1.Changes of Cerebral Haemodynamics in Neonates with Hypoxic-Ischemic Encephalopathy Treated with Shengmai Injection

Objective To study the changes of cerebral haemodynamics in neonates with hypoxic-ischemic encephalopathy(HIE) treated with Shengmai injection compared with citicoline,and explore the clinical interventions of Shengmai injection on HIE.Methods Fifty patients with HIE were randomized into 2 groups: shengmai group treated with Shengmai injection(n=30),and citicoline group treated with citicoline(n=20).The haemodynamic parameters included peak systolic flow velocity(PSFV),end-diastolic flow velocity(EDFV),pulsation index(PI) and resistance index(RI) of anterior and middle cerebral artery were detected by color Doppler ultrasound before treatment and on 7~(th) day after treatment respectively.Control group(n=50) compared with the two groups.Results PSFV,PDFV and RI of HIE patients were significantly lower than those of control group(P
2.Inhibition of NF-kB enhanced X-ray irradiation induced apoptosis in human non-Hodgkin lymphoma

Objective To investigate the role of NF-kB in radiation-induced apoptosis ot human non-Hodgkin lymphoma (NHL) cells and the mechanism involved.Methods Three human non-Hodgkin lymphoma cell lines,Namalwa,Ramos,and Raji cells were divided into control,IR and IR + QNZ ( 10nmol/L) groups,respectively.Annexin-V kit was used to determine cell apoptosis. Protein expression levels of Survivin,Bax,Bcl-2 and cleaved Caspase-3 were evaluated by Western blot.Survivin mRNA was quantified by real-time PCR.Results Inhibition of NF-kB by QNZ pretreatment significantly enhanced X-ray induced apoptosis in human NHL cells in a dose-dependent manner (t =2.93 - 12.52,P < 0.05).At the same time,QNZ significantly reversed the expression levels of Survivin protein and mRNA that were upregulated by radiation(t =3.29 - 16.72,P < 0.05).QNZ also increased the levels of Caspase-3 and proapoptotic protein Bax,but reduced anti-apoptotic protein Bcl-2 expression level and hence the ratio of Bcl-2/Bax.Conclusions Inhibition of NF-kB could enhance radiation-induced cell apoptosis in human NHL cells through down-regulating Survivin expression and decreasing Bcl-2/Bax ratio.
3.Proteomic analysis of the serum from patients with acute rejection after liver transplantation

Objective To investigate the protein markers that specifically expressed in patients with acute rejection (ACR) after liver transplantation, and to explore preliminarily the mechanisms. Methods Serum samples from three patients with pathologically confirmed ACR after liver transplantation in Tianjin First Central Hospital were collected as ACR group. Three serum samples from patients with normal liver function indicators after liver transplantation were collected as No-ACR group. And six serum samples from healthy examination were mixed with equal amount as healthy control group. Isobaric Tags for Relative and Absolute Quantitation (iTRAQ) was employed to separate, screen and identify the differentially expressed proteins between three groups. KEGG and STRING software were applied to deeply analyze the data of three groups. Results A total of 88 differentially expressed proteins were found between ACR group and healthy control group. There were 39 differentially expressed proteins between No-ACR group and healthy control group. Ten differentially expressed proteins were acquired between ACR group and No-ACR group. Comparing 88 and 10 differentially expressed proteins, 9 proteins were the same. Among 88 differentially expressed proteins, 30 of them showed a direct interaction, and can be positioned in 13 signaling pathways based on KEGG and STRING software. Fourteen (46.67%) of the 30 proteins were located in the complement and coagulation cascade pathway. Among 39 differentially expressed proteins, which were detected between No-ACR group and control group, 10 proteins showed a direct interaction including 9 proteins concentrated in the complement and coagulation cascade pathway. Conclusion By proteomic analysis, nine differentially expressed proteins are obtained, which may be regarded as the candidate bio-markers for ACR early diagnosis after liver transplantation. The complement and coagulation cascades system is significantly adjusted after liver transplantation, indicating this pathway plays an important role in the occurrence of ACR.
4.Effect of tetrandrine on proliferation of human pterygium fibroblasts

