1.Surgical treatment of refractory slow-transit constipation

Objective To evaluate the method and outcome of surgical treatment for refractory slow-transit constipation (STC). Methods Clinicopathologic data of 24 patients with severe STC which underwent (colectomy) in recent years were retrospectively analyzed. Results 18 of the patients underwent pancolectomy,and 6 underwent subtotal colectomy. All of the 14 STC patients complicated with outlet obstructive (constipation)(OOC) were corrected preoperatively by related surgery. The curative rate of surgical (treatment) was 95.8%,with an average of(3?1.9) times of bowel motion per day. The incidence of (abdominal) distention decreased from 75% to 12.5%. One patient who underwent ileorectal anastomosis (developed) recurrent constipation. Conclusions Colectomy produces satisfactory results in the majority of (patients) with refractory slow-transit constipation.Patients complicated with OOC should undergo energetic (preoperative) correction therapy,in order to ensure a satisfactory surgical outcome.

Objective To study the effects of pulsed electromagnetic fields(PEMFs)of 15Hz and lmT on osteoblast differentiation of bone marrow mesenchymal stem cells.Methods The bone marrow mesenchymal stem cells were isolated and cultured in vitro,the third passage cells were harvested and half of them was exposed to PEMFs,another half exposed to sham PEMFs served as the control.The activity of ALP was measured.And the ex- pression of osteoblast and adipogenesis was measured with the semi-quantitative RT-PCR,adipogenic differentiation was observed with the Oil Red O Staining.Results When compared with the control,PEMFs of 15 Hz and 1 mT significantly increased the activity of ALP(P

AIM:To compare the higher-order aberrations between implantations of AcrySof and AcrySof ReStor multifocal aspherical intraocular lens(IOL) with various pupil diameters.METHODS:Fifty-four patients(62 eyes)with bilateral senile cataracts were retrospectively selected.Patients were operated with phacoemulsification and IOL implantation.They were divided into two groups based on the IOL implantation of AcrySof IQ and AcrySof ReStor.Wave front aberration:spherical aberration(C12)and the root mean square of the total higher-order aberration(RMSh)were observed 3 months after surgery.RESULTS:The larger the pupil was, the higher the C12 and RMSh were in the eyes (P<0.01).There were no statistical differences in C12 or RMSh between two groups at 5, 6 or 7mm pupil diameters (P>0.05). CONCLUSION:There are no differences between AcrySof IQ group and AcrySof ReStor group at 5, 6 or 7mm pupil diameters.
5.Expression and location of ARMS2 protein in normal eye tissue

Background Researches determined that the alteration of A69S locus of age-related maculopathy susceptibility 2 ( ARMS2 ) gene is closely associated with the pathogenesis and progression of age-related maculopathy ( AM D ).However,the location of ARMS2 protein in normal eye tissue is still in controversy,therefore,its function is below understanding up to now.Objective The goal of this laboratory work was to investigate the distribution,expression and location of ARMS2 protein in normal adult retina and choroid as well as in retinal pigment epithelial (RPE) cells and lay a basis for exploring further its function in the protein level.Methods Ten donor eyeballs of normal adult male with the age from 28-42 years were collected in eye bank of Qingdao Eye Hospital.The frozen sections of the retina and choroid were prepared for the detection and location of ARMS2 in 3 eyes by immunofluorescence under the confocal laser microscope.The retina was isolated for the primary culture of RPE cells using explant culture method.The cells were then identified by CK32 antibody by immunofluorescence.The distribution and expression of the ARMS2 protein in retina,ehoroid and RPE cells were determined by immunofluorescence technique.Results ARMS2 protein was strongly expressed in retinal vessel,RPE cell layer,Bruch membrane and choroidal vessel,but weak expression was in retinal ganglion cell layer,inner nuclear layer,outer plexiform layer,outer nuclear layer and inner plexiform layer in the normal eyes.The primarily cultured cells appeared the polygon shape with the abundant pigment in cytoplasm.The immunofluorescence of the cells showed the positive response for CK32,exhibiting the green fluorescence granules in the cytoplasm.The positive expression of ARMS2 protein also was seen in the cytoplasm of RPE cells,appearing the red fluorescence.Conclusions ARMS2 protein mainly distribute and locate retinal and choroidal vessels,RPE cells and Bruch membrane in normal eye.
6.Protective effect of sodium pyruvate on ischemia/reperfusion injury of rats subjected to hemorrhagic shock.

