachers of special education. METHODS: A total of 344 teachers of special education from Guangxi Zhuang Autonomous Region were selected as study subjects by judgment sampling method. The questionnaires of Workplace Loneliness Scale, Maslach Burnout Inventory and Conscientiousness Scale of Big Five Inventory were used to conduct the survey. The mediating role of job burnout in workplace loneliness and conscientiousness was analyzed. RESULTS: The total score of workplace loneliness was 32.6±8.3. The median total score of job burnout and conscientiousness was 31.0 and 30.6 respectively. Partial correlation analysis showed that the dimensions of job burnout were positively correlated with the total score of workplace loneliness and all its dimension scores, and negatively correlated with conscientiousness score(P<0.01). The degree of explanation for the change of workplace loneliness and job burnout on conscientiousness were 9.0% and 17.0% respectively. CONCLUSION: Job burnout plays a partial mediating role in workplace loneliness and conscientiousness in teachers of special education. Reducing workplace loneliness and job burnout can help improve the conscientiousness of teachers of special education.
4. Chemical constituents of kumquat essential oil analyzed by GC-MS and their acute toxicity in mice

Objective To analyze the chemical constituents of kumquat essential oil(KEO)and the acute toxicity of KEO in mice. Methods The chemical constituents of KEO were analyzed by GC-MS. After preliminary test, KM mice were randomly divided into control and KEO groups with 20 mice in each group(10 male and 10 female). The doses of KEO were designed with ratio 0.758 between the 100% and 0% mortality amount as 10, 7.60, 5.70, 4.40, 3.30 and 2.50 ml/kg. The acute toxicity was evaluated by examination of the mice for 14 days after single administration of KEO. Results The principal constituents in KEO were α-pinene(0.46%), β-phellandrene(0.11%), β-myrcene(2.34%), D-limonene(95.62%), geranyl acetate(0.10%), β-copaene(0.67%), and γ-elemene (0.13%), which accounted for 99.43% of the total chemical content of KEO. The half-lethal dose(LD50)of KEO in mice by the acute toxicity test was 6.24 ml/kg with the 95% confidence limit of(5.39-7.33)ml/kg in female mice and 5.73 ml/kg with the 95% confidence limit of(4.91-6.79)ml/kg in male mice. The possible toxic target organs were liver, spleen and gastrointestinal tract. Conclusion KEO with the content of D-limonene was obtained from kumquat for the first time, and the toxicity of the KEO was low according to the toxicity classification standard of World Health Organization.
5.CD177+ neutrophils and its correlation with mucosal impairment in patients with inflammatory bowel disease

Objective To investigate the percentage of CD177+ neutrophils in peripheral blood and inflamed mucosa of the intestine in patients with inflammatory bowel disease(IBD)and in healthy controls, and to investigate the correlation between CD177+ neutrophils and mucosal impairment as well as clinical characteristics. Methods A total of 54 patients with ulcerative colitis(UC),62 patients with Crohn disease (CD),and 48 healthy controls were enrolled. SYBR-green Realtime PCR was applied to determine the ex-pression proportion of CD177+ neutrophils in peripheral blood and in inflamed intestinal mucosa to investigate the correlation between CD177+ neutrophils and endoscopic and clinical disease activity. Results In periph-eral blood and inflamed mucosa of the intestine,the expression of CD177+ neutrophils was significantly higher than that in health controls or patients in remission stage(P< 0. 05),with higher proportion( P<0. 05). The expression of CD177+ neutrophils in peripheral blood and inflamed mucosa of the intestine was positively correlated with both endoscopic disease activity index and clinical severity of the disease( P <0. 05). Conclusion CD177+ neutrophils increase in peripheral blood and intestinal mucosa of patients with IBD. The percentage of CD177+ neutrophils is a favorable marker for both endoscopic disease activity and clinical severity of the disease. CD177+ neutrophils may play a vital role in the immune response in the intes-tinal mucosa,as a new subset of effector neutrophils.
6.Removal of Shield Needles from Graphene Oxide Thrombus and Preparation.

Atherosclerosis is a chronic inflammatory disease commonly seen in clinical practice. It can lead to thickening of vascular intima, occlusion of lumen stenosis and thrombosis, leading to angina pectoris, hypertension, myocardial infarction and other diseases, posing a serious threat to human life and health. This study provides a method for removing shield needles from graphene oxide thrombus and its preparation. The graphene oxide shield needle mainly includes flexible rotating shaft, radial flexible rod, rotating needle, adsorption main pipe and dosing main pipe, laser measuring device, high definition camera and other structures, which has the following advantages:firstly, it achieves multi-angle rotation grinding thrombosis, precise rotation grinding, avoids vascular damage and infection; secondly, thrombolytic drugs can be applied in the process of rotary grinding and small thrombus can be adsorbed to effectively avoid secondary embolization of blood vessels; thirdly, it a coating of graphene oxide on a rotating needle, which protects against bacteria and infection. This study has practical reference value for the development of thrombotherapy and the application of graphene in the medical field.

