1.Influence of clinic design of the prostheses with ERA attachment for distal extension defect: A three-dimensional finite dement analysis

Objective:To evaluate the stress distribution and displacement pattern in the restoration of Stern-ERA attachment mandible removable partial denture (RPD) with different designs through three-dimensional (3D) finite element (FE) analysis.Methods:3D-FE models were established based on human CT data and manufacturer's catalog.Vertical and 45° lingual oblique forces of 100 N were applied on the artificial teeth of the RPD.Three ERA attachment partial dentures with different bracing arms were designed,namely the basic design,rest bracing arm design and cross-arch bracing arm design.The von Mises stresses and displacements of abutment teeth and mucosa under denture base were analysed.The displacement of denture bases was examined.Results:Under the vertical and oblique loading,the maximum yon Mises stress was found at the distal shoulder on the distal abutment.The maximun displacement was observed at the buccal and distal occlusal surfaces of the distal abutment.The maximum displacement on the abutment was found to be the highest in rest bracing arm design group (14.91 μm and 63.09 μm),followed by the basic design group (9.38 μm and 50.56 μm) and the cross-arch bracing arm design group exhibited the smallest displacement (8.85 μm and 47.31 μm).Conclusion:The basic design Stern_ERA attachment for unilateral mandible distal extension defect appears to be more reasonable compared with rest arm and crossing-arch arm design from biomechanical point of view.
2.Regulation effect of taurochenodeoxycholic acid immunologic function in mice

Aim To study the effect of Taurochenodeoxycholic Acid (TCDCA) on immunologic cell in vitro and in vivo in mice. Methods Five doses were used , including 0.043, 0.065, 0.1, 0.153, 0.189 g?kg-1 in vivo and 0.01, 0.05, 0.1, 1, 10 mg?L-1 in vitro. Effects of TCDCA on peritoneal macrophage in mouse in vitro,splenic lymphocyte, normal organism, immunologic functional inhibiting model and nerve-endocrine-immunoregulation system were investigated. Results TCDCA had the obvious immunoregulation effect on normal organisms in mouse. 0.1 g?kg-1 TCDCA had the obvious anastated effects on the immunological inhibiting model made up by the cyclosporine A and cyclophosphamide. TCDCA may affect the peritoneal macrophage and splenic lymphocyte in vitro directly, meanwhile two kinds of immunological cells showed the obvious immunoregulation effects on the whole. 0.1 g?kg-1 TCDCA can increase the hydrocortisone contents of the nerve-endocrine-immunoregulation system in mouse most remarkably. Conclusion TCDCA had the adjustment effect on the immunoregulation function in mouse.
3.Effects of Intra-Abdominal Implantation of Sustained-Releasing Fluorouracil on Expressions of Survivin, Caspase-3 and CD44V6 in Patients with Colorectal Cancer

Objectives To observe the pre and post-operational changes of the expressions of survivin, caspase-3 and CD44V6 in patients with colorectal cancer after intra-abdominal implantation of sustained releasing fluorouracil. Meth-ods Sixty-four patients with colorectal cancer (Dukes’stage of B and C) were divided into treatment group and control group, 32 patients in each group. The standard radical surgery was performed in two groups of patients. The fluorouracil im-plants were implanted intra-abdominally in treatment group. The peripheral blood levels of surviving and caspase-3 were de-tected by RT-PCR. The level of CD44V6 was detected by flow cytometry in two groups of patients. Results There were no significant differences in levels of survivin, caspase-3 and CD44V6 before surgery between two groups (P>0.05). The level of survivin (0.362 ± 0.183) was significantly lower at 14 days after operation in treatment group than that of control group (0.585±0.207), but the level of caspase-3 (2.001±0.146) was significantly higher than that of control group (1.654±0.111). The levels of CD44V6 were significantly lower in treatment group (1.857±0.535) and control group (3.471±0.496) after opera-tion than those before operation (9.557±1.170 and 9.729±0.943, P<0.05), and the level of CD44V6 was significantly lower in treatment group than that of control group (P<0.05). Conclusion The implant for the sustained release of fluorouracil showed a positive impact on micrometastases and prognosis of colorectal cancer, while improved the long-term efficacy of postoperative colorectal cancer.
4.Mechanism of MCP-1-JAK2/STAT3 signal transduction in spinal cord dorsal horn contributing to rats with type 2 diabetic neuropathic pain