AIM: To investigate the effect of tetrandrine (Tet) on proliferation of human pterygium fibroblasts (HPF) in vitro and to search for a new method to prevent the recurrence after pterygium surgery.METHODS: With different concentrations (0 to 160μmol/L) of Tet acting on HPF cultured in vitro, the impact was observed at 24, 48, 72 hours respectively after Tet intervention. The MTT method was used to assay the biologic activities of Tet and inhibitive rate of cell growth. The expression of proliferating cell nuclear antigen (PCNA) in each group was detected by immunohistoche-mistry before and after Tet intervention.RESULTS:With different concentrations of 20, 40, 80 and 160μmol/L and acting for 24 to 72 hours, Tet could inhibit the proliferation of HPF in a dose- and time-dependent manner (P<0.05). After the intervention of Tet, the expression of PCNA protein declined. When the concentration of Tet was in the range of 20 to 160μmol/L, it was able to inhibit the expression of PCNA in a concentration-dependent manner (P<0.05).CONCLUSION:Tet could significantly inhibit the proliferation of pterygium fibroblasts, and the inhibitive action was in a dose- and time-dependent manner within a certain range of concentration. But in high concen-tration (>160μmol/L), Tet would have cytotoxity.
5.The relationship between juxtapapillary duodenal diverticulum and recurrence of biliary stones

Objective To explore whether the presence of juxtapapillary duodenal diverticula (JPDD) risks biliary stone disease and recurrence.Methods 829 patients undergoing ERCP in our hospital from Aug 2008 to Dec 2012 were divided into four groups:biliary stone disease (n =609) non-stone biliary abnormality (n =124) common bile duct malformation with cholelithiasis (n =38) and normal control group (n =58).There were 206 patients with JPDD and 623 patients without JPDD.Biliary stoneformation,post-ERCP pancreatitis,cannulation failure,and stone recurrence were compared between those with JPDD and those without.Results The incidence of JPDD in biliary stone disease group (27.8%) was significantly higher than in non-stone biliary anatomical abnormality group (18.5 %) (x2 =4.512,P < 0.05).In biliary stone disease group,rates of post-ERCP pancreatitis were significantly higher in JPDD cases (33.7%) compared to those without JPDD (13.8%) (x2 =30.841,P < 0.05).The cannulation failure rate was also higher in patients with JPDD (15.4%) compared to JPDD negative (6.8%) (x2 =0.731,P <0.05).Recurrence rates in biliary stone disease were significantly higher in patients with JPDD (19.5%) when compared to JPDD-lacking individuals (9.3%) (x2 =14.51,P < 0.05).Conclusions JPDD is a risk factor for biliary stone formation.JPDD also is associated with post-ERCP pancreatitis,cannulation failure and biliary stone recurrence.
6.ELECTRON MICROSCOPIC STUDY OF THE CHANGES OF CONNECTIVE TISSUE CELLS DURING THE SKIN WOUND HEALING IN RATS

Open skin wounds were made in 28 Wistar rats, and the changes of the connective tissue cells were observed by electron microscopy during wound healing. The fibroblasts and macrophages were increased in the early stage, later on the fibroblasts might be transformed into myofibroblasts which became preponderant cells in the wound gradually and led to wound contraction and scar formation. The intercellular substances were phagocytosed by fibroclasts simultaneously. The mast cells were closely apposed to the fibroblasts or myofibroblasts and represented active functions, such as degranulation. Considerably they released the granule contents to the extracellular space and effected the other cells functions. It shows that the cells mentioned above interplay and take part in the wound healing.
7.Risk factors for rotator cuff tears with long head of bicep tendon lesions and their effects on preoperative function

