1.Applications of 18F-Fluorodexyglucose Positron Emission Tomography on the Diagnosis and Treatment of Carcinoma of Large Intestine

Objective To introduce the current study on 18F-fluorodexyglucose positron emission tomography (18F-FDG PET) scanning in diagnosis and treatment of carcinoma of large intestine.Methods The literatures about 18F-FDG PET scanning in diagnosis and treatment of carcinoma of large intestine in recent years were reviewed.Results 18F-FDG PET scanning is superior to CT and MRI in identificating carcinoma of large intestine recurrence, metastasis in the early stage after operation and staging carcinoma of large intestine.Conclusion 18F-FDG PET scanning may be one of the accessory examinations in carcinoma of large intestine and may be helpful for the choice of treatment.
2.Effects of bone marrow-derived mesenchymal stem cells engraftment on VEGF in lung tissue and in plasma of rabbits at early stage of smog inhalation injury

Objective To discuss the effects of bone marrow-derived mesenchymal stem cells (MSCs) on vascular endothelial growth factor (VEGF) in lung tissue and in plasma, and extra-vascular lung water at the early stage of smog inhalation injury. Method The rabbit model of smog inhalation injury was established by using home-made smog generator, and the rabbit models were randomly(random number) divided into control group (group S, n = 32) and MSCs treatment group (group M, n = 32). Ten mL phosphate-buffered saline (PBS) was injected into ear marginal vein immediately after injury in rabbits of group S. The third generation of MSCs 1/10/10 mL PBS was injected into ear marginal vein immediately after injury in rabbits of group M. The levels of VEGF in peripheral blood and lung tissue were neasured 0 h,2 h,4 h and 6 hours after injection respectively, and analyzed. The right lung of rabbits was taken out to measure and calculate lung water mass fraction after experiment.Results In lung tissue, the levels of VEGF decreased gradually in group S (P < 0.05), and though the levels of VEGF in lung tissue appeared with significant decreasing trend in group M (P < 0.05), they were still higher than those of group S at corresponding intervals ( P < 0.05). In peripheral blood, the levels of VEGF increased gradually in group S ( P < 0. 05), and the levels of VEGF in group M appeared with markedly increasing trend ( P < 0.05),but they were lower significantly than those in group S at corresponding intervals ( P < 0.05). Conclusions MSCs engraftment to the rabbits with smog inhalation injury could increase VEGF in lung tissue, decrease VEGF in plasma and reduce extra-vascular lung water, showing protective effect on smog inhalation injury to a certain extent.
3.Study of ocular surface macro genome in dry eye patients

Objective To investigate the difference in ocular surface microbiota between dry eye patients and healthy subjects,and discuss the role of microbiota in dry eye.Methods Twenty cases of dry eye patients and 90 cases of healthy subjects were collected in the PLA General Hospital and Zhongshan Ophthalmic Center.The samples of conjunctiva impression cytology were collected from all subjects,and then metagenomic shotgun sequencing was performed following the DNA extraction.The differences in alpha diversity and metabolic pathways of the ocular surface microbiota between dry eye patients and healthy subjects were evaluated.Results There was no significant difference in alpha diversity of the microbial community between dry eye patients and healthy subjects (P =0.13).However,an increase of 15 species and a decrease of 10 species were detected on the ocular surface of dry eye patients.The enriched antibiotic resistance genes in dry eye patients were more than healthy subjects.Conclusion Although the alpha diversity of the microbial community on ocular surface between dry eye patients and healthy subjects are not distinguishable,a significant difference could be found in relative abundance and metabolic pathways,suggest that these specific microbiome may be related to the pathogenesis and disease progression of dry eye.
4.Application of the Requirement of Biological Variation Determination in the Quality Management of Blood Cell Analyzer

