Objective To study the genetic polymorphism of Nanjiang yellow goat and to investigate the feasibility of this research by AFLP. Methods The genomic DNAs of 15 goats were analyzed with RAPD and AFLP molecule marking techniques. The effects of the two methods were compared. Results The rates of the polymorphic loci and the genetic similar index of population by RAPD marker were 26.98% and 0.942 respectively, and that by 31.4% AFLP and 0.874 respectively. Conclusion AFLP and RAPD technique are effective methods in genetic polymorphism monitoring of Nanjiang yellow goats, and supply useful parameter with appraise genetic stability.

Purpose To investigate whether there is a difference in BRCA1 gene single nucleotide polymorphisms (SNPs) in Uighurs and Han Chinese sporadic breast cancer,and to analyze the relationship between SNPs locus and tumor susceptibility.Methods 100 cases of sporadic breast cancer (Uighur and Han 50 cases each) and 100 cases of mammary gland disease (Uighur and Han 50 cases each) were collected as the research object,the BRCA1 gene rs16941 and rs16942 were sequenced.Results The distribution of AA,AG and GG genotypes of rs16941 and rs16942 between Uygur and Han breast cancer groups were statistically significant (P =0.009,P =0.017).Compared with AA genotype of rs16941,AG genotype of rs16941 could reduce the risk of breast cancer in Uighurs (OR =0.964,95％ CI:0.260-3.583,P =0.009).Compared with AA genotype of rs16942,AG genotype of rs16942 could increase the risk of breast cancer in Uighurs (OR =1.017,95％ CI:0.293-3.916,P =0.017).Compared with AA genotype of rs16941,AG genotype could reduce the risk of breast cancer in Han nationality (OR=0.824,95％ CI:0.210-3.234,P =0.044).Conclusion The distribution of AA,AG and GG genotypes of rs16941 and rs16942 in Uygur and Han breast cancer groups are statistically significant,and SNPs is correlated with tumor susceptibility.

[Objective] To explore the effect of low intensity pulsed ultrasound(LIPUS)on the ability of repairing rabbit radius defect by cell-scaffold complex composed with bone marrow stromal cells(BMCs)and ?-tricalcium phosphate(?-TCP).[Method]Bone marrow of a rabbit was drawn and cultured by differentiation culture medium to harvest BMCs and the third generation BMCs were cultivated on ?-TCP for one week in vitro.Bilateral distal radial defects were made using a 1-mm saw in 20 mature New Zealand white rabbits.The cell-scaffold complexs stimulated by LIPUS were implanted in rabbit radial defects(cell-scaffold complexs without stimulation as control).All the rabbits were killed at 4 or 8 weeks after operation.The defect healing state was observed with X-ray and histological technic.The gray density of CA was analyzed by computer image analysis system.[Result]It was found that the callus in the experimental group grew more quickly and apparently than that in the control group.At 4 weeks after operation,there was significantly statistical difference(P0.05).Histological slices in the experimental group showed that the absorption,organization of hematoma,the formation of trabeculae and matrix preceded those in the control group.There were more fibroblast,osteoblasts and callus in the experimental group.[Conclusion]Cell-scaffold complex composed with BMCs and ?-TCP stimulated by LIPUS can repair bone defect more effectively than that without LIPUS stimulation at the early stage of healing,but the effect tend to decrease at the late stage.

OBJECTIVE To investigate and analyze the application of prophylactic antibiotics in perioperative period in hospital and offer the scientific basis for reasonable usage and management of antibiotics through surveying the situation.METHODS Totally 120 cases under operation were investigated prospectively from Jan to Aug in 2007.RESULTS The rate of antibiotics usage was 100% among the 120 cases.40% of the patients received single antibiotic treatment,56% and 4% received 2 or 3 kinds of antibiotics combined therapy respectively.The rate of prophylactic usage was 80.3%,the rate of therapeutic one was 19.7%;50% of the patients treated with antibiotics had the duration of postoperative prophylaxis of 7 days or more,the longest one was 30 days.the antibacterials used in turns were cephalosporins,penicillins,lincomycin,macrolides and nitroimidazole.CONCLUSIONS The duration of antibiotic used after operation is too long and the rate of combined anti-infective drugs is too high.It means that their are some problems existed in prophylactic use of antibiotics during cardiovascular surgery at perioperative period in condition of extracorporeal circulation,it is necessary that the antibiotic administration should be standardized.

