BACKGROUND:Kunming mouse embryonic fibroblasts are the most common feeder layers at present, and there are rare reports addressing C57BL/6 mouse embryonic fibroblasts as feeder layers.
OBJECTIVE:To separate and culture C57BL/6 mouse embryonic fibroblasts in vitro, and produce feeder layers to enlarge the resources of mouse embryonic fibroblasts.
METHODS:C57BL/6 mouse embryonic fibroblasts were isolated and cultured by trypsin digestion method in vitro. The biological characteristics and growth rule of the fibroblasts were investigated, then the feeder layers for the cel culture were produced. The growth of cel colonies on the prepared feeder layer was tested.
RESULTS AND CONCLUSION:C57BL/6 mouse embryonic fibroblasts grew wel with a large amount, by trypsin digestion method at different concentrations. There was no significance in the survival rate after cryopreservation for 1 week, 2 weeks, 1 month, 3 months and 6 months. The cel s were proliferative from the second to fifth passage and declined sharply after the sixth passage. The planted mouse embryonic fibroblasts feeder layers had a high activity within 3 days, but got a sharp decline after 4 days. So it is best to use C57BL/6 mouse embryonic fibroblast feeder layers within 3 days after they’re inactivated. C57BL/6 mouse embryonic fibroblast feeder layer can support embryonic stem cel s and induce pluripotent stem cel s to grow as Kunming mouse embryonic fibroblasts.

Positron emission tomography-computed tomography (PET-CT) plays an important role in the diagnosis and treatment of liver cancer.18F-FDG PET-CT imaging-positive rate is low due to a variety of factors affecting the hepatoma cells.But the joint imaging can significantly improve the diagnostic positive rate and provide the basic information for treatment selection and prognosis.PET-CT is better than conventional imaging techniques in the diagnosis and therapy monitoring for metastatic liver cancer.

Objective To train students'divergent thinking and the ability of innovation in functionalexperimental lesson. Methods Ask students to make the assumptions of animals' death and to verify them by anatomy in the experimental test,then to analyze them in multilateral ways. Results This method could heighten studtns'initiative and improve their ability of ob-servation and analysis. On the other hand,divergent thinking could be trained and interest of in-novation could be improved by this way. Objective: To investigate the anti-tumor efficiency of B16 melanoma HACs in vivo and in vitro.Methods: Tris-HCI extract and Sephacryl S-200 gel filtration were applied to prepare B16 HACs, and the cytolokicity of specific CTL induced by HACs was tested. Results: The 41, 47 and 53 tube HACs obtained by gel filtration could decrease the tumor incidences, delay the time of tumor development and decrease the mortalitites of mice.Conclusion:60 ~ 97 kD HACs from B16 melanoma cytosol have the activites of inhibiting tumor and could be used in effective anti-tumor therapy.

Objective To assess the immune protective effect of donor spleen perfusion on the renal allografts in specific sensitized canine recipients.Methods Male dogs were operated as donors and recipients for kidney transplantation.Eighteen dogs were previously sensitized by a method of multi-infusion of small number of lymphocytes from a paired donor and were then randomly divided into 3 groups equally.In donor specific spleen perfusion group,the artery and vein of the excised do- nor canine spleen were connected to the abdominal aortic artery and inferior vena cava of the recipient with processed fistula tubes,perfused at a speed of 18-25 ml/min for 40 min and then kidney from the same donor was transplanted to the recipient;In non-specific spleen perfusion group,the methods of excised spleen perfusion and kidney transplantation were indifferent to the above group except for the organs which removed from a non-specific donor;In control group,kidney transplantations were per- formed after an exclusion time of 40 min in the open abdominal surgery.Cross-match between paired dogs monitored by means of complement dependent cytotoxicity(CDC)and mixed lymphocyte culture (MLC)test before and after donor spleen perfusion was investigated.Rejection episodes and serum creatinine level in each group were assessed after kidney transplantation.Results Immune sensitization was successfully induced in all 3 groups after 3 to 4 times of donor lymphocyte stimulations.CDC as well as lymphocytes proliferation in MLC and CH50 of recipients was significantly declined after spe- cific spleen perfusion,and white blood cell count in peripheral blood also decreased transiently.After kidney transplantation between the paired sensitized dogs,the advanced rate of renal allograft function deterioration was slower in specific spleen perfusion group than in other two groups.The pathologic investigation suggested that severity of rejection in spleen perfusion group was milder than in other two groups.Conclusions Donor spleen perfusion can present a specific absorption efficacy for pre- formed antibodies and activated lymphocytes in sensitized recipients,as well as simultaneously deplete part of platelets in peripheral blood.So donor spleen perfusion may delay the presenting time of rejec- tion in early phase after kidney transplantation between the paired sensitized dogs,ameliorate the se- verity of rejection so as to improve the renal allograft function.