Objective To study application of the quality management of Blood Cell Analyzer according to the requirement of biological variation determination.Methods Collected the indoor imprecision value(CV%) from 8 items detected by blood cell analyzer during from April to Nov.of 2016,and the bias (Bias%) of 8 items of two EQA (external quality assessment) from the ministry of health in 2016.Then according to the 3 levels of the minium,appropriate and optimal quality specifications derived from the biological variability the rates of imprecision and bias were culculated.The pass rate of the imprecision and bias was calculated.By using mean bias and mean imprecision and biological variation 3 levels of total error (TEa) crite rion,and to calculate the corresponding σ and QGI value,so as to evaluate the performance of whole blood cell analyzer.Then improved the quality.Results For the imprecision value of 8 items,except the MCHC average value,all others were all 100 % meeting the appropriate level of quality requirements.For the bias value (Bias %) from 8 items,except MCH,all others were over 80 % meeting the appropriate level of quality requirements.While for the calculated σ value,based on the best level of quality requirements,except the σ value of WBC was 4.6,the σ value of all other items were all<3.Based on the appropriate level of quality requirements,except the σvalue of MCHC was 1.9,the value of σ of all other items were all> 3,and based on the minimal requirements,the σ value of all 8 items were all >3.After analysis,this blood cell analyzer,except that MCHC should use the minimal quality standard requirements,all other examination items could used the proper quality standard requirements,and the calculated QGI were all <0.8.Conclusion Based on the biological variation determination requirement and calculated σ and QGI value,this method could be used to more accurate quality evaluation of blood cell ana lyzer.Which is a higher levelof quality management,will be more conducive to quality improvement and better serve the clinical.
5.ANALYSIS AND CHARACTERIZATION OF CAROTENOIDS IN GINGKO SEED

Objective: To analyze and identify the carotenoid in gingko seed.Methods: The chemical composition of the pigment from gingko seed was identified as carotenoid by UV absorption spectrum. Its content was determined by spectrophotometric method and its composition was analyzed and identified by thin-layer chromatography and HPLC method.Results: Gingko contained carotenoids, roughly about 489 ?g/100 g in which lutein, ?-carotene and ?-carotene amounted to 69.20%,15.80% and 7.45% respectively. Conclusion: Gingko contained large quantity of carotenoids, mainly lutein, and next in order ?-carotene and ?-carotene.
6.Impact of misregistration between SPECT and CT on attenuation correction of myocardial perfusion imaging

Objective To evaluate the effects of misregistration with different directions and magnitudes between SPECT and CT on image quality and semi-quantification of MPI.Methods The data of 19 healthy volunteers (11 males,8 females ; mean age:(65.3 ± 9.6) years) were retrospectively analyzed.They all had a low pretest likelihood of coronary artery disease according to exercise and rest 99Tcm-MIBI MPI.The CT attenuation correction (CTAC) was performed on a SPECT/CT system.The CT images were manually shifted by 0.5,1.0,1.5,2.0,2.5,3.0,3.5,4.0,4.5 cm distance along the up/down,left/ right,and anterior/posterior axes respectively by using the system built-in software.The counts of the left ventricle were measured using myocardial Bull's eye generated from the reconstructed CTAC images.The image quality and semi-quantification of the CTAC images reconstructed from the raw data with and without shifting were compared and analyzed.Paired t test was used to analyze the data.Results There was no visible artifact with 0.5 cm shifting.The image quality was deteriorated significantly and the counting difference was significant with shifting distance greater than or equal to 1.0 cm.The image artifact of apex wall was mainly due to the upward shift,anterior and apex wall due to the downward shift,septal wall due to the leftward shift,anterior,apex and lateral wall due to the rightward shift,lateral and infero-posterior wall due to the forward shift,anterior,apex and septal wall due to backward shift.The counting difference caused by the downward shift was significantly more severe than that caused by the upward shift ((-9.68±8.06) % and (-2.04±1.83)%,t=6.573,P<0.01) ; and the rightward shift was more severe than the leftward shift ((-9.02± 8.47) % and (-4.38±3.67) % ; t =1.987,P<0.05).The image artifacts in anterior,apex and lateral walls were more severe than those in the infero-posterior and septal walls.Conclusions CTAC image artifacts in myocardial perfusion SPECT/CT studies could be caused by misregistration ≥ 1.0 cm.Different directions and magnitudes of shift could result in different degrees of attenuation artifacts at different locations on the original images.
7.Unconditional logistic regression analysis of risk factors of type 2 diabetes mellitus in Dalian

