1.Changes of Activity of Placental Villus Cytochrome C Oxidase in Pregnant Women with Preeclampsia

To explore the role of cytochrome C oxidase (CCO) in the pathogenesis of preeclampsiais, CCO activity was determined in 32 patients with preeclampsia and 26 normotension pregnancy women by the rate of cyanide-sensitive oxidation of reduced cytochrome c using ultraviolet spectrophotometry. The results showed that CCO activity was significantly lower in the preeclampsia group (0. 30±0. 39/min, n=32) than in the control group (0. 73±0. 54/min, n=26), P<0. 01. The occurrence of IUGR in the preeclampsia group was significantly higher than in the control group (P<0. 05). It was suggested that the decrease of activity of CCO might interfere with the function of electronic chains, result in the reduction of ATP production, leading to the mitochondria dysfunction and placental dysfunction in preeclampsia patients. Mitochondria dysfunction may be involved in the pathogenesis of preeclampsia.
2.Clinical Significance of Detection of Granulocyte Macrophage Colony Stimulating Factor,C Reaction Protein and Tumor Necrosis Factor-?lpha in Infant Cytomegalovirus Hepatitis

Objective To explore the role of granulocyte macrophage colonystimulating factor(GM-CSF),C reaction protein(CRP)and tumor necrosis factor-?lpha(TNF?)in infant cytomegalovirus hepatitis.Methods Cytomegalovirus DNA of 56 cases of children with cytomegalovirus hepatitis and 51 normal controls in blood were detected using FQ-PCR.The levels of GM-CSF and TNF?in serum were assayed with ELISA,and CRP was measured with immunoturbidmemetry assay.Results The levels of M-CSF,CRP and TNF? in serum of infant cytomegalovirus hepatitis were higher obviously than those in serum of the normal control group.Conclution M-CSF,CRP and TNF? participate in pathological immune response and anti-infective immunity of cytomegalovirus hepatitis.
3.The value of multi-slice computer tomography and postprocessing techniques in diagnosis of the intestinal obstruction

Objective To explore the diagnostic value of multi-slice computer tomography (MSCT) in cause and location of agnogenic intestinal obstruction. Methods With light speed 16 multiple slice spiral-CT, 68 patients clinically suggested intestinal obstruction were underwent plain scan and second phase enhanced scan in whole abdomen. The image data was post-processed through GEADW4. 4 station with multi-planar reconstruction (MPR) and maximum intensity projection (MIP). The CT image characters of agnogenic intestinal obstruction were analyzed by comparing CT image suggestions with operation findings. Results MSCT clearly indicate the location, cause and severity of intestinal obstruction, and the diagnose accordance rate with operation findings was 95.6 % (65/68). Conclusions MSCT is a quick, simple and effective means for diagnosis of the location,causes and severity of agnogenic intestinal obstruction. It could provide an important clue for the doctors when making clinical decisions.
4.Interleukin-1β induced transdifferentiation of renal tubular epithelial cells and its effect on the cytoskeleton

Objective:To investigate the effect of interleukin-1β (IL-1β) on epithelial-mesenchymal transition (EMT) and cytoskeleton rearrangement of renal tubular epithelial cells.Methods:Immortalized renal tubular epithelial cell line NRK52E was cultured in vitro with IL-1β (30 μg/L) for 3 days and 6 days,then the cell morphology was observed;The mRNA expressions of α-smooth muscle actin (α-SMA),cytoskeleton components β-actin and α-tubulin were semi-quantitative examined by RT-PCR.The protein expression of α-SMA and arrangements of β-actin and α-tubulin were assessed by immunofluorescent staining.Results:After induced by IL-1β for 3 days and 6 days in vitro,the mRNA and protein expression of α-SMA increased significantly compared with corresponding control cells (P<0.001),it prompted that NRK52E cells underwent EMT;At the same time,the cell morphology also changed,from a typical multilateral paving stone to fibroblast-like appearance,with multiple processes; Cytoskeletal protein β-actin mRNA expression was also slightly increased (P<0.05).The distributions and arrangements of β-actin protein were also changed,from cell membrane transferred to peri-nucleus and cytoplasm,moreover it formed fiber bundle-like structures.However,another cytoskeleton protein α-tubulin in IL-1β induced cells,neither it's mRNA expression nor it's distribution had significant differences compared with the control group.Conclusion:IL-1β can induce NRK52E cells undergoing EMT in vitro,cell morphology changes into fibroblast-like appearance with multiple processes,and also the cytoskeleton protein β-actin expression increases and rearrangement occurrs.However,there was no changes onα-tubulin.
5.The change of Caveolin-3 in the rabbit skeletal muscle when ischemia-reperfusion injury

