1.EXPERIMENTAL OBSERVATION OF TRANSPLANTATION OF CULTURED NEWBORN PIG PANCREATIC ISLET CELLS

In this article, we report the pancreatic islet cells function of newborn pig pancreatic tissues cultured by Ham F_(10), and the condition of graft survived into the bodies of Sprague-Dawley rats, and also study the influence of Glycyrrhiza uralensis to the grafts. The cultured islet cells of newborn pig were transplanted into the mesenteric cavities (the first & second group), kidney cystic(the third group) and brain tissucs (the fourth group) of the 4 groups adult Sprague-Dawley rats. The first group rats drank Glycyrrhiza uralensis water, after 8~11weeks of operation a examination pathological was made with the cut graft The results showed that the first and second group grafts survived over 11weeks, and survived better the group drunk the Glycyrrhiza uralensis water was much better than other. The purpose of this experiment is to find the heteropolar source of pancreatic tissues offering body.
2.Specific loss of heterozygosity of chromosome 3p loci in soft tissue leiomyosarcoma.

OBJECTIVETo analyze the loss of heterozygosity (LOH) at 5 loci on chromosome 3p in soft tissue leiomyosarcoma (LMS).
METHODSLOH was detected in 22 cases of LMS using PCR-silver staining targeting 5 microsatellite sites on 3p14.2-pter. Relation between LOH and LMS clinical pathological features was also analyzed.
RESULTSTen of 22 LMS samples showed LOH at more than one locus (45.4%). Among the 5 loci, LOH occurred more frequently at D3s1295 (36.8%) and D3s1289 (10.5%), but absent at D3s1293. No significant difference was found on LOH incidence between different grade, size and location of LMS.
CONCLUSIONSLOH on chromosome 3p14.2-23 region is relatively frequent in LMS. Region around D3s1295 and D3s1289 may harbor tumor suppressor gene relating to LMS.

3.Preparation of Scutellariae Radix total flavonoids-loaded liposomes and their in vitro anti-tumor activity

AIM To prepare Scutellariae Radix total flavonoids-loaded liposomes and to evaluate their in vitro anti-tumor activity.METHODS Liposomes were prepared by reverse evaporating method.Taking phospholipid concentration,ratio of phospholipid to cholesterol,ratio of organic phase to aqueous phase and total flavonoids concentration as influencing factors,together with encapsulation efficiency as an evaluation index,Box-Behnken design was applied to optimizing the preparation,after which MTT was employed to detect the obtained liposomes' inhibitory effect on human hepatocellular carcinoma cells HepG2.RESULTS The optimal conditions were determined to be 30 mg/mL for phospholipid concentration,3 ∶ 1 for ratio of phospholipid to cholesterol,2 ∶ 1 for ratio of organic phase to aqueous phase,and 5 mg/mL for total flavonoids concentration.The obtained round and uniform liposomes demonstrated an average particle size of (160.7 ± 12.0) nm,Zeta potential of (-41.4 ±2.3) mV,encapsulation efficiency of (86.19 ±0.44)%,drug loading of (5.32 ± 0.04)%,and accumulative release rate of (80.77 ± 2.53) % at 24 h.Compared with total flavonoids,liposomes exhibited stronger inhibitory effect on HepG2 cells with the IC50 of 48.853 μg/mL at 48 h in a time-and dose-dependent manner.CONCLUSION Scutellariae Radix total flavonoids-loaded liposomes stored at low temperature (4 ℃) display sustained-release effect and in vitro anti-tumor activity.
4.The role of ABCG2 in the sensitivity of glioma to pyropheophorbide-amethyl ester-mediated photodynamic therapy

