2.Progress in the application of electroencephalography scores in infantile spasm

Infantile spasm is the most common epileptic encephalopathy in infancy.Without early diagnosis and effective treatment, serious consequences and economic burden maybe caused.One of the main characteristics of infantile spasm is the hypsarrhythmia on electroencephalography (EEG). It is important to analyze EEG accurately and reliably by scoring EEG.In order to further improve the EEG scoring system and better play the role of EEG in the diagnosis, treatment and prognosis implications of infantile spasm, in this paper, the advantages and disadvantages of various EEG scoring systems were analyzed, so as to provide clues by summarizing current EEG scoring systems and their applications in clinical practices.
3.Effect of temsirolimus on bladder cancer cells in vitro and in vivo

ObjectiveTo examine the effects of temsirolimus, an inhibitor of mammalian target of rapamycin, on bladder cancer cell lines T24 and BIU-87 in vitro and in vivo for purpose of evaluating the probability of mTOR targeted therapy for bladder cancer.MethodsAfter being treated by a different concentration of temsirolimus, T24 and BIU-87 cells were tested by MTT assay for cell proliferation activity.Cell cycle and apoptosis analysis were performed with flow cytometer. Wound scratch assay was used for cell migration activity and transwell motility assay. Western blot analysis was used to test the mTOR phosphorylation. Subcutaneous inoculation of 6-week-old nude mice was performed using 1 × 106 T24 cells in 50% matrigel for both control (n = 10) and temsirolimus (n = 10) groups. The volume of tumors was examined and then the expression of Ki-67 was detected by immunohistochemistry.ResultsTemsirolimus significantly inhibited proliferation of T24 and BIU-87 cells in a dose- and time-dependent manner. After administration of temsirolimus on T24 and BIU-87 cell lines for 24 h, the rate of wound healing in 0 nmol/L groups were (88.9 ± 14. 1 ) % and ( 83.6 ± 16.3)% , which were higher than in the 5 nmol/L groups, which were (42.7 ± 11.6) % and ( 36.9 ± 9.7 ) % ( P < 0.05 ). In the transwell motility assay, the number of cells in the 0 nmol/L group was 26.5 ± 5.8 and 28.2 ± 4.6, which was higher than in the 5 nmol/L group ( 19.0 ±3. 8 and 21.3 ± 5.1, respectively) (P < 0. 05). When temsirolimus was administered on T24 and BIU-87 cell lines for 48 h the percentages of cells delayed in phase G0/G1 in 5 nmol/L group were ( 77.46 ±6.11)% and (73. 39 ± 4. 94)% respectively, and higher than in the 0 nmol/L group, which were (65.99 ±5.01 )% 、(60.15 ±3.98)% (P <0.05). There was no statistically significant difference in the apoptosis rate between the two groups (P > 0.05 ). In Western blot analysis, the ratios of p-mTOR/β-actin were 0.92 ±0.09 and 1.01 ± 0.08 in 0 nmol/L group, and higher than in the 5 nmol/L group (0.47 ±0.05、0.04 ±0. 01 ) (P < 0.05 ). After administration of temsirolimus for 21 days, the tumor volume in nude mice in the control group were 351.1 ± 139.9 mm3 , which was larger than 351.1 ± 139.9 mm3 in the temsirolimus group ( P < 0.05 ). The positive rate of Ki-67 expression was ( 67.3 ± 8.4 ) % in the control group, which was higher than in the temsirolimus group ( 35.5 ± 6.7 ) % ( P < 0.05 ).ConclusionsThis study provides in vitro and in vivo evidence that temsirolimus may inhibit the viability of bladder cancer cells and temsirolimus could be exploited as a potential therapeutic strategy in bladder cancer.
4.Effects of Huanglian Jiedu Decoction on phosphatidylinositol-3-kinase expression in target tissues of type 2 diabetic rats

