1.Clinical analysis of 23 primary sinonasal malignant melanoma.

OBJECTIVE:
To review the pathological and clinical features and treatment of sinonasal malignant melanoma.
METHOD:
A retrospective analysis of 23 cases of sinonasal malignant melanoma. All the patients were conformed by histopathology, the most common symptoms were nasal obstruction and epistexis. Eight cases were treated with surgery, 8 with surgery and radiotherapy, 5 with surgery and chemotherapy, 2 with surgery and ra- diotherapy plus chemotherapy.
RESULT:
Twenty patients were followed up, the survival rates of 3 and 5 years were 50% (10/20) and 35% (7/20), respectively.
CONCLUSION
Sinonasal malignant melanoma has an aggressive behavior and easy recurrence and poor prognosis. Early diagnosis and reasonable treatment can increase the survival rate of the disease.

2.A Dimeric Structure of The Scorpion Toxin BmK M1 at 1.4 (A) Resolution: Non-proline cis Peptide Bond and Its Inntrinsic Structural Elements

Non-proline cis peptide bond is rarely found in proteins. The recent surveys revealed that this unusual peptide configuration is by no means a curiosity, but overwhelmingly occurs at functionally important sites. However one still doubts whether it is related to crystal packing interactions, since all non-proline cis peptide bonds identified so far are from crystal structures. A toxin BmK M1 from the scorpion Buthus martensii Karsch have been crystallized as a dimer in space group P212121 with unit-cell dimensions a = 76.39 (A),b=52.77 (A), c=27.12 (A). This dimeric structure was solved by molecular replacement and refined to R=0.109 for all reflections at a resolution of 1.4 (A). The extensively refined structure definitely shows that the cis peptide bond Pro9-His10 equally occurs in both molecule A and molecule B in the dimer. The observation manifested that this striking non-proline cis peptide is not related to crystal packing, but caused by certain intrinsic factors. The detailed analyses and comparison with the structure of BmK M8, which is homologous to M1 but has trans peptide bond 9-10, showed that the five-residue reverse (8 ~12) with a consensus sequence (-KPXNC-) may be the intrinsic structural element for the cis form of this peptide bond. A pair of well organized main-chain hydrogen bond between residues 10 in cis unit and 64 at C-terminus forms main tertiary interactions to stabilize this energetically unfavorable peptide bond.
3.The correlation of cerebral perfusion characteristics and cognitive function on patients with unipolar and bipolar depression

Objective To analyze the correlation of cerebral perfusion characteristics and cognitive function on patients with unipolar and bipolar depression. Methods The r-CBF imaging was detected by SPECT forall subjects. Evaluated cognitive function of subjects using WAIS-CR and WMA, and analysed data using SPSS.Results Data showed that some regions in the brain, e.g. temporal lobe, frontal lobe, hippocampus, and basal ganglia, had significant alterations of r-CBF in depressed patients as compared with healthy controls (P < 0. 05 ).Compared with UPD, BPD showed increased perfusion in basal ganglia ( 0,22.73%; 4.54%, 36. 36% ) (P <0.05 ). Furthermore there was no significant different incognitive deficits between UPD and BPD(P > 0. 05 ) , and correlations between cognitive impairment and hypoperfusion of rCBF (P < 0. 05 ). Conclusion There be exist difference in r-CBF between UPD and BPD. The perfusion of r-CBF in BPD is significantly increased than that in UPD,and it suggest that hyperperfusion in basal ganglia may be the biological basis when bipolar depression turning to manic.

OBJECTIVETo investigate the effect of Actinidia chinensis Planch polysaccharide (ACPS) on the growth and apoptosis of human gastric cancer SGC-7901 cells, and to explore the effect of SGC-7901 cells on p-p38 expression.
METHODSThe inhibition rates at different concentrations of ACPS on SGC-7901 cells at 24, 48, and 72 h were detected using CCK-8 method. Apoptosis ratios in SGC-7901 were determined by flow cytometry after 48-h treatment of different concentrations of ACPS. The expression of pro-caspase-9, PARP, and p-p38 in SGC-7901 cells after treated by different concentrations of ACPS was detected using Western blot. The expression of pro-caspase-9, PARP, and p-p38 was detected after SGC-7901 cells were pre-treated by p38 specific inhibitor.
RESULTSCompared with the control group, the optical density of SGC-7901 cells decreased after treated by 1, 2.5, 5, and 10 mg/mL ACPS (P < 0.05). Meanwhile, the longer the acting time, the lower the optic density (P < 0.01). IC50 was 7.43 mg/mL at 24 h; 3.88 mg/mL at 48 h, and 1.32 mg/mL at 72 h respectively. ACPS suppressed the protein expression of pro-caspase-9 (P < 0.01) and up-regulated the expression of PARP (89KD) (both P < 0.01). Further study showed that the protein expression of p-p38 was up-regulated in SGC-7901 cells treated by ACPS of different concentrations at 24 h (P < 0.05). The expression of phosphorylation p38 and the ACPS induced apoptosis of SGC-7901 cells could be inhibited after treated by specific inhibitor for 2 h.
CONCLUSIONSACPS could inhibit the growth of SGC-7901 cells and induce apoptosis. The underlying mechanism of inducing apoptosis was partially due to activating the p38MAPK path and further activating Caspase9 and PARP, finally leading to cell death.

