2.The effect of eye-acupuncture therapy on the TNF-α levels in rats with cerebral ischemia-reperfusion injury

Objective To investigate the effect of the eye-acupuncture therapy on serum TNF-α levels in rats with cerebral ischemia-reperfusion injury and the related mechanism. Methods Healthy SD rats were randomly divided into four groups: normal group, sham-operated control group, ischemia-reperfusion model group and eye-acupuncture group according to body weight. Sham-operated control group, ischemia-reperfusion model group and eye-acupuncture group were divided into the 3h group, the 24h group and the 72h group, a total of 10 groups (n=8). To establish the rat model of cerebral ischemia-reperfusion by suture method in ischemia-reperfusion model group and eye-acupuncture group. Eye-acupuncture was separately started immediately after reperfusion and at 30 min before sampling in the 3h eye-acupuncture group, besides, eye-acupuncture was separately taken every 12h in the 24h eye-acupuncture group and in the 72h eye-acupuncture group. The rats in normal group were not treated, those in sham-operated control group were inserted the fishing thread 1cm, and the others were identical with those in ischemia-reperfusion model group. At 3h, 24h, 72h after reperfusion, the neurophysical behaviours were accessed by ZeaLonga neurophysical impairment marks in ischemia-reperfusion model group and eye-acupuncture group. The method of ELISA was taken to detect the change of serum TNF-α levels in rats after the eye-acupuncture therapy. Results Compared with the ischemia-reperfusion model group, the neurologic impairment score of the eye-acupuncture group decreased obviously; The levels of serum TNF-αin ischemia-reperfusion model group at 3 h, 24 h, 72 h respectively were(76.803±18.325)pg/ml、(85.511±13.334)pg/ml、(86.831±9.232)pg/ml. The level of serum TNF-α in normal group was(24.304±6.511)pg/ml. The level of serum TNF-α in Sham-operated control group at 3 h, 24 h, 72 h respectively were(24.928±3.792)pg/ml,(27.533±5.362)pg/ml,(29.366±5.874)pg/ml. Ischemia-reperfusion model group compare with normal group and sham-operated control group, the difference were significant(P<0.01). The levels of serum TNF-α in rats after the eye-acupuncture therapy at 3 h, 24 h, 72 h respectively were(40.185±3.335)pg/ml, (48.523±7.687)pg/ml, (51.611± 6.403)pg/ml. Compared with ischemia-reperfusion model group, the levels of serum TNF-α were strongly reduced(P<0.01). Conclusion The eye-acupuncture therapy could play a role in improving cerebral ischemia-reperfusion injury evidently and the mechanism was related to its reducing serum TNF-α levels.
3.Research status of photodynamic therapy combined anti - VEGF drugs to treat exudative age-related macular degeneration

Photodynamic therapy ( PDT ) is a new technique to diagnose and treat diseases with photodynamic effect produced by photosensitizer and light, and is now a main method of treating exudative age - related macular degeneration ( AMD ) . ln recent years, with the development of science and technology, combinations of PDT have become a research hot spot. ln this paper, we reviewed the research status of treatments on exudative AMD with PDT combined anti-VEGF drugs.
4.Effect of the application of fast recovery surgery theory in endoscope neurosurgery

Objective To evaluate the effect of the application of fast track surgery theory, in endoscopic neurosurgery. Methods 198 patients of neuroendoscopic endonasal tranasphenoidal surgery for pituitary edenomas were divided into the experiment group and the control. The fast track surgery theory was applied in the experiment group while traditional methods in the control group. Psychological effect of preoperative bowel preparation, the postoperative recovery time of bowel function, analgesia effect, incidence of complications, postoperative nutrional state and hospital day were compared between the two groups. Results The experiment groups were less anxious than the control(P <0. 01 ). The postoperative recovery time of bowel function and Postoperative hospital day wore fewer than the control ( P < 0. 01). Postoperative pain was eased effectively. Condusions The fast track sugery theory can be applied in endoscopic neurosurgery to reduce the level of preoperative anxiety,increasing patient's comfort and improve the nutritional status,help recovery of patients.
5.Transplantation of mesenchymal derived stem cells followed by G-CSF injection can reconstitute hematopoiesis of lethally irradiated BALB/c mice.