BACKGROUND: Diabetes mellitus is a chronic metabolic disease caused by various factors,such as environmental factor,inherited factor,etc.,and its cause isn't very clear now. This study aims to investigate the risk factors mentioned above in the onset of diabetes mellitus, and is of significance in the first and second grade of prevention of diabetes mellitus.OBJECTIVE: To investigate the risk factors of type 2 diabetes mellitus to provide evidence for the proper intervention of it.DESIGN: Cross-sectional study based on diagnosis.SETTING: Department of epidemiology in a university and department of endocrinology in a university hospital.PARTICIPANTS: The subjects were residents who have resided in town and country of Dalian for more than 5 years and were elder than 40 years old. The method of stratified cluster random sampling was carried out among natural persons who were divided into two groups according to city and country. Totally 2 500 persons were taken in total and 1 250 persons were from each group. The subjects who had been diagnosed as diabetes mellitus were included.METHODS: Questionnaire survey was employed for all the subjects, including physical examinations such as height,weight,waist girth,hip girth,blood pressure,blood glucose,etc. Patients of type 2 diabetes mellitus were taken as the case group and the normal was as the control group. The unconditional univariate and multivariate logistic regression were used.MAIN OUTCOME MEASURES: Unconditional logistic regression analysis of risk factors of type 2 diabetes mellitus with single factor analysis and multivariate analysis.RESULTS: Family history of diabetes mellitus(OR = 2.339),obesity[body mass index(BMI),OR = 1.462],systolic pressure(OR = 1.016),hyperlipidemia(OR = 1.615), age(OR = 1.043) were the major risk factors for type 2 diabetes mellitus.CONCLUSION: Family history of diabetes mellitus, increase of systolic pressure,obesity,high blood lipid and age are the risk factor for type 2 diabetes mellitus. For all communities,control of body mass and high blood pressure is an important way to prevent type 2 diabetes mellitus.
8.Expression and clinical significance of inducible nitric oxide synthase and β-catenin in nasopharyngeal carcinoma tissue

Background and purpose: There have been no reports on the clinical and biological significance of the co-expression of inducible nitric oxide synthase (iNOS) and β-catenin (β-cat) in has.pharyngeal carcinoma(NPC). This study was aimed to investigate the expression of iNOS and β-cat in NPC, to analyze their interrelation, and to explore their roles in the carcinogenesis, development, invasion, and metastasis of NPC. Methods: The expression of iNOS and β-cat was examined in 50 poorly differentiated NPC and 15 normal nasopharyngeal epithelium tissue by immunohistochemical staining (SP method). None of the patients had received radiotherapy and chemotherapy. The results of immunostaining were observed by two pathologists independently, using double blind scoring method. iNOS and β-cat expression were categorized by the extent and intensity of staining using a semiquantitative method. Results: No iNOS expression was observed in 15 normal nasopharyngeal epithelium, but β-cat expression was located in cytomembrane in normal samples. The positive rate of iNOS protein expression was 74.0% (37/50) in NPC tissues. The expression of iNOS was statistically different among the NPC group and normal nasopharyngeal epithelium group, T_(1-2) group and T_(3-4) group, metastatic lymph nodes group and no metastatic lymph nodes group. There was a great quantity of β-cat expression in cytoplasm, little or no β-cat expression in cytomembrane and nucleus in NPC tumorous tissues. Statistical analysis indicated that strong positive expression of β-cat in cytoplasm was significantly higher in NPC than those of normal nasopharyngeal epithelium. The positive expression of β-cat was significantly correlated with clinical stage, invasion and lymph gland metastasis. Neither iNOS nor β-cat was significantly different between different ages and genders. There was a positive relationship between iNOS and β-cat expression in NPC tissues (r=0.394, P=0.005). Conclusion: The positive expressions of iNOS and β-cat in cytoplasm were significantly correlated with clinical stage, invasion and metastasis, and were not significantly different in terms of ages and genders of the patients. There was a positive relationship between the expression of iNOS and β-cat in NPC tissues. Co-overexpression of iNOS and β-cat may play an important role in carcinogenesis, development, invasion, and metastasis of NPC.
9.The value of right lateral decubitus position to decrease artificial defect of cardiac anterior wall in 99Tcm-MIBI SPECT myocardial perfusion imaging for women

