2. The effect of combination of β-catenin and bone morphogenetic protein 9 on osteogenic differentiation of osteosarcoma cell line TE85

Objective: To investigate the effect of β-catenin and bone morphogenetic protein 9 (BMP-9) on osteogenic differentiation of osteosarcoma TE85 cells. Methods: The TE85 cells were infected by recombinant adenovirus Adβ-catenin and AdBMP9 alone or in combination. The expression levels of β-catenin and BMP9 mRNAs in TE85 cells were detected by semi-quantitative reverse transcription-polymerase chain reaction (RT-PCR). The relative β-catenin/TCF4 activity was detected by Luciferase Reported System. Alkaline phosphatase (ALP) staining and numeration were used to detect the early osteogenic potential of TE85 cells, and the alizarin red staining was performed to detect the late osteogenic potential. The expression levels of osteoprotegerin (OPG), osteocalcin (OC) and osteopontin (OPN) mRNAs were detected by semi-quantitative RT-PCR, and the expression levels of OC and OPN proteins were detected by Western blotting. Results: The exogenous mRNAs of β-catenin and BMP9 in TE85 cells were increased by adenovirus-mediated Adβ-catenin and AdBMP9 (both P < 0.05), and the relative β-catenin/TCF4 activity was increased after infection with Adβ-catenin (P < 0.05). Adβ-catenin or AdBMP9 infection could enhance the activity of ALP in a limited range in TE85 cells (P < 0.05), but they could not promote the mRNA and protein expressions of late osteogenic biomarkers OPN and OC as well as the calcium deposition. However, both early and late potential osteogenic abilities were enhanced obviously after infection with combination of Adβ-catenin and AdBMP9 (all P < 0.05). Conclusion: Osteogenic differentiation can be limitedly induced by β-catenin and BMP9 alone in TE85 cells, but this differentiation ability can be remarkably enhanced by combination of β-catenin and BMP9 with a synergistic effect.
3.Effect of comprehensive prehospital management on patients with emergency trauma treatment

Objective To study the effect of comprehensive prehospital management for patients with emergency trauma treatment. Methods Fifty emergency patients who were admitted to the emergency department were investigated respectively before and after the implementation of the comprehensive prehospital management of the people's hospital of Gaozhou City, with the service satisfaction questionnaire, the Anxiety Self-assessment Scale (SAS) and the Depression Self-rating Scale (SDS) . The time of admission, first aid time, emergency treatment effect, satisfaction and mental status change of the two groups were compared. Results There was no statistically significant difference in age, gender, cause of injury and distribution of injury in the two groups (P>0.05) . The admission time and first aid time of the patients in the observation group respectively were (15.6±4.2) and (33.5±8.4) minutes, which were shorter than the control group of (24.3±5.0) and (60.5±10.2) minutes (P<0.05) . The emergency effect and satisfaction of the observation group were superior to the control group (P<0.05) . The difference between SAS and SDS scores in the observation group was 21.3 ±3.4 and 20.4±3.1, both of them were higher than the control group (12.1±2.7 and 12.8±2.5) (P<0.05) . Conclusion The comprehensive pre-hospital management can shorten the time of admission and first aid, and effectively improve the clinical treatment effect and satisfaction of emergency trauma patients.
4.Clinical analysis about 5 cases of actinomycete keratitis

Objective To analyze clinical diagnosis and management of 5 patients with actinomycete keratitis.Design Retro- spective case series.Participants 5 patients (5 eyes) with actinomycete keratitis.Methods The clinical features and microbiologic da- ta of 5 culture-proven cases of actinomycete keratitis recorded between October 2004 to March 2006 were analyzed.Main Outcome Measures clinical characteristics,isolations identification,drug susceptibility test and treatments.Results All patients were males and farmers.Of the 5 cases presented in this study,4 cases were followed by minor trauma as a predominant risk factor,and were pre- sented by a chronic progressive corneal ulcer with a wreath pattern of infiltrate.The diagnosis of all cases was based on laboratory in- vestigations,by which 4 cases of nocardia and one case of streptomyce were identified.A variable drug sensitivities were presented in nocardia isolates,which including TMP-SMZ,amicasin,gentamicin and fluorine-quinolones.Conclusions Nocardia keratitis is mainly followed by a minor trauma.It is identified predominantly by laboratory investigations.Tropical and systemically sensitive biotic are the initial choice,while debridement and amnionic transplantation could be an effective alternative.

OBJECTIVETo study the feasibility of close and three-dimensional reconstruction of maxillary defects combination with pedicled buccal fat pad graft, temporalis myofascial flap and titanium mesh.
METHODSAmong 11 patients with maxillary tumors, 9 were malignant and 2 were benign. 8 cases underwent subtotal maxillectomy and 3 cases underwent total maxillectomy. Maxillary defects were closely reconstructed combination with pedicled buccal fat pad graft (BFP), temporalis myofascial flap (TMF) and titanium mesh to restore the profile of maxilla.
RESULTSAll flaps were alive with satisfactory function and aesthetic appearance. The uncovered BFP and the facial muscle surface of TMF epithelialized within 4-6 weeks. Their degree of mouth opening ranged from 2.5 to 4.0 cm and without nasal foods return. All patients had a good voice quality.
CONCLUSIONMaking use of BFP, TMF and titanium mesh can restore the good contour and function of maxillary. It is an effective method of reconstruction of maxillary defects, and is easy to manipulate. It is worthy to popularization.