OBJECTIVETo explore the hematopoietic reconstitution potential of mesenchymal derived stem like cells.
METHODSWe transplanted bone marrow mesenchymal derived stem like cells into lethally irradiated BALB/c mice. Hematopoietic cells were derived from the non-adherent bone marrow cells 24 hours after initial culture while murine mesenchymal derived stem like cells from bone marrow of donor mice were cultured for 10 days before the transplantation.
RESULTSAll mice of group 1 and 3 died in 7-8 days post irradiation following transplantation, while all the mice from group 2 and 4 survived. The time course of hematopoietic reconstitution was then observed. The peripheral blood and bone marrow cell count recovered in the MSC + G-CSF transplanted group and the BM transplanted group after 3 weeks. Interestingly, CFU-GM number in the MSC + G-CSF transplanted group increased significantly after 2 weeks and even more than that in the BM transplanted group after 3 weeks while as CFU-GM colony dropped 2 weeks after in the BM transplanted group. Spleen colony (CFU-S) number and size of the MSC + G-CSF transplanted group was significantly greater than the BM transplanted group. Furthermore, PCR analysis was performed using peripheral blood cells to determine if any male-derived cells were present. No male-derived cells were found in any of the mice from group 1 and 3. Y-chromosome-specific src gene was found to be dominant in the MSC + G-CSF transplanted group and the BM transplanted group by week 4 post transplantation. In addition, we demonstrated that induction with G-CSF lead to CFU-GM colony formation from MSC compartment in vitro.
CONCLUSIONThese results indicate that under stimulation of G-CSF, mesenchymal derived stem like cells might differentiate into hematopoietic primitive stem cells in vivo and have the capacity to re-establish hematopoiesis in lethally irradiated mice. This study should provide an alternative transplantation treatment for malignancy.

6.Study of biological characteristics of murine dermal mesenchymal stem cells and their enhancement of hematopoiesis recovery.

OBJECTIVETo study the characteristics of dermal mesenchymal stem cells (DMSC), and explore whether they could enhance hematopoiesis recovery in vivo as well as facilitate proliferation and differentiation of hematopoietic cells in vitro.
METHODSMultipotential stem cells from the murine dermal mesenchyme were dissociated and cultured as donor cells. After 2 approximately 3 passages, the growth status, cell cycle, immunophenotype and morphology of DMSC were analyzed. Hematopoietic cells were plated onto a feeder layer formed by DMSC, cell count and CFU-GM yields were observed dynamically. Female mice received 5 Gy (137)Cs radiation were injected with DMSC cultured for 2 - 3 passages via tail vein. Cell count and CFU-GM yields of the bone marrow were observed regularly. Pathological study of the liver, spleen and bone marrow was done to evaluate hematopoiesis recovery.
RESULTSMurine DMSC are adherent cells with a morphology of fibroblastoid and spindle and multiangle in shape. Immunophenotypes showed that CD(45), CD(34), HL-DR positive DMSC were 1 - 3%, CD(44) and CD(13) positive DMSC 75 approximately 95%. Cell cycle assay demonstrated 83% of DMSC being G(0)/G(1) phase. In vitro, the total cell count and CFU-GM yields in the experimental group were higher than those of the long-term culture bone marrow cells by the third week. The DMSC can sufficiently support the proliferation and differentiation of hematopoietic cells for seven weeks. In vivo, peripheral granulocytic count, cells in the bone marrow of one femoral bone and CFU-GM by the third week in the experimental group were much higher than those of controls. Genetic assay of the murine blood demonstrated Y chromosome.
CONCLUSIONThe DMSC have characteristics of stem cells. DMSC sped up hematopoiesis recovery of irradiated mice. DMSC as a feeder layer can support proliferation and differentiation of hematopoietic cells.