7.Suicide and self-injury among college students of Yao nationality in Guangxi and its relationship with psychological adjustment ability

Objective:
To understand the status of suicide and self-injury among college students of Yao nationality in Guangxi and their relationship with psychological adjustment ability, and to support the prevention and intervention of suicide and self-injury behavior of Yao university students.
Methods:
From April to May 2019,a questionnaire survey was conducted among 844 Yao university students in three universities in Guangxi in demographics, psychological resilience, and forgiveness.
Results:
Among the students of the Yao ethnic group in Guangxi, 10.90% had suicidal ideation; 4.62% had suicide plan; 3.55% had suicide preparation; 2.84% had suicidal action; and 12.56% had suicidal behavior. The total scores of students with suicidal ideation in Psychological Resilience, Self-Compassion and Hearland Forgiveness Scale were significantly lower than those without suicidal ideation, which was statistically significant(t=-3.74, -5.54, -4.97,P<0.01). Multivariate Logistic regression analysis showed that a high score for forgiveness was negatively correlated with suicidal behavior(OR=0.28) and self-harm behavior (OR=0.32)(P<0.05).
Conclusion
The psychological resilience, forgiveness, and self-compassion of Yao college students are closely related to suicide-related psychological behaviors and self-harm behaviors. Suicide and self-harm behaviors can be reduced by improving the self-psychological adjustment ability of Yao college students.
8.Effect of low expression of senescence marker protein 30 on proliferation and oxidation of human lens epithelial cells line SRA01/04 under high calcium conditions

AIM: To explore the effect of low expression of senescence marker protein 30(SMP30)on proliferation and oxidative stress of human lens epithelial cells(LECs)line SRA01/04 under high calcium conditions.
METHODS: Three RNAi sequences were designed to knock down SMP30 target gene RGN expression(KD1-3), and the blank-load sequence was used as the negative control group(NCKD), all of which were used to construct lentiviral vectors to infect SRA01/04 cells. Meanwhile, the uninfected SRA01/04 cells was used as the blank control group(CON). After transfecting SRA01/04 cells, the lentiviral vector with the highest knockdown efficiency was selected by RT-PCR for subsequent experiments. Cells were treated with 15mmol/L CaCl2 for 24h to simulate a high calcium conditions. BrdU-Elisa assay was used to measure cell proliferation, superoxide dismutase(SOD)assay kit and oxidized glutathione/total glutathione(GSSG/T-GSH)assay kit were used to detect the level of intracellular oxidative stress.
RESULTS: KD1-3 and NCKD lentiviral vectors were successfully constructed to infect SRA01/04 cells with an infection efficiency of about 80%. The knockdown efficiency of KD1-3 group was 93%, 60% and 74%, respectively, KD1 group was selected for follow-up experiment. Under the high calcium conditions, the activity of relative cell proliferation and SOD in KD1 group \〖(2.42±0.08)and(11.69±0.52U/mg)\〗 were significantly lower than that in NCKD group \〖(2.95±0.08)and(31.10±2.24U/mg)\〗 and CON group \〖(2.96±0.25)and(26.33±1.04U/mg)\〗, the ratio of GSSG/T-GSH in KD1 group(70.80±2.34)was significantly higher than that in NCKD group(15.93±3.47)and CON group(20.05±2.45)(P<0.05); there was no significant difference between NCKD group and CON group(P>0.05).
CONCLUSION: Under high calcium conditions, SRA01/04 cells(HLECs)with low expression of SMP30 mediated by shRNA lentivirus resulted in the decrease of the proliferation activity and antioxidant capacity, suggesting that SMP30 may play a protective role in regulating cell proliferation and anti-oxidative stress in HLECs.
9.Quality of life and its influencing factors of adolescents in Nanning low-income families

Objective:
To understand the quality of life and its influencing factors of adolescent from low-income families in Nanning City.
Methods:
With the help of the community, junior and senior high school students from 1 144 low-income households and 1 067 non-minimum households in 4 districts of Nanning City were randomly selected to complete a questionnaire survey regarding quality of life and study stress source.
Results:
The quality of life and total scores of adolescents in the low-income families were significantly lower than the national norm(P<0.01), and also lower than the scores of adolescents from normal in come families. The scores of boys in low-income households were significantly higher than those of girls in terms of exercise quality, life chance, negative emotions, and other factors (P<0.05). Girls scored significantly higher in teacher-student relationships, parent-child relationships, and self-concepts than boys (P<0.05). Junior middle school students were significantly higher than high school students in terms of convenience, athletic ability, teacher-student relationship, physical feelings, work attitude, life opportunity, negative emotions, peer relationships, self-satisfaction, other and total scores.Differences were of statistical significance (P<0.05). Regression analysis showed that there are 9 factors that affect social and psychological functions; 8 factors that affect living environment; 5 factors that affect physiological and mental health; 5 factors that affect satisfaction with quality of life; and 4 factors that affect total quality of life(P<0.05).
Conclusion
The quality of life of middle school students from low-income families is low, and effective measures should be developed and implemented accordingly.
10.Correlation of high-risk HPV 16/18 infections with prostate cancer.

ObjectiveTo study the correlation of high-risk human papillomavirus 16 and 18 (HPV16/18) infections with the risk of prostate cancer (PCa) and their association with the clinicopathologic indexes of PCa.
METHODSWe collected tissue samples from 75 cases of PCa and 73 cases of benign prostatic hyperplasia (BPH). We detected HPV16/18 infections in the samples by immunohistochemistry and PCR combined with reverse dot blot (RDB) assay.
RESULTSImmunohistochemistry revealed 16 cases of HPV16/18 positive in the PCa (21.3%) and 7 cases in the BPH samples (9.5%), with statistically significant difference between the two groups (P=0.049). PCR combined with RDB assay showed 17 cases of HPV16 infection (22.6%) and 13 cases of HPV18 infection (17.8%), including 4 cases of HPV16/18 positive, in the PCa group, remarkably higher than 6 cases of HPV16 infection (8.2%), 3 cases of HPV18 infection (4.1%) and no HPV16/18 positive in the BPH controls (P=0.001). No significant differences were observed between the result of immunohistochemistry and that of PCR combined with RDB assay (P=0.069). The risk of HPV16/18 infections was found to be correlated with the clinical T-stage and Gleason score of PCa (P<0.05 ) but not with the patient's age, PSA level or lymph node metastasis (P>0.05 ).
CONCLUSIONSHigh-risk HPV16/18 infections are correlated with the risk of prostate cancer.