Objective To determine the efficacy of lifestyle interventions in adults with impaired glucose tolerance (IGT).Methods MEDLINE (Pub Med),EMBASE,Science Citation Index (SCI),and Cochrane Database were retrieved for articles about the relations of lifestyle intervention and IGT.Searches were limited to English language publications.The RCTs outcome evaluated in this study included 2 h plasma glucose level and fasting plasma glucose,meta-analysis was carried oat by Stata 11.0 among articles for the inclusion and exclusion criteria.The difference of effects was expressed as standardized mean deviation(SMD).Results A total of 10 RCTs studies met the inclusion criteria.Lifestyle interventions were associated with a decline in 2 h plasma glucose and fasting plasma glucose level (SMD =-0.67,P＜0.01 ;SMD =-0.33,P＜0.01),but high heterogeneity was identified in this meta-analysis.Conclusion Physical,dietary and both combined interventions can reduce 2-h plasma glucose and fasting plasma glucose levels in adults with impaired glucose tolerance.As high heterogeneity was identified in this meta-analysis,more high quality research is needed.

Objective:To investigate the long-term efficacy of oral appliance treating of obstructive sleep apnea and hypopnea syndrome (OSAHS) in different periods of treatment.Methods:A total of 55 patients were included in the study.Patients were all diagnosed with OSAHS by overnight polysomnography and all received oral appliance (OA) as the therapy.The OA positioned the mandible at 60％-70％ of the maximal mandible advancement position and created a 4-5 mm incisor separation.The patients were instructed to wear the appliance during sleep,6-8 hours per day,for 5-7 days per week.They were divided into four groups by the period of treatment,including less than 1 year group;1-2 years group;2-6 years group and 6-9 years group.The polysomnographic study was used to investigate the efficacy of the four groups.The outcome measures included the score on the apnea-hypopnea index (AHI),the longest apnea time and the lowest oxygen saturation (LSaO2) levels during an overnight sleep.Results:The AHI decreased significantly in all the four groups.The less than 1 year group decreased from 24.50 (14.65,54.05) to 7.40 (2.12,10.00) events/h (P ＜ 0.001);The 1-2 years group decreased from 19.50 (12.15,39.23) to 1.80 (0.70,6.58) events/h (P =0.001);The 2-6 years group decreased from 25.00 (11.41,42.60) to 4.50 (1.35,7.90) events/h (P =0.001);The 6-9 years group decreased from 26.2 (16.95,47.45) to 4.00 (1.90,26.70) events/h (P =0.043).The longest apnea decreased significantly in less than 1 year group,1-2 years group and 2-6 years group.The longest apnea decreased from 57.00 (37.70,61.50) to 25.00 (15.90,33.50)seconds (P＜0.001) in the less than 1 year group,from 41.00 (25.50,62.26) to 13.10 (0.00,22.10) seconds (P =0.001) in the 1-2 year group and from 42.50 (30.35,58.15) to 15.60 (0.00,28.10) seconds (P =0.003) in the 2-6 year group.The LSaO2 levels increased significantly in the less than 1 year group and 2-6 years group.The LSaO2 levels rose significantly in the less than 1 year group,from 74.18％ ±7.96％ to 84.06％ ±7.67％ (P =0.001),and in the 2-6 years group,from 76.71％ ± 10.98％ to 84.06％ ±4.64％ (P =0.006),The LSaO2 levels did not increase significantly in the 1-2 years and 6-9 years groups.Kruskal-Wallis test showed that there were no statistically significant differences in the AHI,longest apnea time and LSaO2 in 4 the groups.Conclusion:The oral appliance is an effective therapy for patients with OSAHS in the long-term treatment.However,it's recommended to make appointments with patients as a follow-up supervision whether there is any efficacy decrease.And the oral appliance should be replaced if necessary.

Samples of skeletal muscle in forearm were taken from 60 sites in each of 10 normal autopsy subjects (male 7, female 3) between 3 and 69 years of age. The distribution and composition of type I and type II fibers in the muscles of the forearm were studied by histochemical method for myofibrillar ATPase. The results showed that the mean percentage of type I fibers ranged between 45% to 59% in the muscles of the forearm. In general, the slow contracting fibers of the forearm muscles were slightly higer than 50%. In the flexors and extensors of the forearm the respective percentage of type I fibers were 49.8% and 55.3%. Objective:In present study we observed the effect of whole body irradiation (WBI) with 75 mGy X-rays on the immune function of tumor-bearing mice.Methods:Lewis lung carcinoma cells were implanted into the right thigh muscle of C57BL/6J mice.Ten days after tumor implantation,the tumor-bearing mice were administrated with 75 mGy X-rays WBI,then the mice were sacrificed 18 h after irradiation to detect the immune parameters including the spontaneous proliferation of thymocytes,the proliferative response of splenocytes to ConA and LPS,the cytotoxic activities of specific cytotoxic lymphocytes (CTL) and natural killer cells (NK),as well as lymphokine activated killer cells (LAK) in spleen.The methods we used were 3H-TdR incorporation or release assay.Results:The immune parameters of exposed tumor-bearing mice were much higher than those of sham-irradiated tumor-bearing mice (P<0.01).Conclusion:These results suggested that low dose radiation (LDR) could enhance the immune function of tumor-bearing mice,which might be of practical significance in the prevention and therapy of cancer.
