1.Recent progress of study in pregnancy and thyroid cancer

Thyroid cancer accounts for the most common endocrine malignancies in women during childbearing age.Many questions remain to be answered regarding the association between thyroid cancer and pregnancy.This review will discuss both the effects of pregnancy on thyroid cancer and vice versa.Recommendations on how to manage thyroid cancer in pregnancy will also be cited from a Chinese academic guideline as listed at the end of this review.
2.Application of flow cytometry to detect cell apoptosis

Flow cytometry (FCM) is a technology for fast qualitation, quantitative analysis and separation of single cells or other biological particles. The method is rapid, sensitive and accurate, and its objective and direct result can be analyzed with multi-parameters simultaneously. The morphological, biochemical and molecular changes during apoptosis include cellular shrinkage, permeability transition of plasma membrane, caspases activation, dissipation of mitochondrial trans-membrane potential, phosphatidylserine redistribution, calcium flux and DNA fragmentation and content. The review outlined the methods to characterize, identify and quantify apoptoUc cells by flow cytometry to further determine cell apoptosis.
3.Clinical significance of direct immunofluorescence assay for rapidly detecting influenza A virus antigen

Objective To study the direct immunofluorescence method for rapidly detecting influenza A virus antigen and its clinical application .Methods The inpatients with clinically diagnosed acute respiratory tract infection were selected .Two hundred samples were collected and divided into two groups :the winter and spring epidemic seasons children group and the summer and au‐tumn non‐epidemic seasons children group ,100 cases in each group .The US D3 Ultra influenza A virus antigen detection reagent was used .The fluorescence labeled monoclonal antibody was adopted to detect influenza A virus antigen in the patients with cold . Results During the winter and spring seasons of influenza prevalence ,8 cases of 100 detected cases were positive with the positive rate of 8% ;during the summer and autumn seasons of non‐influenza prevalence ,2 cases of 100 cases were positive with positive rate of 2% ,the difference in the positive rate between the two groups had statistical significance(P<0 .05) .Conclusion Applying the direct immunofluorescence method for rapidly detecting influenza A virus antigen is an economical ,efficient ,rapid and specific detec‐tion method ,and can guide clinical rapid diagnosis and rational medication .
4.Experimental Study on Antitumor Effect of Volatile Oil of Xihuang Pill and Its Immune Mechanism

This study was aimed to explore the antitumor effect of volatile oil of X ihuang pill and its immune mecha-nism in order to screen the antitumor active site of Xihuang pill. Among 70 female Wistar rats, 10 rats were random-ly selected as the blank control group; and the other 60 rats was used in the establishment of walker 256 breast can-cer cell tumor-bearing rat models. The model rats were randomly divided into the negative control group (model group), high-dose volatile oil group, middle-dose volatile oil group, low-dose volatile oil group, high-dose Xihuang pill group, and lentinan group (positive control group), with 10 rats in each group. The intragastric administration was given twice a day for 14 days. Blood was taken from the abdominal aorta. Tumor tissues was removed and weighed to calculate the tumor inhibitory rate. ELISA method was used to detect the level of IL-2, IL-6, IL-10, IFN-γ and TGF-β in peripheral blood. The flow cytometry was used to detect the content of CD3+ T cell, CD4+ T cell, CD8+ T cell, and B7-1 cell (CD80). The results showed that the tumor inhibitory rates of volatile oil of high-dose group and middle-dose group were 28.4% and 24.1%, respectively. Compared with the model group, the average level of IL-2 and IFN-γ of volatile oil of high-dose group and middle-dose group and CD3+ T cell, CD8+ T cell, B7-1 cell con-tent were obviously increased (P< 0.05). It was concluded that volatile oil of Xihuang pill had certain antitumor ef-fect, which was one of the antitumor active sites of Xihuang pill. The volatile oil of Xihuang pill upregulates the lev-els of IL-2 and IFN-γ, as well as the contents of CD3+ T cell, CD8+ T cell, B7-1 cell in order to increase the im-mune clearance function of tumor-bearing rats.
6.Risk factors analysis of hyperthyroidism complicated hyperthyroid heart disease

