Objective To analyze the relationship between clinical pathology character and expression of E-cadherin，CerbB-2，p53，Ki-67 in breast cancer in order to evaluate its clinical significance.Methods To observe 93 cases of breast cancer.Immunohistochemical methods were used to detect the expression of E-cadherin，CerbB-2，p53，Ki-67.Results We found that the expressions of E-cadherin，CerbB-2，p53，Ki-67 in breast cancer samples were associated to histopathology grade and lymph node metastasis (p<0.05), but the expressions of CerbB-2，p53，Ki-67 were not associated to tumor size and histological type (p>0.05).Compared to other types, it showed low expression of E-cadherin in lobular carcinoma(p<0.05).Conclusion E-cadherin，CerbB-2，p53，Ki-67 can be regarded as molecular indexes of breast cancer, and they can be used to give diagnosis, choose treatment and forecast prognosis in clinical work.

In handling crises in hospitals caused by natural disasters like earthquake, systemati cpractical measures of crisis management are needed, which include: early contact with top managements for support in counter disaster supplies, immediate organization of temporary first aid stations by hospital staff, logistic support by full time personnel to solve problems such as drugs and medical equipments as well as food and drinking water, and psychological consultation to patients and staff members.

An ultra-high performance liquid chromatography-mass spectrometry ( UPLC-MS/MS) method was developed for the determination of 8 kinds of cephalosporins, cefoperazone, cefquinome, cefalonium, cefazolin, cefapirin, Ceftiofur, cefpirome and cefalexin, in edible parts of aquatic products. The samples were extracted with acetonitrile-water and cleaned up by multi-walled carbon nanotubes ( MwCNTs) SPE cartridge. All the target compounds were separated on an Acquity Xselect CSH C18 column with gradient elution by using acetonitrile and 0. 1% formic acid aqueous as eluent, and detected by UPLC-MS/MS under ESI+ ionization and MRM mode. Under optimized conditions, this method had a good linearity (R2≥0. 995) and the limits of quantification were in the range of 2-10 μg/kg ( S/N=10 ) . The recoveries of the method for the target compounds spiked at three different levels were 67. 3%-94. 2% with the relative standard deviations (RSDs) of 3. 3%-14%. The method had the characteristics of low cost, high accuracy and good precision, and could meet the requirements of cephalosporins determination.

Exopolysaccharide La0.1-1 was extracted from the broth of a marine bacterium Lentibacter algarum ZXM100T isolated from the seawater in the coastal region of Qingdao and purified by Q Sepharose Fast Flow ion-exchange chromatography and Superdex 75 gel-permeation chromatography. Its physiochemical properties and primary structural characters were investigated by chemical analysis together with high performance liquid chromatography (HPLC), high performance gel permeation chromatography (HPGPC) and gas chromatography and mass spectrometry (GC-MS). The results show that the total sugar content of the exoploysaccharide La0.1-1 was about 66% with an average molecular weight at 12.0 kDa. La0.1-1 is mainly composed of Gal, Man, GlcN at the ratio of 1.35:1.1:1.0. Results of GC-MS and NMR demonstrate that the exopolysaccharide La0.1-1 mainly exists with the beta configuration. The primary linkage styles are --> 2)-Manp(1 --> and --> 3)-Galp(1 --> with a small amount of --> 4)-Galp(--> 1 and --> 4)-Manp(1 --> linkages. The linkage mode of GlcN is --> 4)GlcN(1 --> and terminal linkage. The exopolysaccharide has mainly a linear sructure with a few branches linked to 0-6 of --> 2)-Manp(1 --> and 0-4 or 0-6 of --> 3)-Galp(1 -->. 1D-NMR data also revealed that La0.1-1 is substituted by certain acetyl; the acetyl is mainly linked to N-2 of GlcN. The exopolysaccharides of the bacterium of Lentibacter genus is reported for the first time, and an exopolysaccharide with novel structure was obtained, which enriched marine polysaccharide resources.

