1. Mechanism research of apatinib-treated breast cancer based on network pharmacology

OBJECTIVE: To study the potential targets and action mechanism of apatinib-treated breast cancer. METHODS: Combination with bioinformatic analysis, PharmMapper reverse docking technology was used to predict potential targets of apatinib and docking was performed using Autodock 4. 0 to examine the interaction of aptinib wih predicted protein targets. RESULTS: Cell division protein kinase 2,GTPase HRas, NONE and cytochrome P450 2C9 were found to be the potential targets of apatinib-treated breast cancer. Stable hydrogen bonds were formed between apatinib and the four predicted targets, and the binding free energy was low. CONCLUSION: Apatinib maybe participate the functional regulation or biological action of cell division protein kinase 2, GTPase HRas, NONE and Cytochrome P4502C9 to treat breast cancer, which provides a theoretical basis and a clue for further exploration of the mechanism research of apatinib-treated breast cancer.
2. Mechanism of gastric carcinoma cell apoptosis induced by chlorogenic acid-like compounds extracted from Broussonetia papyrifera

Objective To explore the mechanism of gastric carcinoma cell SGC-7901 apoptosis induced by chlorogenic acid-like compounds extracted from Broussonetia papyrifera (CALCBP) bark. Methods The SGC-7901 cells were used to evaluate the anti-tumour activity of the extract in vivo, and the proliferation of cells was examined by MTT assay. The cell morphological changes of cells were observed by DAPI staining; The cell apoptosis and the cell cycle were detected respectively by flow cytometry after PI and Annexin V/PI staining; The intracellular ROS were determined under the fluorescence microscope using DCHF-DA probe, the changes of mitochondrial membrane potential were observed by JC-1 staining. The protein expression of p53, Bcl-2, Bax, and Cytochrome C, p-p38, p-JNK, JNK, p-ERK, ERK were analyzed by Western blotting. Results The proliferation of SGC-7901 cells was inhibited significantly by CALCBP in dose-dependent and time-dependent manner, the condensed chromosome and apoptotic body can be observed in the treated cells and the cell cycle was arrested in G2/M phase, the mitochondrial membrane potential was significantly decreased, whereas the cellular ROS levels of the treated cells were significantly increased. Moreover, the protein expression of p53, Bax, Cytochrome C, and p-p38 were significantly up-regulated and p-ERK and Bcl-2 expression were significantly down-regulated. Conclusion The apoptosis of gastric cancer cell SGC-7901 induced by CALCBP was probably related to oxidative stress of the cell mitochondrial via p38-MAPK and ERK-MAPK signal pathways.
3. Preparation of molecular imprinted polymers for solid-phase extraction of L-tetrahydropalmatine

Objective: To prepare the molecularly imprinted polymer (MIPs) of L-tetrahydropalmatine (L-THP) by the molecular imprinting technique and study on solid-phase extraction. Methods: Using L-THP as template, methyl acrylic acid (MAA) as functional monmer, and ehtylene glycol dimethacrylate (EDMA) as a cross-linking agent to prepare the L-THP-MIPs. A test was conducted to investigate the selectivity and the specificity of solid-phase extraction. Results: The experiment showed that the MIPs had the specific adsorption to L-THP, but did not have the specific adsorption to corydaline the structural analogue with L-THP. Conclusion: The L-THP-MIPs have a good selectivity and the specificity of L-THP.
4. Research on preparation technology for Sini Boiling Powder

Objective: To optimize the extracting process for Sini Boiling Powder at the condition of optimum particle size. Methods: The optimum size of particle was prepared, HPLC method was used for the determination of the content of ammonium glycyrrhetate and UV spectrophotometry was used for the determination of the content of total alkaloids. The decoction process was optimized by single factor test with boiling time, amount of water, and boiling times as factors. Then compared with traditional decoction, we can have a basis on clinic. Results: The optimal conditions of decoction process were soaking 20 min,14-fold water, boiling 30 min, once. The results showed that both the yield of dry extract and the content of active ingredient at different time points from the boiling powder granule were greatly more then its traditional one during the whole decoction process, and it suggested that Sini Boiling Powder is more safe then Sini traditional decoction. Conclusion: The advantage of high decoction rate and short preparing time on boiling powder can solve the problems that the traditional Chinese medicine is currently facing and should not be ignored.
5.Effects of glucocorticoid with different dosage on bone mineral density and biomechanical properties in rats