Objective To observe the damage degree and expression pattern of Caveolin-3 mRNA by ischemia-reperfusion injury in rabbits of skeletal muscle cell at different phases.Methods In this study,from April 2013 to December 2013,30 lower limbs of 15 Chinese White Rabbits were used and divided into two groups:all the left lower limbs were experimental group,which were made as an experimental model of ischemia-reperfusion injury by occluding left common iliac artery using noninvasive vascular.All the right lower limbs without surgical treatment were the control group.Gastrocnemius samples were obtained at 4h and 8h after reperfusion and handled by HE staining and observed by optical microscopy.By Real-time PCR,Caveolin-3/GAPDH mRNA were detected.Results HE stain showed:in control group,there was no edema,degeneration and inflammatory cell infiltration; in experi-meatal group,muscle cell degeneration had occured at ischemic 5 h.The edema was aggravated,a large number vacuole were formed and inflammatory cell were infiltrated at 4 h reperfusion.Reperfusion injury at 8h significantly reduced compared to 4 h.The Caveolin-3/GAPDH mRNA expression levels by SPSS 19.0 showed:Control group:1.026 ± 0.065,1.004 ±0.037,1.022 ±0.051,experimental group:1.159 ±0.073,1.445 ±0.053,1.208 ±0.058 at ischemic 5 h,4 h and 8 h reperfusion,respectively.On-line analysis of variance cases of ischemic 5 h and 4 h reperfusion and 8 h reperfusion,the experimental group than the control group were increased,with statistical significance (P < 0.05).The experimental group of ischemic 5 h and 8 h reperfusion was no significant difference (P > 0.05).It showed Caveolin-3 mRNA expression levels in ischemia-reperfusion 8 h group returned to normal.There was significant statistical difference between the ischemic 5 h and 4 h reperfusion (P < 0.05).There was significant statistical difference between the 4 h reperfusion and 8 h reperfusion (P < 0.05).Conclusion The expression of Caveolin-3 in experimental group showed a trend of first increased and then decreased.The expression levels of Caveolin-3 mRNA in skeletal muscle cells after ischemia-reperfusion injury is consistent with the development and progression of muscle cell damage.The results indicate that Caveolin-3 may play a control role in the injury and recovery of skeletal muscle cell.
6.The clinical characteristics of postinfectious irritable bowel syndrome in Qingdao

Objective To investigate the clinical characteristics of postinfectious irritable bowel syndrome (PI-IBS) in Qingdao. Methods Two hundred and four PI-IBS and 2068 non-PI-IBS patients were investigated with questionnaire including general information, symptoms and quality of life scores with microecological study before and after therapy. Results (1) The morbidity rate of PI-IBS in female was 2. times of that in male, which was similar to that in non-PI-IBS. (2) Brainwork labors dominated in both PI-IBS and non-Pl-lBS patients. (3) As to the simultaneous presence of extra-gastrointestinal symptoms,there was no statistical difference between the rate of physical symptoms in PI-IBS and non-PI-IBS patients (X<'2>= 10. 5, P>0.05) ,but the rate of mental symptoms was higher in PI-IBS than in non-PI-IBS patients, and the difference was significant(X<'2>= 28.7, P<0.05). (4)The alteration of intestinal microflora rate in PI-IBS was obviously higher than that in non-PI-IBS patients. (5) The quality of life scores in PI-IBS was improved after treatment with Birid Triple Viable , and there was significant difference(t =3. 8, P<0.01),but there was no statistical difference in non-Pl-IBS (t = 1.5, P>0.05). Conclusion There was some difference in certain clinical characteristics between PI-IBS and non-PI-IBS patients in Qingdao.
7.Liver diseases in adult-onset Still's disease

Clinical data of 108 patients with adult-onset Still's disease(AOSD)were analyzed.The results showed that 56 patients(51.9%)developed liver damage and abnormal liver functioil(alanine aminortransferase and aspartate aminortransferase were significantly increased).Gender,age,time from disease onset to confirmed diagnosis,fever,rash,arthritis,pharyngeal pain,splenomegaly,lymphadenopathy,anemia,leukocvtosis,thrmbocytosis,and increased erythrocyte sedimentation rate,serum C-reactive protein,and serum iton protein showed no significant difierence between the two groups (P>0.05).Clinical symptoms of liver damage were atypical.Among the 56 patients with liver damage,8 were presented with gastrointestinal symptoms,and 24 showed enlarged liver iu B.ultrasound examination.Our results suggest the prevalence of liver damage in AOSD patients may be hiSh,which could attribute to the disease itself and the medication.The patients would expect good response to corticosteroid treatment and satisfactory outcomes.
8.Study on elasticity of ipsilateral external iliac artery with echo-tracking after gunshot wound in pig limbs