OBJECTIVE: To observe the effects of Huanglian Jiedu Decoction (HLJDD), a traditional Chinese compound herbal medicine, on p85 mRNA and protein expressions of phosphatidylinositol-3-kinase (PI-3K) in target tissues (skeletal muscular and adipose tissues) in rats with type 2 diabetes mellitus (T2DM) and to investigate the molecular mechanism of HLJDD in treating T2DM. METHODS: The male Wistar rats were injected with streptozotocin (STZ) 30 mg/kg through tail vein, and fed with high-fat and high-caloric diets to induce T2DM. Then the rats were randomly divided into untreated group, aspirin-treated group and HLJDD group, and treated correspondingly. Meanwhile, a group of normal animals without any treatment was set up for normal control group. Ten weeks later, serum fasting blood glucose (FBG), serum fasting insulin (FINS) and oral glucose tolerance test (OGTT) were routinely determined. The expressions of PI-3K p85 mRNA and protein in skeletal muscle and adipose tissue were determined with RT-PCR and Western blotting before and after insulin treatment. RESULTS: Compared with the untreated group, the FBG and OGTT levels in T2DM rats treated with HLJDD decreased significantly (P<0.05). The FINS in HLJDD group was lower than that in the normal control group (P<0.05), but was not significantly different from that in the untreated group. The PI-3K p85 mRNA and protein expressions in HLJDD group obviously increased, as compared with those in the untreated group. CONCLUSION: The effect of HLJDD in treating T2DM was probably associated with its improvement of PI-3K p85 mRNA and protein expressions in skeletal muscle and adipose tissue of the T2DM rats.
6.Relationship between osteoporosis and carotid artery disease in the elderly

ObjectiveTo study the relationship between osteoporosis and carotid artery disease in the elderly. Methods102 elderly patients were registered for this study and the extraeranial carotid was detected by ultrasound scan. The carotid intima-media thickness (IMT) and the plaque index (PI) were measured and calculated respectively. Meanwhile the patients were divided into two groups according to the results of bone mineral density (BMI) measurement: the osteoporosis group and the non-osteoporosis group. The IMT, PI, carotid stenotic rate and some biochemical parameters were recorded and compared between the two groups. Relationship between these parameters and osteoporosis were evaluated by logistic regression model and partial correlation analysis. ResultsThe differences in serum calcium and the levels of TC,TG,HDL and LDL between osteoporosis group and non-osteoporosis group were statistically significant (all P<0.05), while the level differences in serum phosphorus, fasting blood glucose and uric acid had no statistical significance between the two groups (P>0.05). Moreover, the IMT and PI in osteoporosis group were significantly higher than those in non-osteoporosis group (P<0.05). Among the 102 patients, 48 cases showed the carotid stenotic rate > 50%, including 41 patients in osteoporosis group as well as 7 patients in non-osteoporosis group(P<0.05). Logistic regression showed that age, HDL-C, IMT, PI and carotid stenotic rate>50 % were the correlated factors of osteoporosis and among them, IMT, PI and carotid stenotic rate>50% had higher risks (OR = 17.13,99.33,289.13). There was positive correlation between the carotid artery disease and osteoporosis. ConclusionsThere is a relationship between osteoporosis and carotid artery disease in the elderly. Emphasis should be paid on comprehensive prevention for osteoporosis and carotid artery disease in the elderly.
7.Prognostic predictors of minor stroke

Minor stroke is characterized by the mild symptom and rapid recovery,and does not easily cause disability,but a part of patients with minor stroke are the high risk population of recurrent stroke,and can cause disability,cognitive impairment,and emotional disorders.Early prediction of the prognosis in patients with minor stroke,screening for the high risk population of recurrent stroke and disability,and giving appropriate treatment may prevent recurrent stroke and improve the prognosis of patients.This article reviews the predictors of prognosis in patients with minor stroke.
8.Protective Effects of Cordyceps Sinensis from Different Habitats on Acute Injury of Hepatic Tissue Induced by Carbon Tetrachloride

