1. Preparation of molecular imprinted polymers for solid-phase extraction of L-tetrahydropalmatine

Objective: To prepare the molecularly imprinted polymer (MIPs) of L-tetrahydropalmatine (L-THP) by the molecular imprinting technique and study on solid-phase extraction. Methods: Using L-THP as template, methyl acrylic acid (MAA) as functional monmer, and ehtylene glycol dimethacrylate (EDMA) as a cross-linking agent to prepare the L-THP-MIPs. A test was conducted to investigate the selectivity and the specificity of solid-phase extraction. Results: The experiment showed that the MIPs had the specific adsorption to L-THP, but did not have the specific adsorption to corydaline the structural analogue with L-THP. Conclusion: The L-THP-MIPs have a good selectivity and the specificity of L-THP.
2. Extraction and verification of miRNA from ginseng decoction

Objective: To verify the existence of microRNAs (miRNAs) extracted from fresh ginseng decoction. Methods: Fresh ginseng was prepared into decoction according to the conventional method. The miRNA were extracted from the condensed ginseng decoction by plant microRNA extraction kit. Then miRNA were treated by DNase I and subjected to agarose gel electrophoresis and Agilent 2100 bioanalysis. MiR-159 and miR-6135, which were highly expressed in ginseng, were selected and verified by real-time quantitative PCR to detect the expression in the decoction. Results: Ginseng miRNA were successfully extracted from fresh decoction. MiR-159 and miR-6135 were expressed in fresh decoction with lower levels than those of fresh ginseng. Conclusion: miRNAs stably existed after processing, and retained some stability after high-temperature treatment. The findings provide a valuables basis for the further studies on ginseng miRNAs.
3. Efficacy of Ropivacaine injection at acupoints for labor analgesia and its effect on breastfeeding and serum prolactin

OBJECTIVE: To investigate the efficacy of acupoint injection of Ropivacaine for labor analgesia and its effect on breastfeeding and prolactin secretion. METHODS: A total of 80 primipara who asked to receive labor analgesia were randomized into epidural analgesia group (n=35) and acupoint injection group (n=36), and other 36 primipara who refused to receive labor analgesia were subjected to the control group. The labor analgesia began to be performed when the puerpera's uterine orifice opened about ≥3 cm (the first stage of labor), including puerpera controlled epidural analgesia (PCEA, Sufentainil + 0.1% Ropivacaine hydrochloride, 5 µg/mL at L3-L4 interspace, till the birth of fetus) or acupoint injection of Ropivacaine (0.2%, 1 mL/acupoint) at bilateral Hegu (LI4) and Sanyinjiao (SP6). The delivery situations such as the duration of labor, and number of cases who used oxytocin, obstetric-forceps-aided delivery, cesarean delivery were recorded. The serum prolactin concentration was assayed by using ELISA. The visual analogue scale (VAS) scores at the time points of T0 (about 3 cm widening of the orifice of uterus and before performing analgesia), T1 (30 min after labor analgesia), T2(about 10 cm widening of the orifice of uterus) and T3(coming out of fetal head). The duration of labor, ratio of use of oxytocin, onset time of breastfeeding, and times of breastfeeding within 24 postpartum hours were recorded accordingly. RESULTS: The VAS scores at time-points of T1, T2 and T3 were significantly lower in both epidural analgesia and acupoint injection groups in comparison with their own T0 and the control group (P<0.05), and were also considerably higher in the acupoint injection group than in the epidural analgesia group (P<0.05). The duration of the 2nd stage of labor (from complete opening of the uterus orifice to complete birth of the fetus) was significantly longer, (P<0.05) and the number of oxytocin-using puerpera was obviously bigger in the epidural analgesia group than in the control group (P<0.05). After partum, the 1st breastfeeding time was obviously earlier and the frequency of breastfeeding notably increased in both epidural analgesia and acupoint injection groups than in the control group (P<0.05), the serum prolactin content was remarkably higher in the acupoint injection group than in the epidural analgesia group (P<0.05). No significant differences were found between the acupoint injection and the control groups in the duration of the 1st and 2nd stages of labor, and in the numbers of oxytocin-using puerpera, obstetric forceps-aided birth and cesarean delivery (P>0.05), and between the epidural analgesia group and control group in the serum prolactin levels (P>0.05).. CONCLUSION: Injection of Ropivacaine at LI4 and SP6 is effective for labor analgesia and raising prolactin level, and favorable to breastfeeding in the early postpartum period.
4. Identification of miRNA target genes in Panax ginseng by degradome sequencing analysis

Objective:To analyze and identify ginseng miRNA. Methods: The miRNA and target genes of Shizhu ginseng and Yuan ginseng were detected by the degradation sequencing technology; Functional annotation of Degradome genes was carried out using public databases of KEGG/NR/GO database; The expression of miRNA and target genes of Shizhu ginseng and Yuan ginseng was determinated by real time fluorescence quantitative PCR technique. Results: A total of 13 target genes of eight miRNA families were obtained; The target gene type of miRNA was mainly transcriptional factor, response factor, and signal transduction pathway by means of KEGG/NR/GO database analysis. The results of real time fluorescence quantitative PCR verification of aqc-miR-159, bdi-miR162, cpa-miR319, pgi-miR4376, smo-miR396 and its target genes were basically consistent with the expression of miRNA and target genes from the degradation group. Conclusion: The target genes of partial Panax ginseng miRNA is clarified, which lays the foundation for further study of the possible function of ginseng miRNA.
5.Study on UPLC Fingerprint of Vaccariae Semen before and after Processing and Content Determination of Erythrine and Vaccarin