Background and purpose:Adenosine triphosphate-binding cassette superfamily G member 2 (ABCG2), which has been found over-expressed in a variety of cancer cells, takes part in the drug resistance of cancer through effux of anticancer drugs. The purpose of this study was to investigate the mechanisms of human glioblastoma cells sensitivity to pyropheophorbide-a methyl ester (MPPa)-mediated photodynamic therapy (PDT) eradicating tumour cells and its relationship to ABCG2.Methods:U87 and A172 glioma cell lines in the logarithmic growth phase were selected and exposed to the treatment of MPPa-PDT and MPPa-PDT+fumitremorgin C (FTC) respectively. The cell viability was measured with the use of CCK-8 assay. The expression of ABCG2 was detected by Western blot. The intracellular contents of MPPa in each group without illumination were tested by lfow cytometry. Flow cytometry with AnnexinⅤ-FITC/PI double staining was used to detect the cell apoptotic rate. DCFH-DA staining was used to assess the generation of intracellular reactive oxygen species (ROS).Results:The MPPa-mediated PDT could eradicate A172 and U87 cancer cells in an energy-dependent manner. The light energy density in A172 was 8 times of that in U87 when the cell viability reached median lethal dose after MPPa-mediated PDT. The high expression of ABCG2 in A172 cells affected the accumulation of intracellular MPPa. Inhibition of ABCG2, not only could enhance the eradicating effect of MPPa-PDT on A172 cells, but also could increase the yield of ROS triggered by MPPa-PDT and the accumulation of intracellular MPPa.Conclusion:The human glioblastoma cell line A172 is insensitive to MPPa-mediated PDT. The mechanism may relate to ABCG2, which decreases the MPPa content in cancer cells through effux of MPPa, resulting in decline of cytotoxicity.
5.Application of fast track surgery in the post-operation early drinking and eating of children with under general anaesthesia

Objective Application of fast track surgery to study the safety and feasibility of early drinking and eating of infants and children after non-gastrointestinal surgery under general anesthesia.Methods Totals of 240 cases of children with orthopedic surgery were randomly assigned into experimental group and control group.In experimental group,consciousness of sober and clear implementation of postoperative anesthesia of children were received early dietary guidance.While in control group,children were albert 4 hours and then received diet.Respectively,2 hours after operation and 1h after eating,the hunger,thirst,nausea,vomiting,cough,pain of children were assessed and compared.Results There was no significant difference in the time of consciousness of sober and the time of clear implementation of postoperative anesthesia of two groups (P >0.05).There were 90.0% children had drinking and 77.5% children had eating in 2 hours after operation in the experimental group.The incidence of the sense of thirst and hunger in the postoperative 2 hours of experimental group respectively were 10.8% and 22.5% significantly lower than 95.0% and 99.2% of control group ( x2 =66.5,47.0,respectively;P < 0.01 ).The post-operation pain score of experimental group in 2 hours was (4.73 ±0.13 ),and that of control group was (4.73 ±0.13),and the difference was statistically significant ( t =1.86,P < 0.05 ).No significant difference was found in the incidence of cough,nausea and vomiting in the 2 hours after operation and 1 hour after eating between two groups (P >0.05).Conclusions The regular of paediatric orthopaedic surgery under general anesthesia after surgery in children with 4 hours eating need to amend.It is feasibility to develope early drinking and eating for postoperation children under general anesthesia on the basis of assessing sober and clear of the children.
6.An epidemiological study on the index cases of severe acute respiratory syndrome occurred in different cities among Guangdong province.

OBJECTIVETo analyze the epidemiological features of the index cases of severe acute respiratory syndrome (SARS) occurred in different cities in Guangdong province and to trace for the source of infection.
METHODSStandardized individual case inventory was adopted to conduct investigation on index cases and on persons who had close contact with index cases in Guangdong province. Data on the epidemiological characteristics, secondary cases and the links among index cases were analyzed.
RESULTSBetween November 16, 2002 and April 16, 2003, there had been 13 index cases of SARS including 3 cadres, 3 farmers, 2 retirees, 2 workers and 1 shop attendant, reported from 13 cities in Guangdong province. Between November 2002 and January 2003, there had been 7 cities reported to have identified index cases of SARS with 6 of them being infected in their own cities and 1 imported from Guangzhou city. All of the cases had no close contacts to similar patients but 6 of them later caused 2nd or even 3rd generation cases of SARS. Most cases hit young people (7/13) with a sex ratio of 1:0.6. The fatality rate of index cases was high (4/13).
CONCLUSIONNo evidence showed that there was direct transmission among the index cases. Data regarding the geographical origin of those index cases led to the assumption that the infection had started in six cities of Pearl river delta region and the Hong Kong Special Administrative Region.

7.Transgenic mice produced by intracytoplasmic sperm injection.