Objective To investigate the effects of exposure to glucocorticoids with different dosage on skeleton of normal 3-month-old rats by dual energy X-ray absorptiometry (DXA), biomechanical testing and bone histopathology. Methods Thirty-one 3-month-old female clean level SD rats were randomly divided into 3 GC-treated groups, with tail intravenous injections of dexamethasone (Dex) at the dosage of 1, 2.5, 5 mg/kg twice per week for 8 weeks, respectively, and 1 normal control group treated with saline. At the end of experiment, bone mineral content (BMC) and bone mineral density (BMD) of the femur and the 3rd lumbar vertebrae in rats were measured by DXA. The 3-point bending test of the total femur and compression test of the 5th lumbar vertebrae were also conducted, respectively. Microstructure of the trabecula in proximal metaphysis of the tibia was observed by bone pathological section for quantitative analysis. Results Compared with control group, the body weight was significantly decreased in all Dex-treated groups, while no obvious decrease in vertebral BMC, BMD and maximum compressive loads was found. The total femoral BMC was also reduced significantly in all Dex-treated groups, while the total femoral BMD, proximal and distal femoral BMD were reduced only in Dex 1mg group. The fracture load, maximum load and elastic load for 3-point bending test were all evidently decreased in Dex 1mg group, while the Dex 2.5mg group and Dex 5mg group only showed a decline in elastic load. All Dex-treated groups showed an unevenly spatial distribution of the trabecula, which indicated a low bone metabolic state. Conclusions The exposure to GC for 8 weeks brings negative effects on skeleton of the 3-month old rats, which will cause more bone loss and worse mechanical properties in femur than in lumbar vertebrae. Higher dosage of Dex does not increase bone mass loss or change the mechanical properties. Both the decline in bone mechanics, especially elastic load, and unevenly density distribution of trabecula indicate that Dex will affect more bone quality other than bone mass. The side effects of GCs on skeleton in clinical application should be evaluated with various methods.
6.Assessment of ergonomics risk in typical upper limb operations of electricians based on RULA

Background A large number of power supply workers are involved in live working and are exposed to multiple risk factors for long periods of time during outdoor work, which in turn causes occupational health problems. Objective To identify potential ergonomic factors related to work-related musculoskeletal disorders (WMSDs) and estimate the ergonomic risk of electricians. Methods A total of 31 workers were randomly selected from a training base under a power supply enterprise in Guangxi, and their operation processes were recorded by video. Using the Rapid Upper Limb Assessment (RULA) method, the workers were scored by the posture of their arms, neck, and torso during operation, and the final scores were summarized to assess their risk level. Two independent samples t-tests were used to compare the differences in RULA scores. Results A total of 31 videos for mounting/dismounting insulation mask and 29 videos for breaking/connecting contact terminal were collected. The RULA score for mounting/dismounting insulation mask was 6.6 ± 0.7 (left side) and 7.0 ± 0.2 (right side), most of the workers had a score of 7 (71.0% on the left side and 96.8% on the right side), and the upper limb and overall scores on the right side were higher than those on the left side (P < 0.05). The RULA score for breaking/connecting contact terminal was 6.9 ± 0.4 (both right and left sides), most of workers had a score of 7 (89.7% on the left side and 93.1% on the right side), and the difference of the overall scores between the right and left side for this operation were not statistically significant (P > 0.05). The ergonomic risk levels for both operations were level 3 or higher. Conclusion Mounting/dismounting insulation mask and breaking/connecting contact terminal are typical upper limb operations, and have a high level of ergonomics risk, requiring effective preventive and protective measures.

9.Application of acupoint massage in postpartum hypogalactia in puerpera.

OBJECTIVETo evaluate the effects of acupoint massage for postpartum hypogalactia in puerpera.
METHODSA total of 60 puerpera of cesarean section were randomized into an observation group and a control group, 30 cases in each one. In the observation group, 2 cases were dropped out. Finally, there were 28 cases in the observation group and 30 cases in the control group. In the control group, the routine nursing after operation in obstetrics department was given, guiding maternal breastfeeding and postpartum health education. In the observation group, on the basis of the treatment as the control group, the corresponding acupoints were selected from the breast related meridians for massage. Acupoint massage was given since the 3rd day after operation, 8:00 am to 9:00 am, once a day, 1 min at one acupoint, continuously for 5 days. The lactation volume, breast filling, galactostasis and milk viscosity were compared in the parturient of the two groups. The clinical therapeutic effects were compared between the two groups.
RESULTSFrom the 1st to the 2nd days, the differences were not significant in lactation volume, breast filling and galactostasis score between the two groups (all>0.05). From the 3rd to 5th days, the lactation volume, breast filling and galactostasis score in the observation group were all lower than those in the control group (all<0.01). The difference in milk viscosity was not significant in the first 4 days between the two groups (all>0.05). On the 5th day, the score of milk viscosity in the observation group was lower than that in the control group (<0.01). The cured rate was 64.3% (18/28) in the observation group, which was better than 13.3% (4/30) in the control group (<0.05).
CONCLUSIONMassage at the acupoints from the breast related meridians effectively promotes the lactation secretion in puerpera of postpartum hypogalactia and alleviates breast distention.
10.Progress and prospect of heart transplantation in children

Pediatric heart transplantation is the standard treatment for children complicated with refractory heart failure which is difficult to be treated by conventional surgery or drugs. At present, an increasing quantity of pediatric heart transplantation is being performed worldwide, whereas relevant experience is still lacking in China. In recent 10 years, significant progress has been achieved in pediatric heart transplantation. On one hand, the number of pediatric heart transplantation has been increased year by year. On the other hand, ABO-incompatible heart transplantation, application of ventricular assist device in children, and recipient-donor weight mismatch transplantation have been widely employed to resolve the shortage of donor heart in pediatric heart transplantation.