OBJECTIVE:Compare the fingerprint difference of Vaccariae Semen before and after processed (stir-fried),and to determine the contents of erythrine and vaccarin before and after stir-fried. METHODS :UPLC method was adopted. The determination was performed on YMC Trait C 18 column with mobile phase consisted of acetonitrile-water (gradient elution )at the flow rate of 0.35 mL/min. The detection wavelength was set at 219 nm,and the column temperature was 35 ℃. The sample size was 1 μL. Using vaccarin as reference,the fingerprints of Vaccariae Semen crude product and its processed product (each of 17 batches,S1-S17,CS18-CS34) were drawn. The similarity evaluation and common peak identification were carried out by Similarity Evaluation System of TCM Chromatographic Fingerprint (2012 edition);cluster analysis ,principle component analysis (PCA)and factor analysis were performed by using SPSS 20.0 software. The contents of erythrine and vaccarin in Vaccariae Semen crude product and its processed product were determined by UPLC. RESULTS :There were 5 common peaks in UPLC fingerprints of 17 batches of Vaccariae Semen crude product and its processed product. The similarities were all higher than 0.99. Among them , 2 common peaks were identified ,i.e. erythrine ,vaccarin. Results of cluster analysis showed that S 1-S17 were clustered into one category and CS 18-CS34 were clustered into one category. Results of PCA and factor analysis showed that variance contribution rate of the first principle component was 76.418%;erythrine and vaccarin had higher loading on the first principal component (eigenvalues were 0.976 and 0.966,respectively). The linear ranges of above 2 components were 6.437-321.832 μg/mL and 7.729-386.437 μg/mL,respectively(r>0.999). The limits of detection and quantitation were 0.085,0.284 ng (crude product) and 0.739, 2.465 ng (processed product ), respectively. RSDs of precision ,reproducibility,stability(12 h)and durability tests were all lower than 3%(n=6 or n=5). The contents of the two components were 0.11%-0.20%,0.42%-0.63%(crude product )and 0.08%-0.11%,0.34%-0.50%(processed product ). CONCLUSIONS :UPLC fingerprint of Vaccariae Semen crude product and its processed product are established successfully. Although the chemical constituents in Vaccariae Semen are consistent before and after stir-fried ,the contents of erythrine and vaccarin are all decreased after stir-fried.
6.Effects of glucocorticoid with different dosage on bone mineral density and biomechanical properties in rats

Objective To investigate the effects of exposure to glucocorticoids with different dosage on skeleton of normal 3-month-old rats by dual energy X-ray absorptiometry (DXA), biomechanical testing and bone histopathology. Methods Thirty-one 3-month-old female clean level SD rats were randomly divided into 3 GC-treated groups, with tail intravenous injections of dexamethasone (Dex) at the dosage of 1, 2.5, 5 mg/kg twice per week for 8 weeks, respectively, and 1 normal control group treated with saline. At the end of experiment, bone mineral content (BMC) and bone mineral density (BMD) of the femur and the 3rd lumbar vertebrae in rats were measured by DXA. The 3-point bending test of the total femur and compression test of the 5th lumbar vertebrae were also conducted, respectively. Microstructure of the trabecula in proximal metaphysis of the tibia was observed by bone pathological section for quantitative analysis. Results Compared with control group, the body weight was significantly decreased in all Dex-treated groups, while no obvious decrease in vertebral BMC, BMD and maximum compressive loads was found. The total femoral BMC was also reduced significantly in all Dex-treated groups, while the total femoral BMD, proximal and distal femoral BMD were reduced only in Dex 1mg group. The fracture load, maximum load and elastic load for 3-point bending test were all evidently decreased in Dex 1mg group, while the Dex 2.5mg group and Dex 5mg group only showed a decline in elastic load. All Dex-treated groups showed an unevenly spatial distribution of the trabecula, which indicated a low bone metabolic state. Conclusions The exposure to GC for 8 weeks brings negative effects on skeleton of the 3-month old rats, which will cause more bone loss and worse mechanical properties in femur than in lumbar vertebrae. Higher dosage of Dex does not increase bone mass loss or change the mechanical properties. Both the decline in bone mechanics, especially elastic load, and unevenly density distribution of trabecula indicate that Dex will affect more bone quality other than bone mass. The side effects of GCs on skeleton in clinical application should be evaluated with various methods.

