1.Effects of maternal subclinical hypothyroidism diagnosed in the third trimester on pregnancy outcomes

Objective To investigate the incidence of subclinical hypothyroidism (SCH) during the third trimester of pregnancy and its effects on pregnancy outcomes and neonatal hypothyroidism.Methods A total of 10 695 women in the third trimester of pregnancy (28-42 weeks of gestation) who labored from January 1,to December 31,2012 in Changzhou Maternal and Child Health Care Hospital Affiliated to Nanjing Medical University were enrolled.The levels of thyroid stimulating hormone (TSH),free thyroxine (FT4) and thyroid peroxidase antibody (TPOAb) of the mothers were quantified by electrochemical immunoassay.The time-resolved fluorescence immunoassay was used to detect neonatal thyroid hormone levels.Using t test or x2test,the incidence of adverse pregnancy outcomes was compared between SCH group and euthyroid group and between SCH women with positive (n=40) or negative TPOAb (n=176).Results The incidence of SCH was 2.02% (216/10 695) and the positive rate of TPOAb in SCH women was 18.5% (40/216).No neonatal thyroid dysfunction was found.According to the age matched,222 cases were randomly selected as controls from 7 757 euthyroid women.Compared with the controls,SCH women had a higher incidence of premature rupture of membranes [28.7% (62/216) vs 14.9% (33/222),x2=12.34],anemia [11.6% (25/216) vs 4.1% (9/222),x2=8.65],pregnancy-induced hypertension [9.7% (21/216) vs 4.5% (10/222),x2=4.53],premature labor [8.8% (19/216) vs 3.6% (8/222),x2=5.10] and intrahepatic cholestasis of pregnancy [8.3%(18/216) vs 2.3% (5/222),x2=8.14] (all P<0.05).The overall incidence of adverse pregnancy outcomes was also higher in SCH group than in the controls [69.4% (150/216) vs 49.5% (110/222),x2=17.96,P<0.01].The incidence of fetal growth restriction and still birth in SCH mothers with positive TPOAb was higher than in those with negative TPOAb [7.5% (3/40) vs 0.0% (0/176),x2=13.32,P<0.01; 2.5% (1/40) vs 0.0% (0/176),x2=4.40,P<0.05],but there was no significant difference in the overall incidence of adverse pregnancy outcomes compared with TPOAb-negative mothers [65.0% (26/40) vs 70.5% (124/176),x2=0.46,P=0.50].Conclusions SCH diagnosed in the third trimester may lead to adverse pregnancy outcomes.Early screening for thyroid dysfunction is necessary.
2.Metabolic study of ginsenoside Re in rats.

OBJECTIVETo systemically study the metabolism of ginsenoside Re in rats.
METHODSix SD rats were used divided into 3 groups. After oral administration of ginsenoside Re at a dosage of 100 mg x kg(-1), feces were collected at 12, 24, 36, 48, and 60 h. The microbial transformation metabolites of ginsenoside Re were used as standard references. HPLC-ESI-MS/MS analysis was used in the metabolism studies.
RESULTSix metabolites of ginsenoside Re were detected in rat feces by HPLC-ESI-MS/MS analysis. Their structures were identified as 20(S)-ginsenoside Rg2, 20(S)-ginsenoside Rh1, 20(R)-ginsenoside Rh1, ginsenoside F1, 3-oxo-ginsenoside Rh1 and protopanaxatriol.
CONCLUSIONGinsenoside Re has the same metabolic pathway with microorganisms, to some extent, justified the use of microbial models for mammalian metabolism studies.