AIM:ToexplorewhetherMCP-1-JAK2/STAT3signaltransductioninthespinaldorsalhornin-volves the formation and development of rat type 2 diabetic neuropathic pain (DNP).METHODS: The male Sprague-Dawley rats were fed with a high-fat and fructose diet for 8 weeks,and then received a single intraperitoneal streptozocin in-jection to prepare the type 2 DNP model.The type 2 DNP rats were randomly divided into 4 groups (n=16):DNP group, MCP-1 neutralizing (DM) group, DNP+AG490 (DA) group and solvent control (SC) group.A catheter of PE-10 was placed into the subarachnoid space of the rats in groups DM , DA and SC.After 3 d, the rats in DM,DA and SC groups were injected with MCP-1 inhibitor 10μL at 0.1 mg/L, AG490 10μL at 1 mmol/L and DMSO 10μL at 3.5%once a day for 14 days, respectively.Another 16 normal rats were selected as control (C) group and were fed with common forage. The mechanical withdrawal threshold (MWT) and thermal withdrawal latency (TWL) were measured at 1, 3, 7 and 14 d after subarachnoid injection .The lumbar segments 4-6 of the spinal cord were removed at the same time for determination of the expressions of p-JAK2 and p-STAT3 by Western blot .RESULTS:Compared with C group , MWT was significantly de-creased, TWL was shortened and the expression of p-JAK2 and p-STAT3 in the spinal dorsal horn was up-regulated at 1, 3, 7 and 14 d in DNP and SC groups (P<0.05).Compared with DNP group, MWT was significantly increased, TWL was prolonged and the expression of p-JAK2 and p-STAT3 in spinal dorsal horn was down-regulated at 1, 3, 7 and 14 d in DM and DA groups (P<0.05).No significant difference in the MWT, TWL and expression of p-JAK2 and p-STAT3 between DNP group and SC group was observed (P>0.05).CONCLUSION:The MCP-1-JAK2/STAT3 signal transduction in the spinal dorsal horn involves the formation and development of DNP in rats .
5.ANTI-INFLAMMATORY EFFECTS OF IMMUNE MILK

Objective:To study the anti-inflammatory effect of immune milk. Method:Twenty four strains of pathogenic bacteria (including 12 strains of pathogenic E.coli, 8 strains of Salmonella, 3 strains of Shigella and 1 strain of Yersinia enterocolitica) originated from human intestinal tract were selected as antigens, and dairy cows were immunized. Results:Vaccination could not increase the concentration of IgG in the immune milk, but significantly highten the specificity of milk antibodies. The antibody titers against 24 pathogenic bacteria in immune milk was 128, 64 folds of control regular milk. Conclusion:Feeding of immune milk could inhibit edema of rat footpad induced by carrageenin and formaldehyde and the formation of acestoma of mice caused by cotton embedded hypophloedally. Meanwhile, the PGE2 level in inflammatory footpad of rat fed with immune milk was remarkably lower than that of rat fed with regular milk. These results indicate that immune milk has anti-inflammatory property.
6.Influence of radiosensitivity by mild hyperthermia in a pulmonary adenocarcinoma cell line