Objective To investigate the antioxidant ability and radiosensitivity in the malignant transformed human bronchial epithelial cell line BEP2D induced by α-particle exposure.Methods Glutathione Peroxidase (GPX),Catalase (CAT) and Glutathione (GSH) assay kits were employed to detect GPX and CAT enzyme abilities and the levels of GSH in BEP2D,RH21 ( passage 21 of α-particle-irradiated BEP2D cells),and BERP35T-1 cells (derived from nude mice bearing malignant transformed cells generated from cells of passage 35 of α-particle-irradiated BEP2D cells).MTT assay were used to test the growth rate of BEP2D,RH21 and BERP35T-1 cells treated with 0,30,60,90,120,and 150 μmoL/L H2O2.Colony-forming test and MTT assay were used to examine the cell survival fraction and the growth rate of BEP2D,RH21 and BERP35T-1 cells exposed to 0,2,4,and 8 Gy of γ-rays,respectively.Results GPX and CAT enzyme activities in RH21 and BERP35T-1 cells were obviously lower than in BEP2D( t = 5.75-67.92 ,P ＜ 0.05 ).CAT enzyme activity in BERP35T-1 was lower than that in RH21 cells (t =22.25 ,P ＜0.01 ).Compared to BEP2D cells,decreased level of GSH was detected in BERP35T-1 cells(t = 7.76,P ＜ 0.05 ),but there was no change in RH21.After treatment with 30,60,90,120,and 150 μmol/L H2O2,the growth rates of BEP2D were all higher than those of BERP35T-1 cells(t = 10.37-58.36,P ＜0.01 ).Meanwhile,the growth rates of BEP2D were all higher than those of RH21 cells after treatment with 60,90,120,and 150 μ mol/L H2O2 (t = 29.90-84.68,P ＜ 0.01 ).In addition,compared to BEP2D cells,decreased cell survival fraction and growth rate of BERP35T-1 cells were observed after irradiation with 2,4,and 8 Gy of y-rays ( t = 5.87-34.17,P ＜ 0.05 ).The cell survival fraction and growth rate of RH21 were all lower than those of BEP2D cells at 4 and 8 Gy post-irradition( t =6.33- 45.00,P ＜ 0.05 ).Conclusion The function of antioxidant system decreased in the α-particleinduced transformed cells,which could contribute to the acceleration of cellular malignant transforming process and radiosensitivity.

Objective To study the effect of moxonidine (Mox) on the His bundle electrogram (HBE) of normal rabbits. Methods A total of 24 healthy rabbits were randomly divided into four groups: control group, small dose of Mox (0.1 mg?kg -1), medium dose of Mox (0.3 mg?kg -1) and large dose of Mox (0.9 mg?kg -1). The electrode catheter was inserted from the right carotid artery to record the HBE. The HBE and the synchronism surface ECG were recorded before and after intravenous injection. Results In normal rabbits, the R-R interphase, P-R interphase of the ECG and the H-V interphase of the HBE were prolonged in a dose-dependent manner after intravenous injection of Mox. Mox exerted no significant influence on the A-H interphase. Conclusion ① Mox decreases the heart rate of rabbits in a dose-dependent manner in vivo. ② Mox dose-dependently prolongs the P-R interphase of the surface ECG and the H-V interphase of HBE. This indicates that Mox mainly acts on the intraventricular conducting system.

Objective To investigate the correlation between Gab2 expression and platinum drugs chemotherapy resistance by detecting the expression of Gab2 in ovarian cancer.Methods Immunohistochemical and Western blotting techniques were used to detect the expression of Gab2 in 107 specimens of serous ovarian tumor tissue and 12 specimens of normal ovarian tissues resected during bladder cancer radical operation in the department of gynaecology of our hospital from January 2011 to June 2015.The correlation between Gab2 expression with clinical stage and effect of combined chemotherapy based on platinum drugs was analyzed.Results The Gab2 expression level in ovarian cancer tissue was significantly higher than that in normal ovarian tissue,moreover had a relation with the FIGO clinical stage,the expression level of Gab2 increased with the increase of clinical stage,the difference was statistically significant (P<0.05);61 cases were sensitive to chemotherapy,46 cases were resistant to chemotherapy,the Gab2 expression level in the patients with chemotherapy resistance was significantly higher than that in the patients with chemotherapy sensitivity,the difference was statistically significant (P<0.05).Conclusion Gab2 may become one of effective indexes for predicting platinum drugs chemotherapy sensitivity.

In this article, we report the pancreatic islet cells function of newborn pig pancreatic tissues cultured by Ham F_(10), and the condition of graft survived into the bodies of Sprague-Dawley rats, and also study the influence of Glycyrrhiza uralensis to the grafts. The cultured islet cells of newborn pig were transplanted into the mesenteric cavities (the first & second group), kidney cystic(the third group) and brain tissucs (the fourth group) of the 4 groups adult Sprague-Dawley rats. The first group rats drank Glycyrrhiza uralensis water, after 8～11weeks of operation a examination pathological was made with the cut graft The results showed that the first and second group grafts survived over 11weeks, and survived better the group drunk the Glycyrrhiza uralensis water was much better than other. The purpose of this experiment is to find the heteropolar source of pancreatic tissues offering body.

Objective: To observe effects of Yiangxuefukang Granules on the contractile function of uterine smooth muscles. Methods: Both the uteri of Sprague Dawley rats within 4～6 days postpartum and the uteri isolated from women of caesarean operation were used to study the effects of Yiangxuefukang Granules on the contractile amplitude, frequency and vitality of uterine smooth muscles. Results: The Granules significantly increased the contractile amplitude, frequency and vitality of both rat uterine smooth muscless in vivo and human uterine strips isolated.Conclusion: The Granules can be used in the treatment of the postpartum hemorrhage and the poor uterine involution caused mainly by lacking in the strength of uterine contraction.

Objective To investigate the relationship between in vivo anti-tumor efficacy of Shenqi injection and its effect on immunologic function in H22 tumor-bearing model mice,and explore the cellular immunologic mechanism of Shenqi injection in inhibiting tumor growth.Methods Three different doses of Shenqi injection were given to H22 tumor-bearing model mice in treatment groups.The tumor growth inhibitory rate(IR) and the index of phagocytosis of peritoneal macrophage were calculated.MTT assay was used to observe the effect of Shenqi injection on spleen lymphocytes stimulated by ConA in vitro separated from H22 tumor-bearing mice.Murine serum IL-2 and IFN-? were detected by means of ELISA assay.Results IR was significantly reduced in two treatment groups compared with that of the model mice(60.72%,48.65%,P
