2.The effect of fasudil via Rho/ROCK signaling pathway on the inflammation and fibrosis in human mesangial cells in high glucose medium

Objective To study the effect of fasudil on inhibiting the Rho/ROCK signaling pathway under high glucose in human mesangial cells (HMCs) inflammation and fibrosis. Methods Synchronized HMCs were divided into following groups: (1) Normal glucose control group ( NG, 5. 5 mmol/L glucose) ;(2) High glucose group (HG, 30 mmol/L glucose) ; (3) Mannitol group (Man, 5.5 mmol/L glucose + 24. 5 mmol/L mannitol) ; (4) High glucose + fasudil group ( HG + F, the concentrations of fasudil were 25 ,50 and 100 μmol/L, respectively). Collect the supernatant and cells at 0, 12, 24, 36, 48 and 72 h respectively, and determine the concentration changes of the RhoA, ROCK- Ⅰ, connective tissue growth factor (CTGF)mRNA with real-time PCR method in the cells, then used the ELISA method to check the protein content of the fibronectin ( FN) , CTGF, TNFα in the supernatant. Results ( 1) RhoA, ROCK- Ⅰand CTGF mRNA of the HMCs cultured under the high glucose expressed significantly higher than those in the normal group, and there was certain time-dependence. Besides, there was no statistic significance by comparing Man and NG. (2) Under the high glucose situation, after the fasudil pretreatment with different concentrations and 24 h or 48 h culture with high glucose, RhoA, ROCK- Ⅰ , CTGF mRNA expression was significantly decreased in HG + F, compared with HG, and there was certain concentration-dependence. (3) High glucose increased the FN, CTGF, TNFα protein secretion of HMCs in a time-dependent manner, but normal glucose and mannitol had no such effect. (4) After the fasudil pretreatment with different concentrations and culture with high glucose for 12, 24, 36, 48, 72 h, the FN, CTGF, TNFα protein secretion was significantly reduced compared with HG. Conclusion Fasudil can reduce the secretion of downstream inflammatory factors and cytokines by inhibiting high glucose-activated HMCs Rho/ROCK signaling pathway, and reduce the inflammation and fibrosis of HMCs. This provides a new basis for the therapeutic target in the treatment of diabetic nephropathy.
3.Electronic cigarette use among adolescents in Ningxia Hui Autonomous Region

Objective:
To investigate the status of electronic cigarette use among adolescents in Ningxia Hui Autonomous Region, and to provide evidence for tobacco control in adolescents.
Methods:
Based on the 2019 National Youth Tobacco Epidemic Monitoring Program, multistage proportional sampling method was used to select middle school students from Ningxia Hui Autonomous Region. A questionnaire revised by Chinese CDC was used to collect the general information, the cognition and use of electronic cigarettes, and the access to advertising of electronic cigarettes and related products.
Results:
Totally 9 019 questionnaires were distributed, 8 401 valid ones were recovered, and the response rate was 93.2%. The rates of electronic cigarette use and attempt among students were 4.3% and 13.4%. The rates of electronic cigarette use and attempt in male students were 7.7% and 22.9%, which were higher than that in female students (0.8% and 3.8%, P<0.05) . The rates of electronic cigarette use and attempt varied in different schools ( P<0.05 ), which were higher in vocational high school students ( 11.5% and 26.8% ). Among 246 students who used electronic cigarettes, 30.1% did not thought electronic cigarettes contained nicotine, while 60.2% did not know whether electronic cigarettes contain nicotine. In the past 30 days, 27.0% of the students had seen the advertisements of electronic cigarettes and related products, mainly through TV, store, supermarket, convenience store, grocery store, electronic cigarette experience store or retail store.
Conclusions
The rates of electronic cigarette use and attempt among adolescents in Ningxia Hui Autonomous Region are 4.3% and 13.4%. Boys and vocational high school students have higher rates. Students generally know electronic cigarette and have more access to it.
4.The application of hypothermic perfusion via renal artery balloon catheter in laparoscopic partial nephrectomy

