4.Difference of gene expression between 8-cell early embryos and 8-cell compacted embryos in mice.
Journal of Central South University(Medical Sciences) 2007;32(2):252-258
OBJECTIVE:
To investigate the differences of gene expression before and after the compaction to search for the control mechanism of compaction.
METHODS:
8-cell embryos (626 mice) and 8-cell compacted embryos (437 mice) were collected from Kunmingbai mice, respectively. The cDNA of early 8-cell embryos was used as a tester, and the cDNA of 8-cell compacted embryos as a driver. Differences of gene expression were investigated between early 8-cell embryos and 8-cell compacted embryos using combining SMART (switching mechanism at 5o end of the RNA transcript) -PCR and suppression subtractive hybridization (SSH).
RESULTS:
Four hundreds and seventy eight positive clones were obtained, of which 384 clones with a range of 200~1000 bp and low redundancy were selected for further analysis. Eighty-three ESTs (expressed sequence tags) of the genes expressed differently were gained between early 8-cell embryos and 8-cell compacted embryos. Bioinformatic analysis showed that among the screened 83 putative positive ESTs, 51 ESTs matched 36 known genes, 27 ESTs matched 7 hypothetical genes, and 5 ESTs were new. Genes, which were up-regulated during the compaction, included cytoskeleton(4.8%); enzymes (9.6%); transcriptional factor/regulatory factor (12%); binding protein, protein-protein interactions and transport(13.3%); structural constituent of ribosome (14.5%); pluripotency associated gene(27.7%), and molecular function unknown(18%). Novel ESTs and 17 ESTs which matched the hypothetical genes were put into GenBank with accession numbers.
CONCLUSION
Combining SMART-PCR and suppression subtractive hybridization is an efficient method to investigate the gene expression difference in a few samples. During the compaction, the genes maintaining cell pluripotentiality express actively, which may be related to maintaining the embryonic cell totipotence in the differentation environment. All the 17 ESTs might be novel genes related to the compaction in the compacted embryos.

Objective To investigate the prevalence of endemic fluorosis in Guide county of Qinghai province, in order to provide appropriate measures to monitor and control the disease. Methods Damo, Wenquan, Baoning villages(water source has been changed) and Taiping village(water source has not been changed) in Guide county were involved in the study in 2008. One tap water sample was collected in dry and rainy seasons, respectively. Water fluoride was tested in accordance with the "Standard Test Methods for Drinking Water" (GB/T 5750.5-2006); of all the children aged 8 to 12, dental fluorosis was diagnosed using Dean criteria; 6 copies of urine samples were collected in each age group, urinary fluoride was measured using fluoride ion-selective electrode (WS/T 89-1996). According to the "Clinical Diagnostic Criteria of Endemic Skeletal Fluorosis "(WS 192-2008), clinical skeletal fluorosis was determined in adults over the age of 16 by X-ray examination for 10 people in each selected village. Results The mean water fluoride was 0.58,0.38,2.28,0.37 mg/L in Damo, Wenquan, Taiping, and Baoning villages, respectively, and that of Taiping village exceeded the national standard(1.0 mg/L). One hundred and ninety-three children aged 8-12 were checked, the detection rate of dental fluorosis was 49.74% (96/193); urine samples of 116 children were tested, median urinary fluoride was 1.49 mg/L A total of 1503 adults over the age of 16 were examined, the clinical detection of skeletal fluorosis was 51.63%(776/1503); a total of 82 people were X-rayed, X-ray detection of skeletal fluorosis was 20.73%(17/82). The characteristic of X-rays were degeneration and ossification of interosseous membrane. Conclusions Prevalence of dental fluorosis of children and adult clinical skeletal fluorosis are higher. The endemic fluorosis is still comparatively serious. Prevention efforts need to be further strengthened.