8.Correlation between the types of the constitution in TCM and the sleep status in PLA Navy divers

Objective To investigate the correlation between the types of constitution in TCM and the sleep status in the PLA Navy divers. Methods Eighty-nine PLA Navy divers who performed 10m diving professional training were selected. Constitution in TCM was classified and determined by the standardized standard Constitution in Chinese Medicine Questionnaire, sleep status was evaluated by the Pittsburgh Sleep Quality Index. Results 62.9% of Navy divers were the mild constitution in TCM. The eight kinds of the biased constitution in TCM are ranked with yang deficiency, phlegm-dampness, dampness-heat, qi deficiency, yin deficiency, blood stasis, qi stagnation and special intrinsic quality. Among 78 Navy divers with good sleep quality, there were 49 divers (72.1%) with the mild constitution in TCM and 19 (27.9%) with the biased constitution in TCM. Among 21 Navy divers with poor sleep quality, there were 7 divers (33.3%) with the mild constitution in TCM (accounting for) and 14 (66.7%) with the biased constitution in TCM. For Navy divers with poor sleep quality, the sleep quality scores were positive correlated with the blood stasis constitution in TCM (r=0.481,P<0.05). Conclusion Sleep status is correlated with the types of the constitution in TCM, and regulating constitution in TCM can improve sleep quality in PLA Navy divers.
9.Identification and isolation of mesenchymal stem cells from human fetal pancreas.

OBJECTIVETo isolate and identify the phenotype and biological characteristics of pancreas derived mesenchymal stem cell.
METHODSFresh pancreas of 4-5 months old aborted fetus was dissected free from connective tissue, and was cut into small pieces. The adherent cells were harvested and subcultured, after the third subculture, the cells were used for examination. Cell cycle was analyzed by measuring DNA content by FACScan flow cytometer. Phenotype of MSCs was analyzed by immunohistochemical SA technique and differentiated cells were identified by relevant specific staining.
RESULTSFetal pancreas derived cells gave rise to a population of adherent cells characterized by the presence of a predominant cell type with a typical fibroblast like morphology. By transmission electron microcopy, MSC had few endoplasmic reticulums and mitochondrias. During the log phase of growth, MSC proliferated with a two fold population at 30 h. MSC can be ex vivo expanded by successive cycles of trypsinization, seeding, and culture. Under these conditions, MSC had capability of passaging up to 30 times without displaying significant changes in morphology, with 2-fold increase in cell number after each passage. This indicates the high ex vivo expansion potential of MSC. The results showed that the yield of CFU-Fs was above 200 clones even after the 6th passage. Cell cycle analysis by flow cytometry revealed that more than 83% of cells were in the G0/G1 phases, while a small population of cells were actively engaged in proliferation (S + G2 + M = 17%). We also showed that more than 86% of cells were positive stained by FITC labeled CD44, CD29, CD13, and only about 1% of cells were positive for CD34, HLA-DR. Expression of collagen I, III was positive while vWF was negative. In the differentiation study, we found culture-expanded pancreas MSCs could be directed into the osteogenic lineage as detected by osteoblastic morphology, expression of alkaline phosphatase, modulation of osteocalcin mRNA production and the formation of a mineralized extracellular matrix. We also found that MSCs could give rise to the adipogenic and chondrogenic lineage as evidenced by accumulation of lipid-rich vacuoles within cells and the expression of lipoprotein lipase mRNA or the expression of collagen II and the deposition of proteoglycans.
CONCLUSIONMesenchymal stem cells existing in human pancreas can be isolated by their adherent ability and should be essential to sustain a steady supply of primitive cells in tissue remodeling.

10.Effects of co-administration of growth hormone(GH) and aspirin to women during in vitro fertilization and embryo transfer (IVF-ET) cycles.

OBJECTIVETo study the effects of the co-administration of growth hormone (GH) and aspirin to women with suboptimal response to GnRHa/FSH hyperstimulation protocol during in vitro fertilization and embryo transfer (IVF-ET) cycles.
METHODSForty cases of poor ovarian response in previous IVF-ET cycles were randomly divided into 2 groups: the studied group of GH and aspirin (n = 20), and the control group without GH or aspirin (n = 20).
RESULTSThe co-administration of GH and aspirin significantly increased the rates of retrieved oocytes (P < 0.01), promoted the maturation of oocytes (P < 0.01) and improve the fertilization rates (P < 0.05). However, there were no statistically differences between the two groups in the number of replaced embryos (P > 0.05) and the pregnancy rate (P > 0.05).
CONCLUSIONThe co-administration of GH and aspirin to poor ovarian responders is effective to increase the rates of retrieved oocytes, promote the maturation of oocytes and improve the fertilization rate in IVF-ET.