8.Effects of programmed cell death on human dental follicle cells and changes of programmed cell death under different hydrostatic pressures.

OBJECTIVETooth eruption requires the presence of the dental follicle (DF) around the unerupted tooth. This study is to investigate programmed cell death on human dental follicle cells and changes of programmed cell death under different hydrostatic pressures: 0, 50 and 100 kPa.
METHODSHuman dental follicles from third mandibular molars were surgically removed from adolescents who need for orthodontics treatment after informed content, then trypsinized and cultured. Human dental follicle cells were divided into three groups according to different hydrostatic pressures: 0, 50 and 100 kPa and their programmed cell death were labeled by using TdT-medi-ated-dUTP nick and labeling (TUNEL).
RESULTSDental follicle cells cultured were elongate shape and exhibited fibroblastic characteristics. Compared with 0 kPa, programmed cell death cells on human dental follicle cells were increased 0.23% and 31.65% under 50 kPa and 100 kPa hydrostatic pressures respectively. 100 kPa group increased significantly (P < 0.01).
CONCLUSIONIt suggested that programmed cell death occured in human dental follicle cells cultured in vitro and was influenced by different hydrostatic pressures. Hydrostatic pressure may improve tooth erup-tion through dental follicle.

9.An experiment study and clinical observation of the influence of free-skin-grafted scrotal avulsion injuries on spermatogenesis.

OBJECTIVETo explore the influence of Free-skin-grafted penoscrotal avulsion injuries on spermatogenesis.
METHODSForty-two male New Zealand albino rabbits during child-bearing period were divided into the experimental group (n = 24) and the control group (n = 18) using random digits table, and 24 female rabbits with reproductive history were used for mating experiment. The experimental group animal's scrotum skin were excised, and the split skin from abdominal region was used to repair the skin defect of scrotum. The control group did not any processing. Six rabbits were randomly chosen respectively in control group and on the 3rd and 8th weekend after the model was successfully established in experimental group. The testicular surface temperature was measured in the eighteen rabbits using the method of burying thermometer, then the testicular biopsy were performed for hematoxylin-eosin (HE) staining. On the 8(th) weekend after the model was successfully established in experimental group, matched-pair feed was performed in the other 12 rabbits respectively in experimental group and in control group. Observation of corresponding mother rabbit fertility. Three patients of penoscrotal avulsion injuries were treated using split skin grafts, and the information of sex life and the quality of sperm were obtained by follow up.
RESULTSThe testicular surface temperature was similar on the 3rd and 8th weekend after the model was successfully established in experimental group [(36.15 ± 0.24)°C, (36.77 ± 0.42)°C] with that of the control group. Testis tissue (HE) staining showed the tier of spermatogenic cells was rule arrangement and lot of mature sperms were found in the convoluted seminiferous tubules in control group. The tier of spermatogenic cells was diminished and disposed derangement, the spermatozoa were not seen on the 3(th) weekend of the experiment group. The tier of spermatogenic cells was increased and some spermatozoa were seen on the 8th weekend of the experiment group. Male and female matched-pair feed showed the experimental group conception rate 8/12, and 4.1 ± 3.2 rabbit babies were born averagely, while that of was 12/12 and 6.0 ± 1.3 in control group (P > 0.05). The skin grafts there were some contracture in early stage (1 - 2 months) when the skin grafts applied to repair the avulsing scrotum in three patients. But the skin grafts became loose with downward sagging and there were the good cosmetic result in one year, and without any contracture. The sperm quality was normal after the skin grafts applied to repair the avulsing scrotum in the late stage.
CONCLUSIONSThe skin grafting is little arrest the testicle spermatogenesis in the three methods (skin flap reconstruction scrotum, testicle buried, split skin grafting) that have usually been used to repair scrotum skin lose. For a young male, the best treatment for penoscrotal avulsion injuries is free skin grafting, while skin flaps are not recommended for reconstructing the scrotum.

10.Bevacizumab (Avastin) inhibits lung cancer-induced pulmonary microvascular angiogenesis.

OBJECTIVETo study the effect of bevacizumab (Avastin) on pulmonary microvascular angiogenesis induced by lung cancer.
METHODSThe effects of Avastin on cell growth, cell cycle and apoptosis of lung microvascular endothelial cells were assayed by flow cytometry.
RESULTSAvastin inhibited the growth of lung microvascular endothelial cells and induced increase of cell apoptosis index in a concentration-dependent manner. With the increment of Avastin concentration from 25 to 100 microg/ml, avascular area increased from 0.944-/+0.073 cm square to 5.189-/+0.192 cm square, and the cell apoptosis index increased from 32.5%-/+1.5% to 39.25-/+1.6%.
CONCLUSIONAvastin can inhibit the growth and induce apoptosis of lung microvascular endothelial cells in three-dimensional culture, possibly in association with cell cycle arrest in G(0/1) phase.