By using whole blood selenium, 24 hr urinary selenium and hair selenium contents as the indices of assessing human selenium status, it was found that the populations in the endemic areas of Keshan disease were practically in a selenium poor status. The selenium contents in locally grown staple grains and daily diets in the endemic areas were also lower than those in the non-endemic areas. In an area covering a cross section of Keshan disease geographic belt in our country, the hair selenium contents of agricultural populations were measured. The results indicated that all the hair selenium contents in the endemic sites were always at a lower level, whereas those in the non-endemic sites distant from the endemic areas were generally at a higher level; they decreased gradually until the endemic areas were reached; and finally, along the contiguous region of the endemic and non-endemic areas they were insignificantly different. The hair selenium contents among the agricultural populations were significantly lower than those among the non-agricultural ones in the same endemic areas. However, no regular correlation had been observed between the seasonal prevalence of Keshan disease and the variation of hair selenium contents in the same populations living in the same endemic sites. It is considered that the endemic areas of the disease seem to be a Se-deficiency belt, and Se-deficiency probably might be a pathogenic geo-gen in the prevalence of Keshan disease.

Chinese Journal of Traumatology 2003;6(3):171-173
OBJECTIVETo explore the management of thoracic spinal trauma (TST) associated with closed thoracoabdominal injuries (CTAI).
METHODSA retrospective study was made on 259 patients with TST admitted to our department as an emergency treatment from January 1996 to June 2001. We summarized the clinical features of TST associated with CTAI.
RESULTSAmong 259 patients with thoracic spinal trauma, 112 were associated with CTAI. Traffic accident was the most common cause. The force causing upper TST was more violent than that causing the lower. Pulmonary complications were the leading cause of death in this group. Surgery could not improve neurological function for completely paraplegic patients.
CONCLUSIONSThe reason that upper TST has the tendency to be associated with CTAI is its special anatomical feature. Routine ultrasonic examination can avoid misdiagnosis of latent closed abdominal injuries associated with spinal injury. The presence of potential injuries, especially CTAI, should be considered when deciding whether or not to perform surgery early.

The shape and structure of granules are controlled by the granulation process, which is one of the main factors to determine the nature of the solid dosage forms. In this article, three kinds of granules of a traditional Chinese medicine for improving appetite and promoting digestion, namely, Jianwei Granules, were prepared using granulation technologies as pendular granulation, high speed stirring granulation, and fluidized bed granulation and the powder properties of them were investigated. Meanwhile, synchrotron radiation X-ray computed micro tomography (SR-µCT) was applied to quantitatively determine the irregular internal structures of the granules. The three-dimensional (3D) structure models were obtained by 3D reconstruction, which were more accurately to characterize the three-dimensional structures of the particles through the quantitative data. The models were also used to quantitatively compare the structural differences of granules prepared by different granulation processes with the same formula, so as to characterize how the production process plays a role in the pharmaceutical behaviors of the granules. To focus on the irregularity of the particle structure, the box counting method was used to calculate the fractal dimensions of the granules. The results showed that the fractal dimension is more sensitive to reflect the minor differences in the structure features than the conventional parameters, and capable to specifically distinct granules in structure. It is proved that the fractal dimension could quantitatively characterize the structural information of irregular granules. It is the first time suggested by our research that the fractal dimension difference (Df,c) between two fractal dimension parameters, namely, the volume matrix fractal dimension and the surface matrix fractal dimension, is a new index to characterize granules with irregular structures and evaluate the effects of production processes on the structures of granules as a new indicator for the granulating process control and optimization.

10.Two-step image registration based on control volumes.
Journal of Southern Medical University 2011;31(11):1801-1805
OBJECTIVETo increase the accuracy of beam targeting and improve the dose distributions in radiotherapy using a robust image registration algorithm based on control volumes.
METHODSControl volume mapping and thin-plate spline deformable transformation were combined. The use of the control volumes increased the accuracy for the deformable registration. A bi-directional mapping method was also employed to correctly match the control volumes.
RESULTSThe clinical images were simulated using the proposed two-step image registration algorithm. The simulation results indicated that the registration algorithm was robust and universal, even in cases of obvious deformation.
CONCLUSIONSThe algorithm provides a more convenient and robust resolution as compared to manual landmark-based methods and single-step deformable transformations, and may help in automatic image registration in radiation therapy.
