5.Efficacy and safety of posterolateral knee inverted“L”shaped pathway when placing the supporting plate

BACKGROUND:Studies have shown that tibial plateau posterolateral fractures can be treated by posterolateral knee approach, however, there is no clear anatomical research to deeply evaluate and analyze this approach. OBJECTIVE:To evaluate the efficacy and safety of posterolateral knee inverted“L”shaped pathway in invloving tibial plateau posterolateral fractures. METHODS:Eight fresh frozen adult corpses, 16 knees side, were al dissected using posterolateral inverted“L”shaped pathway. During the dissection, the exposure range was observed and important parameters of anatomical structure were measured. RESULTS AND CONCLUSION:The pathway may be ful y exposed to the posterolateral aspect of tibial plateau and posterior cruciate ligament tibial insertions. The operations completed by out team did not create any obvious interference to superior tibiofibular joint, fibular head and posterolateral corner structure. The exposed mean length of common peroneal nerve in incision was 56.48 mm, with a mean angle of 14.7° tilt towards the axis of the fibula. The mean distance between the neck of the fibular and fibular head tip was 31.26 mm, an average of 42.18 mm to the joint line. The mean distance between the opening of the interosseous membrane and the articular surface was 48.78 mm. The divergence of the fibular artery from the posterior tibial artery was an average of 76.46 mm from articular surface. These results confirm that posterolateral inverted“L”shaped pathway met the requirements of anatomical reduction and buttress fixation for posterolateral tibial plateau fracture. Exposure of the common peroneal nerve can be minimized or even avoided by modifying the skin incision. Because the popliteal artery branches anterior tibial artery passed through interosseous membrane hole and peroneal artery and then separated from the posterior tibial artery, pathways dissection to distal deep area should be carried out careful y. Placement of a posterior buttressing plate carries a high vascular risk if the plate is implanted beneath these vessels.
6.Serum proteomic study on hypertension patients with Gan-Dan damp-heat syndrome.

OBJECTIVETo study the serum proteomics in hypertension patients with Gan-Dan damp-heat syndrome (GDDH) for tentatively find special proteins associated with the syndrome.
METHODSStudy was performed in 60 hypertensive patients and 39 healthy persons as control. In the patients enrolled, 40 were differentiated as GDDH syndrome and the other 20 as non-GDDH syndrome. Their serum proteins were captured by weak cation nano-magnetic beads, and proteomic fingerprint was made by matrix assistant laser demodulation ionizing time-of-flight mass spectrometry (MALDI-TOF-MS) through mapping with protein chip reader type PBS II-C. After all the proteomic fingerprints being analyzed by Biomarker Wizard 3.1, the special expressed proteins for GDDH syndrome were identified by Biomarker Patterns Software 5.0 to create the syndrome decision model.
RESULTSTotally, 182 difference protein peaks between patients of GDDH and healthy persons (P<0.05); and 132 difference protein peaks between patients of GDDH and non-GDDH were detected (P<0.05). A decision model consisted 5 screened out protein peaks with mass-to-charge ratio of 2761.555, 6624.362, 2487.192, 2461.610 and 2744.318 was created, which could well differentiate the GDDH syndrome, with the sensitivity of 96.55%, specificity of 90%, false positive rate of 10% and false negative rate of 3.45%. Further blind test for prospective check showed its sensitivity being 81.82%, specificity 89.66%, false positive rate 10.34% and false negative rate 18.18%.
CONCLUSIONThe differently expressed protein is the material foundation of GDDH syndrome; molecular biological decision model established on the basis of this foundation can offer a tool for making Chinese medicine syndrome differentiation more objectively and accurately.

7.Design, synthesis and pharmacological investigation of isoindoline derivatives as 5-HT/NE double reuptake inhibitors.

A series of isoindoline derivatives were designed, synthesized, and evaluated for their double inhibitory activities. All of them were new compounds, and their structures were confirmed by 1H NMR and HR-MS. Preliminary in vitro pharmacological tests showed that all compounds exhibited 5-HT or NE reuptake inhibition activity. Among the tested compounds, compound I-3 exhibited potent inhibitory activity against 5-HT and NE reuptake in vitro, and exhibited potent antidepressant activity in vivo. These compounds designed can be further optimized for finding more potent 5-HT/NE dual reuptake inhibitors and antidepressant candidates as well.

8.Effects of acupuncture and continuous multiple points pulse stimulation on linguistic frequency hearing loss in the patient of sensorineural hearing loss.

OBJECTIVETo explore effective therapeutic methods for sensorineural hearing loss (SNHL).
METHODSThirty-four cases of mild and moderate SNHL who were definitely diagnosed with pure tone test and by otologists, including 24 cases of insufficiency of the liver and kidney, 10 cases of excess of liver-fire. They were treated with acupuncture at main points, Ermen (TE 21) or Tinggong (SI 19) or Tinghui (GB 2), and Yifeng (TE 17), Shenting (GV 24), Baihui (GB 20), Houding (GV19), Touwei (ST 9), Conger 1-3, Congnao 1-2; and adjuvant acupoints: Huangshu (KI 16), Qihai (CV 6), Guanyuan (CV 4), Taixi (KI 3) were added for insufficiency of the liver and kidney; Zhongwan (CV 12), Tianshu (ST 25), Yanglingquan (GB 34) and Taichong (LR 3) were added for excess of liver-fire. Sequential multipoint pulse stimulation was adopted, twice each week, for 20 consecutive weeks (40 sessions) and then pure tone test was carried out repeatedly.
RESULTSOf 34 cases (58 affected ears), 13 cases were cured and 21 improved, and the various linguistic frequency hearing threshold pure hearing values before and after treatment increased significantly (P < 0.05).
CONCLUSIONAcupuncture and continuous multipoint pulse stimulation has definite therapeutic effect on linguistic frequency hearing loss in the patient of SNHL.

9.Detection of Segregation of Foreign Genes in Transgenic Wheat Lines by Multiplex Polymerase Chain Reaction

The method of multiplex PCR was set up to identify two or three transgenes in one reaction such as uidA and bar; uidA and IDx5 or uidA, bar and 1Dx5 genes. Three sets of primer pairs which was specific to each of these three genes respectively were designed and synthesized. Recombinant plasmids pAHC25 and p1Dx5 harboring uidA + bar and 1Dx5 gene separately were used as template DNA in the process of optimizing an multiplex PCR reaction. The optimal annealing temperature for uidA and bar MPCR is range from 57. 1℃-62. 3℃ , for uidA and 1Dx5 is range from 60℃ to 60. 6℃ , and for uidA、bar and 1Dx5 range from 57. 0℃-58. 4℃. The amount of template for MPCR is twice as much as that for simplex PCR, while the concentration of primers is the same with simplex one. Less than 50bp MPCR products can be separated clearly by 10% non-denaturalized polyacrylamid gel electrophoresis. Fourteen transgenic wheat lines were tested by multiplex and simplex PCR respectively, which shows the same results and hence presents that MPCR is the reliable, rapid and high-effective approach to detect foreign genes from transgenic plant.
10.The Influence of Peptidoglycan of Lactobacillus on Immune Function of Mouse

Mice was stimulated by peptidoglycan from Lactobacillus sp and detect cytokines production。It was found that PG induced the production of inflammatory cytokines (IL-1,TNF-? in peritoneal macrophages,IFN-? in spleen cell)and did not induce the IL-2 production in spleen cell. Affymetrix MOE430A genechip was used to analyze changed gene expression of immune cells. It was found that expression of cytokines and related genes were changed under peptidoglycan administration. This might induced by activation of TLR-NF-?B signal pathway.