Objective:To investigate the radiosensitization and the cell-cycle of mild hyperthermia(≤42℃)on human pulmonary adenocarcinoma cell line SPC-A-1 in vitro. Methods: The human pulmonary adenocarcinoma cell line SPC-A were treated with radiation and the combination of radiation with mild hyperthermia. Radiosensitivity was determined by clonogenic assay and quantified by calculating the thermal enhancement ratio (TER). Flow cytometry was used to observe the cell-cycle. Results: Do, Dq calculated from the dose-response curve for radiation combined with 41.5℃ were 1.390 Gy, 1.426 Gy, whereas 1.693 Gy, 2.453 Gy for radiation alone, respectively. TER was 1.218. The proportion of cells in S phase was found to be 14.81% in the radiation group. The values, after 48 hours and 72 hours, with 6Gy radiation combined immediate 41.5℃ one hour mild hyperthermia, were 5.89% and 9.08%, respectively, versus 18.8% and 31.91% with 6 Gy radiation alone. Conclusion:Radiosensitization of mild hyperthermia in SPC-A-1 cells associated with the hyper-radiosensitization of the cells in S phase.
7.Influence of self-management education on the health status of patients with gastric carcinoma during chemotherapy

Objective To analyze the method of self-management education on the health status of patients with gastric carcinoma during chemotherapy. Methods Patients with gastric carcinoma (60 cases) were divided into the test group and the control group with 30 cases in each group. The two groups both received chemotherapy of complete cycle and general health education. The test group adopted self-management education based upon the above treatment, including collective education, group discussion and individual instruction. The treatment effect was appraised and compared by senior primary nurses. Results The recent total effective rate of chemotherapy in the test group was 76.7%, which was higher than that of the control group (50.0%). The level of life quality of the test group was also better than that of the control group (P<0.05). Conclusion Patients-centered self-management education could improve the health status of patients with gastric carcinoma during chemotherapy.
8.Balloon embolization therapy for traumatic carotid cavernous fistula

Objective To analyze effect of balloon embolization therapy for traumatic carotid cavernous fistula (TCCF),so as to provide references for treatment of this disease.Methods A total of 21 patients with TCCF managed with balloon embolization from April 2006 to March 2012 were collected in the study.There were 17 males aged (32.3 ±5.4) years and 4 females aged (40.6 ±5.8)years.Perioperative clinical presentations and imageologic changes were observed as well as operative outcome was evaluated.Results After 3 months of follow-up,20 patients experienced successful detachable balloon embolization,but the other one patient had to have stent angioplasty due to the failure of the procedure.All patients had some improvements on clinical presentations after operation (P < 0.05).Imageologic examination revealed that all patients had complete occlusion of TCCF following operation.In follow-up,one patient had a relapse,with total curative ratio of 95%.Conclusions Detachable balloon embolization is effective in treatment of TCCF.Presently,the technique is an optimal method for such kind of disease and is worthy of wide clinical use.
9.Activation of the Akt/mTOR pathway confers protection against UVB-induced apoptosis in HaCaT cells

Objective To investigate the role of activation of Akt/mTOR pathway in denfense against UVB-induced apoptosis in cultured human skin keratinocyte cell line HaCaT. Methods HaCaT cells were irradiated with UVB at different doses for various durations. Western blotting was performed to detect dynamic changes of Akt/mTOR pathway-related signaling molecule, such as phosphorylated-epidermal growth factor receptor (EGFR), -Akt, -4EBP1, etc; apoptosis was estimated by staining with DNA dye Hoechst 33342. To evaluate the role of signaling molecules in defense against UVB-induced apoptosis, HaCaT cells were pretreated before irradiation with EGFR inhibitor (PD153035), PI3K inhibitor (LY294002), mTOR inhibitor (rapamycin) followed by the detection of expressions of signaling molecule and apoptosis. Results UVB could activate Akt/mTOR pathway in a dose- (5 ~ 30 mJ/cm2) and time- (5 ~ 30 min) dependent manner. PD153035,LY 294002 and rapamycin could inhibit UVB-induced activation of the Akt/mTOR pathway. The apoptosis rate in HaCaT cells was upregulated by pretreatment with rapamycin and LY294002. Conclusion The activation of Akt/mTOR signaling pathway could inhibit the UVB-induced apoptosis in cultured HaCaT cells.