Objective To evaluate the feasibility and safety of hypothermic perfusion via renal artery balloon catheter before laparoscopic partial nephrectomy.Methods A total of 35 patients,who were arranged to receive laparoscopic partial nephrectomy during the period from March 2013 to December 2016 at the General Hospital of PLA,China,were enrolled in this study.The tumor was located in the left kidney in 22 patients and in the right kidney in 13 patients.The long diameter of the tumors was 2.3-7.0 cm,with a mean of (4.1±1.2) cm.Before laparoscopic partial nephrectomy,the implantation of renal artery balloon catheter was performed in all patients.The intraoperative renal cold-ischemia time,the time spent for operation and the amount of intraoperative blood loss were recorded.The creatinine clearance rate (Ccr) and estimated glomerular filtration rate (eGFR) were determined before operation as well as at one,3 and 7 days after operation.The intraoperative and postoperative complications were analyzed.Results Successful implantation of renal artery balloon catheter was accomplished in all 35 patients,with the technical success rate being 100%.Slipping of balloon catheter before laparoscopic partial nephrectomy occurred in one patient;in 2 patients incomplete occlusion of renal artery was found during operation and artery forceps had to be used to occlude the renal artery.In the other 32 patients,laparoscopic partial nephrectomy under the condition of hypothermic perfusion via renal artery balloon catheter was successfully carried out.No severe complications occurred during and after the operation.The average cold-ischemia time of the diseased kidney was 45 (20-125) min,the mean time for interventional procedure was 28 (20-40) min,the average surgery time was 147(95-235) min,the average amount of intraoperative blood loss was 180(50-1000) ml.The Ccr values determined before and at one,3,7 days after nephrectomy were (96.5±15.1),(75.2±10.5),(54.3±13.8) and (91.8±14.1) ml/min respectively.The eGFR values determined before and at one,3,7 days after nephrectomy were (99.5±15.3),(70.3±12.5),(65.5±11.7) and (96.8±12.3) ml·min-1·m-2 respectively.Statistical analysis indicated that in both groups the preoperative and 7-day postoperative Ccr values and eGFR values were significantly different from their one-day and 3-day postoperative values (P<0.01),but no statistically significant differences existed between preoperative values and 7-day postoperative ones (P>0.05).Conclusion Hypothermic perfusion via renal artery balloon catheter before laparoscopic partial nephrectomy is clinically safe and feasible,it can prolong the safe time of renal ischemia and protect renal function.
5.The nutritional status among children under 60 months year-old after one year of the Earthquake in Wenchuan.

OBJECTIVETo evaluate the effects of Wenchuan Earthquake on the nutritional status, growth, and the prevalence nutritional anemia, vitamin A deficiency (VAD) and vitamin D deficiency among children under 60 months old living in the disaster areas.
METHODSA nutritional survey was conducted in April 2009. The survey recruited 466 under 60 months old children, including 162 children aged 0 months old and 304 children aged 24 - 59 months old. The children's growth status, prevalence of anemia, and the iron deficiency prevalence, vitamin A, D, B(12), folic acid status were measured. The study findings were compared to the results from 2002 Chinese Nutritional and Health Survey.
RESULTSThe exclusive breast milk feeding rate among infants under 6-months was 58.8% (30/51). Among the 0 - 23 months old children, only 10.7% (16/150) got breast feeding within one hour after delivery. Ninety-two per cent (149/162) 0 - 23 months old children never received any nutrient supplements. The average cereals and roots intakes of the 24 - 59 months old children living in the disaster area were (267.2 ± 154.3) g/d, higher than the result of rural children average (178.75 g/d) of 2002 National Nutrition and Health Survey (u = 9.995, P < 0.01). The average intakes of vegetables, aquatic products, meat and poultries were (63.6 ± 56.7), (2.6 ± 7.9), (19.4 ± 24.0) g/d, respectively, significantly lower than 2002 results 135.05, 8.82 and 32.23 g/d (u = 21.971, 13.728 and 9.321, P < 0.01). Fruits, dairy products and legumes intakes were (102.2 ± 110.8), (65.2 ± 123.8) and (20.5 ± 29.0) g/d, respectively, higher than 2002 results (32.81, 2.87 and 6.50 g/d; u = 10.919, 8.778 and 8.417, P < 0.01). The prevalence of vitamin A deficiency and marginal deficiency was 15.4% (29/188) and 30.3% (57/188), respectively. The sum of vitamin D deficiency and insufficiency was 92.0% (183/199). The prevalence of anemia of the 0-months old children and 24 - 59 months old children was 47.5% (77/162) and 21.5% (60/279), respectively. The prevalence of iron and zinc deficiencies was 45.7% (86/188) and 65.5% (127/197). The prevalence of stunt was 13.6% (38/279) among the 24 - 59 months old children.
CONCLUSIONThe results indicated that the dietary structure of the children living in the disaster area was not ideal. Although, the intakes of energy and protein supporting foods could meet the requirements, but the dietary lacks of meats, poultries, dairy products, legume products, aquatic products and vegetables. The vitamin A, vitamin D deficiency, iron and zinc deficiencies are of a high prevalence in the disaster area.