In our previous study, normal and fertile mice were successful produced from oocytes following intracytoplasmic sperm injection (ICSI). In the present study, the possibility of producing transgenic embryos and offspring with this procedure was evaluated. After freezing-thawed once using HEPES-CZB medium without cryoprotectants, the cauda sperm from KM fertile male were exposed to the circular or linear pEGFP-N1 DNA for 1 min and then co-injected into metaphase II oocytes of B6D2F1 strain. When the zygotes with two pronuclei were cultured in CZB medium to day 3.5, 39.1% (9/23) of them, derived from oocytes co-injected with sperm head and pEGFP-N1 plasmid DNA, were expressed GFP protein. After transfer of the ICSI embryos with two pronuclei from co-injection of sperm head and foreign DNA, seven recipients delivered 30 pups (23.8%, 30/126). Southern blot results revealed that three of sixteen offspring integrated with GFP and neomycin genes together (18.8 %). Interestingly, all of them were produced from oocytes co-injected sperm head and linear DNA (33.3%, 3/9), while none of seven ICSI offspring integrated either GFP or neomycin gene in the group of co-injection of sperm head and circular plasmid DNA. These results indicated that the high efficiency of transgenic mouse could be produced by ICSI. It may be shown that linear DNA is more easily to integrate into host genome than circular DNA when ICSI was used to produce transgenic animals.

8.Changes and significance of peripheral blood fibrinogen level in patients with severe acute respiratory syndrome.

OBJECTIVETo investigate variations and significance of plasma fibrinogen in patients with severe acute respiratory syndrome (SARS).
METHODSTotally 148 patients with SARS were divided into the following groups: initial stage group (44 cases) and after the initial stage group (104 cases), common type group (87 cases) and severe type group (61 cases), unilobar lung involvement group (49 cases), bilobar lung involvement group (53 cases) and diffuse lung involvement group (46 cases). The values of plasma fibrinogen of the 148 SARS patients were analyzed and compared among the different groups.
RESULTSThe mean value of plasma fibrinogen (x +/- SD, 522.29 +/- 154.87 mg/dl) of the 148 cases was higher than the normal value (p less than 0.01). There were significant differences between the initial stage and after initial stage groups, between the common and severe type groups, and among the unilobar, bilobar and diffuse lung involvement groups (p less than 0.05).
CONCLUSIONtheses results suggested that elevation of peripheral blood fibrinogen in SARS patients may play an important role in development and progress of the disease and its treatment.

9.Effects of sRNA 00085 on the environmental fitness of Listeria monocytogenes

The purpose of this study was to investigate the regulatory effects of biofilm associated non-coding small RNA(sRNA)00085 on the survival and environmental fitness of Listeria monocytogenes.Homologous recombination technology was used to construct a deletion mutant strain(△sRNA 00085)and a complementary strain(C △sRNA 00085)of the sRNA00085 target gene.The differences in biological characteristics were compared among the standard strain,△sRNA 00085,and C△sRNA 00085.The deletion of sRNA00085 led to a significant decrease in biofilm formation capacity and sensitivity to several antibiotics,including penicillin,piperacillin,doxycycline,tetracycline,vancomycin,and cotrimoxazole.However,only the minimum inhibitory concentration(MIC)of tetracycline exhibited a significant decrease in △sRNA00085.Meanwhile,the decreased biofilm formation and antibiotic resistance of the sRNA00085 mutant were restored in the C△sRNA00085 strain.Furthermore,we investigated the transcription levels of tetracycline resistance-related genes in L.monocytogenes.Down-regu-lated transcription of the tetS gene but no significant difference in transcription of the tetA gene were observed in △sRNA 00085 compared with the standard strain and C△sRNA00085.Moreover,the elimination of sRNA00085 did not affect bacterial growth ability or sensitivity to disinfectants.These findings highlight that sRNA00085 plays an important role in the environ-mental adaptability of L.monocytogenes by affecting bacterial biofilm formation and resistance.
10.Association of-238G/A and -857C/T polymorphisms of tumor necrosis factor-alpha gene promoter region with outcomes of hepatitis B virus infection.

OBJECTIVESTo determine whether -238G/A and -857C/T polymorphisms of tumor necrosis factor-alpha (TNF-alpha) gene promoter were associated with outcomes of hepatitis B virus infection.
METHODSA total of 246 HBV self-limited infected subjects and 443 chronic hepatitis B (HB) patients were recruited in this case-control study. TNF-alpha-238G/A and -857C/T gene promoter polymorphisms were examined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP).
RESULTSThe frequency of TNF-alpha-238 GG (90.7%) in chronic HB group was significantly lower than that (95.1%) in self-limited group (P = 0.041). The frequency of TNF-alpha-857 CC (79.7%) in chronic HB patients was significantly higher than that (70.9%) in self-limited infected subjects (P = 0.021). Multiple logistic regression analysis revealed that both TNF-alpha-238GA and -857CC were independently associated with chronic HB.
CONCLUSIONSTNF-alpha promoter variants are likely to play a substantial role in influencing the outcomes of HBV infection.