Objective:To investigate the risk factors of rotator cuff tears combined with long head of bicep tendon (LHBT) lesion and its effects on preoperative function.Methods:From January 2016 to January 2020, there were 680 patients with rotator cuff tears, including 260 males and 420 females, were included. The average age was 56.1±8.7 (range 27-74 years). There were 250 cases on the left side and 430 cases on the right side. There were 436 cases on the main side, while 274 cases had definite trauma before operation. The following were the Post rotator cuff tears classification, 133 cases in partial injury, 473 cases in small and medium size of rotator cuff tears, 74 cases in large and massive size of rotator cuff tears. The Habermayer-Walch classification of LHBT lesions was as following, 302 cases in type 0 (normal), 216 cases in type 1 (tendonitis), 29 cases in type 2 (dislocation and subluxation), 104 cases in type 3 (partial tear), 20 cases in type 4 (complete tear) and 9 cases in type 5 (SLAP injury above type II). The patient with rotator cuff tears were divided into partial tears group, medium and small tears group, large and massive tears group according to the classification of Post. The visual analogue scale (VAS), simple shoulder test (SST), Constant-Murley function score and the score of the University of California Los Angeles (UCLA) were recorded. The differences in age, sex, main side, trauma and LHBT lesion in patients with three types of rotator cuff tears were analyzed. The preoperative pain and function scores of patients with or without LHBT lesions in various rotator cuff tears were compared among the groups by t-test. The age, sex, main side, with trauma or not, and the classification of rotator cuff tears were analyzed by Logistic regression to investigate the risk factors of patients with rotator cuff tears with LHBT lesions. Results:There were 378 cases (55.6%) with LHBT lesions in 680 patients with rotator cuff tears, including 216 cases of tendinitis (57.1%), 104 cases of partial tear (27.5%), 29 cases of dislocation and subluxation (7.7%), 20 cases of complete tear (5.3%) and 9 cases of SLAP injury (2.4%). The incidence of partial injury, small and medium injury, large and massive rotator cuff injury combined with LHBT lesions were 35.3% (47/133), 57.5% (272/473) and 79.7% (59/74) respectively. The preoperative VAS score, UCLA score, Constant-Murley score and SST score were 5.20±1.52, 14.81±4.12, 41.45±4.93 and 4.56±1.96 respectively in rotator cuff tears group and 5.29±1.65, 14.34±4.01, 41.60±5.88, 4.47±1.97 in LHBT group ( P>0.05). In the partial rotator cuff tears group, the VAS score was 5.16±1.41 in patients with simple rotator cuff tears and 5.68±1.46 in patients with LHBT lesion ( t=2.004, P=0.047). Regression analysis showed that age ≥60 years and rotator cuff tears degree were the risk factors for rotator cuff tears with LHBT ( P<0.001). Conclusion:It is common for patients to have rotator cuff tears with LHBT lesions. The more severe the rotator cuff tears are, the higher the incidence of LHBT lesions will be. Patients with rotator cuff tears aged 60 and above are more likely to have LHBT lesions. However, the complications of LHBT could not affect the preoperative pain and functional scores of patients with rotator cuff tears.
8.EFFECT OF LOW LEVEL SOYBEAN ISOFLAVONES ON THE OXIDATIVE MODIFICATION OF LIPOPROTEINS IN HUMAN SERUM

Objective: To investigate the action of low level soybean isoflavones (genistin, genistein and daidzein) on the oxidative modification of lipoproteins in serum. Methods: After a system of lipoprotein oxidation mediated by Cu 2+ was established in a dilute serum, the effects of soybean isoflavones on the course and the end of lipoprotein oxidation could be reflected by monitoring the production of conjugated dienes and thiobarbituric acid-reactive substances (TBARS) respectively when isoflavones were added. Results: After 0.5-10 ?mol/L genistein, daidzein, genistin or ?-tocopherol was added into the lipoprotein system respectively before the oxidation initiated by Cu 2+ , the production of conjugated dienes or TBARS in the system was significantly reduced with a dose-dependent relationship. When the lipoprotein oxidation was initiated by Cu 2+ at 37 ℃ for 1 h or 1.5 h, soybean isoflavones also revealed strong inhibition on the oxidation in a weakening way. In comparison with soy isoflavones, ?-tocopherol had smaller inhibition on the production of conjugated dienes, but had promotion on the increase of TBARS. Conclusion: Lipoprotein oxidative modification in serum was weakened by low level soybean isoflavones, and its action after the oxidation initiated was more effective than that of ?-tocopherol.
9.Diagnosis,Treatment and Prognosis of Neonatal Intestinal Atresia

Objective To explore diagnosis,treatment and prognosis of intestinal atresia(IA) in neonates,so as to increase the healing rate.Methods Surgical treatment was given to 29 cases of congenital IA from Apr.2004 to Nov.2008.Comprehensive analysis was carried out on clinical manifestations and flat X-ray.All cases were with vomiting,15 cases were with normal fetal faeces.Stomach and intestinal atresia were observed in the upper abdomen in 19 cases,fluid levels in generous stomach and expended intestines were seen in 21 cases by X-ray.All children underwent operation,including ileum atresia(16 cases),duodenal atresia(7 cases),and jejunal atresia(6 cases).The types of atresia were type Ⅰ(9 cases),type Ⅱ(9 cases),type Ⅲa(9 cases),type Ⅲb(1 case) and type Ⅳ(1 case).Nine cases were treated with septum resection and then anastomosed longitudinally,3 cases underwent end-to-end anastomose and 17 underwent end-to-back anastomose.All cases were served in intravenous hyperalimentation.Results All cases lived postoperatively,and 1 child gave up for postoperatively treatment.Twenty-eight cases were cured.There was 1 case of obstruction of anastomatic,and was cured by non-operatively treatment.There was no anastomatic leakage and wound disruption.Twenty-six cases were followed up for 3 months and up to 4 years.These symptoms had no relapse.Conclusion Early diagnosis,appropriate surgical procedure and surrounding operation management are crucial for improving the cure rate.