3.Experimental Study on Stability of Pelvic Ring Reconstruction Using Fibular Autograft for Periacetabular Tumor Type Ⅱ Resection

Objective To evaluate the stability of the pelvic ring reconstruction using fibular autograft for periacetabular tumor type Ⅱ resection. Methods 6 adult cadaveric specimens were tested. The periacetabular tumor resection models were established according to Ennecking's type Ⅱ resection. The resected pelvic rings were reconstructed with double-fibular graft fixed by four internal fixation techniques including plates, pedicle-rods (PR), lateral-rods (LR) or sacral-iliac rods (SIR). Axial loading from the proximal L3 vertebral body was applied by MTS load cell in the gradient of 0~500 N in the double feet standing state. Images in front view were obtained using CCD camera. Based on Image J software, displacement of the first sacral vertebrae (S1) of the reconstructed pelvis and intact pelvis were calculated using digital maker tracing method with center-of-mass algorithm. Results The rotational movements and vertical displacement of S1 around the normal side femoral head of the reconstructed pelvis in coronary plane were found in simulated bilateral leg standing position. The average vertical load-displacement and load-angular rotation curve of S1 in coronary plane were approximately linear behavior under the vertical load 500 N. The average vertical displacement and angle of S1 in coronary plane had not overacted. The stability of axial direction and rotation had not changed significantly when reconstructed by LR or Plates compared with the intact pelvis, but the SIR did. Conclusion Plates and LR fixation were more stabile for periacetabular tumor type Ⅱ resection.
4.Low-field MRI: a quick scan based on the large field of view.

To optimize the configuration of the conventional SE and FSE sequence parameters in low field MRI. With the combination of larger vision, relatively thinner slice thickness, small number of excitation (NEX = 1 or 2), the signal-to-noise ration and the spatial resolution on follow-up images are moderate, the scan time is significantly shorter along with fewer motion artifacts, the check success rate is improved.

5.Distribution study of two constituents in rat after oral administration of Smilax china extract.

OBJECTIVETo investigate the distribution in rat after oral administration of Smilax china extract.
METHODOxyresveratrol and resveratrol were quantitatively determined by HPLC/DAD method in rat tissues including heart, liver, spleen, lung and kidney after oral administration of S. china extract. The separation was performed on a Zorbax SB-C18 column (4.6 mm x 250 mm, 5 microm) with acetonitrile-water using linear gradient elution and UV detection at wavelength of 320 nm. The flow rate was 1.0 mL x min(-1) and the column was kept at 40 degrees C.
RESULTThe HPLC/DAD method for the simultaneous determination of oxyresveratrol and resveratrol in rat tissues was developed.
CONCLUSIONThe method has been successfully applied for the distribution study of two active constituents in rat through oral administration of S. china extract.

6.Preclinical pharmacodynamic research of Juhuang Qingre San

Objective To investigate the efficacy on antibacterial , antipyretic, analgesic, anti-inflammatory and immune reg-ulation of Juhuang Qingre San .Methods Efficacy on antibacterial was evaluated by the method of doubling dilution ,vivo antibacterial effect was observed by inoculating BABL/C mice with Streptococcus pneumonia ,efficacy on antipyretic was investigated by using endo-toxin fever model ,analgesic function was assessed using mice hot plate and torsion analgesic method ,effect of anti inflammatory was as-sessed with rat paw edema model and mice Peritoneal capillary permeability model , the role of regulating immune system was evaluated by mice carbon clearance test .Results Juhuang Qingre San can decrease the death rate of mice in antibacterial test in vitro or vivo,in low and mid dose group of Juhuang Qingre San , heating temperature of rabbits decreased significantly 2-3 h after administration ( P<0.05), and high dosage group decreased significantly 2-4 h after administration (P<0.05),the mice pain threshold increased signif-icantly (P<0.05) after the administration of 1.5, 2, 3 h in every group of Juhuang Qingre San was , and can significantly reduce the number of mice writhing after intraperitoneal injection (P<0.05),rat paw swelling rate decreased significantly 1, 2, 3, 4, 5 h after administration in each dosage group of Juhuang Qingre San (P<0.05), peritoneal capillary permeability decreased in the acute in-flammation model of mice (P<0.05), in carbon particle clearance test of immune regulation , K value of high and medium dose group of Juhuang Qingre San was increased , while the value of alpha increased as well ( P<0 .05 ) .Conclusion The results suggested that Juhuang Qingre San has efficacy of antibacterial , antipyretic, analgesic, anti-inflammatory and immune enhancing .
