Objective:To analyze chlorogenic acid of Chaenomeles speciosa by high performance liquid chromatography-mass spectrometry. Methods:A method for determining chlorogenic acid of Chaenomeles speciosa by HPLC-MS is described. The operating conditions were Diamonsil C18 column (5μm, 4.6×250 mm) at room temperature, acetonitrile-0.05%phosphoric acid solution as mobile phase by step gradients elution at a flow of 0.8 ml/min and UV detection at 325 nm. Results: The content of chlorogenic acid of Chaenomeles speciosa is 1.3 mg/g (RSD 0.4%), respectively. Conclusion:Chlorogenic acid was detected from Chaenomeles speciosa, which may be related to its inhibition of FAS and the potential role of anticancer.

Objective: To detect gallic acid in Paeonia suffruticosa Andr. by high performance liquid chromatography-mass spectrometry. Methods: A method for determining gallic acid in Paeonia suffruticosa Andr. by HPLC-MS is described. The operating conditions were Diamonsil C18 column (5μm, 4.6×250 mm) at room temperature, methanol-0.05% phosphoric acid solution as mobile phase by step gradients elution at a flow of 0.8 ml/min and UV detection at 270 nm. Results:The content of gallic acid is 1.8 mg/g. The relative standard deviation of gallic acid is 1.7%. Conclusion:The content of gallic acid in Paeonia suffruticosa Andr. is high, which may be related to the inhibition of FAS.

Objective To investigate the effects of p-cresol on human umbilical vein endothelial cells.Methods The effects of p-cresol on endothelial cell growth,cell cycle,cell morphological change and p21 protein were detected by the CCK-8 assay,flow cytometry assay,inverted microscope and Western blotting.Results P-cresol could inhibit the growth of human umbilical vein endothelial cells in dose-and time-dependent manners (all P ＜ 0.05).The human umbilical vein endothelial cells treated with p-cresol became elongated processes,cloudy cytoplasm,and irregular shapes.The p-cresol stopped human umbilical vein endothelial cells at cell cycle G1 and had no effect on cell apoptosis.The p-cresol could increase protein expression of p21 in a dose dependent manner (P ＜ 0.05).Conclusion P-cresol can increase protein expression of p21,induce cell cycle arrest at G1 stage and inhibit the proliferation of human umbilical vein endothelial cells.

Ultrasonic microbubbles were used to open blood-brain barriers (BBB) with a reversed and limited behavior feature in the study, which could improve the brain-targeted delivery of anti-tumor drugs. The glioma rat model was prepared. Low-frequency ultrasound was combined with microbubbles to affect the permeability of BBB compared with the permeability of independently administered Evans blue (EB) crossing BBB. Time point and length of ultrasound were investigated whether they affect the permeability of BBB and the damage of brain tissue. The effect of the growth time of glioma on BBB permeability was explored. Only glioma had a very little impact on BBB permeability. However, ultrasonic microbubbles opened the BBB with the features of temporary, limited and reversed behavior and improved EB and magnetic resonance imaging contrast agent penetrating BBB. A length of 30 s ultrasound is appropriate for opening BBB and no damage of brain tissue. Drugs should be injected before ultrasound so that they enter into brain as BBB opening. Ultrasonic microbubbles can open BBB effectively and safely, which improve drugs penetrating BBB under proper time point and length.

Objective:To investigate and evaluate the effect of antibacterial agents clinical use and management training for medi-cal institutions in Hubei province in order to promote rational antibacterial drug use. Methods:A questionnaire was designed, inclu-ding basic conception, medication principle, mechanism and medication feature etc. The data were studied by SPSS 13. 0 software to analyze the knowledge level of antibacterial agents in medical staffs. Results: Medical staffs had good acquisition in the knowledge points involved in choice questions. The correct percentage of each scoring in essay questions differed considerably within the range of 7. 88%-99. 01%. Management rules could be well understood, while the definition of antibacterial drugs and the difference between antibacterial drugs and antibiotics were paid little attention. Conclusion: The overall knowledge level of rational antibacterial agents use in the trainees is promising with comprehensive understand of the management rules. It's necessary to distinguish antibacterial a-gents from antibiotics more clearly.

Signal transducer and activator of transcription 3 (STAT3) is an important regulatory factor of cell proliferation and metastasis, involved in the occurrence and development of a variety of malignant tumors, and it is one of the hot spots in the research of targeted anti-tumor drugs. Our group screened a novel benzobis (imidazole) structure small molecule compound LZJ541 through the screening model of Janus kinase (JAK)/STAT3 pathway inhibitors, which has definite STAT3 inhibitory activity. We examined the effect of LZJ541 on the proliferation of HepG2 and PC-3 cells by MTT assay in vitro, detected the effect of LZJ541 on the expression of STAT3-related proteins in HepG2 cells by Western blot, and measured the effect of LZJ541 on the apoptosis and cell cycle arrest of HepG2 cells via flow cytometry. The results indicated that LZJ541 significantly inhibited the activation of STAT3 signaling pathway and restrained the proliferation of HepG2 cells. Its half maximal inhibitory concentration (IC₅₀) was 13.8 μmol·L^-1, which was much lower than that of PC-3 cells (with low STAT3 expression, IC₅₀: 41.99 μmol·L^-1), LZJ541 can also inhibit the phosphorylation of STAT3 in HepG2 cells, thereby inducing apoptosis and cycle arrest and then exerting anti-tumor effects. In conclusion, LZJ541 has a certain anti-tumor effect in vitro, which provides an experimental basis for the development of new STAT3-targeted anti-tumor drugs around this kind of compounds.

Objective To investigate the anti-inflammatory and analgesic effects of extracts from Garden Balsam seeds in order to provide theoretical basis for rational clinical application. Methods The mouse ear swelling induced by xylene, the body torsion induced by acetic acid or hot plate method, the rat foot swelling induced by egg white and granuloma model were set up to study anti-inflammatory and analgesic effects of extracts from Garden Balsam seeds. Results The water extracts at the middle and high doses reduced the body torsion counts of mice (P<0. 05), alleviated the rat toe swelling, but did not affect mice ear swelling. The extracts at the high dose prolonged pain threshold at 60 min (P<0. 01) and 90 min (P<0. 05); the alcohol ex-tracts at the middle and high doses significantly lowered body twisting counts (P<0. 01), attenuated the ear swelling of mice (P<0. 05), remarkably prolonged the pain threshold (P<0. 01)at 30 min, weakened the rat foot swelling, and elevated 60 min pain threshold at the high doses (P<0. 01), but the extracts exerted no effects on the rats granuloma. Conclusion The extracts from Garden Balsam seeds have anti-inflammatory and analgesic effects.
