1.The Effect of Sertoli Cells Feeders on Mouse Spermatogonial Stem Cells Cultured in vitro

Objective Spermatogonial stem cells(SSCs)dissociated from 2~5 days postpartum mice were cultured on Sertoli cells feeders,to study the effect of Sertoli cells feeders on culture of mouse spermatogonial stem cells.Methods Specific markers CD9 of mouse SSCs cultured serum-free StemPro-34 SFM culture medium were identified by immunohistochemical assay.Result During the first week of culture,on Sertoli cells feeders or STO feeders,the biologica1 behaviors of spermatogonial stem cells showed no obvious difference.After a week of culture,compared with control,there were more number of spermatogonial stem cells remained when they were cultured for 60 days.These cells were expressed CD9 positive.In conclusion Sertoli cells can be used as feeders not only to promote survival but also renewal of spermatogonial stem cells.

3.Reconstruction of medial canthal skin defects with local flaps: 15 case report

Objective To reconstruct skin defects with local flaps after resection of tumors. Methods From October 2006 to December 2009, medial canthal defects in 15 patients were repaired with local flaps in the hospital. The size of the defects varied from 1 cm × 1 cm to 1.8 cm × 2.0 cm. According to the size and location of the defects, different local flaps such as glabellar flaps ,rotation flaps, advancement flaps, nasolabial flaps and combination of these local flaps were selected and designed to repair the skin defects. Results Of the 15 patients, 12 received the reconstruction of skin defects with single flap, the other 3 with two flaps; totally,18 local flaps were designed and applied. All the flaps survived with primary healing postoperatively. A followup in 11 patients for 6 to 12 months showed that the color and texture of flaps were similar to those of surrounding tissues and incision line scar was inconspicuous. Medial canthal contours were restored without distortion of surrounding structures. Satisfactory function and aesthetic outcomes were achieved. Conclusions In the repair of inner canthus skin defects, to maintain and restore the "cosmetic unit" integrity of inner canthus by using suitable local flaps would lead to a satisfying restoration of function and appearance.
4.Rice Straw Degradation with White Rot Fungi and Cellulose Multienzyme Produced by Aspergillus niger

Rice straw degradation with white rot fungi and cellulose multienzyme was studied. The results indicated that the LiP, MnP activity produced by P. chrysosoporium 172 could reach 28.3U/g and 12.6U/g respectively under suitable culture condition. And the lignin was degraded efficiently in contrast with the cellulose and hemicellulose in rice straw solid fermentation. Following treatment with white rot fungi, using multienzyme produced by A. niger NL-1 could greatly accelerate the decomposition of rice straw. The decomposition rate of cellulose, hemicellulose, lignin and the loss rate of dry material were 53.8%,57.8%,44.5% and 46.3% respectivity when hydrolysis rice straw 48h with cellulase multienzyme (including 3 IU /g rice straw) after culture P. chrysosoporium 172 for 10 days. Scanning electron microscope analysis showed the cell wall of rice straw was destroyed severely, and the whole tissue got loosely. These results demonstrated the rice straw had been decomposed efficiently and completely.
5.Decoration reconstruction for finger defect in emergency

Objective To discuss methods of decoration reconstruction for finger defect in emergency and to observe the elinical effects.Methods Of the 41 cases of finger injuries of different degrees,15 were repaired with part of the skin flaps of the big toenails or skin flaps of the second toenalis,8 were repaired with part of the skin flaps of the big toenails,7 were reeonstructed with the second tiptoes,11 were repaired with the abdominal skin flaps of the big toes or lateral flaps of the second toes.Results All the 41 fingers sur- vived.One skin flap of the big toe was somewhat swelling and a decorating operation was performed.The 4~18 months of follow-up visitation of the rest cases revealed good function and shapes.No obvious functional ab- norality was found in the donating feet.Conclusion Various kinds of decoration reeonstruetion for finger defects are available to recover the hand shape and function as much as possible.

Background Stromal cell-derived factor-1_?(SDF-1_?)has been demonstrated to be essential for stern cell mobilization/homing.Recent evidence indicates that SDF-1_? has been expressed in injured carotid arter- ies.Besides,high SDF-1_? plasma levels are clinically associated with stable coronary artery disease.Objective To investigate whether SDF 1 involves in mobilization of endothelial progenitor cells(EPC)and reendothelialization after vascular injury.Methods SDF-1_? was detected by RT-PCR and Western blot in carotid arteries of mice at different time points after wire-induced injury.SDF-1_? determination in peripheral blood samples and BM was per- formed by SDF-1_? enzyme-linked immunosorbent assay(ELISA)kit.EPC in peripheral blood collected at different time points after vascular injury were quantified by flow cytornetry.In subgroup,blocking SDF-1 rnonoclonal anti- body was injected,peripheral blood EPC were quantified after vascular injury and reendothelialization of injured ar- teries was determined 14 days later.Results Expression of SDF-1_? was evident at day 1,and peaked at day 3 after arterial injury.A rise in plasmatic concentration of SDF-1_? and a significant reduction of SDF-1_? in bone marrow concentration was noticed at all time points following injury.The amount of circulating EPC was increased shortly after induction of vascular injury and persisted up to 7 days(P
7.Blood-brain barrier penetration of cefepime after neurosurgery.