Objective To explore the value of echo-tracking technique in the detecting the ipsilateral external iliac artery elasticity after gunshot wound in pig limbs.Methods Two-dimensional ultrasound imagings and elasticity parameters of external iliac artery of fifteen pig limbs were obtained respectively,and the results were compared with pre-injury groups.The animals were sacrificed after ultrasonography.Pathological examinations of adjacent external iliac artery of injured pig limbs were analyzed.Results Two-dimensional ultrasound imagings of external iliac arteries had no significant changes post-injury.The changes of elasticity parameters were significiant differences in the injured group comparing with the pre-injured group (P <0.05),including the increased stiffness parameter(β),pressure-strain elasticity modulus (Ep)and the decreased arterial compliance(AC).Pathological result showed that the internal elastic lamina of artery detected were flat,endothelial cells came off discontinuously and structure of them were undefined.Conclusions Echo-tracking technique can find the elastic changes of adjacent artery indirect injured by gunshot wound sensitively and which can suggest the occurrence of vascular indirect injury.
10.Efficacy of exercise therapy in the treatment of anterior cruciate ligament reconstruction patients:a network meta-analysis

OBJECTIVE:Exercise intervention is considered the cornerstone of rehabilitation after anterior cruciate ligament reconstruction.However,no clear conclusion has been reached regarding which exercise therapy is more effective in improving knee muscle strength and function in patients after anterior cruciate ligament reconstruction.To this end,this study used a network meta-analysis method to compare the efficacy of exercise therapy after anterior cruciate ligament reconstruction,providing evidence-based medical basis for selecting the best exercise therapy. METHODS:A computer search was conducted in PubMed,Web of Science,Embase,The Cochrane Library,and EBSCO to collect randomized controlled trials addressing exercise therapy following anterior cruciate ligament reconstruction.The search time limit was from the establishment of the database to 2023-11-20.Outcome indicators included three continuous variables:quadriceps muscle strength,hamstring muscle strength,and knee joint function score.EndNote X9.1 was used for literature screening.The Cochrane risk of bias assessment tool was used to evaluate the quality of the included literature.The GRADE score was used to rate the strength of evidence for the results of the article.Network meta-analysis was performed using Stata 16.0. RESULTS:A total of 36 randomized controlled trials were included,involving 1 179 patients undergoing anterior cruciate ligament reconstruction.The overall quality of the included literature was moderate.Nine types of exercise therapies were involved:isokinetic training,cross training,eccentric training,aquatic rehabilitation,blood flow restriction training,motor control training,plyometric training,whole-body vibration training,and multimodal training;the control measure was conventional rehabilitation training.The results of the network meta-analysis showed that compared with conventional rehabilitation training,eccentric training[standardized mean difference(SMD)=2.08,95%confidence interval(CI):0.56 to 3.60,P=0.007)had the best improvement effect on quadriceps muscle strength in patients undergoing anterior cruciate ligament reconstruction,followed by multimodal training(SMD=1.69,95%CI:0.11 to 3.27,P=0.249)and whole body vibration training(SMD=0.81,95%CI:0.11 to 1.51,P=0.042).In terms of improving patients'hamstring muscle strength,multimodal training(SMD=2.08,95%CI:0.30 to 3.86,P=0.022)had the best effect,followed by plyometric training(SMD=1.51,95%CI:0.18 to 2.84,P=0.026)and isokinetic training(SMD=1.37,95%CI:0.06 to 2.67,P=0.039).Multimodal training(SMD=4.60,95%CI:2.40 to 6.80,P<0.001)was the most effective in improving knee joint function scores,followed by eccentric training(SMD=1.75,95%CI:0.24 to 3.25,P=0.023)and aquatic rehabilitation(SMD=1.65,95%CI:0.07 to 3.24,P=0.041). CONCLUSION:Evidence of low to moderate strength suggests that multimodal training may be the most effective exercise therapy in improving knee muscle strength and function in patients after anterior cruciate ligament reconstruction,followed by eccentric training,plyometric training,isokinetic training,whole body vibration training,and aquatic rehabilitation.More high-quality clinical randomized controlled trials are still needed in the future to verify the reliability of the conclusions.