Objective To study the risk factors of hyperthyroid heart diseases(HHD) by analyzing clinical features of patients in order to provide a scientific basis for prevention and treatment of HHD. Methods Nine hundred and eighty two cases were selected as objective from in-patient data of Thyroid Disease Treatment Centre of Shandong Province. The cases were divided into hyperthyroidism group and HHD group. The variables of etiology,sex, age, duration of disease, TSH, FT3, FT4 and TRAb were analyzed by comparative analysis. The risk factors were analyzed by logistic regression. Results The prevalence of hyperthyroidism complicated hyperthyroid heart disease was 7.7%(76/982), age, duration of diseases, FT3, TRAb in the HHD group were [(51.4 ± 11.5), (6.3 ±2.1) years, 21.6 pmol/L, 71.6 U/L], in hyperthyroidism group were [(37.9 ± 9.8), (2.6 ± 1.3) years, 14.9pmol/L, 49.6 U/L]. The differences were statistically significant(u = 9.93,15.23, T = 44954,48792.5, P < 0.05)between the two groups. The factors of the older, higher FT3 and TRAb, longer duration, Graves disease (OR =1.751,1.470,1.483,1.445,1.234) increased the risk of HHD. Conclusions Graves disease, longer duration, old age, higher FT3 and TRAb are the risk factors of HHD. Timely prevention and control of risk factors is necessary to reduce the incidence of HHD.
7.Correlation between the types of the constitution in TCM and the sleep status in PLA Navy divers

Objective To investigate the correlation between the types of constitution in TCM and the sleep status in the PLA Navy divers. Methods Eighty-nine PLA Navy divers who performed 10m diving professional training were selected. Constitution in TCM was classified and determined by the standardized standard Constitution in Chinese Medicine Questionnaire, sleep status was evaluated by the Pittsburgh Sleep Quality Index. Results 62.9% of Navy divers were the mild constitution in TCM. The eight kinds of the biased constitution in TCM are ranked with yang deficiency, phlegm-dampness, dampness-heat, qi deficiency, yin deficiency, blood stasis, qi stagnation and special intrinsic quality. Among 78 Navy divers with good sleep quality, there were 49 divers (72.1%) with the mild constitution in TCM and 19 (27.9%) with the biased constitution in TCM. Among 21 Navy divers with poor sleep quality, there were 7 divers (33.3%) with the mild constitution in TCM (accounting for) and 14 (66.7%) with the biased constitution in TCM. For Navy divers with poor sleep quality, the sleep quality scores were positive correlated with the blood stasis constitution in TCM (r=0.481,P<0.05). Conclusion Sleep status is correlated with the types of the constitution in TCM, and regulating constitution in TCM can improve sleep quality in PLA Navy divers.
8.Experimental study on anti-tumor effect of xihuang pill and its immune clearance function.

OBJECTIVETo discuss the anti-tumor effect of Xihuang pill on tumor-bearing rats and its effect on the immune clearance function of tumor-bearing organisms.
METHODWalker256 tumor cells were adopted to establish the tumor-bearing rat model. The rats were randomly divided into five groups: the normal control group, the model control group, the lentinan group and Xihuang pill low dose, middle dose and high dose groups, with 10 rats in each group, and continuously treated and given drugs for 14 d after modeling. Blood and tumors were collected from abdominal aorta to calculate the tumor inhibition rate. The content of CD3+, CD4+, CD8+ T cells and adhesion molecule B7-1 (CD80) in peripheral blood were detected by flow cytometry (FCM). The expressions of IL-2 and IFN-gamma in were determined by ELISA.
RESULTThe tumor inhibition rate of the Xihuang pill high dose group was 33. 1 percent. Compared with the model group, the Xihuang pill large dose group showed significantly low IL-2, IFN-gamma, CD3+, CD4+, B7-1 in peripheral blood, with statistical significance in their differences (P < 0.05).
CONCLUSIONXihuang pill could show its anti-tumor effect by enhancing the immune clearance function and increasing IL-2, IFN-gamma, CD3+ T, CD4+ T, B7-1 in peripheral blood.

