1.Effect and mechanism of miRNA-4465 overexpression in the migration and invasion of breast cancer MDA-MB-231 cells

Objective To investigate the effect of overexpression of miRNA-4465 on invasion and metastasis of breast cancer MDA-MB-231 cells and its mechanism.Methods MiRNA-4465 minics was transfected into the MDA-MB-231 cells,and real-time PCR was used to detect the expression of miRNA-4465.Transwell assay was used to investigate the migration and invasion capability of cells after being transfected.Bioinformatics analysis were performed to predict the potential targets of miRNA-4465,and finally,luciferase reporter plasmids assay and west-ern blot was used to confirm the potential target of miRNA-4465.Results Comparing with negative control group, the expression of miRNA-4465 in miRNA-4465 minics group was increased significantly.Transwell migration assay showed that migration capability of cells in miRNA-4465 minics group was decreased(P =0.001);transwell inva-sion assay showed that invasion capability of cells in miRNA-4465 minics group was decreased(P =0.010).Lucif-erase reporter plasmids assay showed that comparing with the negative control +psiCHECK2-EZH2 3’UTR(wild type),the fluorescence activity of miRNA-4465 minics +psiCHECK2-EZH2 3’UTR(wild type)was decreased by 66%(P =0.001).Moreover,comparing with the negative control group,the expression of EZH2 protein was de-creased after transfecting miRNA-4465 minics into MDA-MB-231 cells.Conclusion MiRNA-4465 can suppress the invasion and metastasis of breast cancer MDA-MB-231 cells,which may be related to the regulation of EZH2.

Objective:To investigate the effect of cPLA 2(cytosolic PLA 2, 85-kD PLA 2) on the signaling pathways of IL-1?、IL-6 release in LPS induced Hela cells.Methods:①LPS stimulated Hela cells were exposed to the complex of cPLA 2 initiation site-directed antisense oligonucleotide and liposome in the indicated doses and time, and then,analyzed the changes of cPLA 2 both in mRNA and in protein by RT-PCR and Western blot.②At the same time, the supernatants were collected to detect the level of IL-1? and IL-6 under radioimmunoassay(RIA).Results:①The transfection led to a dose-dependent decrease in cPLA 2 protein, while the change of mRNA is not notable.②The levels of IL-1?、IL-6 in the transfected cells were reduced following the repression of cPLA 2 compared with that of LPS added only.Conclusion:cPLA 2 probably plays an important role in the signaling mechanisms on IL-1?、IL-6 release in LPS-induced Hela cell. 3.Measurement of Serum INF-2? in Patients with SARS and Its Clinical Significance

Objective To observe the level of serum IFN-2? in patients with severe acute respiratory syndrome(SARS). Methods The level of serum IFN-2? in 66 patients with SARS were measured by RIA, and healthy subjects served as control. Results The results showed that level of serum IFN-2? in patients with SARS in active period were significantly higher than that in control, and level of serum IFN-2? in severe type of patients with SARS either rose or fell. Conclusions The change of serum IFN-2? level indicated that cellular immunity participated in immunoreaction to SARS virus. The change of IFN-2? level in severe type of patients with SARS reflected the difference of immunoreaction in the patients.

5.Endovascular interventional embolization for the treatment of aneurysms located at the bifurcation of middle cerebral artery

Objective To evaluate the therapeutic efficacy of endovascular embolism in treating aneurysms located at the bifurcation of middle cerebral artery. Methods During the period from Jan. 2009 to May 2013 at authors’ hospital, a total of 21 patients with intracranial aneurysms located at the bifurcation of middle cerebral artery were treated with interventional embolization. Of the 21 cases , steel coils were used in 7, stent-assistant embolization was adopted in 12, and dual-microcatheter technique was employed in 2. The results were analyzed. Results Postoperative angiography showed that complete embolization of the aneurysm was achieved in 14 patients, residual neck was detected in 2 patients and subtotal obliteration of the aneurysm was seen in 5 patients. Procedure-related complications occurred in 2 patients, including cerebral bleeding during operation (n = 1) and postoperative small area cerebral infarction. Seventeen patients were followed up for 6 months, and follow-up angiography showed that no recurrence of aneurysm was seen. Conclusion For un-ruptured, or ruptured with no big hematoma, intracranial aneurysms located at the bifurcation of middle cerebral artery, endovascular interventional embolization is the treatment of choice.
6.Meta-analysis of short-term complications and outcome after endoscopic and microscopic pituitary adenoma surgery

Objective To evaluate the efficacy objectively and systematically between endoscopic and microscopic surgery for transsphenoidal pituitary adenoma by a Meta-analysis.Methods A systematic literature search of Medline,the Cochrane Library,the Web of Science and the Google Scholar were performed to recruit studies between January,1990 and May,2015 that compared endoscopic and microscopic approaches in the treatment of pituitary adenomas.A Meta-analysis(R-3.1.3) was conducted to estimate the effect of different operative procedures.Results A total of 22 studies including 3 randomized or quasi-randomized controlled trails and 19 case-control studies met the inclusion criteria.Among 1421 patients studied,733 of them were treated with endoscopic and 688 were treated with microscopic surgery.The rate of gross tumor removal was higher in patients who underwent endoscopic surgery (71%) than those who underwent microscopic surgery (56%)(P < 0.05).The rate of remission of hypersecretion was also higher in patients who underwent endoscopic surgery(60%) than those who underwent microscopic surgery(54%),but the difference was not significantly(P > 0.05).Conclusion The present study indicates that the endoscopic transsphenoidal approach is more effective than microscopic surgery in the treatment of pituitary adenomas.
7.Risk Assessment of Combined Detection of Lipoprotein Associated Phospholipase A2 and Cystatin C for Lupus Nephritis in Patients with Systemic Lupus Erythematosus