Objective To explore the value of right lateral decubitus position MPI for differentiating myocardial perfusion defect from cardiac anterior wall attenuation artificial defect,caused by breast of woman.Methods Forty-nine patients(average age (61.5±8.4) years) who had low likelihood of coronary artery disease and had perfusion defect in the anterior wall after exercise stress 99Tcm-MIBI MPI were included.All underwent supine and right lateral decubitus position during resting SPECT images.The myocardial perfusion SPECT images at left ventricle were reconstructed and were measured by Bull's-eye,based on the counts.Results from both supine position imaging and right lateral decubitus position imaging were compared.Paired t test was used to statistically analyse the data by SPSS 13.0.Results Compared with supine position,the counts of the anterior,inferior,apex and lateral wall in right lateral decubitus position were significantly higher:(71.30±3.53) % vs (66.50±3.85) %,(70.06±4.45) % vs (65.44±4.16) %,(77.90±3.00)% vs (75.81±4.08)%,(79.30±2.26)% vs (72.60±3.87)% (t=6.731,5.286,3.555,10.885,all P<0.01).The counts of septal wall were significantly lower ((66.60±3.98)% vs (70.06±4.51)%,t=-4.625,P<0.01) in right lateral decubitus position than that in supine position.Among the different regions of anterior wall,the counts of the anterior-middle ((76.40 ± 3.80) % vs (68.60 ± 4.76) %) and anterior-apex region ((77.10±3.24) % vs (69.00±3.54) %) were significantly higher (t =9.916,8.870,both P<0.01) in right lateral decubitus position than those in supine position,but there was insignificance ((56.94±6.06)% vs (58.50±4.98) %,t =-1.493,P>0.05) at anterior-basal region.The artificial defect of different degrees in anterior wall was observed in all patients in supine position,23 cases (46.9%,23/49) showed artificial defect in the anterior-middle region and 16 cases (32.7%,16/49) in the anterior-apex region.All artificial defect showed radioactive filling in right lateral position imaging,12 cases(24.5%,12/49) filled markedly and 32 cases(65.3%,32/49) filled completely.Conclusions The artificial defects caused by breast of woman mainly present in anterior-middle region and in anterior-apex region of the anterior wall in supine position imaging.The right lateral decubitus position imaging can markedly improve these artificial defects.
10.Effects of apoptosis of Tca8113 cells induced by adriamycin on telomerase and telomere repeat binding factor proteins.

OBJECTIVETo investigate the role of telomerase and telomere repeat binding factors (TRF) in apoptosis.
METHODSThe proliferative activity of Tca8113 cells was assessed by methyl thiazolyl tetrazolium (MTT) assay. After Tca8113 cells were treated with adriamycin at 5 mg/L, apoptotic morphology was observed under microscope with Giemsa staining and apoptosis examined by flow cytometry; analysis of telomerase activity was performed by TRAP-enzyme-linked immunosorbent assay; expression and expression level of TRF proteins were detected with immunohistochemical staining and immunofluorescence label assay, respectively.
RESULTSAfter Tca8113 cells were treated with adriamycin at 5 mg/L for 5 days and 7 days, the cells apoptosis was found. Telomerase activity dropped in time-dependent manner. Expression of TRF proteins appeared in nucleus of the cells. No statistical difference in expression levels of TRF was observed between the treated and untreated cells.
CONCLUSIONSTca8113 cells apoptosis induced by adriamycin decreased telomerase activity, but did not influence the expression level of TRF proteins.