To study and compare the protective effects of Cordyceps sinensis from different habitats on acute injury of hepatic tissue induced by carbon tetrachloride ( CCl4 ) . Methods:The rat model of acute injury of hepatic tissue induced by CCl4 was established. The biochemical indicators of alanine aminotranferease ( ALT) and aspartate aminotransferease ( AST) in the serum and superoxide dismutase ( SOD) and L-glutathione( GSH) in the liver tissue were detected, and the liver tissue HE staining was used for the histopathology determination. Results:Compared with those in the model group, the levels of AST and ALT in the serum were sig-nificantly reduced, the levels of SOD and GSH in the liver tissue were notably increased, and the necrosis of liver cells was improved in Cordyceps sinensis groups at low, medium and high dose from different habitats (P<0. 05), however, the differences between the two different habitats were not significant (P>0. 05). Conclusion: Cordyceps sinensis from Qinghai and Tibet shows some positive differences in the protective effect on the acute chemical liver injury induced by CCl4 in rats,however, the differences are not remarka-ble.
9.Human dermal fibroblasts as a feeder layer promote the growth of human keratinocytes

Methods Human dermis fibroblasts were isolated and cultured and then treated with mitomycin-C. The expression of type Ⅰand type Ⅲ precollagen mRNA and relevant protein in feeder layer were examined by RT-PCR and Immunohistochemistry. 10.Regulatory effects of hedgehog pathway on intestinal epithelial barrier function under hypoxia

Objective To investigate the regulatory effects of hedgehog pathway on intestinal epithelial barrier function under hypoxia.Methods The IEC-6 cells of rats were divided into 3 groups:the normoxia group (21% oxygen concentration),the hypoxia group (2% oxygen concentration) and the hypoxia + cyclopamine group (cells pretreated by 5 mmol/L of cyclopamine,and then exposed in an atmosphere with 2% oxygen concentration).The mRNA expressions of IHH,PTCH and GLI-1 were detected,and the transepithelial electrical resistance (TER) was determined.The protein expressions of tight junction proteins (ZO-1,Occludin,Claudin-1) and IHH were assayed by using the Western blot.All data were analyzed using the one-way analysis of variance or LSD-t test.Results The relative mRNA expressions of IHH,PTCH and GLI-1 were 0.056 ± 0.009,0.459 ± 0.087,0.142 ± 0.023 in the normoxia group,and 0.303 ± 0.052,0.678 ± 0.073,0.483 ± 0.061 in the hypoxia group,with significant difference between the 2 groups (t =-14.05,-11.85,-6.52,P < 0.05).The relative protein expressions of IHH in the normoxia group and the hypoxia group were 0.39 ±0.06 and 0.91 ±0.15,with a significant difference between the 2 groups (t =-8.08,P < 0.05).The TERs of the normoxia group,the hypoxia group and the hypoxia + cyclopamine group were (134 ± 5) Ohm/cm3,(100 ± 6) Ohm/cm3 and (118 ± 5) Ohm/cm3,with significant difference between the 3 groups (F =1.04,P < 0.05).Compared with the normoxia group,the TER of the hypoxia group was decreased by 27.7% (t =7.84,P < 0.05) ; compared with the hypoxia group,the TER of the hypoxia + cyclopamine group were increased by 16.4%,but it was still significantly lower than the normoxia group (t =4.23,P < 0.05).The expressions of ZO-1,Occludin and Claudin-1 were 1.18 ± 0.24,0.80 ±0.13 and 0.90 ±0.09 in the normoxia group,and 0.58 ±0.08,0.32 ±0.05 and 0.50 ±0.09 in the hypoxia group,and 0.92 ± 0.21,0.43 ± 0.10 and 0.82 ± 0.11 in the hypoxia + cyclopamine group,with significant difference between the 3 groups (F =4.95,2.88,10.09,P <0.05).The expressions of ZO-1,Occludin and Claudin-1 in hypoxia group were decreased by 48.7%,40.0% and 55.6% when compared with the normoxia group (t =12.86,9.35,18.90,P <0.05).The expressions of ZO-1,Occludin and Claudin-1 in the hypoxia + cyclopamine group were increased by 59.9%,35.2% and 65.1% when compared with the hypoxia group (t =5.63,2.92,6.66,P < 0.05).Conclusion Hedgehog signal pathway could be activated under hypoxia,and then the expressions of tight junction proteins are decreased,which finally induces the injury of intestinal epithelial barrier function.