7.Comparative analysis of ultrasoundgraphy and CT guided percutaneous paracentesis for liver abscess

Objective To explore differences of ultrasoundgraphy and CT guided percutaneous paracentesis for liver abscess and provide references for the paracentesis treating of liver abscess .Methods 68 patients with liver abscess arranged ultrasoundgraphy guided percutaneous paracentesis were regarded as ultrasound group ,and 38 patients accepted CT guided percutaneous paracentesis were considered as CT group .The succeed rate of paracentesis ,operation period ,treatment effectiveness ,complications of the 2 groups were compared .Results The succeed rate of paracentesis in ultrasound group (81 .7% ) was not different obviously to CT group(82 .6% ) (χ2 =0 .016 2 ,P=0 .898 6) .The operation period of ultrasound group(22 .3 ± 5 .6)min was shorten than CT group (25 .6 ± 4 .8)min ,t= 3 .057 4 ,P< 0 .05 .The heal rate of the 2 groups(91 .2% ,92 .1% ) was not different (χ2 = 0 .027 1 ,P=0 .869 3) .There was no different of the complications in the 2 groups(5 .88% ,8 .57% )(χ2 =0 .010 1 ,P=0 .920 1) .Conclusion Either ultrasoundgraphy or CT guided percutaneous paracentes is feasible and safe for liver abscess .Compared to CT ,the ultra-soundgraphy is more convenient and economic .
8.Effects of EPO on expression of TGF-β1 and ColⅠin myocardium of liver cirrhosis rats

Aim To observe the effects of liver cirrho-sis on the expression of transforming growth factor-β1 ( TGF-β1 ) and ColⅠin rat myocardium and interven-tion of erythropoietin ( EPO ) . Methods Thirty-six male Sprague-Dasley rats were randomly divided into three groups:control group, liver cirrhosis group and EPO group, then the cardic hemodynamic parameters in vivo and levels of serum lactate dehydrogenase ( LDH ) as well as creatine kinase isoenzyme ( CK-MB) were measured. With Masson′s trichrome stain, changes of collagen formation of myocardial tissue in different groups were observed. Also the mRNA ex-pressions of TGF-β1 and ColⅠin myocardium were de-tected by RT-PCR. Results In contrast to control group, rats in liver cirrhosis group showed a decline in systolic and diastolic function of left ventricule, rising myocardial enzyme, a distinct increase of cardiac colla-gen deposition, as well as an elevation of TGF-β1 and ColⅠmRNA expressions. In contrast to liver cirrhosis group, rats in EPO group demonstrated an improve-ment in systolic and diastolic function of left ventricule as well as in cardiac collagen deposition, and a de-crease in both myocardial enzyme and TGF-β1 and ColⅠmRNA expressions. Conclusion Liver cirrhosis can lead to the changes of myocardial structure and function in rats,and it can accelerate myocardial inter-stitial fibrosis; EPO can protect the myocardial injury in liver cirrhosis rats.
9.Distribution and Drug Resistance of Pathogens in a Hospital During Last Three Years

OBJECTIVE To investigate the distribution and resistance of commonly encountered pathogens,and provide reference of antimicrobial agents.METHODS For the clinical specimens during the last three years,flora identification and bacteriostatic test were operated with Microscan WalkAway-40.RESULTS Gram-negative bacteria were 252,507 and 742 strains,Gram-positive bacteria were 142,166,and 243 strains,fung were 0,26,and 229 strains from 2002 to 2004,respectively,the most commonly encountered Gram-negative bacteria were Escherichia coli,Klebsiella pneumoniae,Pseudomonas aeruginosa,Enterobacter cloacae,and Acinetobacter baumannii;the Gram positive bacteria were Staphylococcus aureus,S.epidermidis,S.haemolyticus,and Enterococcus.CONCLUSIONS It is important for reasonable drug application to know the distribution and resistance of commonly encountered bacteria.
10.Transplantation of bone marrow mesenchymal stem cells affects the proliferation and function of CD4+T cells in mice

BACKGROUND:Mesenchymal stem cells are found to have the immunoregulatory activities and a potential application prospect in the treatment of autoimmune diseases.
OBJECTIVE:To explore the mechanism of transplanting mesenchymal stems cells on the treatment of multiple sclerosis.
METHODS:The mouse mesenchymal stems cells were prepared, and injected into the al ogenic and syngenic normal mice, to detect the frequency of CD4+CD25+Foxp3+T cells in the spleen, thymus, and lymph nodes by flow cytometry, and to detect the Foxp3, transforming growth factor-β1, and interleukin-10 mRNA in the spleen, thymus, and lymph nodes by reverse transcription-PCR.
RESULTS AND CONCLUSION:Transplantation of mesenchymal stem cells on normal mice led to a significant up-regulation of CD4+CD25+Foxp3+T cells, Foxp3, transforming growth factor-β1, and interleukin-10 mRNA in the spleen, thymus, and lymph nodes both in the al ogenic and syngenic transplant groups. Transplantation of mesenchymal stem cells may be an available method in the treatment of autoimmune diseases, and CD4+CD25+Foxp3+T cell, Foxp3, transforming growth factor-β1, and interleukin-10 may be involved in this process.