AIMTo study the protective effect of sodium pyruvate on ischemia/reperfusion injury following hemorrhagic shock.
METHODSRat models of hemorrhagic shock were built up. When the shed blood was infused, the rats were also randomly provided by one of normal saline, glutathione and sodium pyruvate. Rats were killed 3 hours after the reperfusion, the activity of plasma lactate dehydrogenase (LDH) and glutamic-oxaloacetic transaminase (GOT), the level of tissue malondialdehyde (MDA) and the activity of tissue myeloperoxidase (MPO) were detected. Biopsy specimens were obtained to investigate morphological changes of the myocardial, hepatic, lung and renal tissue.
RESULTSThe activity of plasma LDH and GOT, the level of MDA of hepatic, lung and renal tissue and the activity of MPO of myocardial, lung and renal tissue decreased remarkably in group given sodium pyruvate compared with group given normal saline, and the effect of group given sodium pyruvate was more remarkable than group given glutathione.
CONCLUSIONThese data support the view that sodium pyruvate shows protective effect on ischemia/reperfusion injury following hemorrhagic shock. It is possibly relevant to scavenging of oxygen free radicals, reduction of neutrophil, and anti-inflammatory response.

7.Occupational stress status and influential factors of the peasant-workers in factory.

OBJECTIVETo investigate the occupational stress status and influential factors of the peasant-workers in factory.
METHODS410 peasant-workers in factory were tested by revised work requirement-control table and occupational stress measurement tools, and data were statistically analyzed by analysis of covariance.
RESULTSThe scores of work requirement, mental health in male peasant-workers were higher than those in female workers. The scores of everyday sense of stress in female workers were higher than those in male peasant-workers (P < 0.05). Workers with higher education level scored more in decision making and less in mental health than ones with lower education levels (P < 0.05). The workers divorced scored more in occupational stress factors, everyday nervous sense of stress and work future prospects than other marriage status workers (P < 0.05). Living with the family scored more in self-respect sense, mental health than other living styles (P < 0.05). The workers exposed to higher levels of noises have more scores in occupational stress factors, everyday sense of stress, and have less scores in decision making and work requirement than those exposed to lever levels of noises (P < 0.05 or P < 0.01). The workers exposed to higher level of powder or chemical toxin had more scores in occupational stress factors, work requirements and less scores in mental heath than those exposed to lever of powder or chemical toxin (P < 0.05 or P < 0.01).
CONCLUSIONSGender, educational level, marriage status, living styles and work environment have some influence on occupational stressors and strains. Self-respect sense and occupational stress factors can impact mental health and depression symptoms.

8.The impact of air temperature variation on the visits to emergency room in Shanghai

Objective To assess the association between air temperature and emergency room visits among patients covered by medical care program from ' third-grade' hospitals in Shanghai.Methods Generalized additive model (GAM) was used to analyze time series,and AR(P) was used to deal with auto correlation of time series.After controlling factors as both medium-term and long-term trends,day of the week,vocation,typical pneumonia and pollutants,the association between air temperature and emergency room visits in virtue of quadratic curve and differential coefficient principle were estimated.Results When air temperature was below 14.71℃,the increase of 95% confidence interval to relative risk in corresponding emergency room visits along with 1℃ increase of air temperature,was less than 1.However,when air temperature was above 19.59℃,the relative risk' s 95% confidence interval was greater than 1.When air temperature varied at the range of 14.71℃-19.59℃,the 95% confidence interval of the relative risk would include 1.Hence,air temperature range between 14.71℃-19.59℃,was called the optimum temperature range.Conclusion Our findings indicated that the current air temperature had an acute impact on the number of emergency room visits among patients covered by medical care program visiting those third grade hospitals in Shanghai.
9.Expression of CAS in hepatocellular carcinoma tissues and its relationship with HBV infection.