9.Transplantation of mesenchymal derived stem cells followed by G-CSF injection can reconstitute hematopoiesis of lethally irradiated BALB/c mice.

OBJECTIVETo explore the hematopoietic reconstitution potential of mesenchymal derived stem like cells.
METHODSWe transplanted bone marrow mesenchymal derived stem like cells into lethally irradiated BALB/c mice. Hematopoietic cells were derived from the non-adherent bone marrow cells 24 hours after initial culture while murine mesenchymal derived stem like cells from bone marrow of donor mice were cultured for 10 days before the transplantation.
RESULTSAll mice of group 1 and 3 died in 7-8 days post irradiation following transplantation, while all the mice from group 2 and 4 survived. The time course of hematopoietic reconstitution was then observed. The peripheral blood and bone marrow cell count recovered in the MSC + G-CSF transplanted group and the BM transplanted group after 3 weeks. Interestingly, CFU-GM number in the MSC + G-CSF transplanted group increased significantly after 2 weeks and even more than that in the BM transplanted group after 3 weeks while as CFU-GM colony dropped 2 weeks after in the BM transplanted group. Spleen colony (CFU-S) number and size of the MSC + G-CSF transplanted group was significantly greater than the BM transplanted group. Furthermore, PCR analysis was performed using peripheral blood cells to determine if any male-derived cells were present. No male-derived cells were found in any of the mice from group 1 and 3. Y-chromosome-specific src gene was found to be dominant in the MSC + G-CSF transplanted group and the BM transplanted group by week 4 post transplantation. In addition, we demonstrated that induction with G-CSF lead to CFU-GM colony formation from MSC compartment in vitro.
CONCLUSIONThese results indicate that under stimulation of G-CSF, mesenchymal derived stem like cells might differentiate into hematopoietic primitive stem cells in vivo and have the capacity to re-establish hematopoiesis in lethally irradiated mice. This study should provide an alternative transplantation treatment for malignancy.

10.Study of biological characteristics of murine dermal mesenchymal stem cells and their enhancement of hematopoiesis recovery.

OBJECTIVETo study the characteristics of dermal mesenchymal stem cells (DMSC), and explore whether they could enhance hematopoiesis recovery in vivo as well as facilitate proliferation and differentiation of hematopoietic cells in vitro.
METHODSMultipotential stem cells from the murine dermal mesenchyme were dissociated and cultured as donor cells. After 2 approximately 3 passages, the growth status, cell cycle, immunophenotype and morphology of DMSC were analyzed. Hematopoietic cells were plated onto a feeder layer formed by DMSC, cell count and CFU-GM yields were observed dynamically. Female mice received 5 Gy (137)Cs radiation were injected with DMSC cultured for 2 - 3 passages via tail vein. Cell count and CFU-GM yields of the bone marrow were observed regularly. Pathological study of the liver, spleen and bone marrow was done to evaluate hematopoiesis recovery.
RESULTSMurine DMSC are adherent cells with a morphology of fibroblastoid and spindle and multiangle in shape. Immunophenotypes showed that CD(45), CD(34), HL-DR positive DMSC were 1 - 3%, CD(44) and CD(13) positive DMSC 75 approximately 95%. Cell cycle assay demonstrated 83% of DMSC being G(0)/G(1) phase. In vitro, the total cell count and CFU-GM yields in the experimental group were higher than those of the long-term culture bone marrow cells by the third week. The DMSC can sufficiently support the proliferation and differentiation of hematopoietic cells for seven weeks. In vivo, peripheral granulocytic count, cells in the bone marrow of one femoral bone and CFU-GM by the third week in the experimental group were much higher than those of controls. Genetic assay of the murine blood demonstrated Y chromosome.
CONCLUSIONThe DMSC have characteristics of stem cells. DMSC sped up hematopoiesis recovery of irradiated mice. DMSC as a feeder layer can support proliferation and differentiation of hematopoietic cells.
