5.Effects of Icariin on MK-801-induced Schizophrenia Model in Mice

Objective To investigate the effects of icariin, a major active component of traditional Chinese herb Epimedium on schizo-phrenia animal model. Methods MK-801, a N-methyl-D-aspartate (NMDA) receptor antagonist, was intraperitoneally injected into mice to establish a schizophrenia animal model. The open field behavior test infrared detection system was used to measure the mobility of mice in order to find the best optimal dose. 40 mice were randomly divided in to control group, model group, risperidone group and icariin group with 10 mice in each group. The risperidone group and the icariin group were given 0.1 mg/kg risperidone and 50 mg/kg icariin respectively. All groups were injected with 0.6 mg/kg MK-801 0.1 ml intraperitoneally, except the control group. The total distance and the central dis-tance within 210 minutes were tested. Results The total distance and the central distance were significantly longer in the model group than in the control group (P<0.001), and were shorter in the risperidone group than in the control group (P<0.05). The total distance was shorter in the icariin group than in the model group, but there was no significant difference (P=0.065); and the central distance was shorter (P=0.037). There was no significant difference in both total distance and the central distance between the risperidone group and the icariin group (P>0.05). Conclusion Icariin may improve the positive and negative symptoms of schizophrenia mice, which may be beneficial to schizo-phrenia therapy.

7.Expression changes of DNA repair enzymes and DNA damage in diabetes rat hearts

Objective To identify if there was increased DNA damage in cardiomyocytes of rats with diabetic cardiomyopathy and also to evaluate the change in gene expression of DNA repair enzymes 8-oxoguanine DNA glycosylase 1(OGG1) and AP endonuclease 1(APE1).Methods Total DNA,RNA and proteins of hearts were isolated in diabetes rat hearts.DNA damage was examined with quantitative polymerase chain reaction(Q-PCR) assay.mRNA and protein expressions of OGG1 and APE1 were detected with real time-polymerase chain reaction(RT-qPCR) and Western blot analysis.The levels of 8-hydroxy-2′-deoxy-guanosine(8-OHdG) in DNA were studied with ELISA.ResultsThe damage of mtDNA was increased in rat hearts(P<0.05) and there was not significant damage in nDNA.The amount of 8-OHdG in DNA was significantly increased(P<0.05),mRNA and protein expressions of OGG1 and APE1 were increased in diabetic rat hearts(P<0.01).Conclusions The damage of mtDNA increased in diabetic rat hearts.Although the expression of OGG1 and APE1 increased in diabetic rat hearts,which fails to repair the damage of the mtDNA.The increased mtDNA damage may contributes to myocardium damage.
8.Experimental Study of Contrast Enhanced Ultrasound for Evaluation of Blunt Hepatic Trauma in Rabbits

Purpose To investigate the value of contrast enhanced ultrasound (CEUS) in evaluating blunt hepatic trauma and to provide experimental reference. Materials and Methods Fifteen healthy New Zealand white rabbits were randomly divided into groups A, B and C, with 5 rabbits in each group. Hepatic area of the rabbits in each group was stroked using a self-made mini striker with forces of 127.8 N, 213.0 N and 298.2 N, respectively, to establish rabbit models of blunt hepatic trauma. Gray-scale ultrasound (US) and CEUS were performed within 20 minutes before and after trauma. Results Hepatic capsular rupture was observed in 10 rabbits on US. Ill-defined hypoechoic areas in various degrees were observed. CEUS and time-signal intensity curve analysis showed that the normal hepatic tissue began to enhance after contrast agent injection, well-defined against non-enhanced hepatic lesions. The sensitivity of US and CEUS for blunt hepatic trauma were 66.7% (10/15) and 100.0% (15/15), respectively (P<0.05). The agreement with specimen was 40.0% (6/15) for US and 86.7% (13/15) for CEUS, respectively (P<0.05), with Kappa=0.815 for CEUS (P<0.001). Conclusion CEUS improves diagnostic accuracy of blunt hepatic trauma and accurately assesses the severity, which shows good agreement with gross specimen.
9.Estrogen receptor-β expression in laryngeal carcinoma: correlation with the expression of epithelial-mesenchymal transition specific biomarkers.

OBJECTIVE:
To detect the expression of ERβ in laryngeal carcinoma and the its correlation with the expression of epithelial-mesenchymal transition(EMT) specific biomarkers.
METHOD:
Picture MT-Pv9000 was used to detect ERβ and EMT in 72 cases of human aqueous laryngeal carcinoma and 8 cases of adjacent non-neoplastic laryngeal mucosa by immunohistochemistry.
RESULT:
The positive rates of ERβ in tumors and adjacent non-neoplastic laryngeal mucosa were 27.78% and 25.00%, respectively. The differences were not significant (P > 0.05). The abnormal expression rates of E-cadherin and β-catenin were 61.11% and 76.39% respectively. The expression of ERβ correlated negatively with the loss of E-cadherin, nuclear translocation of β-catenin and increased TNM stage. The differences were significant (P < 0.05).
CONCLUSION
The positive expressions of ERβ suggest a good prognosis in the differentiation, clinical stages and lymphatic metastasis of the laryngeal carcinoma. The underlying mechanism may be related with the abnormal expressions of E-cadherin and β-catenin.

10.Fragmentation pathways of five estrogens using electrospray ionization quadrupole time-of-flight mass spectrometry.

The fragmentation pathways of five estrogens (estradiol, estrone, equilin sulfate, 17 a-dihydroequilin sulfate and equilenin sulfate) have been studied with high resolution and high mass accuracy using electrospray ionization quadrupole time-of-flight mass spectrometry (ESI-Q-TOF/MS) in the negative ion mode. Molecular weights were obtained from [M-H](-) ions in the product ion spectra. The results indicate that the five structurally similar estrogens have similar fragmentation pathways. Using their stable isotope forms as internal reference compounds, the accurate mass and composition of the fragment ions were determined. During collision-induced dissociation (CID), cleavage is initiated by loss of oxygen atoms from carbon-17, after which D and C rings cleave sequentially and rearrange to finally form stable conjugate structures with highly abundant characteristic fragment ions at m/z 183 (accompanied by m/z 181), m/z 169 and m/z 145 (accompanied by m/z 143). Understanding these characteristic fragmentation pathways of estrogens will be helpful in identifying the structures of steroid hormones in general.
