Thyroid peroxidase antibody (TPOAb) play an important role in the diagnosis of autoimmune thyroid disease.Chemiluminescence immunoassay (CLIA) is a new method been strongly recommended in recent years.The definition of positive value of TPOAb is inconsistent.The definition of TPOAb positive value is important for exploring the development of autoimmune thyroid disease.TPOAb is significantly increased in Hashimoto's disease and the titer is associated with the degree of infiltration and destruction of thyroid follicular.It is essential to monitor TPOAb in postpartum thyroiditis and early pregnancy.The positive of TPOAb is closely related to the adverse pregnancy outcomes caused by various risk factors.Graves disease combined with Hashimoto's must to be considered.

[Summary] A dozens of variants in intron 1 of FTO gene have been found to be significantly associated with obesity by a serious genome-wide association studies (GWAS) in past eight years.Very recently,The New England Journal of Medicine published an important genetic study of obesity,which challenged the point of FTO causing obesity directly.The researchers find one of the variants changes the binding ability of upstream regulator to IRX3/IRX5 and thus promotes IRX3/IRX5 expression instead of FTO.Which one is the real causal gene of common obesity,FTO,IRX3/5,both,or neither? All these questions need to be answered.

Objective: To observe emodin-induced molecular changes in PI3K/AKT signaling pathway in human leukemia K562 cells transplanted into BALB/c nude mice, and to explore whether emodin induces the apoptosis of K562 cells through PI3K/AKT signaling pathway. Methods: The subcutaneously transplanted tumor model of human K562 cells in nude mice was established. After continuously intraperitoneal injection with different doses of emodin for 12 d, the mice were sacrificed. Then the tumor weight and volume were measured, and the tumor inhibition rate was calculated. Emodininduced apoptotic morphological changes of K562 cells were detected by HE stain and scanning electron microscopy. RT-PCR and Western blotting were used to detect the expressions of PI3K, AKT and FoxO 3a mRNAs and proteins, respectively. Results: The relative tumor volumes (V/V0) were significantly smaller in the low-, moderate- and high-dose emodin-treated groups (8.90±0.24, 5.62±0.17 and 2.06± 0.31, respectively) than that in the untreated group (11.83±0.47; P <0.01). Significant apoptosis of K562 cells was found in emodin-treated groups under a light microscope and an electron microscope. RT-PCR revealed down-regulation of PI3K and AKT mRNAs expression and up-regulation of FoxO 3a mRNA expression induced by different concentrations of emodin in a dose-dependent manner. Western blotting analysis showed that the expression levels of PI3K and AKT proteins were markedly decreased and the expression level of FoxO 3a protein was significantly elevated in xenografted tumors treated with emodin. Conclusion: Emodin can significantly inhibit the growth of K562 cell xenografts in nude mice. The underlying mechanism may be associated with inhibition of PI3K/AKT signaling pathway. BackgroundJunctional adhesion molecule-1 (JAM-1) is intercellular transmembrane protein newly discovered and associated with the tight junction.Tight junction plays an important role in keeping the transparency of cornea,but there are few studies about JAM-1 in cornea tight junction.ObjectiveThis study was to determine the expression of JAM-1 in corneal epithelium,stroma,endothelium,and locate the distribution of JAM-1.MethodsThe corneas of two SFP Wistar rats were obtained,and the samples of epithelial lamella,corneal stroma and endothelium with Descemet membrane were prepared respectively for the detection of the expression of JAM-1 mRNA using reverse transcription polymerase chain reaction(RT-PCR).Primers were designed according to the genes as RGD web provided,and the objective genes were JAM-1,occludin and claudin-1.Products of PCR were examined by 1.5％ agarose gel electrophoresis and assayed with GelDoc-lt UVP Imaging System.The corneal paraffin sections and stretched preparations of epithelium and endothelium of corneal sections from other two SFP Wistar rats were prepared for the examination of JAM-1protein expression and location by immunohistochemistry.The use of experimental animals followed the Statement of ARVO. Results JAM-1,occludin and claudin-1 mRNA were expressed in rat cornea epithelium,stroma and endothelium.PCR melting curves showed the limpid unimedality.The expression level of JAM-1 mRNA was similar to occludin mRNA and higher than that of claudin-1 with the highest level in the epithelium layer.Immunohistochemistry assay revealed that there was definite JAM-1 antibody staining in the cornea epithelium,stroma and endothelium layers.However,the basal layer of corneal epithelium presented with the strongest staining in comparison with stromal and endothelial layers.Stretched preparations of corneal epithelium and endothelium showed that JAM-1 protein appeared at the junction site of epithelial cells and endothelial cells.The basal layer of the corneal epithelium showed the strongest response,and the staining of corneal endothelium was extensive and diffuse. Conclusions JAM-1 is a composition of intercellular tight junction which expresses in cornea epithelium,endothelium and stroma.However,its appearance and level vary from different corneal layers.

Objective To set up a new method of mild hypothermia via lateral ventricle in rabbits following traumatic brain injury (TBI). Methods Twenty-one New Zealand rabbits were used for this study. TBI was pro- duced with all the rabbits in anesthesia by using free-falling impact model. Animals were randomly divided into three groups: a TBI group, a mild hypothermia group (by perfusion of 25℃Ringer's solution via the lateral ventricle) and a control group. The contents of water and total sodium, potassium of the brain region close to traumatic brain tissue were detected and pathological changes were observed in three groups.