BACKGROUNDIt has been confirmed that the concentration of cefepime in cerebrospinal fluid (CSF) could reach the 10% of its concentration in plasma, exceeding the inhibitory concentration to 90% of organisms (MIC(90)) for common bacteria. However, the blood-brain barrier (BBB) penetration ability of cefepime is still unclear. The aim of this study was to measure the CSF concentration of cefepime in patients after neurosurgical operations, and to determine the penetration of the drug through an incomplete BBB.
METHODSEight patients who received ventricular drainage (VD group) and 5 who underwent lumbar puncture drainage (LPD group) were enrolled into this study. Cefepime (2 g) was injected intravenously in 30 minutes after the neurosurgeries. The concentrations of cefepime in the CSF and plasma were measured by high-pressure liquid chromatography (HPLC) at different time points.
RESULTSThe CSF concentrations of cefepime at different time points in the VD group were significantly higher than those in the LPD group (P < 0.05). In the VD group, the concentration of cefepime in CSF reached the peak ((22.54 +/- 14.06) microg/ml) at 1 to 2 hours after the injection, while in the LPD group at 4 hours ((5.61 +/- 3.73) microg/ml). In both groups, the peak was higher than the MIC(90) of most common bacteria in intensive care unit. The ratio of CSF to plasma cefepime concentrations ranged from 0.30 to 2.14 in the VD group and 0.03 to 1.14 in the LPD group.
CONCLUSIONAfter neurosurgeries, CSF concentration of cefepime can reach a therapeutic level. Thus, the drug could be used to prevent and treat postoperative intracranial infection.

8.Effect on synthesis of nitric oxide in myocardium by local cryoablation.

OBJECTIVETo study the effect on synthesis of nitric oxide in myocardium by local cryoablation and to investigate its mechanism.
METHODSMyocardium was cryoablated locally by a probe cooled to -60 degrees C and rewarmed by normal salt solution, nitric oxide and its synthesis enzyme were measured before and after cryoablation. L-arginine or methylene blue was added before and during cryoablation and the effect of these drugs on synthesis of nitric oxide was studied.
RESULTSNitric oxide and its synthesis enzyme decreased after cryoablation; L-arginine preserved the synthesis of nitric oxide and methylene blue inhibited the synthesis of nitric oxide. However, nitric oxide in serum did not change.
CONCLUSIONNitric oxide and its synthesis enzyme in myocardium decrease after cryoablation.

9.Synchronous or metachronous primary cancers of other organs in 74 patients with gastric cancer.

OBJECTIVETo study the incidence, clinicopathological characteristics, diagnosis, treatment, and prognosis of synchronous or metachronous primary cancers in patients with gastric cancer.
METHODSClinical data of 4426 patients with gastric cancer in our hospital from 1996 to 2007 were reviewed.
RESULTSSeventy-four (1.7%) patients had synchronous or metachronous primary cancer of other organ, of whom 10 were synchronous and 64 were metachronous. Colorectal cancer was the most common type of primary cancer in other organs (43.8%), followed by breast cancer (16.3%). The mean time interval between gastric cancer and metachronous primary cancer was 82.2 (3-354) months. The mean age at the diagnosis of gastric cancer was 61.2 (33-84) years. The 5-year overall survival rate was 42.3%. The 5-year survival rates in patients with synchronous cancer, pre-metachronous cancer or post-metachronous cancer were 15.2%, 42.9% and 51.3%, respectively. Causes of death were primary cancers of other organ in 11 patients, gastric cancer in 24, and renal failure in 1 patient.
CONCLUSIONSPrimary cancer of other organ should be considered in the management of gastric cancer. Aggressive treatment should be used for the second primary cancer. Gastric cancer is the main cause of death in these patients.

10.New method of left ventricular intubation in anaesthetised rats.

AIMTo search for a new method of left ventricular intubation in rat.
METHODSThe wire of percutaneous transluminal coronary angioplasty (PTCA) was put into carotid artery through external carotid artery. Then supported by guide wire, the PE50 tube was advanced into left ventricle.
RESULTSLeft ventricular intubation with PTCA guide wire could be performed in 30 rats and successfully repeated in 27 animals.
CONCLUSIONThe new left ventricular intubation technology in rats is simple and provides a reproducible method.
