1.Conjugated agent insulin-antisense-c-myb-PS-ODN enhances the inhibitory effect on proliferation of rat aortic artery smooth muscle cells

AIM:Vascular smooth muscle cell (SMC) proliferation and migration from the arterial wall media into the intima are believed to play a critical role in the pathogenesis of restenosis. Several studies have demonstrated that phosphothioate (PS) oligodeoxynucleotides targeted against genes involved in SMC proliferation inhibits in vitro SMC proliferation and migration. However, the therapeutic effect of antisense ODN on the individual who receives the treatment of delivery of the agent depends on the efficacy of this agent in great degree. We investigated the inhibition effect of a novel agent, insulin-antisense-c-myb-PS-ODN on SMC proliferation in vitro. METHODS:The rat aortic artery SMCs were cultured in Dulbecco's modified Eagel's medium. The passage 8 to 13 were used as the experiment. Cell surface receptor binding assay was quantified through counting gamma particles emitted from 125 I labeled insulin. SMC rapid proliferation was brought by stimulation of high concentration of fetal bovine serum (FBS). The novel agent of insulin conjugated to the antisense-c-myb-PS-ODN was obtained via incubation of both in condition of certain reagents, pH, temperature, and ion concentration. The characterization and purification of the agent was performed through HPLC. Inhibition of SMC proliferation was reflected by incorporation rate of trillium labeled thymidine deoxyribonucleotide.RESULTS:The binding efficacy of insulin to the receptor was remarkably increased in SMC cultured in supplement of 20% FBS. The inhibition effect of conjugator insulin-c-myb-antisense-PS-ODN was stronger than that of the simple c-myb-antisense-PS-ODN. The inhibition rate of conjugator and simple form on SMC proliferation were 48.34% and 29.54%, respectively. CONCLUSION:The binding efficacy and specificity of c-myb-antisense-PS-ODN to SMC may be enhanced by the insulin receptor mediation through the insulin-insulin receptor interaction. The insulin-receptor targeted method may be a potential and specific therapeutic pathway for restenosis.

3.Pet ownership and lung function in children without asthma

Objective To investigate the association between pet ownership and lung function in children without asthma,and to provide scientific basis for the improvement of lung function in children. Methods Data was drawn from the cross-sectional study ,the clusters were randomly selected from 1 to 2 junior middle schools and primary schools in 24 administrative regions of seven cities in Liaoning Province. The ATS questionnaire survey was performed,and lung function including the forced expiratory volume in 1 second(FEV1),forced vital capacity (FVC),maximal mid-expiratory flow(MMEF)and peak expiratory flow(PEF)was measured by utilizing portable electronic spirometers. Results In this study ,about 6280 children without asthma aged 7 ~ 14 years were enrolled,49.47 % of the children was male. The average lung function of FVC,FEV1,PEF and MMEF was (2.63±0.75)L,(2.47 ± 0.70)L,(4.80 ± 1.42)L/s and(3.37±1.05)L/s,respectively. By adjusting confounding fac-tors,we found pet ownership in the first 2 years of life was significantly associated with the predicted lung function impairment of FVC<85%(aOR=1.30;95%CI:1.01~1.67);current pet ownership was significantly associated with the predicted lung function impairment of FVC < 85%(aOR = 1.32;95% CI:1.09 ~ 1.61),the predicted FEV1<85%(aOR=1.47;95%CI:1.19~1.83),the predicted PEF<75%(aOR=1.48;95%CI:1.16~1.88) and the predicted MMEF<75%(aOR=1.35;95%CI:1.09~1.66). The in utero exposure was not related to lung function impairment. Conclusion Pets ownership has damaging effects on lung function in children without asth-ma,and it reduces FVC,FEV1,PEF and MMEF in children.
4.Efficacy of freeze-dried M. vaccae vaccine in treatment of patients with multi-drug resistant pulmonary tuberculosis