OBJECTIVE:
To investigate the inhibitory effects of novel phosphodiesterase 4 inhibitor ZL-n-91 to the proliferation of leukemia cells L1210 and K562.
METHODS:
CCK-8 method was used to detect the effect of ZL-n-91 to the proliferation of L1210 and K562 cells, and the proliferation rate, IC
RESULTS:
ZL-n-91 showed a significant inhibitory effect to the proliferation of leukemia cells L1210 and K562 in a dose-dependent manner (P<0.001). After treated by ZL-n-91, the leukemia cells L1210 and K562 in the S-phase in cell cycle decreased significantly compared with those in control group (P<0.01). The apoptosis of leukemia cells L1210 and K562 could be induced by ZL-n-91 (P<0.001), and the expression level of apoptosis related protein BAX significantly increased. In the animal experiment, the result showed that ZL-n-91 could significantly inhibit the growth of subcutaneously transplantation tumor (P<0.05).
CONCLUSION
The novel phosphodiesterase 4 inhibitor ZL-n-91 can effectively inhibit the proliferation of leukemia cells L1210 and K562, which has the potential of anti-leukemia drug development.

8. Mediating effect of general self-efficacy between stress coping personality and perceived professional benefits among nursing interns

OBJECTIVE: To explore the mediating effect of the general self-efficacy(GSE), stress coping personality(SCP) and perceived professional benefits(PPB) among nursing practice students. METHODS: A total of 836 nursing interns from six grade A hospitals in six cities were selected as the research subjects using convenience sampling method. The GSE, SCP and PPB were investigated by the General Self-Efficacy Scale, Scale of Stress Coping Personality for College Students and Questionnaire of Nurses Perceived Professional Benefit. RESULTS: The average scores of GSE, SCP, and PPB were(24.6±5.8),(183.1±28.7) and(139.5±18.0), respectively. The scores of GSE and SCP were positively correlated with that of PPB [correlation coefficients(r) were 0.31 and 0.38 respectively, both P<0.01], and a positive correlation was found between GSE and SCP(r=0.41, P<0.01). The hierarchical regression results showed that the sense of control, tenacity and tolerance of SCP of the interns had a predictive effect on their PPB(all P<0.05); but the effect of SCP on PPB was weakened after inclusion of GSE(P<0.01). The structural equation model analysis results showed that both SCP and GSE of interns had a direct positive predictive effect on PPB(all P<0.01), GSE played a partial mediating role between SCP and PPB, accounting for 20.3% of the total effect. CONCLUSION: The SCP of nursing interns can directly or indirectly affect their PPB, and GSE plays a partial mediating effect between SCP and PPB.
9. Relationship between happiness and psychological resilience among sanitation workers

OBJECTIVE: To understand the current situation of psychological resilience of sanitation workers and its impact on their happiness. METHODS: A total of 277 sanitation workers from Dongguan City of Guangdong Province were enrolled as study participants by stratified random sampling method. The psychological resilience and happiness were measured by Connor-Davidson Resilience Scale and Campbell Index of Well-being Scale respectively. RESULTS: The median scores of psycological resilience and happiness of the study subjects were 49. 0 and 7. 8,respectively. Psycological resilience was positively correlated with happiness( P < 0. 01). The multivariate linear regression analysis showed that the types of work,education level,personal monthly income and daily working time were the influencing factors of sanitation workers 'psychological resilience( P < 0. 05). The types of work and psychological resilience can significantly predict happiness(P < 0. 05). After controlling the demographic variables such as age,type of work,education level,personal monthly income,and daily working time,the psychological resilience prediction happiness contribution rate increased by 34. 1%(P < 0. 01). Age has a regulating effect on psychological resilience and happiness. Psychological resilience is easier to predict in the second generation( born after 1978) of sanitation workers in the sense of happiness. CONCLUSION: Psycological resilience of sanitation workers affects their happiness. Enhancing psychological resilience can help improve the happiness of sanitation workers.
10. Influence of social support and self-efficacy on job burnout in sanitation workers

OBJECTIVE: To assess the influence of social support and self-efficacy on job burnout in sanitation workers. METHODS: A convenient sampling method was used to select 405 sanitation workers as study objects. The Maslach Burnout Inventory General Survey, Perceived Social Support Scale and General Self-Efficacy Scale were used to investigate the three dimensions of emotional exhaustion,cynicism and reduced personal accomplishment in job burnout, as well as the social support and self-efficacy. RESULTS: The cynicism was positively correlated with self-efficacy [partial correlation coefficient(r)=0.17, P<0.01]. Reduced personal accomplishment was negatively correlated with self-efficacy and social support respectively(r were-0.32 and-0.20, P<0.01). Hierarchical regression analysis indicated that the demographic variables can explain 30.4%, 29.9%, 22.0% of the three dimensions of sanitation workers′ emotional exhaustion, cynicism and reduced personal accomplishment respectively(P<0.01). After controlling for the demographic variables, self-efficacy could positively predict cynicism(P<0.01), the social support and self-efficacy could negatively predict reduced personal accomplishment(P<0.01). CONCLUSION: The institutional factor is the main reason affecting job burnout of sanitation workers. Cynicism is positively correlated with self-efficacy. Improving social support and self-efficacy can help alleviate the reduced personal accomplishment.