OBJECTIVETo explore whether the cellular apoptosis susceptibility (CAS) protein could serve as a pathologic marker for HCC diagnosis and the roles of CAS expression in HBV infection associated HCC.
METHODSThe expression of CAS protein in HCC and its paracarcinoma tissues, non-tumor liver cirrhosis and hepatitis tissues were detected by immunohistochemistry. Meanwhile, HBsAg, HBcAg and HBV DNA in HCC tissues with HBV infection were examined by immunohistochemistry and in situ hybridization respectively.
RESULTSThe expression of CAS protein was significantly higher in HCC than in its paracarcinomas tissues (P < 0.01), and higher in paracarcinomas tissues than in non-tumor liver cirrhosis and hepatitis tissues (P < 0.01). Poorly differentiated tumors immunochemically stained stronger than moderately or well differentiated (P < 0.01). CAS protein expression was significantly higher in HBV-infected HCC tissues than that of in non-HBV infection (P < 0.01). Meanwhile, in HBV-infected HCC tissues, the staining intensity score of CAS protein in HBV DNA positive HCC tissues was significantly higher than HBV DNA negative tissues (P < 0.05).
CONCLUSIONSHigher expression of CAS protein is found in HCC tissues,and the intensity of CAS protein expression is related closely to tumor differentiation. We suggested that CAS protein might serve as a marker for HCC diagnosis and differentiation estimation, and deduced that CAS might play an important role in the initiation of HBV infection associated HCC through upregulating expression of CAS.

10.Effects of corticosterone, cAMP, cGMP, Ca2+, and protein kinase C on apoptosis of mouse thymocytes induced by X-ray irradiation.

OBJECTIVETo observe the effects of signal factors of corticosterone (CS), cAMP, cGMP, Ca2+ andprotein kinase C (PKC) on lymphocyte apoptosis in mouse thymus induced by X-rays of 4 Gy in vitro.
METHODSThe DNA lytic rate for thymocytes was measured by fluorospectrophotometry.
RESULTSThe DNA lyric rate for thymocytes 4-8 hours after irradiation with 2-8 Gy was significantly higher than that in the control (P<0.01). As compared with the control, the DNA lytic rate for thymocytes treated with 0.01 micromol/L CS (P<0.01), 50 ng/mL cAMP (P<0.01), 0.05-0.4 microg/mL ionomycin (Iono, P<0.05 or P<0.01) or 0.05-0.4 ng/mL phorbol myristate acetate (PMA, P<0.05 or P<0.01), respectively, was significantly increased, while the rate for thymocytes treated with 50 ng/mL cGMP was not significantly increased. The DNA lytic rate for thymocytes treated with 0.01 micromol/L CS (P<0.01), 50 ng/mL cAMP (P<0.01), 0.2 and 0.4 microg/mL Iono (P<0.05), and 0.2 and 0.4 ng/mL PMA (P<0.05) plus 4-Gy irradiation, respectively, was significantly higher than that treated with single 4-Gy irradiation, while the rate for thymocytes treated with 50 ng/mL cGMP plus 4-Gy irradiation was not increased. When both 0.4 microg/mL Iono and 0.4 ng/mL PMA acted on the thymocytes, the DNA lytic rate for thymocytes was significantly higher than that in the control (P<0.01), the DNA lytic rate for thymocytes treated with both 0.4 microg/mL Iono and 0.4 ng/mL PMA plus 4-Gy irradiation was significantly higher than that treated with single 4-Gy irradiation (P<0.05), but was not significantly higher than that treated with 0.4 microg/mL Iono plus 4-Gy irradiation or 0.4 ng/mL PMA plus 4-Gy irradiation.
CONCLUSIONCS, cAMP, Ca2+, and PKC signal factors can promote thymocyte apoptosis induced by larger dose X-rays.