AIM: To explore the immunotheraptical drugs for multi-drug resistant pulmonary tuberculosis (MDR-TB). METHODS: 79 patients with MDR-TB were randomly assigned to two groups. 46 cases in M.vaccae group were treated with "3AkPaThLevL/15PaThLevL" and M. vaccae, and 33 cases in control group were treated only with "3AkPaThLevL/15PaThLevL". The clinical effect and T-lymphocyte subsets in patients were observed after being treated for 1,2 and 3 months, respectively. Objective To investigate the clinical significance and changes of neuron-specific enolase (NSE) and S100 protein in blood serum in patients with enterovirus 71 infection.Methods A total of 176 children with enterovirus 71 infection admitted from March 1,2012 through October 31,2012 were enrolled for a prospective and control study.According to diagnostic criteria of the enterovirus 71 (EV71) infection instituted by expert consensus for treatment of severe patients sets in 2011,the patients were divided into three groups:mild group (n =62),severe group (n =65) and critically care group (n =49),and another 30 healthy children served as control group.The demographics of patients including age and sex were comparable between control group and the sick children groups.Four milliliter of peripheral blood were taken from ill children on the first day before treatment and on the first,second,third day after treatment.The blood samples of healthy children were taken on the first day after physical examination.At the same time,the clinical data of blood routine,blood biochemistry,myocardial enzymes and C-reactive protein during the first 24 hours were collected.Immunohistochemical technique was used to study the change of NSE and S100 levels in serum.Data were expressed in mean ± standard deviation ((x) ± s) and were statistically analyzed by using SPSS 13.0 statistical software.Comparisons were carried out among different groups with one way analysis of variance (ANOVA) and between groups were performed with the Student t test.Changes were considered as statistically significant if P values was less than 0.05.Results ①Compared with mild group and control group,the levels of NSE and S100 protein were significantly higher in severe group and critically care group (P <0.05).②The serum levels of NSE and S100 protein in severe group were higher than in those in mild group with better outcomes (P < 0.05).Conclusion The serum levels of NSE and S100 protein as biomarkers can be used to evaluate the severity of EV71 infection,and can also be used to determine the efficacy of treatment.

Objective To explore the effect of the treating left colon cancer,Compare left colon irrigation at one-stage radical colon resections were performed with the way which is given colon bracket insertion operation then two-stage radical colon resections were performed.Methods 32 cases sigmoid and rectum cancer patients with obstruction were received.And 14 cases were treated by colon irrigation at one-stage radical colon resections; 18 cases were treated by colon bracket insertion operation then two-stage radical colon resections were performed.To analyze the complications of after-operation、the average time in hospital.Results No occurence of anastomotic leakage was found in the two-stage radical colon resections.There was only one case was infected; the operation time(115 ± 26)min.The drainage in 3 days after the operation(127 ± 66)ml,length time(12.5 ± 3.2)d.The group of one-stage radical colon resections:the operation time(210 ± 39)min.The drainage in 3 days after the operation.length time(14.8 ±2.6)d,There was two cases of anastomotic leakage and 12 cases were infected and two dead were found in this group.Conclusion The way which was given colon bracket insertion operation then two-stage radical colon resections were performed,was better than colon irrigation at one-stage radical colon resections were performed,and with the first way patients life quality enhanced.
7.The use of acetabular fossa as a reference for achieying the anticipated inclination of the acetabular cup