6.The nutritional status of reproductive women at one year after the disaster of Earthquake in Wenchuan.

OBJECTIVETo evaluate the effects of Wenchuan Earthquake on the nutritional status and the prevalence of nutritional anemia, vitamin A deficiency (VAD) and vitamin D deficiency among reproductive women (15 - 44 years old) in the disaster areas one year after the Earthquake.
METHODSA nutritional survey was conducted in 3 counties in April 2009, one year after the Earthquake. Two towns from each county were selected as study sites, and this survey recruited 58 pregnant, 66 lactating and 242 non-pregnant-non-lactating women. A comparison was made to the results of 2002 Chinese Nutrition and Health Survey.
RESULTSThe cereals and roots intakes of the pregnant, lactating and non-pregnant-non-lactating women living in the disaster area were (426.8 ± 271.8), (568.0 ± 306.1), and (483.0 ± 277.7) g/d respectively, which were almost the same results (486.8, 509.3 and 495.1 g/d, respectively) from 2002 National Nutrition and Health Survey. The fat and oil intakes were (41.9 ± 51.6), (55.5 ± 69.2), and (66.9 ± 125.7) g/d, respectively, which were also the same ad the results (45.2, 43.9 and 41.4 g/d, respectively) from 2002 National Nutrition and Health Survey. The intakes of meats and poultries were only (58.1 ± 67.7), (76.3 ± 218.7), and (23.9 ± 29.6) g/d respectively, which were much lower than the recommended food intakes from the Branch of Maternal and Child Nutrition of Chinese Nutrition Society. The vitamin A deficiency and marginal deficiency prevalence were 6.9% (24/347) and 18.2% (63/347), respectively. The deficiency and insufficiency of vitamin D was sum to 93.9% (323/344). The prevalence of anemia was 32.6% (112/344). 51.0% (171/335) reproductive women were iron deficient, and 61.6% (210/347) women were suffering zinc deficiency.
CONCLUSIONThe study findings indicated that the dietary structure was seriously effected by the Earthquake. The sources from animal and legume products were relatively low. The micronutrients nutritional status was poor. The vitamin A, vitamin D, and iron, zinc deficiencies were highly prevalent in the disaster area.

7.Clinical application of 18 F-FDOPA PET/CT imaging in the diagnosis of pheochromocytoma/para-ganglioma

Objective To explore the efficiency of 6-18 F-fluoro-L-3,4-dihydroxyphenylalanine (18 F-FDOPA) PET/CT imaging in the diagnosis of pheochromocytoma/paraganglioma (PPGLs). Methods Twenty-six patients (15 males, 11 females;age:13-76 years) who were suspected of PPGLs with adrenal or retro-peritoneal mass were enrolled into this prospective study. All patients underwent 18 F-FDOPA PET/CT ima-ging. The clinical data, laboratory data and imaging results were collected. Region of interest ( ROI) was drawn on the liver and lesions, and the mean standardized uptake value ( SUVmean ) of the liver and the max-imum standardized uptake value ( SUVmax ) of lesions were calculated, as well as the ratio of tumor/lesion ratio ( T/L) . Based on the pathological results considered as the gold standard, the diagnostic efficiency of 18 F-FDOPA PET/CT imaging in PPGLs was calculated and compared with that of CT/MR. Besides, receiv-er operating characteristic ( ROC) curve analysis of T/L was used. Results Twenty-five patients were path-ologically confirmed, including 12 patients with PPGLs and 13 patients with non-PPGLs. The sensitivity, specificity, accuracy, positive predictive value and negative predictive value of 18 F-FDOPA PET/CT ima-ging was 11/12, 12/13, 92.00%(23/25), 11/12 and 12/13, respectively. The area under the ROC curve of T/L was 0.978, with the cut-off value of 1.55. 18 F-FDOPA PET/CT imaging totally detected 29 positive foci, including 12 of pheochromocytoma (PCC) and 17 of paraganglioma (PGL). Among those 18F-FDOPA positive PGL foci, 11 were detected by CT/MR, while 6 with diameter less than 1 cm were missed. Conclu-sion 18 F-FDOPA PET/CT imaging can detect more small PGL lesions than CT/MR, suggesting that it may be a proper imaging modality of PPGLs.
8.Effects of chronic stress during pregnancy on sex-specific offspring emotional damage