Objective To investigate the value of combinedevaluation of serum lipoprotein associated phospholipase A2 (LP-PLA2) and cystatin C (CysC) on systemic lupus erythematosus (SLE) patients with lupus erythematosus.Methods A total of 177 patients with SLE were enrolled in the General Hospital of People's Liberation Army from June 2000 to February 2017,87 cases in lupus group (SLE group),90 cases in lupus nephritis group (LN group),60 cases in healthy control group.The concentration of LP-PLA2 and CysC were measured.The ROC curves was established for estimation of the cutoff values of two indexes in SLE patients with LN,and the diagnostic efficacy of different diagnostic criteria was compared.Results The LP-PLA2 and CysC concentration in the LN group were higher than those in the SLE group (Z=-2.512,-5.688,all P<0.05).The cutoff value of LP-PLA2 was 245.5 U/L and the cutoff value of CysC was 1.235 mg/L on the risk assessment of in SLE patients with LN.When using the parallel method for combined LP-PLA2 and CysC to evaluate the risk of LN,the sensitivity and negative predictive value would be significantly improved (x2 =9.461,4.377,all P< 0.05),the efficiency was better.Conclusion Parallel assessment of serum LP-PLA2 and CysC have clinical value on assessment of the risk of LN in SLE patients.

9.A study of brain-computer interface paradigm based on mental arithmetic.

In the traditional P300 brain-computer interface (BCI) system, the electroencephalogram (EEG) signals can only provide limited information with a low signal-to-noise ratio. A BCI paradigm under visual stimulus was proposed in our study aiming to effectively activate the related brain areas and response signal while dealing with specific cognitive task (mental arithmetic task), so as to enhance the EEG signals. The result was compared with the traditional P300 counting task paradigm. Then the collected EEG data were preprocessed including extracting signal features with coherent averaging method, and analyzing the influences of different experimental paradigms on main components of event related potential (ERP). In the improved paradigm experiments the average increasing rate of P300 amplitude was 6. 83MV (73. 94%). The brain activity from 400ms was more active and lasted longer. Besides, unlike traditional counting task, mental arithmetic task appeared to have apparent activation at 650ms. The results showed that the improved paradigm could activate the related brain areas better and enhance the characteristics of signal. This provides a new system paradigm for BCI.

10.Risk factors for bone mineral density changes in patients with rheumatoid arthritis and fracture risk assessment

Objective:To verify the fracture risk assessment tool ( FRAX) to estimate the probability of osteoporotic fracture in patients with rheumatoid arthritis ( RA ) with or without bone mineral density (BMD), and identify associated risk factors of osteoporosis .Methods: In the study, 200 patients with rheumatoid arthritis aged more than 40 years in Peking University First Hospital from Dec .2009 to Dec. 2012 were recruited.Clinical information was obtained from a questionnaire of their case history and medical records.FRAX tool was administered.Their lumber spine and left femoral BMD were determined by dual energy X ray absorptiometry.The gender, age, disease duration, menopause status, body mass index ( BMI) and accumulative dose of glucocorticoid were obtained in retrospect .Correlation analysis was conducted between the BMD and clinical information .Results:The study population ( female, 77.5%) had a mean age of 59.4 years, in which 10 (13%) patients showed a normal BMD, 67 (87%) were osteopenia or osteoporosis , while 32 patients (16%) had fragile fracture.Compared with the patients with normal BMD, the subjects with low BMD had significantly older age , longer period for corticoids usage , higher day dose and accumulated dose of corticoids .The 10-year fracture risk of sustai-ning major osteoporotic fractures and hip fracture was higher .No significant difference was observed be-tween the 10-year fracture risks calculated with BMD and without BMD .The values of the different area under the receiver operating characteristic ( ROC) curve ( AUC) for major and hip fractures calculated in three ways:without BMD, with the femoral neck BMD, and with T-score.The best result was for FRAX tool for hip fracture with the T-score ( AUC 0 .899 ) .A stepwise multivariate linear regression model was constructed to explore the relationship between the different clinical factors studied and a low BMD . Three statistically significant variables for lumber BMD were pain on visual assessment scale ( VAS ) (P=0.02), fracture history (P=0.003) and a higher steroid accumulated dose (P=0.008).Three statistically significant variables for left hip BMD were age (P<0.001), fracture history (P=0.05) and lower BMI ( P=0.03) .Conclusion:Low BMD is a common complication in RA patients .Risk factors for major fracture and hip fracture are increased .There is a positive correlation between FRAX calculated with and without BMD or T score .FRAX with the femoral neck T score or BMD presents a discriminatory capacity better than FRAX without BMD , according to the AUC ROC .