ObjectiveTo evaluate the feasibility of gadolinium-enhanced dual energy CT pulmonary angiography (CTPA) in detecting pulmonary embolism (PE).MethodsIn vitro dual energy CT of phantoms of gadolinium and iodinated contrast agents with different diluted ratio was performed,and CT values were measured at different tube voltages.Ten rabbits which were grouped into 3 ml/kg and 5 ml/kg groups underwent dual energy CT scan.CT values of pulmonary artery trunk and the first branch of pulmonary artery were measured.Sponge gelatin were injected into the femoral vein of 6 rabbits to make PE model next day,then lungs were re-imaged with dual energy CT 2 h after embolization.Creatinine was repeatedly measured before and one day after injection of gadolinium via ear marginal vein or femoral vein sampling.One-way ANOVA test and independent student t test were used to analyze the difference of pulmonary artery enhancement between different groups.Results ( 1 ) Compared with iodinated contrast agent,CT value of gadolinium-based contrast agent at 80 kV was higher than those at 140 kV and averageweighted 120 kV.(2) At 140,80,and average weighted 120 kV,CT values of pulmonary artery trunk [CT values were (463.1 ± 118.0),(664.2 ± 188.0),(522.9 ± 137.7) HU] and of the first branch of pulmonary artery [ CT values were (445.1 ± 82.3 ),(606.7 ± 207.2),(493.4 ± 117.3 ) HU ] were higher than those at 3 ml/kg [ CT value of pulmonary artery trunk was ( 258.1 ± 55.1 ),( 384.0 ± 92.3 ),(295.4 ± 73.6) HU,CT value of the first branch of pulmonary artery (245.0 ± 73.2 ),( 309.1 ± 94.2),(263.8 ±78.5) HU;all P ＜0.05].CT values of pulmonary artery trunk and the first branch of pulmonary artery at 80 kV were higher than those at 140 kV and average-weighted 120 kV ( pulmonary artery trunk:F =6.004,P =0.005 ; the first branch of pulmonary artery: F =4.374,P =0.018).In 6 rabbits,CTPA showed the enhancement cut-off of bilateral pulmonary arteries,gadolinium mapping showed decreased perfusion in the corresponding lung lobes,manifested as blue on color-coded map,while normal lung was color coded as red or yellow.Creatinine was higher by 6.7％ and 20.6％ for group 3 ml/kg and 5 ml/kg.ConclusionsWith similar X-ray attenuation characteristics as iodine,gadolinium-based contrast agent can be used to pulmonary contrast-enhanced dual energy CT imaging,simultaneously providing both CTPA and gadolinium maps to detect PE.

OBJECTIVETo study the effect of different seminal rhizomes on the growth, quality and quantity of Curcuma longa root.

METHODSingle factor randomized block design was applied, plant samples were collected and investigated periodically, and dry weight, production and the main active ingredient content were measured.

RESULTThe difference seminal rhizomes affected the growth, quality and quantity of C. longa root.

CONCLUSIONThe bigger and stronger rhizomes should be chosen as seeds.

BACKGROUND AND OBJECTIVEExcision repair cross-complementing 1 (Excision-Repair Cross-Complementing 1, ERCC1), an important member of the DNA repair gene family, plays a key role in nucleotide excision repair and apoptosis of tumor cells. Protein kinase C-alpha (Protein kinase C, PKCalpha), an isozyme in protein kinase C family, is an important signaling molecule in signal transduction pathways of tumors, which has been implicated in malignant transformation and proliferation. The aim of this study was to explore the clinical significance of ERCC1 and PKCalpha in non-small cell lung cancer (NSCLC).

METHODSThe expression of ERCC1 and PKCalpha were examined by immunohistochemistry (IHC) in the specimens of 51 cases of NSCLC patients tissue and 21 cases of paracancerous tissue. The relationship between detected data and patients' clinical parameters was analyzed by SPSS 13.0 software.

RESULTSThe positive expression rate of ERCC1 and PKCalpha in NSCLC tissues was significantly higher than paracancerous tissues (P < 0.05). Expression of ERCC1 was closely related to clinical stage and N stage. The positive rate of ERCC1 was higher in III+IV or N1+N2 stage patients compared with I+II or N0 stage (P = 0.011, P = 0.015). We also found that 5-year survival of negative group of ERCC1 was remarkably higher than that of positive group by chi2 test (P < 0.05). Expression of ERCC1 was positively correlative to PKCalpha by Spearman's correlation analysis (r = 0.425, P = 0.002) in NSCLC.