Background Studies have shown that stress during pregnancy can affect the growth and development of fetuses and offspring, and this effect has sex differences, but the results are controversial, and there are few studies on the emotional damage of offspring of different sexes caused by stress during pregnancy. Objective This experiment is designed to observe the effect of chronic stress during pregnancy on emotional damage of offspring of different sexes. Methods Thirty-two SD female rats were randomly divided into a model group and a control group (16 rats in each group), 24 male rats were divided into a model mating group (n=16) and a control mating group (n=8). Each rat of the model group was reared in a single cage and received chronic unpredictable mild stress (CUMS) for 28 d, including hot water swimming for 5 min, cold water swimming for 5 min, tail pinching for 2 min, crowding for 24 h, moist bedding for 24 h, cage shaking for 30 min, and space restriction for 2 h. One stressor was administered daily and the same stressor did not repeat within 7 d. Blood was collected from the endocanthal vein of the two groups of female rats 1 d before and 1, 7, 14, 21, and 28 d after stress, the plasma was separated by centrifugation, and 131I radioimmunoassay was used to measure plasma corticosterone concentration. On postnatal day 21 (PND21), 16 offspring rats (half male and half male) were randomly selected from each group, their plasma corticosterone concentration was measured on PND28 and PND42, and their emotional damage was measured on PND42. Results The plasma corticosterone levels of dams in the model group on the 14th, 21th, and 28th days of stress [(394.02±97.40), (444.12±90.43), and (463.71±107.75) μg·L−1] were higher than those in the control group [(285.63±81.64), (341.78±48.39), and (320.42±84.76) μg·L−1] (all P< 0.05). On PND28 and PND42, the plasma corticosterone levels in the female model offspring group [(543.30±90.21) and (530.76±83.10) μg·L−1] were higher than those in the female control offspring group [(397.77±64.27) and (325.78±61.03) μg·L−1] (both P<0.05). In the sugar water preference test, the total fluid consumption [(10.74±1.28) mL], sugar water consumption [(5.50±1.30) mL], and 1% sucrose preference percentage [(20.36±3.41) %] in the female model offspring group were lower than those in the female control offspring group [(13.74±2.06) mL, (8.56±2.04) mL, and (62.11±8.05) %] (all P<0.05). In the open field test, the horizontal score, vertical score, and cleaning times of the male model offspring group were lower than those of the male control offspring group (all P<0.05). In the tail suspension test, the immobility time of the female and male model offspring groups [(126.95±39.88) and (70.24±28.98) s] was longer than the control offspring groups of the same sex [(54.30±24.99) and (38.63±18.91) s] (both P<0.05), and the duration of immobility time in the female model offspring group was longer (t=3.253, P=0.006). In the forced swimming test, the immobility time of the female model offspring group [(7.97±6.66) s] was longer than that of the female control offspring group [(1.85±2.12) s] (t=2.478, P=0.037). On PND42, the plasma corticosterone level of female offspring was negatively correlated with total fluid consumption, sugar water consumption, and 1% sucrose preference percentage (r=−0.621, r=−0.728, r=−0.699; P<0.05), and positively correlated with immobility time in the tail suspension test and immobility time in the forced swimming test (r=0.571, r=0.712; P<0.05), However, there was no correlation between plasma corticosterone and emotional indicators on PND42 in male offspring (P>0.05). Conclusion Chronic stress during pregnancy causes emotional damage to the offspring, and female offspring show depression-like behaviors.

OBJECTIVETo study the association between high fat-low carbohydrate diet score and newly diagnosed type 2 diabetes in Chinese population.
METHODSData about 20 717 subjects aged 45-59 years from the cross-sectional 2002 China National Nutrition and Health Survey were analyzed. High fat-low carbohydrate diet was scored according to the energy of carbohydrate, fat, and protein.
RESULTSOf the 20 717 subjects, 1 332 were diagnosed with hyperglycemia and 662 were newly diagnosed with type 2 diabetes. Multivariate adjusted analysis showed that the highest score of type 2 diabetes patients was 2.75 (95% CI: 2.09-3.61). The score of type 2 diabetes patients was 1.87 (95% CI: 1.35-2.58) after further adjustment for their socioeconomic status and physical activity. No significant difference was found in the odds ratio after further adjustment for BMI, blood pressure, lipid level, and energy intake. No evidence was observed for the relation between high fat-low carbohydrate-diet score in type 2 diabetes patients due to high family income, less education, physical activity, overweight, hypertension, high TG, or low HDL level.
CONCLUSIONHigh fat-low carbohydrate diets, far different from traditional Chinese diets, are associated with the high incidence of type 2 diabetes in Chinese population.