Objective To discuss the anatomical and radiographic characteristics of acetabular fossa,and evaluate their value as a reference for achieving the anticipated inclination of the acetabular cup.Methods Sixteen adult normal pelvic specimens were studied.Central axis of acetabular fossa and its intersection with fossa edge and acetabular rim were marked.The radiographic appearance of the two intersections was evaluated.With radiographic templating,the relationship between the top of acetabular fossa and the central axis of acetabular cup at anticipated inclination was identified.Implantation of cementless acetabular cup was performed bilaterally on the pelvic specimens.The acetabular fossa was used as a reference for implantation on one side,and the acetabular positioning device was used on the other side.The discrepancy of acetabular inclination was recorded before and after operation in the two groups.Results Intersection of central axis of acetabular fossa with fossa top edge and acetabular rim corresponded to the most medial and lateral aspect of acetabular sourcil respectively on standard anteroposterior pelvic radiograph.There was close correlation between most medial aspect of acetabular sourcil and central axis of the acetabular cup at anticipated inclination of 40°±5°.For the group of using acetabular fossa as reference,the mean difference of inclination was 0.19°±3.14°(-6°-5°)before and after operation,and the discrepancy was 2.75°±2.89°(-2°-8°)for the control group.The difference between the two groups was statistically significant(t=-2.453,P=0.027).Conclusion In primary total hip replacement,if normal anatomy of acetabulum was found,the anticipated inclination of acetabular cup can be accurately obtained,based on the relationship between the top of acetabular fossa and anticipated inclination of acetabular cup on preoperative templating.
8.Clinical Observation on Hepatoprotective Effect of Compound Glycyrrhizin in Treating Patients with Multidrug Resistant Pulmonary Tuberculosis

OBJECTIVE:To observe the hepatoprotective effect of compound glycyrrhizin in treating patients with multidrug resistant pulmonary tuberculosis(MDR-TB).METHODS:A total of 68 MDR-TB cases were enrolled:36(treatment group) were randomly assigned to receive additional glycyrrhizin,and 32(control group) to receive additional glucurolactone and legalon,the hepatoprotective effects and side effects between 2 groups were compared before and after treatment.RESULTS:The total effective rates of the treatment group and the control group were 94.4% and 62.5%,respectively 1month after hepatoprotective treatment,and significant difference was noted between 2 groups(P0.05).CONCLUSION:Compound glycyrrhizin has a reliable hepatoprotective efficacy and few adverse drug reactions in treating MDR-TB patients complicating with lesion of liver function.
9.Re-emergence of Schistosomiasis in Dali City after the Criteria of Transmission Control Were Met

Objective To analyze the epidemic situation on schistosomiasis in five years after reaching the criteria of transmission control in 1995 in Dali City. . Methods. Epidemiological data of schistosomiasis during 1995-2000 were collected and analyzed. . Results . Stool positive rate in the population was increasing yearly, new cases of infection including cases under 14 years old were detected each year. Acute infection occurred during 1996-1998 consecutively. Stool positive rate in cattle was unstable. Snail-ridden area increased significantly in 1996 and spots with infected snails stayed at a higher level during 1997-2000. . Conclusion . Schistosomiasis in Dali city has been re-emerging though it was once deemed under transmission control. Possible reasons include a low drug-taking rate though distributed, frequent movement of cattle population, and less activity on snail control.
10.Clinical study of endoscopic mucosal dissection treating digestive tract diseases

Objective To demonstrate the clinical significance of endoscopic mucosal dissection (ESD) in the treatment of digestive tract diseases.Methods A total of 112 patients with ESD treatment were selected.Preoperative localization endoscopy gastrointestinal ultrasound showed the lesion had not yet invaded muscular layer by ESD.Results Ninety-six cases had complete stripping and removal of lesions,postoperative pathology showed basal and margin had no lesions and membrane was intact integrity,complete resection rate was 85.7%(96/112).Surgery did not appear to uncontrollable bleeding without titanium clip,also not be clipped digestive tract perforation,abdominal infection,subcutaneous emphysema etc.There was no uncontrollable bleeding,digestive tract perforation that couldn't be clipped by titanium clip,abdominal infection,subcutaneous emphysema etc in surgery.For 1 year in 12 patients with early stage cancer of postoperative follow-up,no residual and recurrence.Conclusion ESD is a safe,effective,less painful,can be a one-time operation lesions complete resection for gastrointestinal lesions.