CONCLUSIONThe results suggest ERCC1 and PKCalpha might be correlated with the development of NSCLC. ERCC1 might be related to prognosis of NSCLC. There might be existed a mechanism of coordination or regulation between ERCC1 and PKCalpha.

Bladder cancer is the most common malignancy of the urinary system. Compound Kushen Injection (CKI) is a Chinese medicinal preparation that has been widely used in the treatment of various types of cancers in the past two decades. However, the pharmacological effect of CKI on bladder cancer is not still completely understood. In the current study, network pharmacology combined with bioinformatics was used to elucidate the therapeutic mechanism and potential targets of CKI in bladder cancer. The mechanism by which CKI was effective against bladder cancer was further verified in vitro using human bladder cancer cell line T24. Network pharmacology analysis identified 35 active compounds and 268 target genes of CKI. Bioinformatics data indicated 5500 differentially expressed genes associated with bladder cancer. Common genes of CKI and bladder cancer suggested that CKI exerted anti-bladder cancer effects by regulating genes such as MMP-9, JUN, EGFR, and ERK1. Functional enrichment analysis indicated that CKI exerted therapeutic effects on bladder cancer by regulating certain biological processes, including cell proliferation, cell migration, and cell apoptosis. In addition, Kyoto Encyclopedia of Genes and Genomes enrichment analysis implicated pathways related to cancer, bladder cancer, and the PI3K-Akt signaling pathway. Consistently, cell experiments indicated that CKI inhibited the proliferation and migration of T24 cells, and induced their apoptosis. Moreover, RT-qPCR and Western blot results demonstrated that CKI was likely to treat bladder cancer by down-regulating the gene and protein expression of MMP-9, JUN, EGFR, and ERK1. CKI inhibited the proliferation and migration, and induced the apoptosis of T24 bladder cancer cells through multiple biological pathways and targets. CKI also exhibited significant effects on the regulation of key genes and proteins associated with bladder cancer. Overall, our findings provide solid evidence and deepen current understanding of the therapeutic effects of CKI for bladder cancer, and further support its clinical use.

Three 2,3-diketoquinoxaline alkaloids were isolated from Heterosmilax yunnanensis Gagnep. Their structures were determined through 1D and 2D NMR, HR-ESI-MS, UV, and IR as 1-[5′-(3″-hydroxy-3″-methyl) glutaryl] ribityl-2,3-diketo-1,2,3,4-tetrahydro-6,7-dimethylquinoxaline (1), 1-[2′-(3″-hydroxy-3″-methyl) glutaryl]ribityl-2,3-diketo-1,2,3,4-tetrahydro-6,7-dimethylquinoxaline (2), and 1-ribityl-2,3-diketo-1,2,3,4-tetrahydro-6,7-dimethylquinoxaline (3). Compounds 1 and 2 are novel compounds, and 3 was isolated from H. yunnanensis for the first time. The hepatoprotective activity of these three compounds was evaluated, with compound 3 showing promising hepatoprotective activity.

In order to study the pesticide residues of the medicinal Crataegi Fructus,this study aims to establish an analysis method for pesticide residues( mainly containing insecticides and fungicides) suitable for the actual situation of medicinal Crataegi Fructus based on the survey of the pesticides of the Crataegi Fructus base,combined with the blind screening results of the LC-ESI-MS/MS pesticide screening platform established by the research team in the early stage. Then,the pesticide residues in medicinal Crataegi Fructus from Shandong,Hebei,Henan,Shanxi,and Liaoning( main cultivation areas) were analyzed. The samples were pretreated by the modified Qu ECh ERS method,i.e.,extracted with acetonitrile-water( 9 ∶1),purified by PSA,C_(18),GCB,silica gel. The detection of pesticides was performed by LC-MS/MS. The ion source was ESI with positive scanning mode,and the linearity of 11 kinds of pesticides in the range of 5-300 μg·kg~(-1) was acceptable( R~2>0. 996 9). All the recoveries of pesticides were within 70. 02%~(-1)12. 0% in the low,medium and high levels,with RSD≤17%. The results showed that the detection rate of carbendazim,chlorpyrifos and difenoconazole is 79%,82%,56%,respectively. Besides,the prohibition pesticide carbofuran were detected in some of the batches,indicating the security risk. This study provides methodological references and basic data for risk assessment of Crataegi Fructus and government regulation.
