International Eye Science 2008;8(2):226-229
AIM: To investigate the effect of tetrandrine (Tet) on proliferation of human pterygium fibroblasts (HPF) in vitro and to search for a new method to prevent the recurrence after pterygium surgery.METHODS: With different concentrations (0 to 160μmol/L) of Tet acting on HPF cultured in vitro, the impact was observed at 24, 48, 72 hours respectively after Tet intervention. The MTT method was used to assay the biologic activities of Tet and inhibitive rate of cell growth. The expression of proliferating cell nuclear antigen (PCNA) in each group was detected by immunohistoche-mistry before and after Tet intervention.RESULTS:With different concentrations of 20, 40, 80 and 160μmol/L and acting for 24 to 72 hours, Tet could inhibit the proliferation of HPF in a dose- and time-dependent manner (P<0.05). After the intervention of Tet, the expression of PCNA protein declined. When the concentration of Tet was in the range of 20 to 160μmol/L, it was able to inhibit the expression of PCNA in a concentration-dependent manner (P<0.05).CONCLUSION:Tet could significantly inhibit the proliferation of pterygium fibroblasts, and the inhibitive action was in a dose- and time-dependent manner within a certain range of concentration. But in high concen-tration (>160μmol/L), Tet would have cytotoxity.

Objective To explore a simple and effective way of establishing a mice orthotopic tracheal transplantation model in studying bronchiolitis obliterans to reduce operation time. Methods Tracheas from C57BL/6 mice (n=30) were im?planted into Balb/c mice (n=30). Grafts and hosts were matched according to body weight.Orotracheal intubation and endo-stentwas used for trachea anastomose and animal model was establish. Operating time was also noted. Grafts were harvest?ed on Days 15, 30 and 60 after transplantation, using HE staining to observe pathological changes of allograft. Trachea block?ing rate was also calculated. Results Until survival of trachea transplantation, the operative time of donor′s and the recipi?ent′tracheas resection were (7.0±1.0) min and (7.0±1.0) min respectively.Orotracheal intubation the endo-stentestablish?ment cost(1 ± 0.5)min. Trachea anastomose took(20.0 ± 4.0)min and the operation time last(43.0 ± 10.0)min. Conclusion The improvement of orthotopic tracheas transplantation usingOrotracheal intubation the endo-stenfor trachea anastomose is esay-operated and did required complicate skills and instruments with high successful rate. So it is an ideal and stable model in studying bronchiolitis obliterans.

Journal of Guangzhou University of Traditional Chinese Medicine 2015;(3):430-433
Objective To study the effect of Zusanli and Guanyuan electroacupuncture ( EA) on inflammatory reaction in patients with sepsis. Methods A prospective and randomized trial was carried out in 58 sepsis patients, and the patients were randomly divided into EA group and control group. Patients in both groups were given conventional western medicine treatment, and EA group received EA at acupoints of Zusanli and Guanyuan additionally. The hospitalization time in intensive care unit ( ICU) of the two groups was compared. The changes of serum lactic acid (LA) level and inflammatory indexes including white blood cell and neutrophil count, C- reactive protein (CRP), erythrocyte sedimentation rate (ESR) and procalcitonin (PCT) of the two groups were observed before and after treatment. Results The hospitalization time in ICU of EA group was less than that of the control group, and the difference was statistically significant ( P<0.05) . On the seventh day after treatment, the serum LA level of EA group was significantly lower than that of the control group ( P<0.05) . Inflammatory indexes of EA group were decreased after treatment compared with those before treatment ( P<0.05) , and on seventh day after treatment, the inflammatory indexes of EA group were significantly lower than those of the control group ( P<0.05) . Conclusion EA at acupoints of Zusanli and Guanyuan is effective for the treatment of sepsis through reducing the inflammatory reaction of sepsis patients.

Objective To study the expression of signal transducers and activators of transcription 3(STAT3) in airway epithelial cells of asthmatic mouse model and its association with airway remodeling, explore the role of signal-transduction pathway in airway remodeling. Methods Thirty Balb/c mice were randomly divided into normal control group(n=10),asthma without intervention group(n=10) and AG490 intervention group(n=10).The mice were sensitized with ovalbumin to establish the asthmatic model. The histological changes were evaluated by HE staining, total brochial wall thickness(Wat)and smooth muscle thickness(Wam) were measured by image analysis system, the percentages of collagen deposition were detected by Masson′s trichrome staining, the expression of STAT3 in airway were detected by immunohistochemistry technique;lung tissue extracts were analyzed for phosphorylation of STAT3(p-STAT3)by Western blot.SPSS 13.0 software was used to analyze the data. Results 1.The histological changes including airway thickness, airway smooth cell proliferation and excessive collagen deposition in subepithelial aera were found under light microscope in asthmatic mice.2.The level of STAT3 and p-STAT3 in asthma without intervention group were significantly higher than those in normal control group(Pa

Objective To observe the effects of inhaled budesonide (BUD) in early phase on the airway inflammation in asthmatic mouse, and its effects on the IL-6/signal transducers and activators of transcription 3(IL-6/STAT3 )signaling pathway in airway, explore the therapeutic target of BUD.Methods Forty Balb/c mice were randomly divided into control group(n=10),asthma group(n=10),BUD group(n=10) and AG490 group(n=10).The mice were sensitized with ovalbumin to establish the asthmatic model. The histological changes were evaluated by HE staining. The levels of IL-4,IL-5 and IL-6 in bronchoalveolar lavage fluid (BALF) were measured by enzyme-linked immunosorbent assay.Lung tissue extracts were analyzed for total STAT3 and phospho-STAT3(p-STAT3) by Western blot. Results 1.The levels of inflammatory cells,EOS%, IL-4,IL-5 and IL-6 levels in the BALF in BUD group were significantly lower than those in asthma group (Pa0.05).Conclusions Inhaled corticosteroid can apparently ameliorate airway inflammation in early phase in asthmatic mouse model, and it can downregulate the expressions of IL-6 and STAT3,inhibit the signal-transduction pathway of STAT3.The IL-6 /STAT3 signaling pathway of airway may be one of the potential therapeutic targets of inhaled corticosteroid.
