4.Study of biomarker panel and system biology analysis in human superficial bladder transitional cell carcinoma

Objective To study the biomarker panel of superficial bladder transitional cell carcinoma(SBTCC)and analyze the biological pathway in tumorigenesis by Shotgun proteomics strategy.Methods Normal urothelium cells and cancer cells were harvested by laser capture microdissection from clinical specimen and the proteomic expression profile was identified by two-dimensional liquid chromatography tandem mass spectrometry.The isoelectric point,molecular weight,grand average of hydropathicity,transmembrane helices were analyzed by using proteomics tools.Gene ontology was used to comment the identified proteins.The pathway analysis was performed by ArrayTrack software,and visualized by GenMAPP.Results There were 440 and 218 proteins expressed in cancer cells and normal cells respectively,among them 388 proteins were differerntially expressed.All the database about identified proteins was deposited in an accessible form to researchers at http://www.Proteome-SBTCC.org.cn and http://www.Proteome-NHTE.org.cn.There were 267(68.8%)differentially expressed proteins which had GO biological process comments.The biological pathwavs of these proteins included MAPK signaling pathway,focal adhesion,oxidative phosphorylation,ECMreceptor interaction,etc.Conclusion Shotgun strategy proteomies database of normal transitional epithelium and SBTCC is successfully constructed.And the basis for the understanding of cell biology and discovery of biomarker panel for SBTCC iS provided.

8. Comparison on quality of wild Ziziphi Spinosae Semen between marketed and produced in Tianjin

To analyze and evaluate the quality of Ziziphi Spinosae Semen produced in Tianjin and comparing with Ziziphi Spinosae Semen traded on the market in different places. Twelve kinds of wild Ziziphi Spinosae Semen samples collected from the four represent regions were gathered by systematic combination of regional stratified sampling method and HPLC method was used to determine spinosin, jujuboside A, jujuboside B, and betulinic acid. The contents of spinosin, jujuboside A, jujuboside B, and betulinic acid from wild Ziziphi Spinosae Semen produced in Tianjin are 0.810-1.925, 0.695-1.708, 0.201-0.390, and 0.651-0.789 mg/g; Ziziphi Spinosae Semen produced in Chengde, Hebei province has the highest betulinic acid contents of 1.654 mg/g. The quality of wild Ziziphi Spinosae Semen produced in Tianjin is excellent. What's more, the wild Ziziphi Spinosae Semen produced in Baxian Mountain, Tianjin takes particularly prominent advantages of better quality. Quality of wild Ziziphi Spinosae Semen produced in other three regions is equal to the average level of that traded on the market.
9.Detection of nodular thyroid diseases among 35 495 adult underwent health examinations

Objective To explore the detection rate and major risk factors of nodular thyroid diseases among adults.Methods A total of 35 495 adults who carried out health checkup during January 1st,2009 and December 31th,2010 in our center were assessed for the occurance of nodular thyroid diseases and the size and number of thyroid nodules by age and gender.Chi-square test,rank sum test and Chi-square trend test were used for data analysis.Results The total detection rate of nodular thyroid diseases was 33.87% (female vs male:41.76% vs 29.68%).The prevalence of single thyroid nodule was decreased with age,as opposed to multi-thyroid nodules.Single thyroid nodule was more commonly seen in males,which was reversed in multi-thyroid nodules.Small,middle and large thyroid nodules were found in 77.49%,18.19% and 4.32% males or 72.57%,21.59% and 5.84% females,respectively.Moreover,thyroid cancer was detected in 0.4‰ of those with nodular thyroid diseases.Conclusions The detection rate of nodular thyroid diseases among adult health checkup receivers was higher,which calls on the needs of intensive monitor and effective prevention of the condition.
10.The Protective Effect of Co-Treatment with Antioxidants against IH-Induced Pancreatic Injury in Mice

Objective To explore the protective effect of co-treatment with antioxidants 4-hydroxy-2,2,6,6-tetra-methylpiperidine 1-oxyl (TEMPOL) and reduced glutathione (GSH) in pancreatic injury induced by intermittent hypoxia (IH) in mice. Methods Fifty male C57BL/6J mice were randomly divided into control group(CON), IH group (IH), IH+TEM-POL group (IH+TEMPOL), IH+GSH group (IH+GSH) and IH+TEMPOL+GSH group (IH+TEMPOL+GSH). After successful-ly prepared animal model, the insulin resistance (IR) level, pancreatic malondialdehyde (MDA) level,superoxide dismutase (SOD) activity,glutathione reductase (GR) concentration and pancreatic β-cell apoptosis were detected in five groups. Re-sults The blood glucose level was significantly increased in IH group than that in CON group (P<0.01), but which was sig-nificantly decreased in IH+TEMPOL+GSH group than that of IH group (P<0.01). The MDA content was significantly higher in the IH group than that in CON group (4.48±0.25 vs 1.94±0.21, P<0.01), but SOD activity and GR content were signifi-cantly lower in IH group than those of CON group (61.52±3.33 vs 100.05± 7.26,107.81±7.54 vs 170.54±4.90,P<0.01). The level of MDA was significantly lower in IH+TEMPOL+GSH group (2.07±0.35) than that in IH group. The levels of SOD and GR were significantly higher in IH+TEMPOL+GSH group (96.68±5.85 and 166.87±5.75) than those of IH group (P<0.01). The apoptotic rate of pancreaticβcells was significantly increased in IH group than that in CON group (2.43±0.07 vs 0.54± 0.06, P<0.01), but it was significantly lower in IH+TEMPOL+GSH group (0.56 ± 0.06) than that in IH group (P<0.01). There were no significantly differences in all above indexes between IH group, IH+TEMPOL group and IH+GSH group ( P>0.05). Conclusion The co-treatment with the antioxidant TEMPOL and GSH can obviously protect IH-induced pancreatic injury in mice. However, there was no significant protective effect of TEMPOL or GSH alone on pancreatic injury.
